Smn, the spinal muscular atrophy-determining gene product, modulates axon growth and localization of beta-actin mRNA in growth cones of motoneurons.

Spinal muscular atrophy (SMA), a common autosomal recessive form of motoneuron disease in infants and young adults, is caused by mutations in the survival motoneuron 1 (SMN1) gene. The corresponding gene product is part of a multiprotein complex involved in the assembly of spliceosomal small nuclear ribonucleoprotein complexes. It is still not understood why reduced levels of the ubiquitously expressed SMN protein specifically cause motoneuron degeneration. Here, we show that motoneurons isolated from an SMA mouse model exhibit normal survival, but reduced axon growth. Overexpression of Smn or

Measurements of growth cone adhesion to culture surfaces by micromanipulation.

Neurons were grown on plastic surfaces that were untreated, or treated with polylysine, laminin, or L1 and their growth cones were detached from their culture surface by applying known forces with calibrated glass needles. This detachment force was taken as a measure of the force of adhesion of the growth cone. We find that on all surfaces, lamellipodial growth cones require significantly greater detachment force than filopodial growth cones, but this differences is, in general, due to the greater area of lamellipodial growth cones compared to filopodial growth cones. That is, the stress (forc

Live visualization of protein synthesis in axonal growth cones by microinjection of photoconvertible Kaede into Xenopus embryos.

Photoconvertible fluorescent proteins, such as Kaede, can be switched irreversibly from their native color to a new one. This property can be exploited to visualize de novo mRNA translation, because newly synthesized proteins can be distinguished from preexisting ones by their color. In this protocol, Kaede cDNA linked to the 3' untranslated region (UTR) of beta-actin is delivered into cells fated to become the retina by injection into Xenopus blastomeres. Brief exposure (6-10 s) to UV light (350-410 nm) of Kaede-positive retinal axons/growth cones efficiently converts Kaede from its native gr

Developmental expression and biochemical analysis of conulin, a protein secreted from a subset of neuronal growth cones.

In this report, we analyze the developmental pattern of expression of a new grasshopper protein, Conulin, using the monoclonal antibody 7D2 on whole-mount embryos and dissociated neurons. We also have examined its biochemical properties by immunoblot analysis. Conulin is a protein expressed by a subset of neurons in the grasshopper embryo. The monoclonal antibody 7D2 recognizes Conulin as an M(r) 190 x 10(3) protein that is found in both the soluble and membrane-bound fractions of embryonic proteins. The membrane association is disrupted by alkaline pH and high ionic strength. Conulin first is

Regulation of neuronal growth cone filopodia by nitric oxide.

Nitric oxide (NO) has been proposed to play an important role during neuronal development. Since many of its effects occur during the time of growth cone pathfinding and target interaction, we here test the hypothesis that part of NO's effects might be exerted at the growth cone. We found that low concentrations of the NO-donors DEA/NO, SIN-1, and SNP caused a rapid and transient elongation of filopodia as well as a reduction in filopodial number. These effects resulted from distinct changes in filopodial extension and retraction rates. Our novel findings suggest that NO could play a physiolog

The expression of cell adhesion molecules on the growth cones of chick cutaneous and muscle sensory neurons.

In the developing chick hindlimb, axons that will project along a given peripheral nerve sort out together, as they traverse the plexus region at the base of the limb, and become segregated from axons that will project along other peripheral nerves. This sorting out may involve, at least in part, the differential expression of various cell adhesion molecules (CAMs). To begin to explore this possibility, we have compared the relative levels of immunofluorescent labeling on the growth cones of two populations of sensory neurons whose axons become segregated from one another: cutaneous and muscle

Myelin-associated glycoprotein inhibits neurite/axon growth and causes growth cone collapse.

We have previously shown that myelin-associated glycoprotein (MAG) inhibits neurite growth from a neuronal cell line. In this study we show that 60% of axonal growth cones of postnatal day 1 hippocampal neurons collapsed when they encountered polystyrene beads coated with recombinant MAG (rMAG). Such collapse was not observed with denatured rMAG. Neurite growth from rat embryonic hippocampal and neonatal cerebellar neurons was also inhibited about 80% on tissue culture substrates coated with rMAG. To investigate further the inhibitory activity of MAG in myelin, we purified myelin from MAG-defi

Dynamic subtype- and context-specific subcellular RNA regulation in growth cones of developing neurons of the cerebral cortex.

Neurons of distinct subtypes compartmentalize subtype-specific function in part by differentially localizing and translating specific RNAs, but underlying mechanisms are not understood. Here we investigate messenger RNA localization and stability within subtype-specific growth cones (GCs), leading tips of growing axons, of long-range projection neurons (PNs) of the developing cerebral cortex. Comparison of GC-localized transcriptomes between two subtypes of PNs (interhemispheric-callosal and corticothalamic) across developmental stages identified both distinct and shared subcellular machinery

Spontaneous acetylcholine secretion from developing growth cones of Drosophila central neurons in culture: effects of cAMP-pathway mutations.

We describe a novel bioassay system that uses Xenopus embryonic myocytes (myoballs) to detect the release of acetylcholine from Drosophila CNS neurons. When a voltage-clamped Xenopus myoball was manipulated into contact with cultured Drosophila "giant" neurons, spontaneous synaptic current-like events were registered. These events were observed within seconds after contact and were blocked by curare and alpha-bungarotoxin, but not by TTX and Cd(2+), suggesting that they are caused by the spontaneous quantal release of acetylcholine (ACh). The secretion occurred not only at the growth cone, but

The growth cones of identified motoneurons in embryonic zebrafish select appropriate pathways in the absence of specific cellular interactions.

Developing motoneurons in zebrafish embryos follow a stereotyped sequence of axonal outgrowth and accurately project their axons to cell-specific target muscles. During axonal pathfinding, an identified motoneuron pioneers the peripheral motor pathway. Growth cones of later motoneurons interact with the pioneer via contact, coupling, and axonal fasciculation. In spite of these interactions, ablation of the pioneer motoneuron does not affect the ability of other identified motoneurons to select the pathways that lead to appropriate target muscles. We conclude that interactions between these cel

Structure and locomotion of adult in vitro regenerated spiral ganglion growth cones-- a study using video microscopy and SEM.

Neuronal development and neurite regeneration depends on the locomotion and navigation of nerve growth cones (GCs). There are few detailed descriptions of the GC function and structure in the adult auditory system. In this study, GCs of adult dissociated and cultured spiral ganglion (SG) neurons were analyzed in vitro utilizing combined high resolution scanning electron microscopy (SEM) and time lapse video microscopy (TLVM). Axon kinesis was assessed on planar substratum with growth factors BDNF, NT-3 and GDNF. At the nano-scale level, lamellipodial abdomen of the expanding GC was found to be

A protein expressed in the growth cones of embryonic vertebrate neurons defines a new class of intermediate filament protein.

We have isolated and characterized cDNAs that encode a protein expressed in the axons and growth cones of a subset of Xenopus embryonic neurons. The protein is also expressed in a subset of cells of the brain, including cells in even-numbered rhombomeres, the eye, and the heart. The sequence of the cDNA suggests that the protein belongs to a new class of neural-specific intermediate filaments. Both the RNA and the protein are expressed in the neurula and persist during embryogenesis in the brain, cranial nerves, and spinal cord. Because of the predicted structure of the protein, we have named

Pathfinding by growth cones of commissural interneurons in the chick embryo spinal cord: a light and electron microscopic study.

To investigate putative axonal guidance mechanisms used by commissural interneurons in the chick embryo spinal cord, we have examined growth cone morphology, the microenvironment through which the growth cones advance, and interactions between growth cones and their surroundings. Growth cones of both early and late developing commissural interneurons were examined. The growth cones were visualized by injection of either horseradish peroxidase (HRP) or the fluorescent dye Di-I. Unlabelled growth cones as well as HRP-labelled growth cones were also examined by electron microscopy. The early deve

Axonal Regeneration, Growth Cone, and the FIGN Gene Family: A Comprehensive Review.

Mature mammalian central nervous system (CNS) neurons exhibit limited regenerative capacity, resulting in severe and often irreversible functional deficits following injuries such as spinal cord injury or stroke. Both inhibitory extrinsic environments and intrinsic neuronal constraints govern this regenerative failure. Growth cones-cytoskeleton-rich structures at axonal tips-are critical for axon extension and pathfinding during neural repair, with their structural integrity and motility heavily dependent on microtubule (MT) stability and dynamics. Pharmacological stabilization of MTs has been

Laminin-332 coordinates mechanotransduction and growth cone bifurcation in sensory neurons.

Laminin-332 is a major component of the dermo-epidermal skin basement membrane and maintains skin integrity. The transduction of mechanical force into electrical signals by sensory endings in the skin requires mechanosensitive channels. We found that mouse epidermal keratinocytes produce a matrix that is inhibitory for sensory mechanotransduction and that the active molecular component is laminin-332. Substrate-bound laminin-332 specifically suppressed one type of mechanosensitive current (rapidly adapting) independently of integrin-receptor activation. This mechanotransduction suppression cou

Axonal protein synthesis and degradation are necessary for efficient growth cone regeneration.

Axonal regeneration can occur within hours of injury, the first step being the formation of a new growth cone. For sensory and retinal axons, regenerative ability in vivo correlates with the potential to form a new growth cone after axotomy in vitro. We show that this ability to regenerate a new growth cone depends on local protein synthesis and degradation within the axon. Axotomy in vitro leads to a fourfold to sixfold increase in 3H-leucine incorporation in both neurones and axons, starting within 10 min and peaking 1 h after axotomy. Application of protein synthesis inhibitors (cycloheximi

Differential localization of WAVE isoforms in filopodia and lamellipodia of the neuronal growth cone.

The formation and extension of filopodia in response to an extracellular stimulus by guidance cues determine the path of growth cone advance. Actin-filament bundling and actin polymerization at the tips supply the driving force behind the formation and elongation. We tried to clarify how signals in response to extracellular cues are transformed to induce filopodial generation and extension. Observations on the formation process of filopodia at growth cones in the neuroblastoma cell line NG108 showed that WAVE (WASP (Wiskott-Aldrich syndrome protein)-family verprolin homologous protein) isoform

From the growth cone surface to the cytoskeleton: one journey, many paths.

The mechanisms that guide axons through a complex cellular landscape to reach appropriate target cells are central to our understanding of neural development. Decades of work suggest that guidance information is interpreted by signaling machinery that controls the complex and dynamic cytoskeleton at the growth cone leading edge. Recent insights from the areas of signal transduction and cell biology have identified a number of key components that play central roles in this chain of command, including members of the Ena/VASP and WASP family of proteins. Although our understanding of the precise

In vivo regulation of axon extension and pathfinding by growth-cone calcium transients.

Growth cones at the tips of extending neurites migrate through complex environments in the developing nervous system and guide axons to appropriate target regions using local cues. The intracellular calcium concentration ([Ca2+]i) of growth cones correlates with motility in vitro, but the physiological links between environmental cues and axon growth in vivo are unknown. Here we report that growth cones generate transient elevations of [Ca2+]i as they migrate within the embryonic spinal cord and that the rate of axon outgrowth is inversely proportional to the frequency of transients. Growth cone guidance in insects: fasciclin II is a member of the immunoglobulin superfamily.

The cellular cues that guide neuronal growth cones toward their targets are highly conserved in such diverse organisms as insects and vertebrates. Evidence presented here suggests that the molecular mechanisms underlying these events may be equally conserved. This article describes the structure and function of fasciclin II, a glycoprotein expressed on a subset of fasciculating axons in the grasshopper embryo. Antibody perturbation experiments suggest that fasciclin II functions in mediating one form of neuronal recognition: selective fasciculation. Fasciclin II is a member of the immunoglobul
