Chromosome analysis of multipronuclear human oocytes after in vitro fertilization.

Multipronuclear human eggs are frequent after in vitro fertilization. Their chromosome analysis can provide useful information. Before cleavage it can confirm the suspected polyploidy. Among the cleaved multipronuclear eggs it provides an estimation of the incidence of the possible return to diploidy. Ninety-four multipronuclear eggs were fixed at the first, second, or third cleavage according to the air-drying method of Tarkowski with or without colchicine exposure: 60 were successfully analysed. Twelve were stopped before cleavage (six without colchicine treatment and six with colchicine tre

Long telomere inheritance through budding yeast sexual cycles.

The ends of linear eukaryotic chromosomes are protected from being recognized as DNA double-strand breaks by telomeres, containing repetitive DNA sequences that bind specific proteins. In humans, mutations in telomere regulatory genes lead to short or long telomere syndromes. These syndromes often show genetic anticipation, where the disease has an earlier onset and a more severe manifestation in each new generation. Later generations inherit not only the mutation affecting telomere length, but also abnormal telomere length. Many aspects of telomere length homeostasis are conserved between mam

Maternal ploidy shapes reproductive pathways in the triploid hybrid Chrosomus eos × eos-neogaeus.

Elements transferred from a mother to her eggs may strongly influence the phenotype of her offspring. Such maternal effects depend on the genotype of the mother, and while multiple ploidy levels occur naturally in some vertebrate species, studies evaluating the impact of maternal ploidy on offspring are scarce. This paper aimed to test whether maternal ploidy is responsible for the two reproductive phenotypes observed in the triploid fish Chrosomus eos × eos-neogaeus. Indeed, these hybrids have two different maternal origins (diploid or triploid) and display two reproductive phenotypes, ameiot

Culture-space control is effective in promoting haploid cell formation and spermiogenesis in vitro in neonatal mice.

The classical organ culture method, in which tissue is placed at the gas‒liquid interphase, is effective at inducing mouse spermatogenesis. However, due to reginal variations in the supply of oxygen and nutrients within a tissue, the progress of spermatogenesis was observed only in limited areas of a tissue. In addition, haploid cell formation and its differentiation to spermatozoon, i.e. spermiogenesis, were infrequent and inefficient. Here, we show that the polydimethylsiloxane (PDMS)-chip ceiling (PC) method, which ensures a uniform supply of nutrients and oxygen throughout the tissue by pr

Chromosome anomalies in human oocytes in relation to age.

Cytogenetic analysis of oocytes remaining unfertilized after in-vitro fertilization showed that the source of data obtained could be divided into degenerating and 'Работает' oocytes. The degenerating oocytes, which showed different degrees of chromosome breakage, accounted for a quarter of the total. They were found in older patients with a mean age of 35.0 years. The healthy oocytes without chromosome breaks were mostly haploid and fell into two main groups, those with a normal MII,23,X chromosome complement, and those abnormal in which single chromatids replaced a whole chromosome. No oocytes

Cytogenetic and molecular genetic studies of follicular and papillary thyroid cancers.

Cytogenetic studies have shown frequent clonal abnormalities in papillary carcinoma (PTC) and follicular carcinoma (FTC). Loss of heterozygosity (LOH) may suggest the presence of tumor suppressor genes and has not been reported in these neoplasms. These studies were undertaken to determine if consistent chromosomal abnormalities are associated with thyroid cancer, to determine likely regions for molecular genetic investigations, and to determine if there is allelic loss in thyroid tumors. Cytogenetic analysis of 26 PTC and 5 FTC showed clonal abnormalities in 9 and included -Y, +5, or inv(10)(

Bread Wheat Microspore Culture.

Microspore culture is one of the doubled-haploid-producing techniques that can speed up the breeding process and shorten the time for releasing new cultivars. Growing world population and changing climate demand to expedite the development of new resilient and high-yielding cultivars for staple crops such as bread wheat (Triticum aestivum L.). Here, we describe a comprehensive protocol for obtaining bread wheat doubled haploids through microspore culture. Key steps including donor plant growth and spike pretreatment, microspore isolation and density-gradient-based purification, microspore-ovar

Nonylphenol reduced the number of haploids in in vitro spermatogenesis of the endangered cyprinid Gnathopogon caerulescens.

Nonylphenol (NP), an endocrine disrupting chemical, is widely used in industrial and agricultural processes, causing NP influx into aquatic environments. NP induces hormonal imbalance, and male feminization, and reduces germ cell production during spermatogenesis; however, the mechanism by which it affects spermatogenesis remains unknown. Here, we investigated the effect of NP on spermatogenesis in honmoroko (Gnathopogon caerulescens), an endangered fish endemic to Lake Biwa, Japan, using an in vitro differentiation system. We collected spermatogonia from the testes of non-spawning G. caerules

Apparent diameter and cell density of yeast strains with different ploidy.

Optical density at 600 nm (OD600) measurements are routinely and quickly taken to estimate cell density in cultivation and to track cell growth. The yeast Saccharomyces cerevisiae is one of the microorganisms most used in industry, and the OD600 values are frequently adopted as the indicator of yeast cell density, according to the Beer-Lambert law. Because the OD600 value is based on turbidity measurement, the Beer-Lambert law can be applied only for microbial cultivation with low cell densities. The proportionality constants strongly depend on several parameters such as cell size. Typically,

Effects of polyploidization and their evolutionary implications are revealed by heritable polyploidy in the haplodiploid wasp Nasonia vitripennis.

Recurrent polyploidization occurred in the evolutionary history of most Eukaryota. However, how neopolyploid detriment (sterility, gigantism, gene dosage imbalances) has been overcome and even been bridged to evolutionary advantage (gene network diversification, mass radiation, range expansion) is largely unknown, particularly for animals. We used the parasitoid wasp Nasonia vitripennis, a rare insect system with heritable polyploidy, to begin addressing this knowledge gap. In Hymenoptera the sexes have different ploidies (haploid males, diploid females) and neopolyploids (diploid males, tripl

Evolution of karyotype in haploid cell lines of Drosophila melanogaster.

Seven continuous cell lines have been established in vitro from lethal embryos produced by the female sterile mutant mh 1182 of Drosophila melanogaster. Six lines show haploid metaphases. Karyotype analysis revealed a high level of aneuploid cells with frequent chromosome fragments. In three lines, haploid cells were quickly overgrown by diploid cells. Two lines were more stable but the proportion of haploid cells decreased with time. One line was stable, showing 80-90% of haploid cells for over 1 000 cell generations. Stable haploid clones have been isolated from two lines. Crossing of mh 118

Possible linkage between the major histocompatibility complex and the immune response to factor VIII in classic haemophilia.

The pattern of HLA segregation was analyzed in 10 sibships of haemophilia A patients, each of which included 1 or 2 haemophilic brothers with an antibody to factor VIII. The observed HLA distribution is suggestive of a linkage between the major histocompatibility complex and a gene responsible for immune response to isologous factor VIII.

Evidence for genetic etiology of heteroploidy in embryos of the Japanese quail (Coturnix coturnix japonica).

The Japanese quail was used as an experimental system to detect the effects of genes that affect chromosome behavior and distribution. From a random-bred population, three inbred generations were produced by full-sib matings in 36 families. The expectation from such a breeding scheme was that embryos bearing aberrations induced by recessive mutant genes would cluster within families and recur in particular lineages. Chromosomal aberrations caused by errors during fertilization, cleavage mitosis, and gametogenesis were scored in 2,037 16- to 18-h embryos from 107 families. Comparisons of the ob

Relationship between chromosome fragility, aneuploidy and severity of the haematological disease in Fanconi anaemia.

Fanconi anemia (FA) is a chromosome instability syndrome, characterized by progressive pancytopenia and cancer susceptibility. Other cellular features of FA cells are hypersensitivity to DNA cross-linking agents and accelerated telomere shortening. We have quantified overall genome chromosome fragility and euploidy as well as chromosomes 7 and 8 aneuploidy in peripheral blood lymphocytes from a group of FA patients and age-matched controls that were previously measured for telomere length. The haematology of FA samples were also characterized in terms of whole blood cell, neuthrophil and plate

Reverse Breeding: A Breakthrough Technique for Genetic Fixation and Hybrid Reconstruction.

Reverse breeding (RB), a novel plant breeding approach, helps to identify the parental homozygous lines for any uncharacterized but useful heterozygous plant that a plant breeder needs utmost. For this, genetic engineering at meiotic cell division, particularly meiosis-I, must achieve the right combination and complement homozygous parental lines for recreating the desired hybrid. In this technology, meiotic cross-over is suppressed to stop the recombination at the chromosome level in the selected heterozygous plants. The success of RB in crops depends on the efficiency and effectiveness of do

First description of heterogeneity in 18S rRNA genes in the haploid genome of Cryptosporidium andersoni Kawatabi type.

The Apicomplexan Cryptosporidium andersoni, is a species of gastric Cryptosporidium, is frequently detected in older calves and adult cattle. Genotyping analyses based on 18S ribosomal RNA gene sequences have been performed on a novel C. andersoni genotype, namely the Kawatabi type, and the oocysts were classified into two distinct groups genotypically: Type A (the sequence in GenBank) and Type B (with a thymine nucleotide insertion not in Type A). This study analyzed 3775 cattle at a slaughterhouse and 310 cattle at a farm using microscopy and found 175 Cryptosporidium-positive animals: 171 f

Imaging of SHOX-associated anomalies.

Human growth is a multifactorial trait influenced by environmental, hormonal, and genetic factors. Although it is clear that multiple factors contribute to an individual's final height and limb development, genetic factors play a crucial role. One such gene is the short stature homeobox ( SHOX) containing gene. Knowledge about the SHOX gene has rapidly increased since its discovery in 1997, and we now know that SHOX haploinsufficiency affects the development of the extremities and is an important cause of short stature. Currently, SHOX mutations occur with an estimated incidence of roughly 1 i

Male infant with paternal uniparental diploidy mosaicism and a 46,XX/46,XY karyotype.

A male patient with mosaic paternal uniparental diploidy (PUD) is presented. After birth, the patient presented with hypoglycemia, hemihypertrophy, umbilical hernia, and hepatomegaly. Afterward pancreatic hypertrophy, liver hemangiomas, and cysts were detected sonographically. At the age of 3.5 months, hepatoblastoma was diagnosed. To investigate suspected Beckwith-Wiedemann syndrome (BWS), extensive genetic analyses were performed using DNA from chorionic villus sampling, amniocentesis, and peripheral blood lymphocytes (chromosome analysis, methylation-specific multiplex ligation-dependent pr
