Passive air-pouch anaphylaxis in rats. II. Pharmacological characterization.

The effects of some antiallergic agents on passive air-pouch anaphylaxis (PAPA) in the dorsal skin of rats were investigated by measuring plasma exudation and histamine content in the pouch fluid. Antiallergic agents, disodium cromoglycate (DSCG), tranilast and ketotifen, dose dependently inhibited both plasma exudation and histamine release, except that ketotifen showed a dose-unrelated inhibition of histamine release. An antihistamine, pyrilamine, suppressed plasma exudation without affecting histamine release. A cyclooxygenase inhibitor, indomethacin, and a 5-lipoxygenase inhibitor, nordihy

Effect of histamine on ganglionic transmission.

Histamine has been shown to depress transmission through the perfused superior cervical ganglion of the cat when doses of 150 mug. or more were administered. The intensity of the ganglionic block was related to the dose of histamine employed. In one-third of the experiments, a slow contracture of the nictitating membrane occurred after histamine had been injected; the contracture lasted up to 10 min., and subsequent injections of histamine gave rise to progressively smaller responses. The blocking action of histamine was evident in all experiments and was the most prominent feature observed. H

Role of histamine in the aetiology of byssinosis. II. Lung histamine concentrations in guinea pigs chronically exposed to cotton and flax dusts.

Data presented in this study support the finding that cotton and flax dusts contain agents which potentiate the formation or accumulation of histamine or both in the lungs of guinea pigs exposed to dust, and that such agents are present at much higher levels in cotton dust than in flax dust. The potentiating effect may be through the recruitment of mast cells into the lung. Both cotton and flax dusts contain methylating enzyme inhibitory agents, whereas cotton dust also contains agents that inhibit histaminase activity; flax dust contains agents potentiating histamine activity. These agents wo

Cholinergic influence of K(+)-evoked release of endogenous histamine from rat hypothalamic slices in vitro.

The effects of cholinergic agents on the K(+)-evoked release of endogenous histamine from hypothalamic slices of rats were examined by a superfusion method in vitro. Acetylcholine and carbamylcholine significantly inhibited K(+)-evoked release of histamine from the slices, and the effect of acetylcholine was antagonized by the muscarinic antagonist atropine. On the other hand, nicotine significantly enhanced the K(+)-evoked release and its effect was antagonized by the nicotinic antagonist hexamethonium. The effects of carbamylcholine and nicotine on histamine release from slices of whole hypo

[Effect of antihistamine preparations and histamine on the development of allergic processes].

The effect of antihistaminic drugs, histamine and their combinations on the development of anaphylactic shock and the period of repeated resolution doses was studied in rabbit experiments. It was established that administration of dimedrol during sensibilization aggravated while diazolin and histamine attenuated the course of anaphylactic shock and periods of repeated resolution injections. Simultaneous administration of antihistaminic agents and histamine enhanced the allergization of the body. Under conditions reported dimedrol prevented hypertrophy of the adrenals that occurred in response

Regulation of intestinal goblet cell secretion. II. A survey of potential secretagogues.

The factors that regulate the rate of mucus secretion in intestinal goblet cells are only partially defined. Autoradiographic and ultrastructural studies demonstrated that muscarinic cholinergic agents accelerate the exocytosis of mucus from goblet cells in the crypts throughout the small and large intestine, both in vivo and in mucosal organ culture. The present study seeks to identify other factors that may alter mucous secretory rates. Mucosal explants were exposed to potential secretagogues and inhibitors in the organ-culture system and analyzed by light and electron microscopy, alpha- and

The interaction of histamine with gastrin and carbamylcholine on oxygen uptake by isolated mammalian parietal cells.

Using oxygen uptake as an index of the physiological response of isolated parietal cells, the interactions between histamine and gastrin and between histamine and carbamylcholine and the effects of atropine and metiamide on these interactions have been studied. Parietal cells were isolated from canine fundic mucosa by sequential exposure of separated mucosa to collagenase and EDTA. In previous studies carbamylcholine, isobutyl methyl xanthine, gastrin, and histamine have each been shown to increase oxygen uptake by these cells. Isobutyl methyl xanthine greatly enhanced the histamine effect. Ca

[Effect of histamine-blocking substances on mitogen-induced lympho proliferation in rheumatic diseases].

Peripheral blood lymphocytes of patients with rheumatoid arthritis, psoriatic arthritis and healthy controls, isolated by a ficoll density gradient, were incubated with different concentrations of histamine, cimetidine and diphenhydramine (DPH) and simultaneously stimulated with phytohemagglutinine (PHA). In comparison to healthy control persons the lymphoproliferation rate in arthritic patients could be increased about 20% by addition of the histamine-blocking agents cimetidine and DPH. This suggests the existence of in vivo histamine-induced suppressor cells in arthritic patients, which can

Histamine food poisoning: toxicology and clinical aspects.

Histamine poisoning can result from the ingestion of food containing unusually high levels of histamine. Fish are most commonly involved in incidents of histamine poisoning, although cheese has also been implicated on occasion. The historic involvement of tuna and mackerel in histamine poisoning led to the longtime usage of the term, scombroid fish poisoning, to describe this food-borne illness. Histamine poisoning is characterized by a short incubation period, a short duration, and symptoms resembling those associated with allergic reactions. The evidence supporting the role of histamine as t

Anti-scratching behavioral effects of N-stearoyl-phytosphingosine and 4-hydroxysphinganine in mice.

N-Stearoyl-phytosphingosine (SPS) and 4-hydroxysphinganine (phytosphingosine, PS), which are sphingolipids frequently found in mammalian skin, plants, and yeast, have been used as ingredients in cosmetics. In mice, treatment with SPS and PS inhibited histamine-induced scratching behavior and vascular permeability. These agents inhibited the expression of the allergic cytokines, IL-4 and TNF-alpha, and the activation of the transcription factors, NF-kappaB and c-jun, in histamine-stimulated skin tissues. These agents also showed potent anti-histamine effects in the Magnus test using guinea pig

Histamine mediates myocardial damage by an H1 mechanism independent of coronary blood flow.

Administration of histamine to rabbits may result in myocardial damage similar to that produced by catecholamines and the anthracycline antibiotics. To explore the mechanisms involved in histamine-mediated myocardial damage, conscious New Zealand white rabbits were pretreated with H1 and H2 receptor blocking agents, alone and in combination, and then administered histamine. Coronary artery blood flow was measured with radiolabeled microspheres in rabbits that received histamine alone, and in those that received an H1 blocking agent and histamine. Rabbits that received an H1 blocking agent had

The histamine releasers crotamine, protamine and compound 48/80 activate specific proteases and phospholipases A2.

Crotamine, a basic, myonecrotic, histamine-releasing neurotoxin, was isolated from Crotalus durissus terrificus venom. Carboxypeptidase A was shown to be activated by crotamine when acting upon N-carbobenzoxyglycil-L-phenylalanine. However the activity of carboxypeptidase B upon the substrate hippuryl-L-arginine was not enhanced by this toxin. Teh basic histamine releasers protamine and compound 48/80 also activated carboxypeptidase A. These three agents activated both alpha-chymotrypsin when acting upon acetyl-L-tyrosine ethyl ester and also five snake venom phospholipase-like myotoxins actin

Histamine provocation of clinical coronary artery spasm: implications concerning pathogenesis of variant angina pectoris.

Twelve patients with nonexertional chest pain and nonobstructive fixed coronary disease (less than 50% luminal diameter narrowing) were given histamine to investigate the potential role (coronary artery H1 receptor agonism) of the endogenous agent in producing coronary artery spasm (CAS). Histamine, at intravenous dose of 0.5 to 1.0 microgram/kg/min, provoked CAS in four patients. In six patients neither histamine nor ergonovine provoked spasm, and these patients were considered by chronic follow-up evaluation to have noncardiac etiology for their chest pain syndrome. In one patient CAS was pr

Stimulation of central histaminergic transmission attenuates diazepam-induced motor disturbance on rota-rod and beam walking tests in mice.

Diazepam administration has been shown to influence the release of histamine in various brain areas involved in motor behavior. Therefore, the present study explored the plausible regulatory role of the central histaminergic system in diazepam-induced deficits in motor performance in mice using the rota-rod and beam walking tests. In this study, several doses of diazepam (0.5, 1, 2, and 3 mg/kg, i.p.) were assessed in mice for changes in motor performance on the rota-rod and beam walking test. In addition, the brain histamine levels were determined after diazepam administration, and the diazep
