[Serotonin modulation of acetylcholine-induced currents in mollusk neurons as dependent on cyclic guanosine monophosphate].

It was found that a serotonin-induced decrease of the acetylcholine-induced chloride current in a group of right parietal ganglion neurons of the snail Helix pomatia was mimicked neither by dibutyryl cAMP nor by isobutyl methylxanthine. In contrast it was mimicked by the extracellular application of sodium nitroprusside as a guanylate cyclase activator, and by intracellular injection of cyclic guanosine monophosphate.

[Critical role of intracellular calcium in plasticity mechanisms of defense behavior command neurons LPl1 and PPl1 of Helix lucorum during nociceptive sensitization].

The role of intracellular calcium in changes in excitability and responses of defense behavior command neurons LP11 and PP11 of Helix lucorum to sensory stimulation was investigated in semi-intact preparation of a snail during nociceptive sensitization. It was found that application of sensitizing stimuli onto the snail's head initiated membrane depolarization, increase in its excitability as well as depression of neural responses evoked by sensory stimuli in short-term period of sensitization and significant facilitation of neural responses in long-term period of sensitization. Cytoplasmic Ca2+ activity regulation as measured by a calcium-activated current.

Calcium-activated non-selective cation (CAN) currents were activated by quantitative injections of Ca2+ into voltage clamped bursting neurons of the snails Helix aspersa or Helix pomatia. Membrane potential was held at the potassium equilibrium potential and CAN currents were fit with a rising and falling exponential function. Ca2+ transporters and pumps of the cell membrane, endoplasmic reticulum, and mitochondria were selectively blocked with pharmacological agents. Bath solutions containing 0 Na Ringers, chlorpromazine, Na3VO4, or thapsigargin did not significantly change the CAN current de

A Biomphalaria glabrata peptide that stimulates significant behaviour modifications in aquatic free-living Schistosoma mansoni miracidia.

The human disease schistosomiasis (or bilharzia) is caused by the helminth blood fluke parasite Schistosoma mansoni, which requires an intermediate host, the freshwater gastropod snail Biomphalaria glabrata (the most common intermediate host). The free-swimming parasite miracidia utilise an excellent chemosensory sense to detect and locate an appropriate host. This study investigated the biomolecules released by the snail that stimulate changes in the behaviour of the aquatic S. mansoni miracidia. To achieve this, we have performed an integrated analysis of the snail-conditioned water, through

Functional morphology of the neuropeptidergic light-yellow-cell system in pulmonate snails.

The light yellow neuropeptidergic cell system of the basommatophoran snail Lymnaea stagnalis is homologous to the R3-R14 system of the opisthobranch Aplysia californica, and produces three different neuropeptides. Systems homologous to the light yellow cells of Lymnaea stagnalis have been investigated morphologically in two Basommatophora (Lymnaea ovata, Bulinus truncatus) and three Stylommatophora (Helix aspersa, Cepaea nemoralis, Deroceras reticulatum). To this end, an antibody to synthetic light-yellow-cell peptide-II and oligonucleotides to mRNAs encoding parts of peptide-I and peptide-III

Snails as indicators of pesticide drift, deposit, transfer and effects in the vineyard.

This paper presents a field-study of real pesticide application conditions in a vineyard. The objective was to measure the exposure, the transfer and the effects of pesticides on a non-target soil invertebrate, the land snail Helix aspersa. There was no drift of the herbicides (glyphosate and glufosinate) whereas the fungicides (cymoxanil, folpet, tebuconazole and pyraclostrobin) were detected up to 20 m from the treated area. For folpet and particularly tebuconazole, spray deposits on soil (corresponding to losses for the intended target i.e. the vine leaves) were high (41.1% and 88.8% loss o

Optical recording of odor-evoked responses in the olfactory brain of the naïve and aversively trained terrestrial snails.

Regular spontaneous oscillations were recorded both electro- and optophysiologically using a voltage-sensitive absorption dye in the olfactory part of the brain (procerebral lobe of the cerebral ganglia) of the gastropod mollusk Helix lucorum. Odor application caused transient changes in procerebral oscillations, and an odor-evoked potential was recorded in the procerebrum (PC). The wave of evoked potential originated near the place of olfactory nerve entrance into the PC and propagated via the procerebral neuropile toward the cell body layer. The spread of the odor-evoked potential correspond

Histaminergic neurons in the central and peripheral nervous system of gastropods (Helix, Lymnaea): an immunocytochemical, biochemical, and electrophysiological approach.

Distribution, chemical-neuroanatomy, concentration, and uptake-release properties of histamine (HA)-containing neurons and the possible physiological effects of HA in the central and peripheral nervous system of the pulmonate snails, Helix pomatia and Lymnaea stagnalis, are described. In the CNS of both species, the distribution pattern of HA-immunoreactive (HA-IR) neurons was similar. In both species the majority were located in the buccal, cerebral, and pedal ganglia. In Helix, approximately 400 HA-IR neurons were seen, whereas in Lymnaea approximately 130 labeled cells were visualized. The

Antibacterial and antibiofilm properties of Helix aspersa mucus towards multidrug resistant Staphylococcus aureus.

The increasing prevalence of multidrug-resistant bacteria necessitates the exploration of novel antimicrobial agents. This study aims to investigate the antibacterial and antibiofilm properties of mucus from Helix aspersa, a species of terrestrial snail, against multidrug resistant Staphylococcus aureus strains. The antibacterial effect was assessed using well diffusion, microdilution, and time kill assays. The antibiofilm effect was assessed using crystal violet staining. Protein profiling was conducted through SDS-PAGE Electrophosis to determine the molecular weights of the mucus proteins. T

[ATl- and calcium-dependent mechanisms of salicy- lates influence on electric potentials of neurons in mollusk Helix albescens].

The article is devoted to investigation of mechanisms of salicylates influence on electric potentials in neurons of mollusk Helix albescens Rossm. It is shown that adenosinetriphosphate (5 x 10(-4) M) at the combined bath application with salicylates substantially modifies their neurotropic effects. It removes nonselective inhibition of impulse activity of neurons by acetylsalicylic acid and strengthens activating effects of cobalt and zinc acetylsalicylates. It is shown that at the blockade of Ca2+ entry by CdCl2 or BaCl2 (5 x 10(-5) and 5 x 10(-4) M) in a neuron from extracellular space or f

Nitric oxide increases excitability by depressing a calcium activated potassium current in snail neurons.

In gastropods, the interneuronal messenger, nitric oxide (NO), modulates spike frequency and synaptic transmission. We have characterized the effect of NO on ion currents underlying neuronal excitability, using current-clamp and two-electrode voltage-clamp techniques. Identified neurons of the pulmonate snail, Helix pomatia, respond to the NO donor sodium nitroprusside (SNP) by increasing the firing frequency and decreasing the latency. Voltage-clamp experiments revealed that SNP or S-nitro-N-acetylpenicillamine (SNAP) depressed the macroscopic outward current, while the control compound N-ace

4-aminopyridine activates a cholinergic chloride conductance in isolated Helix neurons.

Isolated Helix neurons were internally perfused under voltage-clamp, with drugs applied by the 'concentration clamp' which allowed an extremely rapid switching of the external solution. 4-Aminopyridine (4-AP) at more than 3 X 10(-4) M induced a chloride current (ICl) which could be blocked by D-tubocurarine. Acetylcholine (ACh) also induced ICl which was similarly blocked. Simultaneous application of 4-AP and ACh resulted in potentiation of ICl over the sum of respective currents. When preceded by either one, the other ICl was reduced in size, showing cross-desensitization. These data indicate

Neural control of olfaction and tentacle movements by serotonin and dopamine in terrestrial snail.

We investigated the role of serotonin (5HT) and dopamine (DA) in the regulation of olfactory system function and odor-evoked tentacle movements in the snail Helix. Preparations of the posterior tentacle (including sensory pad, tentacular ganglion and olfactory nerve) or central ganglia with attached posterior tentacles were exposed to cineole odorant and the evoked responses were affected by prior application of 5HT or DA or their precursors 5-hydroxytryptophan (5HTP) and L: -DOPA, respectively. 5HT applications decreased cineole-evoked responses recorded in the olfactory nerve and hyperpolari

Neural circuit mediating tentacle withdrawal in Helix aspersa, with specific reference to the competence of the motor neuron C3.

The tentacle withdrawal reflex in the terrestrial snail Helix aspersa involves bending and retraction of the tentacles. When elicited by mechanical stimulation of the tentacle, the reflex is mediated by the conjoint action of the central and peripheral nervous systems. The neural circuit underlying the stimulus-response pathways was studied in vitro using a combination of morphological and physiological techniques. Sensory input caused by stimulation of the nose (situated at the superior tentacle's tip) first passes into the tentacle ganglion. Motor fibers are likely excited in the tentacle ga

[Blocking action of calmidazolium and chlorpromazine on the potassium outward current which depends on intracellular calcium ions].

Effects of calmidazolium (R 24571) and chlorpromazine on the delayed potassium outward current in the somatic membrane were studied on nonidentified intracellularly perfused neurons of the snail Helix pomatia. Extracellular application of these substance evoked depression of the outward current. Inhibition of IK occurs at concentrations of calmodulin inhibitors 10(-9)-10(-8) mol/l. These agents inhibit primarily a component of the potassium current depending on the intracellular Ca2+ ions (IK(Ca in)). The inhibitory effect of these drugs can be explained by calmodulin-like structures of the re

Field evaluation of a novel molluscicide (niclosamidate) against Oncomelania hupensis, intermediate host of Schistosoma japonicum.

The molluscicidal activity of a novel molluscicide (niclosamidate) was evaluated in field trials against Oncomelania hupensis, the intermediate host of Schistosoma japonicum. The environmental safety of niclosamidate for local fishes was also studied under field conditions. The results showed that, at the dosages of 8.0 g/m2 and 4.0 g/m3, niclosamidate exhibits highly potent molluscicidal activity in the spraying and immersion trials, resulting in mortality rates of up to 81.8 and 72.7%, respectively. Its performance seems to be target-specific, with good molluscicidal ability observed for Onc

The mechanisms of memory traces storage and interaction in snails: Role of DNA methylation and protein synthesis.

Understanding the molecular mechanisms underlying the storage and interaction of different memory traces remains an important, yet underexplored, topic in neurobiology. We addressed this using grape snails trained to reject two food-conditioned stimuli (CS). Our results indicate that memory storage mechanisms for distinct CS are independent, as selective impairment of memory reconsolidation with an NMDA receptor antagonist affected only one CS. When both CS reconsolidations were simultaneously disrupted, the snails developed amnesia, which manifested as anterograde amnesia on late amnesia stag
