Hemin suppresses interferon responses in periodontal cell models through JAK2-STAT1 inhibition.

INTRODUCTION: Hemin, an oxidized heme species released during erythrocyte breakdown, is detectable in gingival crevicular fluid during periodontal inflammation, but its effects on local immune signaling remain unclear. We investigated whether hemin modifies cytokine-induced interferon responses in HSC-2 oral squamous cell carcinoma cells and primary human gingival fibroblasts. METHODS: Cells were stimulated with interleukin-1β and tumor necrosis factor-α, or with interferon-γ, in the presence or absence of hemin. Transcriptomic changes were assessed by RNA sequencing and pathway analysis, and

Photo-responsive Ce-hemin nanozyme-based portable colorimetric sensor for tetracyclines assay.

A cerium ion-modified hemin polymer (Ce-hemin) was synthesized via a facile route, exhibiting photo-responsive oxidase-like activity and excellent stability. The Ce doping effectively prevented hemin aggregation, facilitated electron transfer, enhanced photocatalytic activity, and created a high adsorption capacity for tetracyclines (TCs). Taking advantage of the photo-sensitivity of TCs, a "turn-on" colorimetric sensing platform based on Ce-hemin nanozymes and smartphone-readout was constructed for visual analysis of TCs. This sensor offers a wide detection range and a low limit of detection

Smart Nanocomposites of Cu-Hemin Metal-Organic Frameworks for Electrochemical Glucose Biosensing.

Herein, a smart porous material, Cu-hemin metal-organic-frameworks (Cu-hemin MOFs), was synthesized via assembling of Cu2+ with hemin to load glucose oxidase (GOD) for electrochemical glucose biosensing for the first time. The formation of the Cu-hemin MOFs was verified by scanning electron microscopy, X-ray powder diffraction, Fourier transform infrared spectroscopy, N2 adsorption/desorption isotherms, UV-vis absorption spectroscopy, fluorescence spectroscopy, thermal analysis and electrochemical techniques. The results indicated that the Cu-hemin MOFs showed a ball-flower-like hollow cage st

Caseoperoxidase, mixed β-casein-SDS-hemin-imidazole complex: a nano artificial enzyme.

A novel peroxidase-like artificial enzyme, named "caseoperoxidase", was biomimetically designed using a nano artificial amino acid apo-protein hydrophobic pocket. This four-component nano artificial enzyme containing heme-imidazole-β-casein-SDS exhibited high activity growth and k(cat) performance toward the native horseradish peroxidase demonstrated by the steady state kinetics using UV-vis spectrophotometry. The hydrophobicity and secondary structure of the caseoperoxidase were studied by ANS fluorescence and circular dichroism spectroscopy. Camel β-casein (Cβ-casein) was selected as an appr

Hemin inhibits the interferon-beta-induced antiviral state in established cell lines.

Hemin and other metalloporphyrins are known as very versatile compounds in nature, because they are able to carry out numerous functions in a free state or in association with specific proteins. When Friend murine erythroleukemia cells are treated with IFN-beta plus 100 microM hemin, the antiviral state is not observed, whereas the antiviral effect of IFN-gamma is unaffected by hemin treatment. This inhibitory effect of hemin is not restricted to erythroid cells. In fact, it is also observed in murine L929 and in human cell lines treated with IFN-beta. Neither trivalent iron in other forms nor

The interaction of ascorbic acid with hemoglobin: Relevance to hemin release and lipid peroxidation.

The interactions between L-ascorbic acid (i.e., Vitamin C, VC) and bovine hemoglobin (Hb) were comprehensively analyzed using fluorescence, circular dichroism, UV-vis absorption spectra and molecular docking method, to elucidate the structural mechanisms through which VC altered Hb redox states and stabilities. VC interacted with the central cavities of Hb to form Hb-VC complex via static quenching mechanism involving nonradiative energy transfer, with van der Waals forces and hydrogen bonds playing predominant roles in the binding processes. VC was surrounded by residues such as α1-Thr137, α1

On the photophysical properties of Al-, Ga- and In-hemin complexes: a quantum chemical investigation.

Photophysical properties of the recently synthesized Al(III), Ga(III) and In(III)-hemin complexes have been studied, in particular, the most important properties relevant to their proposed use as new photosensitizers for photodynamic therapy (PDT). Minimum energy structures, excitation energies, singlet-triplet energy gaps (ΔES1-T), spin-orbit coupling constants (SOCs), and intersystem kinetic constants were computed at a Gaussian variant of B3LYP coupled with x2c-SVPall and D4 Grimme's dispersion in SMD simulated solvent. To verify whether intersystem crossing here can efficiently occur, the

A rabbit reticulocyte model for the role of hemin-controlled repressor in hypochromic anemias.

Hemin allows maximal protein synthesis in intact rabbit reticulocytes and their cell-free lysate preparations by retarding the formation of a translational repressor (HCR) found in the postribosomal supernate. In order to evaluate the role of HCR in the pathogenesis of hypochromic anemias, HCR was isolated and partially purified from intact rabbit reticulocytes incubated in vitro with either 0.1 mM alpha,alpha-dipyridyl (an iron-chelating agent) or 0.1 M ethanol. Both of these agents inhibit reticulocyte protein synthesis. Hemin (50 muM) protects against the inhibition by both agents. A ferrou

Hemin Recapitulates the Labile Iron Pool in the Cellular Fenton Reaction with DNA.

The iron-Fenton reaction in biology is influenced by diverse cellular molecules that coordinate with redox-active ferrous ion. Contemporary research proposed that hydroxyl radical or a ferryl species (HO•/Fe=O2+) are the primary oxidants; however, this was challenged by the observation that physiological bicarbonate redirects the reaction to form carbonate radical anion (CO3•-). Questions remained regarding the roles of O2 concentration, ascorbate, and iron speciation in CO3•- formation. Accordingly, in cellulo studies were conducted under physiological O2 (∼25 μM) with ascorbate replenishment

Function, regulation, and transcriptional organization of the hemin utilization locus of Bartonella quintana.

Bartonella quintana is a gram-negative agent of trench fever, chronic bacteremia, endocarditis, and bacillary angiomatosis in humans. B. quintana has the highest known hemin requirement among bacteria, but the mechanisms of hemin acquisition are poorly defined. Genomic analyses revealed a potential locus dedicated to hemin utilization (hut) encoding a putative hemin receptor, HutA; a TonB-like energy transducer; an ABC transport system comprised of three proteins, HutB, HutC, and HmuV; and a hemin degradation/storage enzyme, HemS. Complementation analyses with Escherichia coli hemA show that H

Characterization of a novel outer membrane hemin-binding protein of Porphyromonas gingivalis.

Porphyromonas gingivalis is a gram-negative, anaerobic coccobacillus that has been implicated as a major etiological agent in the development of chronic periodontitis. In this paper, we report the characterization of a protein, IhtB (iron heme transport; formerly designated Pga30), that is an outer membrane hemin-binding protein potentially involved in iron assimilation by P. gingivalis. IhtB was localized to the cell surface of P. gingivalis by Western blot analysis of a Sarkosyl-insoluble outer membrane preparation and by immunocytochemical staining of whole cells using IhtB peptide-specific

Anticancer Effect of Hemin through ANO1 Inhibition in Human Prostate Cancer Cells.

Anoctamin1 (ANO1), a calcium-activated chloride channel, is overexpressed in a variety of cancer cells, including prostate cancer, and is involved in cancer cell proliferation, migration, and invasion. Inhibition of ANO1 in these cancer cells exhibits anticancer effects. In this study, we conducted a screening to identify novel ANO1 inhibitors with anticancer effects using PC-3 human prostate carcinoma cells. Screening of 2978 approved and investigational drugs revealed that hemin is a novel ANO1 inhibitor with an IC50 value of 0.45 μM. Notably, hemin had no significant effect on intracellular

Spectral demonstration of semihemoglobin formation during CN-hemin incorporation into human apohemoglobins.

The incorporation of CN-hemin into three human adult apohemoglobin species (apohemoglobin, alpha-apohemoglobin, and apohemoglobin modified at its beta93 sulfhydryl with p-hydroxymercuribenzoate) has been monitored at micromolar concentrations in 0.05 M potassium phosphate buffer, pH 7.0, at 10 degrees C. In all cases, Soret spectral blue shifts accompanied CN-protohemoglobin but not CN-deuterohemoglobin formation. This finding in conjunction with isofocusing studies provided evidence of a CN-protosemi-alpha-hemoglobin intermediate, the formation of which appeared to be a direct consequence of

Amphiphilic hemin-based nanoparticles with hypoxia tolerance and inflammation inhibition for osteoarthritis therapy.

Osteoarthritis (OA) remains a major clinical challenge, driven by persistent cytokine-based inflammation and poor regenerative capacity of hypoxic microenvironment and avascular cartilage. Despite recent advances, strategies that concurrently address inflammation and tissue repair remain elusive. Here, we report a dual-functional, injectable nanoparticle system based on amphiphilic hemin, a naturally derived hemoglobin prosthetic group, for the synergistic treatment of OA. Harnessing its amphiphilic properties, hemin self-assembles into nanocarriers capable of encapsulating hydrophobic anti-in

Hemin-Coated Nanoparticles as Drug Delivery Vehicles for Chemotherapeutic Agents in Cancer Therapy.

Chemotherapy holds a prominent position in contemporary cancer treatment modalities; however, poor water solubility, rapid clearance, and unintended side effects remain significant challenges. To mitigate these issues, various nanoparticle-based delivery systems have been devised to improve the pharmacokinetic and pharmacodynamic profiles of chemotherapeutic drugs. While biocompatible, most of these drug delivery systems include materials that are foreign to the human body. Consequently, it is critical for these materials to be effectively removed from the body to prevent potential toxicity. I

Hemin ameliorates influenza pneumonia by attenuating lung injury and regulating the immune response.

The anti-influenza activity of hemin, an inducer, activator and the substrate of heme oxygenase-1 (HO-1), was examined both in vitro and in vivo. The human lung carcinoma cell line A549 was used to evaluate the in vitro effect of hemin on influenza A virus (IAV) replication. A mouse model was used to examine the in vivo activity of hemin. Observation indexes included survival rate and body weight of mice, virus load and pathological examination of the lungs, and characterization of the systemic and local immune responses. The results showed that hemin could induce HO-1 expression in A549 cells

Sensitive voltammetric and amperometric responses of respiratory toxins at hemin-adsorbed carbon-felt.

A hemin [iron-Fe(III) protoporphyrin IX chloride] was adsorbed onto a carbon-felt (CF), which is a microelectrode ensemble of micro carbon fiber (ca. 7 microm diameter). The resulting hemin-adsorbed-CF (hemin-CF) showed a well-defined redox wave based on the hemin-Fe(III)/Fe(II) redox process with the formal potential of -0.225 V vs. Ag/AgCl in deoxygenated phosphate/citrate buffer solution (0.1 mol/L, pH 5.0). The apparent heterogenous electron transfer rate constant was estimated to be 8.6 sec(-1). In air-saturated electrolyte solution, the hemin-CF exhibited an excellent electrocatalytic ac

Burkholderia pseudomallei known siderophores and hemin uptake are dispensable for lethal murine melioidosis.

Burkholderia pseudomallei is a mostly saprophytic bacterium, but can infect humans where it causes the difficult-to-manage disease melioidosis. Even with proper diagnosis and prompt therapeutic interventions mortality rates still range from >20% in Northern Australia to over 40% in Thailand. Surprisingly little is yet known about how B. pseudomallei infects, invades and survives within its hosts, and virtually nothing is known about the contribution of critical nutrients such as iron to the bacterium's pathogenesis. It was previously assumed that B. pseudomallei used iron-acquisition systems c

Isolation of an outer membrane hemin-binding protein of Haemophilus influenzae type b.

Haemophilus influenzae is a heme-dependent bacterium. However, little is known of the heme-iron uptake mechanism in this organism. By using a batch ligand affinity chromatography method, a hemin-binding protein of 39,500 molecular weight was isolated from total membranes derived from H. influenzae type b grown under iron-depleted but not under iron-sufficient conditions. Detection of the hemin-binding protein in a whole-cell binding assay demonstrated a surface-exposed location. Competition binding experiments indicated that this hemin-protein interaction was specific, since only hemin or heme
