Anoxia and capillary filtration coefficient in the isolated cat hindlimb.

Isolated cat hindlimbs were perfused from a reservoir with an albumin-blood mixture at a constant flow of 20 ml X min-1 X 100 g muscle-1 while alternately bubbling the perfusate with either 95% O2-5% CO2 gas mixture or a 95% N2-5% CO2 mixture for 50- to 60-min periods. Capillary filtration coefficient (CFC), vascular resistance (R), and perfusate O2 content were measured in each period. The arterial O2 content fell from fully equilibrated to 0.25 vol% during the use of N2. R fell from about 6 mmHg X min X 100 g X ml-1 during O2 bubbling to approximately 1.0 with N2. CFC averaged 0.012 +/- 0.00

Effects of enkephalins on perfusion pressure in isolated hindlimb preparations.

A series of studies were conducted to determine the effects of leucine-(leu-) enkephalin and methionine-(met-) enkephalin on perfusion pressure. These experiments utilized isolated perfused femoral arterial preparations in pentobarbital-anesthetized cats. The enkephalins were administered intraarterially into the femoral artery and changes in perfusion pressure recorded. Leu-enkephalin in doses of 1 microgram to 320 micrograms produced significant dose-dependent decreases in perfusion pressure (4.0 +/- 1.3% with 1 microgram to 19.1 +/- 2.1% with 320 microgram). Similar declines in perfusion pr

The functional anatomy of the hindlimb of some African Viverridae (Carnivora).

The functional anatomy of the hindlimb of 12 species of viverrids was studied with relation to locomotion. The animals were allocated to primary locomotor categories on the basis of their anatomy and locomotion. The climbing, arboreal walking category (Nandinia binotata) is characterized by a small sacroiliac articulation, the iliopsoas inserts onto a medially located lesser trochanter and the femoral condyles are not posteriorly placed. The hindfoot is plantigrade and its structure permits considerable movement. The pads are soft and the claws retractile. Representatives of the arboreal and t

Discharges of single hindlimb afferents in the freely moving cat.

1. Implanted semimicroelectrodes were used to record single afferent fiber discharges from L7 dorsal roots during unrestrained walking in the conscious cat. 2. A series of tests were used to identify an afferent during a short period of anesthesia following each recording session. The majority of afferents were from muscle spindle primary endings in hindlimb muscles. 3. Ankle extensor spindle primaries generally showed their highest firing rates during that phase of stepping in which they were passively stretched. During active muscle contraction there was evidence of fusimotor drive, although

Pinoresinol diglucoside alleviates hindlimb unloading-induced bone loss in mice.

Microgravity-induced bone loss has long been a critical issue in space exploration. While countermeasures have been suggested, none have achieved the desired effects. Pinoresinol diglucoside (PDG), an effective constituent of the medicinal herb Eucommia ulmoidis Oliv, was reported to increase peak bone mass in growing rats and mitigate dexamethasone-induced osteoporosis. However, it is unknown if it affects microgravity-induced bone loss. Here, PDG effects were investigated in cultured rat calvarial osteoblasts exposed to simulated microgravity (SMG) and in hindlimb-unloaded mice. PDG (1 × 106

Trimetazidine ameliorates hindlimb ischaemic damage in type 2 diabetic mice.

BACKGROUND: Ischaemia caused by lower extremity artery stenosis is the main cause of peripheral artery disease (PAD) in patients with diabetes. Trimetazidine (TMZ) has traditionally been used as an anti-ischaemic drug for coronary artery disease. The effect of TMZ on PAD in a diabetic animal model and the underlying molecular mechanisms remain unclear. METHODS: The db/db mice were challenged with femoral artery ligation (FAL), followed by TMZ treatment for 2 weeks. Scores on hindlimb ischaemia and function were evaluated. Histological and capillary density analyses of gastrocnemius were perfo

Hindlimb torsional alignment changes in growing rabbits after patellar dislocation.

BACKGROUND: Torsional malalignment has been considered as a risk factor for patellar dislocation. But the influence of patellar dislocation for torsional alignment development remains unknown. The present study aims to investigate whether the torsional alteration of the hindlimb occurs after patellar dislocation in growing rabbits. METHODS: In the present study, 30 one-month-old rabbits were included. The experimental group consisted of 30 left knees of rabbits which underwent patellar lateral dislocation. The control group consisted of 30 right knees of the rabbits which no surgical procedur

Mitochondrial therapy improves limb perfusion and myopathy following hindlimb ischemia.

Critical limb ischemia is a devastating manifestation of peripheral arterial disease with no effective strategies for improving morbidity and mortality outcomes. We tested the hypothesis that cellular mitochondrial function is a key component of limb pathology and that improving mitochondrial function represents a novel paradigm for therapy. BALB/c mice were treated with a therapeutic mitochondrial-targeting peptide (MTP-131) and subjected to limb ischemia (HLI). Compared to vehicle control, MTP-131 rescued limb muscle capillary density and blood flow (64.7±11% of contralateral vs. 39.9±4%), a

Chromium supplement inhibits skeletal muscle atrophy in hindlimb-suspended mice.

Skeletal muscle atrophy and whole-body glucose intolerance are consequences of muscle disuse associated with conditions leading to prolonged bed rest. Nutritional supplementation with chromium has been shown to prevent weight loss and improve glucose tolerance in malnourished subjects on long-term total parenteral nutrition. The objective of this study was to evaluate the effect of oral supplementation with a novel chromium complex, chromium (d-phenylalanine)(3) [Cr(d-phe)(3)] at 45 microg/kg/day for 5 weeks, on skeletal muscle atrophy and glucose intolerance in a hindlimb suspension mouse mod

No contralateral neutrophil (PMN) stimulation after rabbit hindlimb ischemia/reperfusion.

Polymorphonuclear neutrophils (PMNs) have been implicated as mediators of the reperfusion injury that occurs in skeletal muscle after ischemia. Our laboratory has previously shown that the venous effluent from rabbit hindlimbs that have undergone 2 or 3 hours of ischemia followed by 1 hour of reperfusion contains PMNs that demonstrate increased phagocytosis and chemotaxis when assayed. It was the purpose of this study to evaluate PMNs from the contralateral rabbit hindlimb utilizing the same periods of ischemia and reperfusion to determine whether this PMN-induced effect is localized to the is

The impact of hindlimb disuse on sepsis-induced myopathy in mice.

Sepsis induces a myopathy characterized by loss of muscle mass and weakness. Septic patients undergo prolonged periods of limb muscle disuse due to bed rest. The contribution of limb muscle disuse to the myopathy phenotype remains poorly described. To characterize sepsis-induced myopathy with hindlimb disuse, we combined the classic sepsis model via cecal ligation and puncture (CLP) with the disuse model of hindlimb suspension (HLS) in mice. Male C57bl/6j mice underwent CLP or SHAM surgeries. Four days after surgeries, mice underwent HLS or normal ambulation (NA) for 7 days. Soleus (SOL) and e

Assessing Therapeutic Angiogenesis in a Murine Model of Hindlimb Ischemia.

Critical limb ischemia (CLI) is a serious condition that entails a high risk of lower limb amputation. Despite revascularization being the gold-standard therapy, a considerable number of CLI patients are not suited for either surgical or endovascular revascularization. Angiogenic therapies are emerging as an option for these patients but are currently still under investigation. Before application in humans, those therapies must be tested in animal models and its mechanisms must be clearly understood. An animal model of hindlimb ischemia (HLI) has been developed by the ligation and excision of

A Revised Method for Inducing Secondary Lymphedema in the Hindlimb of Mice.

Animal models are of paramount importance in the research of lymphedema in order to understand the pathophysiology of the disease but also to explore potential treatment options. This mouse model allows researchers to induce significant lymphedema lasting at least 8 weeks. Lymphedema is induced using a combination of fractioned radiotherapy and surgical ablation of lymphatics. This model requires that the mice get a dose of 10 Gray (Gy) radiation before and after surgery. The surgical part of the model involves ligation of three lymph vessels and extraction of two lymph nodes from the mouse hi

Suspected eccrine adenocarcinoma on footpad of the right hindlimb in a dog.

A 12-year-old, male miniature dachshund has an ulcer on the footpad of the right hind limb. Despite treatment for longer than 6 months, the ulcer did not heal. Biopsy of the lesion was done to make a definitive diagnosis. Histologically, there were lumens containing weakly eosinophilic fluid surrounded by tumor cells with a similar circular pale nucleus and distinct nucleoli that showed some variation in size. Immunohistochemically, the tumor cells were positive for cytokeratin (AE1/AE3) and vimentin, were negative for S100 and p63. A poorly differentiated eccrine adenocarcinoma was diagnosed.

Bilateral hindlimb superficial digital flexor tendinopathy in a sports horse.

Superficial digital flexor tendinopathy is a common cause of forelimb lameness in Thoroughbred racehorses and sports horses; however, this condition is rarely observed in the hindlimb. A 16-year-old Irish sports horse gelding was presented with bilateral swelling in the plantar metatarsal region and severe bilateral hindlimb lameness visible at the walk. The lameness occurred after the cross-country phase of an eventing competition. The clinical suspicion of superficial digital flexor tendon (SDFT) injury was confirmed after ultrasonographic examination, which revealed an extensive core lesion

Spinal interneuronal populations encode static hindlimb posture in the cat.

Proprioceptive signals from primary afferents often correlate with single-joint states (but can also reflect multi-joint configuration), whereas neuronal populations in the cerebral cortex represent limb endpoint postures. Where and how this transformation emerges along the somatosensory axis from peripheral proprioceptive receptors remains unclear. We simultaneously recorded lumbosacral spinal neurons in two decerebrate, immobilized cats while a robotic device held the hindlimb at 16 static endpoint positions spanning hip-knee configurations. Using high-density multielectrode recordings, we a

Nano-curcumin mitigates muscle impairment in hypoxic hindlimb-unloaded mice.

BACKGROUND: Skeletal muscle atrophy due to hindlimb unloading (HU) is worsened under hypoxic conditions, mimicking clinical scenarios such as prolonged bed rest and chronic cardiopulmonary disease. Curcumin has therapeutic potential but suffers from poor bioavailability. This study evaluated the efficacy of curcumin encapsulated in zein-casein nanoparticles (Cur Z-CS NP) in mitigating muscle deterioration in HU mice under hypoxia. METHODS: Cur Z-CS NP were synthesized and characterized for physicochemical properties and drug release. Male C57BL/6 J mice were divided into normoxic and hypoxic

Mechanisms of Rat Gastrocnemius Muscular Atrophy Induced by Hindlimb Unloading.

INTRODUCTION: Muscle atrophy is a critical health challenge during spaceflight. This study investigated the mechanism of muscle atrophy induced by 21-d hindlimb unloading (HLU) and metabolomic changes in rat gastrocnemius muscle (GM) and plasma. METHODS: The rat tail-suspended model was used to simulate microgravity. The effects of HLU on GM atrophy were assessed by measuring morphological changes, levels of oxidative stress factors and proinflammatory cytokines, and the expression of key proteins in the insulin-like growth factor 1 receptor/phosphatidylinositol 3-kinase/Ak strain transformin
