Male rat specific nephrotoxicity resulting from subchronic administration of hexachlorobenzene.

Male rats are more sensitive to the nephrocarcinogenic effect of hexachlorobenzene (HCB) than are female rats. The purpose of this study was to shed light on this phenomenon by investigating mechanisms of subchronic nephrotoxicity of HCB. Groups of rats were administered HCB in corn oil (po) at 100 mg/kg, 5 days per week for 15 days or at 50 mg/kg, 5 days per week for 50 days. Urine was collected on Days 1, 8, and 15 for the 15-day treatment and on Day 50 for the 50-day treatment. Glucosuria, proteinuria, and enzymuria (gamma-glutamyl transpeptidase) were measured to assess renal function. Twe

Investigation of obesogenic effects of hexachlorobenzene, DDT and DDE in male rats.

Obesity has become a very important public health problem and is increasing globally. Genetics, individual and environmental factors play roles in the etiology of this complex disorder. Recently, several environmental pollutants have been suggested to have obesogenic activities. Peroxisome proliferator activating receptor gamma (PPARγ), uncoupling protein-1 (UCP1) and their expression in white adipose tissue (WAT) and brown adipose tissue (BAT) play key roles in adipogenesis. UCP3 and irisin were reported to play roles in non-shivering thermogenesis. Our primary aim was to investigate obesogen

Hexachlorobenzene, a pollutant in hypothyroidism and reproductive aberrations: a perceptive transgenerational study.

Hexachlorobenzene (HCB), a widespread environmental pollutant, contributes to endocrine disruption resulting in hypothyroidism. We investigated the effect of chronic exposure of HCB to explore the functional interconnection between hypothyroidism and infertility. All observations were made through the F1 and F2 generations. Thyroidectomy was also performed to evaluate the contribution of the thyroid gland in affecting ovarian dysfunction and reproductive aberrations. We confirmed that the preconception exposure of HCB leads to hypothyroidism which was reflected by an increase in the body weigh

Hexachlorobenzene exposure and the proportion of male births in Turkey 1935-1990.

Hexachlorobenzene (C(6)Cl(6), HCB) is a chemical that has been associated with significant immediate and long term adverse health effects in humans. It has been associated with both porphyria cutanea tarda and spontaneous abortions among survivors of widespread exposure in the 1950s in southeastern Turkey. HCB binds to the Ah receptor, albeit with lower affinity than dioxin. Dioxin exposure has been reported to lower human secondary sex ratio, putatively through a male mediated effect. We therefore wished to evaluate the impact of the HCB environmental event on the sex ratio of the progeny of

The relation between the oxidative biotransformation of hexachlorobenzene and its porphyrinogenic activity.

The relation between the major toxic effect of hexachlorobenzene, hepatic porphyria, and its oxidative biotransformation was studied in vivo, by observing the effect of modulating its biotransformation on the expression of porphyria. This modulation was achieved by selective in vivo inhibition of the major cytochrome P450 isoenzyme involved in both the hydroxylation of hexachlorobenzene and its primary oxidative metabolite, pentachlorophenol. The involvement of this isoenzyme, cytochrome P450p, was established by in vitro biotransformation studies using microsomes derived from rats treated wit

Sex differences in the metabolism of hexachlorobenzene by rats and the development of porphyria in females.

Male and female F 344 rats were dosed every other day for 103 days with 50 mumole of hexachlorobenzene (HCB)/kg. Females developed a hepatic porphyria, the urine and liver levels of porphyrins being 40- and 310-fold higher respectively than those of males. Urine was periodically hydrolysed and analysed for the three metabolites pentachlorophenol, 2,3,5,6-tetrachlorobenzene-1,4-diol and pentachlorothiophenol (derived from the mercapturate). The combined urinary excretion of these was greater in females than males, especially during the first 10 weeks. Pentachlorothiophenol was particularly high

[Induction of mixed microsomal oxidase in experimental hexachlorobenzene porphyria in rabbits].

The chlororganic pesticide--hexachlorbenzol (C6Cl6), given to rabbits, induces the clinical picture and biochemical changes of porphyria cutanea tarda. The experiments are carried out on 40 rabbits of Chinchilla strain. Microsomal protein and the enzymes cytochrom P-450, aniline-hydroxylase, aminopyrine-dimetilase are determined in liver microsomes, obtained by differential centrifugation. The mitochondrial enzyme Deltaaminolevuline acid-syntatese (DALA-syntatase) is determined in liver homogenate as well as the enzyme gamma-glutamyl-transpeptidase (GGTP) in serum and uro- and coproporphyrines

Hexachlorobenzene toxicity in the monkey primordial germ cell without induced porphyria.

Hexachlorobenzene is a persistent chlorinated organic chemical that has been detected in many tissues from a variety of species including human ovary and human ovarian follicular fluid. When administered in high dosage to nonhuman primates, hexachlorobenzene causes destruction of ovarian primordial germ cells in association with systemic toxicity. The purpose of these experiments was to assess relative ovarian germ cell sensitivity at much lower dosages of hexachlorobenzene that do not produce systemic effects and additionally to evaluate oocyte function by means of the response to superovulat

Hexachlorobenzene levels in multimedia environmental samples from selected chemical production plants.

A sampling and analysis program was conducted to determine hexachlorobenzene (HCB) levels in air, water, soil and sediment. Nine industrial plants were selected, representing six major industries: perchloroethylene, trichloroethylene, carbon tetrachloride, chlorine, triazine herbicides and pentachloronitrobenzene. Of the six industries sampled, higher concentrations of HCB were associated with the production of perchloroethylene, trichloroethylene and carbon tetrachloride. In the one plant that produced only carbon tetrachloride, HCB levels were quite low. HCB concentrations were relatively hi

Transmembrane potential of liver mitochondria from hexachlorobenzene-and iron-treated rats.

The respiratory parameters and the membrane potential of liver mitochondria from rats treated with either hexachlorobenzene, iron or hexachlorobenzene plus iron, to induce experimental porphyria, have been studied. Partial uncoupling of oxidative phosphorylation has been observed in mitochondria from hexachlorobenzene- and hexachlorobenzene plus iron-treated rats. Direct evidence has been presented that this uncoupling is due to the action of pentachlorophenol endogenously formed by metabolism of hexachlorobenzene. No irreversible damage of mitochondria membrane has been revealed under both th

Contrastive analysis of the Raman spectra of polychlorinated benzene: hexachlorobenzene and benzene.

Detection of persistent pollutants such as polychlorinated benzene in environment in trace amounts is challenging, but important. It is more difficult to distinguish homologues and isomers of organic pollutantd when present in trace amounts because of their similar physical and chemical properties. In this work we simulate the Raman spectra of hexachlorobenzene and benzene, and figure out the vibration mode of each main peak. The effect on the Raman spectrum of changing substituents from H to Cl is analyzed to reveal the relations between the Raman spectra of homologues and isomers of polychlo

Urinary porphyrin excretion in a human population highly exposed to hexachlorobenzene.

BACKGROUND: Data from an epidemic reported in Turkey (1955-1959) is the only information about the relationship between hexachlorobenzene (HCB) intake and porphyria cutanea tarda in humans. No information is available on the HCB threshold exposure level to induce porphyria cutanea tarda. OBJECTIVES: To study HCB serum levels and urinary porphyrin excretion in the inhabitants of a village located near an organochlorine compound factory with high HCB concentrations in the air and to detect possible alterations in urinary porphyrin excretion and examine their relationship with HCB serum levels.

A review of porphyria and cancer and the missing link with human exposure to hexachlorobenzene.

A literature review shows that porphyria cutanea tarda (PCT), and possibly acute intermittent porphyria also, tend to occur more often than expected among cases of primary liver cancer, especially when cirrhosis is present. Lymphomas and PCT may be related also, although this is less well documented. Furthermore, there is an interesting report on a case of PCT, sarcoma and probable exposure to 2,3,7,8-tetrachlorodibenzo-para-dioxin. Exposure to hexachlorobenzene (HCB) as a cause of cancer in humans does not seem to have been reported, however, but exposure to this compound and other chlorinate

Porphyrins and pentachlorophenol in rat-liver mitochondria in hexachlorobenzene-induced porphyria.

This study investigated the extent of impairment in functional parameters of liver mitochondria from rats treated for 60 days with hexachlorobenzene (HCB). A constant amount of mitochondrial uncoupling was found throughout the treatment period. At the same time a nearly constant amount of pentachlorophenol was detected in these mitochondria. In contrast, the level of mitochondrial porphyrins increased progressively. There was good correlation between the concentration of mitochondrial pentachlorophenol and the degree of uncoupling of oxidative phosphorylation.

Melatonin formation in pineal gland from rats with hexachlorobenzene experimental porphyria.

Hexachlorobenzene produces an experimental hepatic porphyria in rats, which is similar to human porphyria cutanea tarda, with hyperpigmentation as one of its characteristic features. Alterations in tryptophan metabolism have been previously observed in this chronic porphyria. Melatonin formation from tryptophan via serotonin shows diurnal rhythmicity in the pineal gland, and higher values are observed during the dark phase of an imposed light-dark cycle. The purpose of this study was to determine the contents of tryptophan and its metabolites in pineal gland of normal and hexachlorobenzene-tre

Combined effects of pentachlorophenol and its byproduct hexachlorobenzene on endocrine and reproduction in zebrafish.

Pentachlorophenol (PCP) and its byproduct hexachlorobenze (HCB) are two co-existing persistent environmental chemicals, but their combined toxicity remains unclear. In this study, adult zebrafish were exposed to 5 (low dose) and 25 μg·L-1 (high dose) of PCP, HCB or their combination for 21 days, and the impact on endocrine and reproduction was investigated. Results showed that combined exposure to 25 μg·L-1 PCP and 25 μg· L-1 HCB significantly increased the plasma estradiol (E2) and testosterone (T) levels, altered the expressions of genes along the hypothalamic-pituitary-gonadal-liver (HPGL)

Environmental exposure to hexachlorobenzene (HCB) and risk of female breast cancer in Connecticut.

Earlier studies have provided inconclusive results relating hexachlorobenzene (HCB), an organochlorine fungicide, to female breast cancer risk. The current study, with a total of 304 breast cancer cases and 186 controls recruited in Connecticut between 1994 and 1997, examined the association by directly comparing breast adipose tissue levels of HCB between incident breast cancer cases and noncancer controls. The cases and controls were patients who had breast biopsies or surgery at the Yale-New Haven Hospital (New Haven, CT) and histologically diagnosed either as breast cancer or benign breast
