Mutations in the TM4SF2 gene, which encodes TSPAN7, cause a severe form of intellectual disability (ID) often comorbid with autism spectrum disorder (ASD). Recently, we found that TM4SF2 loss in mice affects cognition. Here, we report that Tm4sf2-/y mice, beyond an ID-like phenotype, display altered sociability, increased repetitive behaviors, anhedonic- and depressive-like states. Cognition relies on the integration of information from several brain areas. In this context, the lateral habenula (LHb) is strategically positioned to coordinate the brain regions involved in higher cognitive funct

Neurotransmitter co-transmission contributes to diverse physiological processes throughout the mammalian brain, including sensory integration, motivational control, and social behaviors. Projections from the globus pallidus internus (GPi; the entopeduncular nucleus, EPN, in rodents) to the lateral habenula (LHb) are well-characterized by the co-transmission of both GABA and glutamate. These dual-release inputs modulate behavioral states in chronically learned helpless (cLH) rats, influencing both the onset and recovery of pathological phenotypes. Here, we employed confocal 3D reconstructions t

BACKGROUND: Our human volumetric MRI study (Dai et al., 2024) demonstrated that habenula (Hb) volume is associated with psychological resilience, a key protective factor against depression. However, the biological mechanisms underpinning this relationship remain unclear. A recent animal study highlighted that neuronal activity in the Hb modulates rapid eye movement (REM) sleep, influencing depressive behaviors during wakefulness. Based on this, we hypothesized that sleep quality mediates the relationship between Hb volume and psychological resilience in humans.
METHODS: We utilized a deep lea

BACKGROUND: Hypersensitivity to punishment is one of the core features of major depressive disorder (MDD). Hypersensitivity to punishment has been proposed to originate from aberrant aversive learning. One of the key areas in aversive learning is the habenula. Although evidence for dysfunctional aversive learning in patients with depression is well established, whether this dysfunction and its neural correlates persist during symptomatic remission of depression remains largely unexplored.
METHODS: Functional magnetic resonance imaging data from 36 medication-free remitted patients with recurr

General anesthetics were first used over 170 years ago; however, the mechanisms of how general anesthetics induce loss of consciousness (LOC) remain unclear. Ciprofol, a novel intravenous anesthetic, has been developed by incorporating cyclopropyl into the chemical structure of propofol. This modification offers the benefits of rapid onset and minimal injection pain. Recent studies have revealed that the glutamatergic neurons of the lateral habenula (LHb) play a crucial role in modulating the LOC induced by propofol and sevoflurane. Nevertheless, the specific involvement of LHb in the anesthet

BACKGROUND: Previous studies have suggested that the habenula (Hb) may be involved in the mechanism of obsessive-compulsive disorder (OCD). However, the specific role of Hb in OCD remains unclear. This study aimed to explore the structural and functional abnormalities of Hb in OCD and their relationship with the clinical symptoms.
METHODS: Eighty patients with OCD and 85 healthy controls (HCs) were recruited as the primary dataset. The grey matter volume, resting-state functional connectivity (FC), and effective connectivity (EC) of the Hb were calculated and compared between OCD group and HC

Drugs of abuse can persistently change the reward circuit in ways that contribute to relapse behavior, partly via mechanisms that regulate chromatin structure and function. Nuclear orphan receptor subfamily4 groupA member2 (NR4A2, also known as NURR1) is an important effector of histone deacetylase 3 (HDAC3)-dependent mechanisms in persistent memory processes and is highly expressed in the medial habenula (MHb), a region that regulates nicotine-associated behaviors. Here, expressing the Nr4a2 dominant negative (Nurr2c) in the MHb blocks reinstatement of cocaine seeking in mice. We use single-n

Depression is a devastating disorder with a combination of diverse symptoms such as low self-esteem, lack of motivation, anhedonia, loss of appetite, low energy, and discomfort without a clear cause. Depression has been suggested to be the result of maladaptive changes in specific brain circuits. Recently, the lateral habenula (LHb) has emerged as a key brain region in the pathophysiology of depression. Increasing evidence from rodent, nonhuman primate, and human studies indicates that the aberrant activity of the LHb is associated with depressive symptoms such as helplessness, anhedonia, and

The treatment of internet gaming disorder (IGD) poses a significant challenge. Light therapy has demonstrated efficacy in preventing cocaine-driven reinstatement in a rodent model. The potential of light therapy, along with its underlying neural mechanisms, in treating IGD remains largely uninvestigated. A total of 104 participants are included in the RCT and 69 in the fMRI analysis. The intervention involves 14 daily sessions of light therapy, light placebo, or cognitive training. At post-intervention, light therapy group shows treatment effects comparable to those of cognitive training group

The lateral habenula (LHb) plays essential roles in behavioral responses to stressful events. Stress is tightly linked to autonomic responses such as cardiovascular responses, yet how the LHb regulates these responses is not well understood. To address this issue, we electrically stimulated the LHb in rats, measured its effects on heart rate (HR) and mean arterial pressure (MAP), and investigated the neural circuits that mediate these LHb-induced cardiovascular responses via the autonomic nervous system. We observed that stimulation of the LHb induced bradycardia and pressor responses, whereas

Chronic nicotine upregulates nicotinic acetylcholine receptors (nAChRs) throughout the brain, and reducing their activity may promote somatic and affective states that lead to nicotine seeking. nAChRs are functionally upregulated in animal models using passive nicotine administration, but whether/how it occurs in response to volitional nicotine intake is unknown. The distinction is critical, as drug self-administration (SA) can induce neurotransmission and cellular excitability changes that passive drug administration does not. In this study, we probed the question of whether medial habenula (

Disproportionate anger and reactive aggression in response to provocation are core symptoms of intermittent-explosive disorder (IED). Previous research shows a link between the propensity for aggression in healthy individuals and altered functioning of prefrontal-limbic and default-mode networks (DMN) at rest when no provocation is present. In a pilot study, we used resting-state functional magnetic resonance imaging to investigate the effects of pronounced reactive aggression in men, exemplified by IED, on the functional organization of resting-state brain networks including subcortical nodes

Lateral habenula (LHb) hyperactivity plays a pivotal role in the emergence of negative emotional states, including those occurring during withdrawal from addictive drugs. We have previously implicated cocaine-driven adaptations at synapses from the entopeduncular nucleus (EPN) to the LHb in this process. Specifically, ionotropic GABAA receptor (R)-mediated neurotransmission at EPN-to-LHb synapses is reduced during cocaine withdrawal, due to impaired vesicle filling. Recent studies have shown that metabotropic GABAB R signaling also controls LHb activity, although its role at EPN-to-LHb synapse

Choroid plexus, pineal gland, and habenula tend to accumulate physiologic calcifications (concrements) over a lifetime. However, until now the composition and causes of the intracranial calcifications remain unclear. The detailed analysis of concrements has been done by us using X-ray diffraction analysis (XRD), X-ray diffraction topography (XRDT), micro-CT, X-ray phase-contrast tomography (XPCT), as well as histology and immunohistochemistry (IHC). By combining physical (XRD) and biochemical (IHC) methods, we identified inorganic (hydroxyapatite) and organic (vimentin) components of the concr

Ketamine is an NMDA receptor antagonist with rapid antidepressant properties when administered at a subanesthetic dose. Preclinical models indicate that a direct injection of ketamine into the lateral habenula (Hb), a small midbrain structure with an evolutionarily preserved role in aversive learning across mammals, can rapidly relieve depression-like behavior. However, there is limited evidence to explain how ketamine acts on the function of the human Hb. In a translational computational neuroscience study, 70 healthy adult volunteers were randomized in a 1:1 ratio to receive ketamine (0.5 mg

Research has implicated disrupted autophagy initiation in the pathophysiology of depression. However, the role of Unc-51-like Kinase 1 (ULK1), a key regulator of autophagy initiation, in depression remains poorly understood. Here, using behavioural, molecular, and genetic approaches, we show that serum ULK1 levels are decreased in both patients with depression and mice susceptible to chronic social-defeat stress, and that both ULK1 and p-ULK1 (Ser-555) are downregulated in the lateral habenula (LHb) of susceptible mice. In addition, ULK1 deficiency in the LHb is sufficient to induce depressive

BACKGROUND: Electroconvulsive therapy (ECT) remains a highly effective intervention for acute episodes of major depressive disorder, offering rapid and robust antidepressant effects. However, its underlying mechanisms remain unclear, as prior studies focusing on conventional brain regions (e.g., the hippocampus) have not fully accounted for ECT's distinct therapeutic profile compared to slow-acting antidepressants. Emerging evidence implicates the lateral habenula (LHb) in mediating rapid antidepressant responses. Nevertheless, its role in ECT's efficacy and the involvement of key molecular ta

Substance P (SP) levels are closely related with the pathogenesis of depression. Recent work has focused on antidepressive effect of substance P receptor antagonist (SPA), however, its action site and mechanism remain largely unresolved. Our previous results showed that the lateral habenula (LHb) plays a key role in the pathogenesis of depression. The current study investigated the effects of SPA microinjected into LHb on the behavioral responses of two rat models that exhibit depression-like behavior. To produce adult rats that exhibit depression-like behavior, rats were either exposed to chr

Itch is an aversive sensory experience inextricably linked to a desire to scratch.1,2 While recent progress has advanced our understanding of itch sensation and transmission, the neural mechanisms underlying how scratching is sensed and controlled remain largely elusive. Here, we show that the left lateral habenula (LHb), which responds to negatively valent somatosensory stimuli,3,4,5,6,7,8 preferentially engages and negatively regulates chloroquine (CQ)-evoked itch and scratching. Calcium activity in left LHb neurons increases when scratching is terminated, and their activation is sufficient

OBJECTIVE: Irritability significantly impacts the mental health and well-being of youth and their families and is associated with adverse concurrent and future outcomes. Brain abnormalities in the reward system are a potential mechanism underlying irritability. However, the habenula, a region closely linked to reward processing, has not been investigated in youth irritability. Additionally, previous research has rarely explored how inconsistent parenting interacts with neurobiological factors to contribute to youth irritability. Thus, this study examined the associations of habenula activity a
