Flu-CNN: identifying host specificity of Influenza A virus using convolutional networks.

Influenza A viruses (IAVs) have historically posed significant public health threats, causing severe pandemics. Viral host specificity is typically constrained by host barriers, limiting the range of species that can be infected. However, these barriers are not absolute, and occasionally, cross-species transmission occurs, leading to human outbreaks. Early identification of changes in IAV host specificity is, therefore, critical. Despite advancements, identifying host susceptibility from genomic sequences during outbreaks remains challenging. Timely predictions are critical for effective real-

MOLECULAR GENETICS UNVEIL HOST SPECIFICITY OF TREMATODES IN TWO COGENERIC BATILLARIA SNAILS.

Host specificity is essential to understanding the ecology and evolution of parasites, and it is often complicated to estimate because of the presence of morphologically similar but genetically distinct species. Morphological identification of larval trematodes is often challenging because of the lack of diagnostic characteristics that only appear in the sexually mature adult stage. The difficulty in accurate species identification may obscure their compatibility with the host species. We investigate the parasites infecting 2 Asian mud snail species, Batillaria multiformis and Batillaria attra

For Someone, You Are the Whole World: Host-Specificity of Salmonella enterica.

Salmonella enterica is a bacterial pathogen known to cause gastrointestinal infections in diverse hosts, including humans and animals. Despite extensive knowledge of virulence mechanisms, understanding the factors driving host specificity remains limited. In this study, we performed a comprehensive pangenome-wide analysis of S. enterica to identify potential loci determining preference towards certain hosts. We used a dataset of high-quality genome assemblies grouped into 300 reference clusters with a special focus on four host groups: humans, pigs, cattle, and birds. The reconstructed pangeno

Genome-wide insights into population structure and host specificity of Campylobacter jejuni.

The zoonotic pathogen Campylobacter jejuni is among the leading causes of foodborne diseases worldwide. While C. jejuni colonises many wild animals and livestock, persistence mechanisms enabling the bacterium to adapt to host species' guts are not fully understood. In order to identify putative determinants influencing host preferences of distinct lineages, bootstrapping based on stratified random sampling combined with a k-mer-based genome-wide association was conducted on 490 genomes from diverse origins in Germany and Canada. We show a strong association of both the core and the accessory g

Host-specificity of monogenean (platyhelminth) parasites: a role for anterior adhesive areas?

Monogeneans (flatworms) are among the most host-specific of parasites in general and may be the most host-specific of all fish parasites. Specificity, in terms of a restricted spatial distribution within an environment, is not unique to parasites and is displayed by some fungi, insects, birds, symbionts and pelagic larvae of free-living marine invertebrates. The nature of cues, how "habitats" are recognised and how interactions between partners are mediated and maintained is of interest across these diverse "associations". We review some experiments that demonstrate important factors that cont

Motor and sensory specificity of host nerve axons influence nerve allograft rejection.

Previous studies have shown both survival and loss of regenerated host axons within nerve allograft segments after withdrawal of Cyclosporin A (CsA) immunosuppression. We hypothesized that the nature of end-organ reinnervation may influence the response of the axon, with survival of axons for appropriate innervation vs degeneration for inappropriate innervation. The rat femoral nerve model was chosen, as it has approximately equal sensory (S) and motor (M) divisions. Four ACI rat peroneal nerve allografts were sutured in straight (right leg: MM and SS) or switched (left leg; MS and SM) orienta

Life history and host specificity of Mediorhynchus centurorum Nickol 1969 (Acanthocephala: Gigantorhynchidae).

Examination of 1905 arthropods from a swamp and surrounding pasture in sothern Louisiana revealed larval specimens of Mediorhynchus centurorum in 8 of 228 woodroaches, Parcoblatta pensylvanica, but in no other species. The life cycle was confirmed by feeding eggs of M. centurorum to laboratory-reared woodroaches. Cystacanths later recovered from the woodroaches developed into mature worms when pipetted into esophaguses of red-bellied woodpeckers, Centurus carolinius: red-headed woodpeckers, Melanerpes erythrocephalus; yellow-shafted flickers, Colaptes auratus; and a hairy woodpecker, Dendrocop

Subcuticular symbionts of intertidal brittle stars: diversity, host specificity, and functional potential.

Echinoderms, widely distributed and abundant marine invertebrates, host diverse microbial communities, including subcuticular symbiotic bacteria (SCB). However, the diversity and functional roles of these bacteria remain largely unexplored in intertidal brittle stars. Here, we utilized both culture-dependent and -independent methods to investigate SCBs in three different intertidal brittle star species. Amplicon sequencing revealed distinct subcuticular microbiota among the three brittle star species, with Endozoicomonadaceae dominating in Ophiarachnella gorgonia, Spirochaetota prevalent in O.

Host specificity of gastrointestinal parasites in free-ranging sloths from Costa Rica.

The diversity and host specificity of gastrointestinal parasites infecting free-ranging sloths is poorly known. We compared gastrointestinal parasites of two sloth species from Costa Rica-three-fingered sloths (Bradypus variegatus) and two-fingered sloths (Choloepus hoffmanni)-for the first time in both a primary forest and an urban habitat. We asked whether host-parasite interactions were predominantly structured by host identity, the habitats in which hosts occurred, or both. Coproparasitology revealed protozoa and nematode eggs from both host species, but cestode eggs were recorded only in

Progress on viral and host determinants of influenza A virus species specificity.

Influenza A viruses have a wide range of hosts and are highly infectious, which can cause zoonotic diseases and pose a serious public health threat to human safety. An influenza pandemic could outbreak if new strains gain the ability of human-to-human transmission, either by genetic mutation or by gene reassortment. It is an urgent issue for the scientific community to reveal the genetic basis and molecular mechanisms underlying the interspecies transmission of influenza viruses, which will provide important implications for the effective monitoring and prevention of potential influenza pandem

Pathogenic Leptospira and their animal reservoirs: testing host specificity through experimental infection.

Leptospirosis is caused by pathogenic Leptospira transmitted through contact with contaminated environments. Most mammalian species are infectable by Leptospira but only few act as efficient reservoir being capable of establishing long term kidney colonization and shedding Leptospira in urine. In Madagascar, a large diversity of pathogenic Leptospira display a tight specificity towards their endemic volant or terrestrial mammalian hosts. The basis of this specificity is unknown: it may indicate some genetically determined compatibility between host cells and bacteria or only reflect ecological

Insights into the Host Specificity of Mosquito-Borne Flaviviruses Infecting Wild Mammals.

Mosquito-borne flaviviruses (MBFVs) are of public and animal health concern because they cause millions of human deaths annually and impact domestic animals and wildlife globally. MBFVs are phylogenetically divided into two clades, one is transmitted by Aedes mosquitoes (Ae-MBFVs) associated with mammals and the other by Culex mosquitoes (Cx-MBFVs) associated with birds. However, this assumption has not been evaluated. Here, we synthesized 79 published reports of MBFVs from wild mammals, estimating their host. Then, we tested whether the host specificity was biased to sampling and investigatio

The 150-Loop Restricts the Host Specificity of Human H10N8 Influenza Virus.

Adaptation of influenza A viruses to new hosts are rare events but are the basis for emergence of new influenza pandemics in the human population. Thus, understanding the processes involved in such events is critical for anticipating potential pandemic threats. In 2013, the first case of human infection by an avian H10N8 virus was reported, yet the H10 hemagglutinin (HA) maintains avian receptor specificity. However, the 150-loop of H10 HA, as well as related H7 and H15 subtypes, contains a two-residue insert that can potentially block human receptor binding. Mutation of the 150-loop on the ba

The lyssavirus host-specificity conundrum-rabies virus-the exception not the rule.

Lyssaviruses are a diverse range of viruses which all cause the disease rabies. Of the 16 recognized species, only rabies viruses (RABV) have multiple host reservoirs. Although lyssaviruses are capable of infecting all mammals, onward transmission in a new host population requires adaptation of the virus, in a number of stages with both host and virus factors determining the outcome. Due to an absence of recorded non-RABV host shifts, RABV data is extrapolated to draw conclusions for all lyssaviruses. In this article, we have focused on evidence of host shifts in the same insectivorous bat res

Patterns of cryptic host specificity in duck lice based on molecular data.

Documenting patterns of host specificity in parasites relies on the adequate definition of parasite species. In many cases, parasites have simplified morphology, making species delimitation based on traditional morphological characters difficult. Molecular data can help in assessing whether widespread parasites harbour cryptic species and, alternatively, in guiding further taxonomic revision in cases in which there is morphological variation. The duck louse genus Anaticola (Phthiraptera: Philopteridae), based on current taxonomy, contains both host-specific and widespread species. Mitochondria

Morphologic, host specificity, and molecular characterization of a Hungarian Cryptosporidium meleagridis isolate.

This study was undertaken in order to characterize Cryptosporidium meleagridis isolated from a turkey in Hungary and to compare the morphologies, host specificities, organ locations, and small-subunit RNA (SSU rRNA) gene sequences of this organism and other Cryptosporidium species. The phenotypic differences between C. meleagridis and Cryptosporidium parvum Hungarian calf isolate (zoonotic genotype) oocysts were small, although they were statistically significant. Oocysts of C. meleagridis were successfully passaged in turkeys and were transmitted from turkeys to immunosuppressed mice and from

Morphologic, host specificity, and genetic characterization of a European Cryptosporidium andersoni isolate.

This study was undertaken in order to characterize a Cryptosporidium muris-like parasite isolated from cattle in Hungary and to compare this strain with other Cryptosporidium species. To date, the large-type oocysts isolated from cattle were considered as C. muris described from several mammals. The size, form, and structure of the oocysts of the Hungarian strain were identical with those described by others from cattle. An apparent difference between the morphometric data of C. muris-like parasites isolated from cattle or other mammals was noted, which is similar in magnitude to the differenc

Host-Specific Adaptation of Legionella pneumophila to Single and Multiple Hosts.

Legionella pneumophila is an endosymbiotic bacterial species able to infect and reproduce in various protist and human hosts. Upon entry into human lungs, they may infect lung macrophages, causing Legionnaires' disease (LD), an atypical pneumonia, using similar mechanisms as in their protozoan hosts, despite the 2 hosts being separated by a billion years of evolution. In this study, we used experimental evolution to identify genes conferring host specificity to L. pneumophila. To this end, we passaged L. pneumophila in 2 different hosts-Acanthamoeba castellanii and the human macrophage-like ce
