scRNA-seq of Penaeus japonicus hemocytes under environmentally-induced restriction of sand-diving behavior.

Penaeus japonicus is an economically important marine shrimp species, but its sand-diving habit affects the further improvement of their culture yield. To investigate the molecular response to the restriction of sand-diving behavior, we designed three culture systems: the sandy group, the sandless group, and the sandless stress group (transfer the individuals to a sandless pond after a week in the sandy group). First, we analyzed the expression differences of stress-related (HSP60) and circadian rhythm genes (period, timeless) at different time points after stress induction, identifying 6 h po

WSSV-induced reversal of the malate-aspartate shuttle facilitates viral replication in shrimp hemocytes.

BACKGROUND: White spot syndrome virus (WSSV), one of the most devastating pathogens in global shrimp aquaculture, has been shown to hijack and reprogram host metabolic pathways to support its replication. Among the various host metabolic circuits, the malate-aspartate shuttle (MAS) is a key redox-balancing mechanism that facilitates the translocation of cytosolic NADH into mitochondria, thereby sustaining glycolysis and mitochondrial function. To date, however, the involvement of MAS in WSSV pathogenesis has not been documented. METHODS: In this study, we investigated the role of the MAS path

Localization of nitric oxide-producing hemocytes in Aedes and Culex mosquitoes infected with bacteria.

Mosquitoes are significant vectors of various pathogens. Unlike vertebrates, insects rely solely on innate immunity. Hemocytes play a crucial role in the cellular part of the innate immune system. The gaseous radical nitric oxide (NO) produced by hemocytes acts against pathogens and also functions as a versatile transmitter in both the immune and nervous systems, utilizing cyclic guanosine monophosphate (cGMP) as a second messenger. This study conducted a parallel comparison of NO synthase (NOS) expression and NO production in hemocytes during Escherichia coli K12 infection in four vector spec

Cell competition time line: winners kill losers, which are extruded and engulfed by hemocytes.

Cell competition is a mechanism that eliminates slow dividing cells from a growing population. It is believed that the genes wasp, psr, and draper are active in the cells that win the competition ("winner cells") and that they are essential in the winner cells for the induction of apoptosis and for the elimination of the "loser cells." Here, we show that lack of those genes in winner cells appears to be dispensable for cell-competition-induced apoptosis and during dmyc-induced supercompetition. Moreover, winner clones do not need those genes in order to preserve their growth advantage. Finally

Discovery of Plasmodium modulators by genome-wide analysis of circulating hemocytes in Anopheles gambiae.

Insect hemocytes mediate important cellular immune responses including phagocytosis and encapsulation and also secrete immune factors such as opsonins, melanization factors, and antimicrobial peptides. However, the molecular composition of these important immune cells has not been elucidated in depth, because of their scarcity in the circulating hemolymph, their adhesion to multiple tissues and the lack of primary culture methods to produce sufficient material for a genome-wide analysis. In this study, we report a genome-wide molecular characterization of circulating hemocytes collected from t

Taking damage as an advantage: Adaptation of Drosophila hemocyte functions upon DNA damage.

Immune cells in invertebrates and vertebrates are constantly exposed to environmental stressors, such as oxidative stress, UV radiation, toxins or infection, causing cellular damage of lipids, proteins but also DNA. Unresolved DNA damage in immune cells compromises genome integrity and stability, and can result in cellular dysfunction, apoptosis, or senescence and thereby contributing to immunodeficiencies, cancer or chronic inflammatory diseases. To counteract the effects and maintain genome integrity, cells employ a highly conserved DNA damage response (DDR) machinery. Beyond its canonical r

Impact of egg incubation on hemocyte recruitment and susceptibility of Galleria mellonella larvae to pathogens.

INTRODUCTION: Galleria mellonella is widely used as an in vivo model in microbiological and toxicological studies due to its reproducibility, low cost, and ease of handling. However, maintaining homogeneous laboratory colonies presents challenges. This study evaluated whether different incubation methods influence the larval immune response against Staphylococcus aureus, Candida albicans, and Escherichia coli. METHODS: Four experimental groups were established: (1) Traditional group (TG), eggs stored at 27°C; (2) Immediate group (IG), eggs stored at 16°C for 120 days before incubation at 27°C

An Engineered Solidified Peptide Hemocyte Sponge as Nanomotor Storage to Combat Bacterial Colitis.

The emergence of drug-resistant bacteria has significantly heightened the urgency for effective interventions within the global public healthcare system. Furthermore, the structural diversity of endotoxins across different bacterial species poses a major limitation on the clearance efficiency of structure-specific anti-endotoxin antibodies. In this study, a peptide-based material with membrane-disrupting functionality is developed. The topological configuration of the peptide is immobilized through Ni2+ coordination, and the thrust force generated by a redox reaction is harnessed to facilitate

Hemocyte alterations during melanotic encapsulation of Brugia malayi in the mosquito Armigeres subalbatus.

The involvement of hemocytes in melanotic encapsulation reactions against Brugia malayi was assessed in Armigeres subalbatus. Hemocyte populations, epitope changes, phenol oxidase (PO) activity, and the presence of an 84-kDa polypeptide were investigated in mosquitoes exposed to a B. malayi-infective bloodmeal (= immune-activated), in mosquitoes given a noninfective bloodmeal (= controls), in nonbloodfed mosquitoes (= naive), or in some combination of these. Total hemocyte populations in immune-activated mosquitoes significantly decreased at 24 hr postbloodmeal (PB) as compared with controls.

Hitchhikers in bivalve immune system: Mixed microplastics and nanoplastics triggers hemocyte autophagy.

Natural microplastic and nanoplastics (MNPs) mixtures generally consist of multiple sizes, but how their co-existence influences the immune system of aquatic organisms remains elusive. Here, we quantitatively tracked and demonstrated that such heterogeneity dynamically reshaped bivalve hemocyte burden in non-additive modes with subpopulation-specific consequences for immune function. Kinetic modeling revealed distinct internalization patterns and selectivity among hemocyte subpopulations, driven by particle proportions and uptake dynamics. Granulocytes displayed indiscriminate capacity for MNP

The Drosophila deubiquitinating enzyme dUSP36 acts in the hemocytes for tolerance to Listeria monocytogenes infections.

Listeria monocytogenes is a facultative intracellular pathogen which can infect Drosophila melanogaster. Upon infection, Drosophila mounts an immune response including antimicrobial peptide production and autophagy activation. A set of previously published results prompted us to study the role of the deubiquitinating enzyme dUSP36 in response to L. monocytogenes infections. We show in this report that flies with dUsp36-specific inactivation in hemocytes are susceptible to L. monocytogenes infections (as are flies with autophagy-deficient hemocytes) but are still able to control bacterial growt

Parallel Recovery Dynamics of Circulating and Tissue-Resident Hemocytes Following Hemolymph Withdrawal in Pomacea canaliculata.

In vertebrates, circulating and tissue-resident immune cells contribute to host defense and tissue homeostasis. Their balance is maintained through hematopoiesis and anatomical reservoirs. In mollusks, this balancing is poorly investigated. Thus, the dynamics of circulating and tissue-resident hemocytes after a hemolymph withdrawal were analyzed in Pomacea canaliculata, a snail that allows repeated hemolymph withdrawals and possesses posterior kidney (PK) cell aggregates known as hemocyte islets. In the absence of specific hemocyte markers, flow cytometry data revealed dynamic changes in circu

Pan-neuronal expression of human mutant huntingtin protein in Drosophila impairs immune response of hemocytes.

Huntington's disease (HD) is a late-onset; progressive, dominantly inherited neurological disorder marked by an abnormal expansion of polyglutamine (poly Q) repeats in Huntingtin (HTT) protein. The pathological effects of mutant Huntingtin (mHTT) are not restricted to the nervous system but systemic abnormalities including immune dysregulation have been evidenced in clinical and experimental settings of HD. Indeed, mHTT is ubiquitously expressed and could induce cellular toxicity by directly acting on immune cells. However, it is still unclear if selective expression of mHTT exon1 in neurons c

Imaging clathrin dynamics in Drosophila melanogaster hemocytes reveals a role for actin in vesicle fission.

Clathrin-mediated endocytosis (CME) is essential for maintaining many basic cellular processes. We monitored the dynamics of clathrin in live Drosophila melanogaster hemocytes overexpressing clathrin light chain fused to enhanced green fluorescent protein (EGFP) using evanescent wave microscopy. Membrane-associated clathrin-coated structures (CCS) constitutively appeared at the peripheral filopodial membrane, moved centripetally while growing in intensity, before being eventually endocytosed within a few tens of seconds. This directed CCS traffic was independent of microtubules but could be bl

Changes in hemocytes of Biomphalaria glabrata infected with Echinostoma paraensei and exposed to glyphosate-based herbicide.

The immune system of snails is highly sensitive to pollutants, which can suppress its immune response. We investigated the effects of exposure to the glyphosate-based herbicide Roundup® Original on the snail Biomphalaria glabrata infected by the platyhelminth Echinostoma paraensei by evaluating changes in the snail's internal defense system. Four cohorts were studied: control group, infected snails, snails treated with Roundup®, and snails infected and treated with Roundup®. The hemocyte viability was assessed, morphological differentiation of cells was observed and flow cytometry was performe

Activation of Imd pathway in hemocyte confers infection resistance through humoral response in Drosophila.

Upon microbial invasion the innate immune system of Drosophila melanogaster mounts a response that comes in two distinct but complimentary forms, humoral and cellular. A screen to find genes capable of conferring resistance to the Gram-positive Staphylococcus aureus upon ectopic expression in immune response tissues uncovered imd gene. This resistance was not dependent on cellular defenses but rather likely a result of upregulation of the humoral response through increased expression of antimicrobial peptides, including a Toll pathway reporter gene drosomycin. Taken together it appears that Im

Patterns of Hemocyte Production and Release Throughout the Molt Cycle in the Penaeid Shrimp Sicyonia ingentis.

The production and release of hemocytes was evaluated throughout the molt cycle in the shrimp Sicyonia ingentis. Hematopoiesis occurs in paired epigastric hematopoietic nodules (HPN) which consist of an extensive network of vessels. Hemocytes are produced within the walls of these tubules and released into the vessel lumens. During molt stage C (intermolt), few cells were present in the tubule wall; most of these were hematopoietic stem cells. Elevated mitotic rates during stages C to D1-2 (2-4%) led to the production and rapid release of individual hemocytes, primarily granulocytes. Although

The host-parasite interface in molluscan schistosomiasis: biotin as a probe for sporocyst and hemocyte surface peptides.

Hemocytes of Biomphalaria glabrata mediate the internal defensive response which, in resistant snail strains, kills sporocysts of Schistosoma mansoni. Lacking a gut, the sporocyst has only its tegument to interact with the host milieu (hemolymph). We have, therefore, focused our study on the surface-exposed proteins of hemocytes and sporocyst tegument. Using gentle biotinylation of living systems, labelled proteins were studied after SDS-PAGE electrophoresis and transfer to nitrocellulose. Results validate the utility of surface biotinylation in studies of host and parasite interfaces. A low d

Transcriptomics reveals different response mechanisms of Litopenaeus vannamei hemocytes to injection of Vibrio parahaemolyticus and WSSV.

As the most important cultural crustacean species worldwide, studies about Pacific white shrimp (Litopenaeus vannamei) have received more attention. It has been well-documented that various pathogens could infect L. vannamei, resulting in huge economic losses. The studies about the responding mechanism of L. vannamei to sole pathogens such as Vibrio parahaemolyticus and white spot virus (WSSV) have been extensively reported, while the studies about the differently responding mechanisms remain unclear. In the present study, we identified the differently expressed genes (DEGs) of L. vannamei hem
