Vaccines against gastrointestinal nematodes are one potential option for the control of parasitic gastroenteritis in ruminants. Excretory/secretory (E/S) and hidden antigens are being studied as candidates for vaccines against Haemonchus spp., which is a major parasite in cattle and small ruminants that are raised in warm climates. Protection has been observed after vaccination with some E/S proteases, particularly cysteine proteases and with some glycans that are abundant on the surfaces and in the secretory products of helminths. However, the most promising results are being obtained with gl

Albendazole at dose rates of 11 to 54 mg/kg (mean of 25 +/- 11 SD) of body weight was highly effective (greater than 99%) against Haemonchus contortus, Ostertagia odocoilei, O mossi, Trichostrongylus askivali and Nematodirus odocoilei in 22 white tailed deer (Odocoileus virginianus) in southern Texas. It was 38% effective against mature and immature deer liver flukes (Fascioloides magna). Toxicoses associated with treatment were not observed. Large numbers of abomasal nematodes (mean of 2,949 +/- 3.147 SD) were recovered from 16 untreated control deer, indicating these may have had a potential

Variable resistance to Haemonchus contortus infection and the associated immune mechanisms have been investigated and characterized in different sheep breeds adapted to tropical climates. In a recent study, we compared the local immune responses of the resistant Santa Inês with those of the susceptible White Dorper and Texel sheep breeds under chronic haemonchosis, and evaluated the correlations of immune-related transcripts and different parasitic stages and sexes. The aim of present study was to further elucidate the immune mechanisms among these sheep breeds by comparing local responses, mi

The development of anthelmintic resistance due to long-term and irregular use of drugs such as ivermectin (IVM) poses a major challenge to the prevention and control of haemonchosis caused by Haemonchus contortus. However, the mechanism of IVM resistance in H. contortus remains incompletely understood. Reactive oxygen species (ROS) generated under oxidative stress conditions serve as key regulators of autophagy. Reactive oxygen species induce autophagy, and autophagy mitigates oxidative stress-mediated damage, thereby enhancing cell survival. Although autophagy is known to contribute to H. con

BACKGROUND: Pathogenic blood-feeding nematodes, such as Haemonchus contortus and the invasive Ashworthius sidemi, infect a wide range of wild and domestic ruminants. While the spread of A. sidemi among European cervids has been studied, its presence in chamois (Rupicapra rupicapra) remains poorly documented. Conversely, H. contortus is known to infect chamois, but previous research has relied mainly on cross-sectional necropsy studies, offering only a limited view of infection dynamics. In this study, we used a longitudinal molecular approach to assess the seasonal occurrence and transmission

BACKGROUND: Arthrobotrys species are nematophagous fungi that secrete extracellular nematocidal products (ECP). The individual and combined effects of ECP from Arthrobotrys oligospora (Ao) and A. musiformis (Am) growth in liquid media against Haemonchus contortus L3 (HcL3) were assessed.
METHODS: The isolation, morphological (MI) and molecular identification (Mol-I), assessment of nematocidal activity (NA) of fungal liquid culture filtrates (LCF) in two liquid media alone and in combination and the myco-compound profile identification (MCP) were performed.
RESULTS: The MI suggested that the

Nematophagous fungi (NF) form part of the soil microbiota and are natural enemies of nematodes, helping to regulate nematode populations. A verticillate NF isolated from soil from Tepalcingo, Mexico, was morphologically and molecularly characterised. This fungus was cultured in two different liquid media-Czapek-Dox broth (CzDoxB) and sweet potato dextrose broth (SPDB)-for 21 days. The ovicidal (OA) and larvicidal (LA) activities of fungal liquid culture filtrates (LCFs) were assessed in 96-well microtitre plates at different concentrations against Haemonchus contortus after 48 h. The morpholog

In this study, a three-step experiment was conducted to evaluate the effectiveness of orange essential oil (OEO) to reduce the severity of parasitic infection in lambs naturally infected with gastrointestinal parasites. In the first and second stages of the experiment, the lambs were distributed in groups (n = 10): without treatment (control), treated with levamisole (7.5 mg/kg), and treated either with 200, 300 or 400 mg/kg OEO in the first stage and with two doses of 300 or 350 mg/kg each of OEO with a six-hour interval in the second stage. In the third stage of the experiment, 20 lambs were

Haemonchus contortus is a parasite species that affects the health and production of grazing small ruminants in different parts of the world. Scanning electron microscopy (SEM) is an important tool for the study of parasites' morphology and taxonomy as it generates images that appear 3D and are generally easier to interpret than optical microscopy images. This study used the SEM to describe the vulval types of adult H. contortus from a Mexican isolate. A total of 14 adult H. contortus females were obtained from two artificially infected goats. Females were fixed and processed by critical point

BACKGROUND: The Barber's Pole worm, Haemonchus contortus is of major concern to sheep producers, particularly in the southern hemisphere. This nematode is also commonly found in many sheep flocks in Northern hemisphere countries but is generally not associated with acute clinical pathology. As with other nematode species, the pattern of disease is changing in the United Kingdom. Changes in management practices, climate, anthelmintic resistance prevalence and parasite adaptation are possible factors thought to be responsible for this.
METHODS: In the present study, a combination of traditional

Heterologous antibody‑mediated immunity was evaluated in cattle naturally infected with Ostertagia ostertagi (Oo) and Haemonchus sp. (Hp). Antibody responses were assessed using ovalbumin (OVA) as unrelated model antigen in three groups of eight heifers, matched for eggs per gram of feces (EPG) and allocated to independent, nematode‑free pastures. Anti‑OVA titers were determined in serum by ELISA. EPG counts and nematode genera were determined in feces using the modified McMaster technique and pooled cultures, respectively. All variables were analyzed using mixed models. One group was previous

The objective of the study was to determine the efficacy of white wormwood on helminthes in beef cattle production. Water extracts of white wormwood of different levels of phytotoxicity were used to treat female adult H. contortus over 8 h under controlled laboratory conditions. The experiment was designed in a completely randomized design with six treatments replicated three times. Treatments 3-6 showed a reduction in worm motility over time (P < 0.05) while it did not change much for T1 and T2 (P < 0.05). Even after 8 h of incubation, more than 50% of the worms were still active in T1, T2 an

This paper examines the impact of climate change in Africa on the transmission potential (Q0) of Haemonchus contortus (H. contortus), a highly pathogenic haematophagous gastrointestinal nematode (GIN) parasite that has significant implications for the health and productivity of small ruminants. Changes in Q0 were assessed over the period 1981-2070 under a high emission scenario (RCP 8.5). Climate data was sourced from bias-adjusted Coordinated Regional Climate Downscaling Experiment (CORDEX) models for Africa, which was split into 13 subregions based on areas within each having approximately s

The most natural areas typically host a variety of plant species that serve as nutritional centers for grazing ruminants. We aimed to determine the growth parameters, histology, and ruminal microbial characteristics of lambs with endoparasites grazing on meadow pasture with experimentally sown chicory (Cichorium intybus). Lambs infected with the larvae of the endoparasite Haemonchus contortus were divided into two groups: lambs grazing in the meadow grassland plot (CON) and lambs grazing in the experimental plot recultivated with chicory (CHIC). Similar initial weights of lambs of both groups

Although benefits of selection for host resistance to gastro-intestinal nematodes have long been recognized, its costs on production traits remain unclear. A main difficulty when studying those costs is to disentangle genetic effects due to selection from plastic responses induced by infection. Putative costs of host resistance have been extensively investigated in growing sheep. However, while most of those studies have relied on live weight to assess body growth, more comprehensive assessments accounting for body composition are advocated to detect trade-offs. In this study we used 90 female

Haemonchus contortus is a constraint to sheep production. Seeking to reduce the use of hosts and produce parasitic stages in large-scale, a 42-day in vitro culture protocol of H. contortus third-stage larvae was optimized using Dulbecco's modified Eagle's medium (DMEM). In cell-free culture, larvae were maintained at 39.6°C, in acidic media (pH 6.1) for 3 or 6 days with Δ4-dafachronic acid followed by DMEM pH 7.4 supplemented or not with Fildes' reagent. In DMEM pH 7.4 at 37°C, supplementation with Caco-2 cells was compared to Fildes. On Day 14, fourth-stage larvae (L4) development rates in ac

The control of gastrointestinal nematodes in livestock is becoming increasingly difficult due to the limited number of available drugs and the rapid development of anthelmintic resistance. Therefore, it is imperative to develop new anthelmintics that are effective against nematodes. Under this context, we tested the potential toxicity of three compounds in mice and their potential anthelmintic efficacy in Mongolian gerbils infected with Haemonchus contortus. The compounds were selected from previous in vitro experiments: two diamine (AAD-1 and AAD-2) and one benzimidazole (2aBZ) derivatives. 2

Widespread resistance in parasitic nematodes to most classes of anthelmintic drugs demands the discovery and development of novel compounds with distinct mechanisms of action to complement strategic or integrated parasite control programs. Products from nature-which assume a diverse 'chemical space'-have significant potential as a source of anthelmintic compounds. In the present study, we screened a collection of extracts (n = 7616) derived from marine invertebrates sampled from Australian waters in a high throughput bioassay for in vitro anti-parasitic activity against the barber's pole worm

Haemonchus contortus is a pathogenic gastrointestinal nematode of small ruminants and, in part due to its capacity to develop resistance to drugs, contributes to significant losses in the animal production sector worldwide. Despite decades of research, comparatively little is known about the specific mechanism(s) driving resistance to drugs such as ivermectin in this species. Here we describe a genome-wide approach to detect evidence of selection by ivermectin treatment in a field population of H. contortus from Sweden, using parasites sampled from the same animals before and seven days after
