Horseradish peroxidase-catalyzed sulfoxidation of promethazine and properties of promethazine sulfoxide.

Promethazine sulfoxide was obtained with a quantitative yield in a horse radish peroxidase-catalyzed reaction of promethazine and hydrogen peroxide and was also prepared by direct chemical synthesis. The enzymatic sulfoxidation of promethazine was studied in vitro as a function of pH, promethazine, and hydrogen peroxide concentration. Promethazine sulfoxide inhibits with an apparent K(i) of 59.7 microM at pH 5.5 the enzymatic reaction, followed spectrophotometrically, polarographically, potentiometrically, and luminometrically. The reaction was also inhibited by ascorbic acid (K(i) 26.8 microM

[Permeability to horseradish peroxidase of the ciliary muscle vessels of squirrel monkeys].

The morphology and permeability to horseradish peroxidase (HRP) of the ciliary muscle capillaries of squirrel monkeys were studied. The endothelial cells of the capillaries were of the continuous type and the interendothelial clefts were closed by a zonula occludens. Many vesicles were present in the cytoplasm of the endothelial cells. In addition to these vesicles, large vacuole-like structures, 100-500 nm in diameter, were observed in the cytoplasm of capillary endothelium, especially in the circular and radial ciliary muscles. When HRP was perfused into the anterior chamber, the 3,3'-diamin

Afferent innervation of the gallbladder in the cat, studied by the horseradish peroxidase method.

Following injection of horseradish peroxidase (HRP) into the wall of the gallbladder of cats, HRP-positive cells were found bilaterally in dorsal root ganglia T2-L3 (T2-L2, and T3-L2/L3 also observed in a few cats) and nodose ganglia. In about 33% of animals labelled cells were also distributed in cervical dorsal root ganglia C5-C7. Labelled cells were more frequently localized on the right side than the left. There was no apparent change in numbers of labelled cells in the nodose ganglion (NG) on either side following greater and lesser splanchnicotomy or section of the right phrenic nerve or

The autonomic innervation in the pharynx. A study by the horseradish peroxidase tracer method.

Autonomic innervation of the mucosa in the pharynx was investigated by the horseradish peroxidase retrograde tracer method. The pharynx was divided into three regions, the epipharynx, mesopharynx, and hypopharynx. The epipharynx was sympathetically innervated by the fibers originating in the ipsilateral superior cervical ganglion and parasympathetically innervated by the fibers originating in the bilateral pterygopalatine ganglia. The mesopharynx and hypopharynx were sympathetically innervated by the fibers originating in the superior cervical, middle cervical, and stellate ganglia but not inn

Reabsorption of horseradish peroxidase by proximal tubules in rats with Heymann nephritis.

The distribution of the histochemical protein tracer, horseradish peroxidase, was studied in proximal tubules of rats with Heymann nephritis. Peroxidase reabsorption was substantially reduced in stage 2 of Heymann nephritis, a period during which the brush border of proximal tubules is severely damaged by specific antibodies. Impairment of the reabsorption function could not be attributed either to proteinuria or disturbances of proximal tubule metabolism and appeared to result from loss of microvilli. Recovery of brush border membrane morphology in stage 4 of Heymann nephritis was not accompa

Thalamic afferents to the motor cortex in the cat. A horseradish peroxidase study.

Thalamic projections to the motor cortex have been studied in the cat by injecting small amounts of horseradish peroxidase in different parts of areas 4 and 6. The nuclei which more abundantly project to the motor cortex are the ventral anterior-ventral lateral complex, the ventral medial and the posterior complex. These projections are topographically organized. Other projections arise from the ventral postero-lateral, ventral postero-inferior, central lateral, central medial, medial dorsal, lateral posterior-pulvinar, suprageniculate and centrum medianum-parafascicular nuclei.

Magnetically Recoverable ICT-Functionalized Fe3O4 Nanoparticles for Efficient Horseradish Peroxidase Immobilization.

The formation of interfacial charge transfer (ICT) complexes between phenolic ligands and metal oxide surfaces enables surface functionalization strategies with potential applications in catalysis and bioconjugation. In this study, magnetite (Fe3O4) nanoparticles were modified with two phenolic ligands, 5-aminosalicylic acid (5ASA) and caffeic acid (CA), to generate ICT complexes capable of covalent or non-covalent enzyme immobilization, respectively. The modified nanomaterials were structurally characterized using X-ray diffraction (XRD), transmission electron microscopy (TEM), and Fourier-tr

Hydrogen bond network in the distal site of peroxidases: spectroscopic properties of Asn70 --> Asp horseradish peroxidase mutant.

The distal His in peroxidases forms a hydrogen bond with the adjacent Asn, which is highly conserved among many plant and fungal peroxidases. Our previous work [Nagano, S., Tanaka, M., Ishimori, K., Watanabe, Y., & Morishima, I. (1996) Biochemistry 35, 14251-14258] has revealed that the replacement of Asn70 in horseradish peroxidase C (HRP) by Val (N70V) and Asp (N70D) discourages the oxidation activity for guaiacol, and the elementary reaction rate constants for the mutants was decreased by 10-15-fold. In order to delineate the structure-function relationship of the His-Asn couple in peroxida

Immobilization of horseradish peroxidase with zwitterionic polymer material for industrial phenolic removal.

Horseradish peroxidase (HRP) is a hemoglobin composed of a single peptide chain that catalyzes the oxidation of various substrates such as phenol and aniline in the presence of hydrogen peroxide via its iron-porphyrin catalytic center. This enzyme is widely used in industrial phenol removal, food additives, biomedicine, and clinical test reagents due to its rapid reaction rate and obvious reaction outcomes. However, the large-scale use of HRP in industrial applications still faces numerous challenges, including activity, stability, and sustainability. This study demonstrates that when peroxida

Electrochemically mediated reduction of horseradish peroxidase by 1,1'-ferrocenedimethanol in organic solvents.

Cyclic voltammetry is an efficient means of analyzing the catalytic reduction of H2O2 at immobilized horseradish peroxidase (HRP)-Eastman AQ 55 electrodes in the presence of 1,1'-ferrocenedimethanol as a one-electron reversible cosubstrate. This system was employed to study the kinetics of the reduction of compound II of HRP in a number of organic solvents. An electrocatalytic response was detected in methanol, ethanol, 1-propanol, 2-propanol, 1-butanol, acetone, 2-butanone, 1,2-propanediol, acetonitrile, ethyl acetate, and ethylene glycol. Unusual bell-shaped variations of the peak or plateau

Rescue of the catalytic activity of an H42A mutant of horseradish peroxidase by exogenous imidazoles.

His-42 plays a critical role in the H2O2-dependent catalytic turnover of horseradish peroxidase (HRP). This is clearly illustrated by the finding that an H42A mutation decreases the rate of Compound I formation by a factor of approximately10(6). As shown here, the addition of 2-substituted imidazoles partially rescues both the rate of formation of Compound I and the peroxidase activity of the H42A mutant. 2-Substituted imidazoles are the most effective because they do not coordinate to the iron. In contrast to native HRP, which exhibits a parabolic pH profile, and the H42A mutant, for which th

Permeability of the round window membrane to horseradish peroxidase in experimental otitis media.

The permeability of the round window membrane (RWM) was studied by using horseradish peroxidase (HRP) in the different stages of otitis media developed in guinea pigs, and it was compared with normal RWM under transmission electron microscopic examination. The experimental model of otitis media was developed by inoculation of a Pseudomonas aeruginosa suspension. When HRP could penetrate into the inner layer of a normal RWM, the duration of HRP instillation into the round window niche was 10 minutes. The permeability of the RWM in otitis media was as follows: 3-day animals showed penetration of

[Anatomy of the central auditory pathway. Demonstration with horseradish peroxidase in the guinea pig].

The neurones of the afferent and efferent auditory pathways were labelled by axonally transported horseradish peroxidase. The first neurone of the ascending system is the acoustic nerve with the spiral ganglion. These fibres terminate mainly in the ventral cochlear nucleus (VCN). The neurones of the latter chiefly run to the contralateral superior olivary complex (SOC), whereas the neurones of the dorsal cochlear nucleus (DCN) terminate mainly in the central nucleus of the contralateral inferior colliculus (IC). The nerve cells of the superior olivary complex and nuclei of the lateral lemniscu

Morphological effects of VIth nerve section in the kitten as revealed by horseradish peroxidase.

The effects of the chronic section of the VIth nerve have been studied in kittens with the help of the horseradish peroxidase intra-axonal retrograde tracing method. Two months following the section of the VIth nerve, only 50% of the abducens motoneuron population was left. However, shapes, somata sizes and cell distribution through the nucleus of the remaining neurons were similar to those shown by controls. Functional implications of the findings are discussed.

Aerobic oxidation of rifamycin SV to rifamycin S catalyzed by horseradish peroxidase.

The aerobic oxidation of rifamycin SV to rifamycin S was catalyzed by horseradish peroxidase. The oxidation rate increased with increasing pH. The oxidation was inhibited by catalase, but superoxide dismutase enhanced the oxidation rate of rifamycin SV. Superoxide inhibited the oxidation and superoxide dismutase relieved the inhibition of oxidation by superoxide. An oxidation mechanism of rifamycin SV catalyzed by horseradish peroxidase which accounts for these data is proposed and a convenient positive assay for superoxide dismutase, using the rifamycin SV-horseradish peroxidase-O2 system, is

Vascular permeability to horseradish peroxidase in brainstem lesions of thiamine-deficient rats.

In the early brainstem lesions of acute dietary thiamine deficiency in rats, an outstanding feature is the occurrence of edema. This study, using horseradish peroxidase as a marker, confirms and extends our previous observations using fluorescent dye-labeled albumin that vascular permeability to proteins remains essentially intact during this phase, and becomes altered only when necrosis and hemorrhage supervene. In addition, however, although a minor degree of pinocytotic transendothelial transport and pericytic uptake of horseradish peroxidase was found in both normal animals and in the earl

3D Bioprinting of Sugar Beet Pectin through Horseradish Peroxidase-Catalyzed Cross-Linking.

Horseradish peroxidase (HRP)-mediated hydrogelation, caused by the cross-linking of phenolic groups in polymers in the presence of hydrogen peroxide (H2O2), is an effective route for bioink solidification in 3D bioprinting. Sugar beet pectin (SBP) naturally has cross-linkable phenols through the enzymatic reaction. Therefore, chemical modifications are not required, unlike the various polymers that have been used in the enzymatic cross-linking system. In this study, we report the application of SBP in extrusion-based bioprinting including HRP-mediated bioink solidification. In this system, H2O

Influence of Cu(II) on the interaction between sulfite and horseradish peroxidase in vitro.

This paper discussed the quantitative influence of Cu(II) on the interaction between horseradish peroxidase (HRP) and sulfite (SO3(2-)), which is a derivate of sulfite dioxide in human bodies, by using fluorescence spectrum and ultraviolet (UV) absorption spectrometry in vitro. The results show that under the conditions of physiological pH and room-temperature, Cu(II) can bind strongly with both the protein part and the ferroporphyrin part in HRP at a low concentration (10(-4) mol L(-1)), and the combination constants are 2.047 x 10(3) and 7.66 x 10(2) L mol(-1), respectively. The afferent connections of the parvocellular reticular formation were systematically investigated in the rat with the aid of retrograde and anterograde horseradish peroxidase tracer techniques. The results indicate that the parvocellular reticular formation receives its main input from several territories of the cerebral cortex (namely the first motor, primary somatosensory and granular insular areas), districts of the reticular formation (including its contralateral counterpart, the intermediate reticular nucleus, the nucleus of Probst's bundle, the dorsal paragigantocellular nucleus, the al
