THE SECRETION OF DIGESTIVE ENZYMES AND CAECAL SIZE ARE DETERMINED BY DIETARY PROTEIN IN THE CRICKET Gryllus bimaculatus.

In Gryllus bimaculatus, the size of the caecum decreases in the latter half of each instar to a stable minimal size with a steady minimal rate of digestive enzyme secretion until feeding resumes after ecdysis. The higher the percent protein in the newly ingested food, the faster and larger the caecum grows, and as a consequent the higher the secretion rate of trypsin and amylase. When hard boiled eggs (40% protein) are eaten the caecum is 2× larger, the trypsin secretion is almost 3× greater, and amylase 2.5× greater then when fed the same amount of apples (1.5% protein). Only dietary protein

Effect of dietary carotenoids on vitamin A status and skin pigmentation in false tomato frogs (Dyscophus guineti).

Several species in captivity develop nutritional diseases including vitamin A deficiency; cases of this disease have been documented in amphibians, which may be linked to an insectivorous diet lacking in vitamin A or carotenoids. Adults and young of Dyscophus guineti were fed three diets over 9 weeks to evaluate effects on carotenoids and vitamin A status and skin pigmentation. Feeder crickets were either supplemented with soy oil (control, CON), soy oil enriched with β-carotene (BC) or mixed carotenoids (MIX) by direct injection of known dosages. Vitamin A from feeder crickets (measured as re

Copulation in the cricket is performed by chain reaction.

The male and female genitalia are finely designed to match each other for copulation in the cricket Gryllus bimaculatus. Copulatory acts of the male, stereotyped and time-fixed, are elicited by stimulation of mechanoreceptors on particular regions of the abdomen, cerci and genitalia. Sequential execution of each motor act proceeds as a chain reaction in which one act stimulates some receptors which in turn elicits another act and so on, while the female remains immobile on the male's back. Each key stimulus for a motor act appears as a result of the male's own act, except for copulatory papill

Spider peptides from Brachypelma smithi with slightly different amino acids at their C-terminal loops exert different insecticidal activities.

The insecticidal molecules of spiders persistently evolve to ensure rapid paralysis of their prey, and the best molecules are transmitted to their progeny. Here, we cloned two insecticidal peptides, Bs2 and Bs3, from the venom glands of the theraphosid Brachypelma smithi. Bs2 and Bs3 are 90.2% identical, but they exhibit interesting structural differences at their C-termini, including a connecting disulfide bond (residues Cys15-Cys36 for Bs2 and Cys15-Cys30 for Bs3). The genomic origin of Bs2 and Bs3 may be a cause for gene duplication events. Moreover, Bs2 differs in two residues from Tal1 (9

Food availability influences adult body mass variability and reproductive traits in a spider.

All animals require food to survive, grow, reproduce, and thus optimize fitness in nature. Food availability can profoundly affect demographic parameters such as longevity and fecundity. Here, we compared reproductive parameters in the false widow spider, Steatoda grossa (Araneae, Theridiidae), when the availability and size of prey (the house cricket, Acheta domesticus) were manipulated. Adult mated female spiders that were fed weekly (constant prey treatment) produced more progeny during their lifetime than females fed every 3 weeks (intermittent prey treatment). Furthermore, the monitoring

Ion transport peptide and ion transport peptide-like regulate ecdysis behavior and water transport during ecdysis in Gryllus bimaculatus.

Ion transport peptide (ITP) and ITP-like (ITPLs) are pleiotropic bioactive peptides in insects. Although the contribution of these peptides to ecdysis has been studied, the precise regulatory mechanisms remain poorly understood. Here, we characterized the functions of itp and itpl variants in the two-spotted cricket, Gryllus bimaculatus. Reverse transcription-quantitative PCR and whole-mount in situ hybridization revealed that itp was expressed in the brain and terminal abdominal ganglion, whereas itpl variants were expressed in all ganglia of the central nervous system. Simultaneous knockdown

The Compound Eye Regulates Free-Running Period and Stability of the Circadian Locomotor Rhythm in the Cricket Gryllus bimaculatus.

The circadian system of many multicellular organisms consists of a hierarchical structure of multiple clocks, including central and peripheral clocks. The temporal structure has been analyzed in terms of central-to-peripheral regulation but rarely from the opposite perspective. In this study, the potential control of the central clock in the optic lobe by the peripheral clock in the compound eye was investigated in the cricket Gryllus bimaculatus. The locomotor activity rhythm of crickets in which one of the two bilateral optic lobe clocks was surgically removed was tested in constant darkness

Radiation induces stress and transgenerational impacts in the cricket, Acheta domesticus.

PURPOSE: Radiation exposure of crickets during their fourth juvenile molt inflicted ionizing radiation damage and altered growth rate, adult size at sexual maturity. High levels of ionizing radiation also impacted the subsequent generation, likely via heritable epigenetic mechanisms. Using radiation as a proxy for external stress, we aim to understand the transgenerational impacts of stress on non-irradiated offspring. METHODS AND MATERIALS: We assess the impacts of ionizing radiation on maturation mass and growth rate in F0 male and female house crickets (Acheta domesticus). We also assessed

SECRETION OF LIPASES IN THE DIGESTIVE TRACT OF THE CRICKET Gryllus bimaculatus.

Little is known concerning the sites and the ratios of the lipase secretions in insects, therefore we undertook an examination of the lipase secretion of fed and unfed adult female Gryllus bimaculatus. The ratio of triacylglyceride lipase, diacylglyceride lipase, and phosphatidylcholine lipase secreted by fed females in the caecum and ventriculus is 1:1.4:0.4. These activities decrease in the caecum by 30-40% in unfed females. The total lipase activity (TLA) in the caecum is about 10 times that in the ventriculus. Minimal lipase secretion occurs before and during the final moult, and remains a

Crickets evade bats via olfaction beyond acoustic cues.

The evolutionary arms race between insectivorous bats and their insect prey is a classic paradigm of acoustic predation and evasion, with insects having evolved sophisticated auditory countermeasures. Both bats and insects also rely heavily on olfaction for key behaviors, such as social communication. Moreover, predator-derived odors are well established as risk cues in many other predator-prey systems. However, whether olfaction plays a role in the bat-insect arms race remains unknown. Here, we unveil a previously unknown olfactory dimension to this interaction. We demonstrated that the body

Genomic signatures of adaptation across a landscape of crickets following the introduction of a parasitoid fly.

Novel species interactions provide an opportunity to assess the earliest stages of genetic adaptation. We combined population genomics and field selection surveys to explore the genomic and geographic landscape of adaptation in small, fragmented Hawaiian cricket populations, which are parasitized by larvae of an introduced fly that targets singing males. Multiple protective male-silencing cricket morphs have recently spread under this novel selection pressure, despite song's important roles in mate attraction. We find evidence of sharp declines in cricket effective population sizes following t

New insights into the flavoring potential of cricket (Acheta domesticus) and mealworm (Tenebrio molitor) protein hydrolysates and their Maillard products.

Insect proteins have an earthy-like flavor and have not shown great flavor potential for food applications so far. In this study, insect proteins of cricket Acheta domesticus and mealworm Tenebrio molitor larvae were first enzymatically hydrolyzed using two peptidase preparations (Flavourzyme1000L and ProteaseA "Amano"2SD). Xylose was then added to facilitate Maillard reactions (30 min, T = 98 °C, 1% (w/v) xylose). A comprehensive sensory evaluation showed that both the hydrolysis and the Maillard reactions changed the flavor description of the samples significantly to more complex and savory-

The effects of chlordane and rotenone on the alimentary tract of the field cricket.

Cricket nymphs (Acheta pennsylvanicus) were treated with the publicly available, commercial forms of 2 generically termed stomach poisons; chlordane and rotenone, and the resulting tissue changes were histologically compared against a control group. Examination utilized stained and unstained slides of the insect's fat body, alimentary tract, and malpighian tubules. The 2 dissimilar operating insecticides shared common effects such as the storage of dark bodies in the fat body, and the structural disruption of the caecal lining and the malpighian tubules. In addition, a unique effect of chlorda

From mating to sperm storage: density-dependent plasticity in pre- and post-copulatory shared mating traits.

Mating interactions depend on traits expressed jointly by males and females, yet the extent to which each sex controls variation in these shared mating traits remains unclear. Because the expression of such traits (like mating latency, copulation duration, and sperm transfer) depends on both partners, their evolution is constrained by intersexual correlations yet facilitated by behavioral plasticity that allows each sex to adjust trait expression across environments. In this study we investigated whether shared mating traits are determined by male or female control or whether the observed outc

Simulation of insect enriched bread consequences on gut microbiota.

Insect powders are newly regulated in Europe and can be used to fortify protein-enriched bakery products. Because protein sources influence gut microbial balance, assessing their impact on the human microbiome is essential. This study investigated how cricket (Acheta domesticus) powder affects colon microbiota and whether sourdough fermentation improves protein-enriched breads. Using an in vitro gut model combining Infogest® digestion with MICODE© colon microbiota fermentation, four bread prototypes were examined. Metabolomic and microbiomic analyses showed that sourdough breads with insect po

Oral administration of GnRH via a cricket vehicle stimulates spermiation in tiger salamanders (Ambystoma tigrinum).

More than 50% of caudates are threatened with extinction and are in need of ex-situ breeding programs to support conservation efforts and species recovery. Unfortunately, many salamander populations under human care can experience reproductive failure, primarily due to missing environmental cues necessary for breeding. Assisted reproductive technologies (ARTs) are a useful suite of techniques for overcoming or bypassing these missing environmental cues to promote breeding. Exogenous hormones are used to stimulate natural breeding behaviors or gamete expression for in-vitro fertilization or bio

Aversive social learning with a dead conspecific is achieved by Pavlovian conditioning in crickets.

Social learning, learning from other individuals, has been demonstrated in many animals, including insects, but its detailed neural mechanisms remain virtually unknown. We showed that crickets (Gryllus bimaculatus) exhibit aversive social learning with a dead conspecific. When a learner cricket was trained to observe a dead cricket on a drinking apparatus, the learner avoided the odor of that apparatus thereafter. Here we investigated the hypothesis that this social learning is achieved by first-order Pavlovian conditioning of an odor (conditioned stimulus) and a dead conspecific (unconditione

Changes in purine and uric acid content in edible insects during culinary processing.

Recently, edible insects were proposed to be promising alternative foods combining nutritional, environmental, and economic benefits. While composition of the basic nutrients of insects is quite well known, little is known about other compounds contained in them such as purines. From this point of view, raw insects were reported to belong among purine-rich foods. However, they are generally consumed after culinary processing, which is known to affect nutritional composition of foods. Therefore, we aimed to analyze the effect of culinary processing (including various combinations of boiling, ro

The effects of high-monosaccharide diets on development and biochemical composition of white-eyed mutant strain of house cricket (Acheta domesticus).

Tryptophan (TRP) is one of the essential amino acids in the animal body. Its exogenicity and low concentrations mean that it can be regarded as one of the key regulatory molecules at the cellular as well as physiological level. It has been shown to have a number of essential functions, such as in the production of other biologically active molecules. The main objective of this project was to investigate the effects of a high monosaccharide diet (HMD) on a hemimetabolic insect-house cricket (Acheta domesticus) and a mutant strain with impaired visual pigment synthesis (closely related to the tr
