Creation of the first ultra-low gluten barley (Hordeum vulgare L.) for coeliac and gluten-intolerant populations.

Coeliac disease is a well-defined condition that is estimated to affect approximately 1% of the population worldwide. Noncoeliac gluten sensitivity is a condition that is less well defined, but is estimated to affect up to 10% of the population, and is often self-diagnosed. At present, the only remedy for both conditions is a lifelong gluten-free diet. A gluten-free diet is often expensive, high in fat and low in fibre, which in themselves can lead to adverse health outcomes. Thus, there is an opportunity to use novel plant breeding strategies to develop alternative gluten-free grains. In this

Enhancement of Photosynthetic Iron-Use Efficiency Is an Important Trait of Hordeum vulgare for Adaptation of Photosystems to Iron Deficiency.

Leaf iron (Fe) contents in Fe-deficiency-tolerant plants are not necessarily higher than that in Fe-deficiency-susceptible ones, suggesting an unknown mechanism involved in saving and allowing the efficient use of minimal Fe. To quantitatively evaluate the difference in Fe economy for photosynthesis, we compared the ratio of CO2 assimilation rate to Fe content in newly developed leaves as a novel index of photosynthetic iron-use efficiency (PIUE) among 23 different barley (Hordeum vulgare L.) varieties. Notably, varieties originating from areas with alkaline soil increased PIUE in response to

Impact of Salinity Duration on Growth, Photosynthesis, Water Relations, and Recovery Capacity Reveals Adaptive Responses in Hordeum maritimum.

Although the physiological mechanisms underlying salt tolerance in wild barley (Hordeum maritimum) are relatively well understood, its recovery after salt stress remains poorly characterized. In this pot experiment, plants were exposed to 200 mM NaCl for 0, 15, 30, 45, or 60 days to evaluate changes in growth and water status. Wild barley exhibited high tolerance during the early stages of salt exposure, maintaining normal growth and water balance for up to 30 days. However, longer exposure reduced plant growth and leaf water content, indicating that prolonged salinity eventually compromises p

A consortium of Hordeum vulgare and gut microbiota against non-alcoholic fatty liver disease via data-driven analysis.

Despite many recent studies on non-alcoholic fatty liver disease (NAFLD) therapeutics, the optimal treatment has yet to be determined. In this unfinished project, we combined secondary metabolites (SMs) from the gut microbiota (GM) and Hordeum vulgare (HV) to investigate their combinatorial effects via network pharmacology (NP). Additionally, we analyzed GM or barley - signalling pathways - targets - metabolites (GBSTMs) in combinatorial perspectives (HV, and GM). A total of 31 key targets were analysed via a protein-protein interaction (PPI) network, and JUN was identified as the uppermost ta

PEG treatment is unsuitable to study root related traits as it alters root anatomy in barley (Hordeum vulgare L.).

BACKGROUND: The frequency and severity of abiotic stress events, especially drought, are increasing due to climate change. The plant root is the most important organ for water uptake and the first to be affected by water limitation. It is therefore becoming increasingly important to include root traits in studies on drought stress tolerance. However, phenotyping under field conditions remains a challenging task. In this study, plants were grown in a hydroponic system with polyethylene glycol as an osmotic stressor and in sand pots to examine the root system of eleven spring barley genotypes. T

Effect of Fe3O4 and CuO Nanoparticles on Morphology, Genotoxicity, and miRNA Expression on Different Barley (Hordeum vulgare L.) Genotypes.

Metal nanoparticles (NPs) have an influence on plant growth and development. They can alter plant shoot and root length, fresh biomass production, and even influence the genome. Nanoparticles are also able to affect expression levels of plant microRNAs. MicroRNAs are able to protect plants from biotic stress, including pathogens which cause powdery mildew. In this study, Hordeum vulgare L. varieties "Marthe" and "KWS Olof" were grown in hydroponics with magnetic iron oxide (Fe3O4) and copper oxide (CuO) NPs added at 17, 35, and 70 mg/L. Plant morphology, genotoxicity, and expression of miR156a

Solute flows from Hordeum vulgare to the hemiparasite Rhinanthus minor and the influence of infection on host and parasite nutrient relations.

Using the facultative root hemiparasite Rhinanthus minor and Hordeum vulgare L. as a host, the flows and partitioning of mineral nutrients within the host, the parasite and between host and parasite have been studied during the study period 41-54 d after planting, i.e approximately 30-43 d after successful attachment of the parasite to the host. In parasitising Rhinanthus shoot growth was 12-fold, but root growth only 2-fold increased compared to non-parasitising plants. Conversely, in the Hordeum host, shoot dry matter growth was clearly reduced, by 33% in leaf laminae and by 52% in leaf shea

Reconstitution of cyanogenesis in barley (Hordeum vulgare L.) and its implications for resistance against the barley powdery mildew fungus.

Barley (Hordeum vulgare L.) produces a leucine-derived cyanogenic beta-D-glucoside, epiheterodendrin that accumulates specifically in leaf epidermis. Barley leaves are not cyanogenic, i.e. they do not possess the ability to release hydrogen cyanide, because they lack a cyanide releasing beta-D-glucosidase. Cyanogenesis was reconstituted in barley leaf epidermal cells through single cell expression of a cDNA encoding dhurrinase-2, a cyanogenic beta-D-glucosidase from sorghum. This resulted in a 35-60% reduction in colonization rate by an obligate parasite Blumeria graminis f. sp. hordei, the ca

Interactive effects of soil acidity and fluoride on soil solution aluminium chemistry and barley (Hordeum vulgare L.) root growth.

A greenhouse study was conducted to determine if concentrations of fluoride (F), which would be added to acid soils via P fertilisers, were detrimental to barley root growth. Increasing rates of F additions to soil significantly increased the soil solution concentrations of aluminium (Al) and F irrespective of the initial adjusted soil pH, which ranged from 4.25 to 5.48. High rates of F addition severely restricted root growth; the effect was more pronounced in the strongly acidic soil. Speciation calculations demonstrated that increasing rates of F additions substantially increased the concen

Changes in organic compounds secreted by roots in two Poaceae species (Hordeum vulgare and Polypogon monspenliensis) subjected to iron deficiency.

Despite their economic and ecological interests, Poaceae are affected by the low availability of iron in calcareous soils. Several studies focused on the capacity of this family to secrete phytosiderophores and organic acids as a mechanism of tolerance to iron deficiency. This work aimed at studying the physiological responses of two Poaceae species; Hordeum vulgare (cultivated barley) and Polypogon monspenliensis (spontaneous species) to iron deficiency, and evaluate especially the release of phytosiderophores and organic acids. For this purpose, seedlings of these two species were cultivated

Silicon nutrition enhances CYP450s- and GST-mediated metabolism of sulfosulfuron in wheat and an invasive weed, Hordeum vulgare subsp. spontaneum.

BACKGROUND: Although silicon nutrition in crops has been reported to improve growth and herbicide tolerance, the response of crop-associated weeds has not been studied. RESULTS: In glasshouse studies, co-applying potassium silicate (K2SiO3) + silicate-solubilizing bacteria (SSB) synergistically increased wheat and spontaneous barley's silicon content by 2.2- and 2.6-fold, respectively. Based on the median effective dose, it increased wheat (2.2-2.8-fold) and spontaneous barley (3.5-3.7-fold) tolerance to sulfosulfuron. Identifying stable and resilient advanced barley (Hordeum vulgare L.) lines analyzed using AMMI and GGE biplot models for grain yield.

The primary objective of this study was to quantify genotype-by-environment interactions for grain yield, and to evaluate drought tolerance in newly developed advanced barley lines. Sixteen barley lines were evaluated over a four consecutive year (2020-2023) across five distinct agro-ecological zones in Iran under rainfed field conditions. Grain yield ranged from 0.210 t ha-1 (2022) to 0.376 t ha-1 (2023), with an overall mean of 1.883 t ha-1. Analysis of variance indicated that environmental effects were the dominant source of variation, accounting for 87.47% of the total sum of squares and e

Genome wide identification, evolution and expression analysis of NHX gene family in cultivated barley (Hordeum vulgare. L) grown under salinity.

BACKGROUND: The Na⁺/H⁺ antiporter (NHX) gene family in plants encodes proteins that maintain ion homeostasis, particularly under salt stress, by exchanging Na⁺ or K⁺ for H⁺ across cellular membranes. In cultivated barley (Hordeum vulgare L.), NHX genes have been only partially characterized. METHODS AND RESULTS: In this study, we conducted a genome-wide identification of NHX genes in barley, examining their evolutionary relationships, gene structure, and expression patterns under salt stress (200 mM NaCl) in two genotypes differing in salinity tolerance. Seven genes were identified in the H.
