PURPOSE: Chimeric antigen receptor (CAR) T-cell immunotherapy in acute myeloid leukemia (AML) remains challenging because of the lack of specific cell surface antigens that are highly expressed on leukemic blasts but largely absent in hematopoietic stem/progenitor cells (HSPC) and healthy tissues. Targeting intracellular antigen via T-cell receptor (TCR)-like CAR T cells offers a promising alternative. Using the cell-mediated lympholysis (CML) technique, we studied lymphocytes of six individuals with discrepancies between the karyotypic and phenotypic sex. Two sets of cytotoxic T cells (CTLs) obtained from two multitransfused female aplastic anemia patients were used as typing reagents.
