Integrative Mapping of SNHG1 RNA-Chromatin Contacts onto the Cancer-Specific Super-Enhancer Landscape in HCT116 Colorectal Cancer Cells.

Long non-coding RNAs (lncRNAs) interact with chromatin and recruit epigenetic complexes to specific genomic loci, yet their relationship with super-enhancers (SEs), key regulatory elements frequently reprogrammed in cancer, remains unexplored. We developed an integrative pipeline that combines RNA-chromatin contact data (RNA-Chrom), histone modification-lncRNA expression correlation profiles (HiMoRNA peaks), and super-enhancer annotations (SEdb 3.0) to map lncRNA-SE regulatory axes. Applying this framework to SNHG1 in HCT116 colorectal cancer cells, we identified 21 SNHG1-reactive super-enhanc

Palladin, an actin-associated protein, is required for adherens junction formation and intercellular adhesion in HCT116 colorectal cancer cells.

Palladin is a scaffold protein involved in the formation of actin-associated protein complexes. Gene expression array analysis on the poorly metastatic HCT116 colon cancer cell line and a metastatic derivative cell line (E1) with EMT (epithelial-mesenchymal transition) features showed a down-regulation of palladin gene expression in the latter. Knockdown of palladin expression in the HCT116 cells suppressed junctional localization of E-cadherin, reduced intercellular adhesion and collective cell migration, showing that palladin plays an important role in maintaining the integrity of adherens j

Design, Optimization and Physicochemical Characterization of Pectin-Based Nanoparticles for In Vitro Anticancer Evaluation Against HCT116 Colon Cancer Cells.

INTRODUCTION: Cancer is a major global health problem characterized by uncontrolled cell proliferation, dysregulated cell cycle progression, and the ability of malignant cells to invade and metastasize to distant organs. Natural flavonoids such as rutin and hesperidin have demonstrated promising anticancer properties; however, their therapeutic application is limited due to poor aqueous solubility and low bioavailability. METHOD: Therefore, the present study aimed to enhance the solubility and colon-targeting efficiency of rutin and hesperidin by formulating them into pectin-based nanoparticl

The selective degradation of PDE4B shortform, using a PROTAC, leads to inhibition of several hallmarks of cancer in HCT116 cells.

BACKGROUND AND PURPOSE: Colorectal cancer is one of the most prevalent cancers worldwide, and novel precision medicine approaches are rapidly improving patient outcomes. Phosphodiesterase 4B (PDE4B) is a member of an enzyme family (PDE4) that hydrolyses intracellular cAMP to control spatial and temporal signalling cues. The PDE4B family, particularly PDE4B shortforms, has been implicated in the progression of colorectal cancer and has emerged as a novel drug target. However, clinically available PDE4 inhibitors are limited therapeutically by their active site-directed action, which lacks subfa

Protective effect assessment of Moringa oleifera against cadmium-induced toxicity in HCT116 and HEK293 cell lines.

The cadmium (Cd) is considered one of the widespread toxic metals in the aquatic and terrestrial environments, which is due to its long half-life, non-degradable characteristic, and toxicity. Aqueous extract of freeze-dried Moringa oleifera (Moringaceae family) leaves was examined for protective effect and antioxidant power against Cd toxicity. The results revealed that Moringa aqueous extract (MAE) has contents of total polyphenols and flavonoids about 30.14 mg GAE/g and 18.35 mg QE/g respectively. Furthermore, phenolic compounds in leaves of Moringa were studied using a high-performance liqu

Notch Receptor 2 Drives Epithelial-Mesenchymal Transition and M2-Like Macrophage Polarization in Human Colorectal Cancer Tissues and HCT116 Cells.

Despite advances in understanding colorectal cancer (CRC) biology, how tumor-cell signaling coordinates epithelial-mesenchymal transition (EMT) with macrophage polarization remains incompletely defined. We investigated whether NOTCH2 expression is associated with EMT and with a GATA3/IL-4-dependent macrophage-polarizing phenotype in CRC. NOTCH2 expression and prognostic associations were analyzed using TCGA data and validated in 20 paired CRC and adjacent normal tissue (ANT) specimens. Gain- and loss-of-function experiments in HCT116 cells assessed proliferation, migration, invasion, EMT marke

N, N-dimethyl-D-erythro-sphingosine increases intracellular Ca2+ concentration via Na+-Ca2+-exchanger in HCT116 human colon cancer cells.

N,N-dimethyl-D-erythro-sphingosine (DMS), an N-methyl derivative of sphingosine, is an inhibitor of protein kinase C (PKC) and sphingosine kinase (SK). In previous reports, DMS-induced intracellular Ca2+ increase concentration ([Ca2+]i) was studied in T lymphocytes, monocytes, astrocytes and neuronal cells. In the present study, we studied DMS-induced increase of [Ca2+]i in HCT116 human colon cancer cells. We found that the DMS-induced increase of [Ca2+]i in colon cancer cells is composed of Ca2+ release from intracellular Ca2+ stores and subsequent Ca2+ influx. The Ca2+ release is not related

Apoptosis Induction in HepG2 and HCT116 Cells by a Novel Quercetin-Zinc (II) Complex: Enhanced Absorption of Quercetin and Zinc (II).

Quercetin forms complexes with various metals due to its structural attributes. It predominantly exhibits chelating activity at the 3-hydroxy/4-carbonyl group. Previously, coordination in synthetically obtained quercetin-zinc (II) complexes has been limited to this group. However, the expanded coordination observed in quercetin-iron complexes has opened avenues for diverse applications. Thus, synthesizing novel quercetin-zinc complexes with different coordination positions is a significant advance. In our study, we not only synthesized and comprehensively characterized a new quercetin-zinc (II

Induction of genotoxic damage and ROS production in HCT116 TP53+/+ and HCT116TP53-/- colorectal cancer cell lines by anticancer drugs.

Reactive oxygen species (ROS) play a dual role in cancer biology, contributing to both tumor progression and therapeutic responses. Many chemotherapeutic agents exert their cytotoxic effects through ROS generation, although the extent and biological relevance of this process remain influenced by cellular context, including the functional status of tumor suppressor genes such as TP53. In this study, we investigated the induction of genomic damage and oxidative stress in two isogenic human colorectal cancer cell lines-HCT116TP53+/+ and HCT116TP53-/--after exposure to four commonly used anticance

An extract from the frass of swallowtail butterfly (Papilio machaon) larvae inhibits HCT116 colon cancer cell proliferation but not other cancer cell types.

BACKGROUND: The frass of several herbivorous insect species has been utilised as natural medicines in Asia; however, the metabolite makeup and pharmaceutical activities of insect frass have yet to be investigated. Oligophagous Papilionidae insects utilise specific kinds of plants, and it has been suggested that the biochemicals from the plants may be metabolised by cytochrome P450 (CYP) in Papilionidae insects. In this study, we extracted the components of the frass of Papilio machaon larvae reared on Angelica keiskei, Oenanthe javanica or Foeniculum vulgare and examined the biological activit
