Structures of Langya Virus Fusion Protein Ectodomain in Pre- and Postfusion Conformation.

Langya virus (LayV) is a paramyxovirus in the Henipavirus genus, closely related to the deadly Nipah (NiV) and Hendra (HeV) viruses, that was identified in August 2022 through disease surveillance following animal exposure in eastern China. Paramyxoviruses present two glycoproteins on their surface, known as attachment and fusion proteins, that mediate entry into cells and constitute the primary antigenic targets for immune response. Here, we determine cryo-electron microscopy (cryo-EM) structures of the uncleaved LayV fusion protein (F) ectodomain in pre- and postfusion conformations. The Lay

Structural and functional analyses reveal promiscuous and species specific use of ephrin receptors by Cedar virus.

Cedar virus (CedV) is a bat-borne henipavirus related to Nipah virus (NiV) and Hendra virus (HeV), zoonotic agents of fatal human disease. CedV receptor-binding protein (G) shares only ∼30% sequence identity with those of NiV and HeV, although they can all use ephrin-B2 as an entry receptor. We demonstrate that CedV also enters cells through additional B- and A-class ephrins (ephrin-B1, ephrin-A2, and ephrin-A5) and report the crystal structure of the CedV G ectodomain alone and in complex with ephrin-B1 or ephrin-B2. The CedV G receptor-binding site is structurally distinct from other henipav

A human lung xenograft mouse model of Nipah virus infection.

Nipah virus (NiV) is a member of the genus Henipavirus (family Paramyxoviridae) that causes severe and often lethal respiratory illness and encephalitis in humans with high mortality rates (up to 92%). NiV can cause Acute Lung Injury (ALI) in humans, and human-to-human transmission has been observed in recent outbreaks of NiV. While the exact route of transmission to humans is not known, we have previously shown that NiV can efficiently infect human respiratory epithelial cells. The molecular mechanisms of NiV-associated ALI in the human respiratory tract are unknown. Thus, there is an urgent

Current knowledge on the host-pathogen interactions of henipaviruses and novel platforms to enable further characterisation.

Henipaviruses, particularly the species Nipah (NiV) and Hendra (HeV), are emerging viral threats with potential to cause a public health emergency of international concern due to their high virulence and absence of approved preventative and therapeutical countermeasures. Consequently, research of NiV and HeV is restricted to high-containment laboratories and relies heavily on in vitro models. Despite NiV and HeV initial characterisation >25 years ago, significant gaps remain in the knowledge of the host-pathogen interactions, which are an important research focus for design of therapeutics and

Jamaican fruit bats are susceptible to henipaviruses but rapidly control infection.

Henipaviruses (HNV), Hendra (HeV) and Nipah (NiV) virus, cause severe pulmonary and neurologic disease in humans and other mammals. Bats in the genus Pteropus naturally host HeV and NiV, but their feasibility in experimental studies is limited due to their large size, low fertility rate, and unavailability outside of their native range. Understanding bat-henipavirus interactions that regulate shedding and replication could improve mitigation of spillover events and illuminate the factors that differentiate severe and controlled HNV infection. Here, we assessed the suitability of the Jamaican f

Henipaviruses at the threshold: preparedness in the era of recurrent spillover.

Henipaviruses remain high-consequence zoonotic pathogens characterized by recurrent spillover, severe disease, and limited but consequential human-to-human transmission. In this commentary, we frame henipaviruses as "threshold pathogens": agents repeatedly approaching, but not yet consistently crossing, the boundary into sustained community transmission. Expanding viral diversity, widening ecological interfaces, and episodic outbreaks challenge conventional preparedness models, particularly for surveillance, countermeasure evaluation, and regulatory decision-making. We argue that future prepar

Unveiling the RNA viral diversity in three organs of the Asian house shrew (Suncus murinus) from Tropical Hainan, China: a previously underappreciated key zoonotic reservoir.

Shrews represent an important reservoir of diverse human-pathogen viruses with implications for human infectious diseases. As the most populous shrew species, the Asian house shrew-Suncus murinus (Su. murinus) is widely distributed across South and Southeast Asia-particularly tropical and subtropical regions-yet its virome remains poorly studied. In this study, we collected 249 Su. murinus from 18 cities/counties (excluding Sansha) across Hainan Island and conducted RNA sequencing on gut, spleen, and lung tissues. We identified 192 RNA viruses, comprising 120 known viral species and 72 novel v

Discovery and genome characterization of six new orthoparamyxoviruses in small Belgian mammals.

In the future, zoonotic spillover events are expected to occur more frequently. Consequences of such events have clearly been demonstrated by recent outbreaks of monkeypox, Ebola virus, and the well-known severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Virus discovery has proven to be an important tool in the preparation against viral outbreaks, generating data concerning the diversity, quantity, and ecology of the vertebrate virome. Orthoparamyxoviruses, a subfamily within the Paramyxoviridae, are important biosurveillance targets, since they include several known animal, human,

Infectome analysis of bat kidneys from Yunnan province, China, reveals novel henipaviruses related to Hendra and Nipah viruses and prevalent bacterial and eukaryotic microbes.

Bats are natural reservoirs for a wide range of microorganisms, including many notable zoonotic pathogens. However, the composition of the infectome (i.e., the collection of viral, bacterial and eukaryotic microorganisms) within bat kidneys remains poorly understood. To address this gap, we performed meta-transcriptomic sequencing on kidney tissues from 142 bats, spanning ten species sampled at five locations in Yunnan province, China. This analysis identified 22 viral species, including 20 novel viruses, two of which represented newly discovered henipaviruses closely related to the highly pat

Emerging and re-emerging viral infections of the central nervous system in Australasia and beyond.

Viral infections of the central nervous system (CNS) have been emerging and re-emerging worldwide, and the Australasia region has not been spared. Enterovirus A71 and enterovirus D68, both human enteroviruses, are likely to replace the soon-to-be eradicated poliovirus to cause global outbreaks associated with neurological disease. Although prevalent elsewhere, the newly emergent orthoflavivirus, Japanese encephalitis virus (genotype IV), caused human infections in Australia in 2021, and almost certainly will continue to do so because of spillovers from the natural animal host-vector life cycle

A systematic review of case reports on mortality, modes of infection, diagnostic tests, and treatments for Nipah virus infection.

BACKGROUND: First identified in Malaysia in 1998, the Nipah virus is a paramyxovirus related to the Hendra virus. The clinical manifestation can vary from a silent infection to a life-threatening encephalitis. The World Health Organization (WHO) has documented 25 outbreaks in South Asia, resulting in 429 cases and 307 deaths to date. Currently, there are no approved treatments for the deadly Nipah virus infection, which is a serious threat to public health worldwide. Consequently, a review was conducted to examine the geographic distribution of the Nipah virus, mortality, transmission pathways

Emerging Vector-Borne Nipah Virus Infection: Unexplored Hazards, Diagnostic Challenges, and the Potential of Phytomedicine-Based Therapeutics.

INTRODUCTION: Millions of people have died from zoonotic illnesses, like COVID-19 and Nipah virus infection (NiV), throughout history. Fruit bats (Pteropus sp.) are the main reservoir host for NiV, an RNA virus belonging to the Henipavirus group, which causes extremely infectious illnesses, such as COVID- 19. NiV outbreaks have posed significant public health concerns, especially in South and Southeast Asia. The Nipah virus (NiV) infection is caused by a virus that belongs to the Paramyxoviridae family's Henipavirus genus. It is the source of zoonosis, which causes respiratory and neurological

Vertical transmission of Nipah virus in an experimentally infected nonhuman primate; a brief report and description of histopathological lesions.

Nipah virus (NiV) is a highly pathogenic henipavirus which causes severe, often fatal disease in humans and animals. African green monkeys (AGM) have been used extensively to investigate NiV disease and are the only nonhuman primate (NHP) model that accurately recapitulates human NiV pathogenesis. Here, we describe histopathological lesions in the reproductive tissues of an incidentally pregnant female AGM that succumbed to NiV disease. We employed multiplex immunohistochemistry techniques to profile specific cell populations which identified NiV antigen in both maternal and fetal cells of the

Vertical transmission and fetal replication of Nipah virus in an experimentally infected cat.

A female adult cat developed clinical disease 13 days after subcutaneous inoculation with Nipah virus (NiV) and was discovered to be pregnant at necropsy. Viral genome was detected in a variety of specimens, including blood, serum, tonsil swabs, and urine, up to 3 days before the onset of disease. Samples collected postmortem, including placenta, uterine fluid, and fetal tissues, were also positive for NiV genome, and the placenta and uterine fluid contained high levels of recoverable virus. The high levels of viral shedding in the adult combined with fetal viral replication suggests that both

Analysis of Recombinant Cedar Virus Infection and Cross-Protection Against Related Henipaviruses in African Green Monkeys.

Cedar virus (CedV), related to the highly pathogenic bat-borne henipaviruses, Hendra virus (HeV) and Nipah virus (NiV), is non-pathogenic in small animal models, likely due to the inability to produce interferon-antagonist proteins. We evaluated the pathogenesis of recombinant CedV (rCedV) in the African green monkey (AGM) model and determined if prior infection conferred cross-protective immunity against a lethal challenge with NiV Bangladesh (NiV-B) or HeV. AGMs infected with rCedV remained asymptomatic, with no clinical signs of disease or detectable viremia. The rCedV infected animals deve

Protection against henipaviruses in swine requires both, cell-mediated and humoral immune response.

Hendra virus (HeV) and Nipah virus (NiV) are members of the genus Henipavirus, within the family Paramyxoviridae. Nipah virus has caused outbreaks of human disease in Bangladesh, Malaysia, Singapore, India and Philippines, in addition to a large outbreak in swine in Malaysia in 1998/1999. Recently, NiV was suspected to be a causative agent of an outbreak in horses in 2014 in the Philippines, while HeV has caused multiple human and equine outbreaks in Australia since 1994. A swine vaccine able to prevent shedding of infectious virus is of veterinary and human health importance, and correlates o

A highly potent, stable, and safe dePEGylated lipopeptide against Nipah virus and related henipaviruses.

Nipah virus (NiV) poses a significant threat owing to its high mortality and the lack of approved therapeutics. Targeting the conserved heptad repeat 1 (HR1) domain of the viral fusion (F) protein constitutes a promising antiviral strategy. Also, lipopeptides derived from the human parainfluenza virus 3 (HPIV3) heptad repeat 2 (HR2) regions inhibit NiV fusion by blocking formation of the critical six-helix bundle (6-HB); however, their efficacy has been impeded by the controversial use of PEGylation. To resolve these limitations, we employed our proprietary heptad repeat 2 C-terminal fragment

Heterologous Sequential mRNA Vaccination of Indian Rhesus Macaques Elicits Broad Binding and Neutralizing Antibody Responses Against Diverse Henipaviruses.

Henipaviruses (HNVs), including Nipah virus (NiV) and Hendra virus (HeV), are highly pathogenic and often lethal zoonotic viruses with broad species tropism and no approved human vaccines. The emergence of genetically divergent HNVs-including Ghana virus (GhV), Langya virus (LayV), and Mojiang virus (MojV)-emphasizes the need for broadly protective countermeasures. Here, we evaluated the antibody (Ab) responses to sequential mRNA vaccines encoding the membrane-bound attachment glycoprotein (gG) from NiV, GhV, and/or LayV in a pilot study with Indian rhesus macaques. Serum binding Ab responses

Synthetic protocells interact with viral nanomachinery and inactivate pathogenic human virus.

We present a new antiviral strategy and research tool that could be applied to a wide range of enveloped viruses that infect human beings via membrane fusion. We test this strategy on two emerging zoonotic henipaviruses that cause fatal encephalitis in humans, Nipah (NiV) and Hendra (HeV) viruses. In the new approach, artificial cell-like particles (protocells) presenting membrane receptors in a biomimetic manner were developed and found to attract and inactivate henipavirus envelope glycoprotein pseudovirus particles, preventing infection. The protocells do not accumulate virus during the ina
