Patience is not always a virtue: effects of terrain complexity on the host-seeking behaviour of adult blacklegged ticks, Ixodes scapularis, in the presence of a stationary host.

Blacklegged ticks, Ixodes scapularis Say (Acari: Ixodidae), are the primary vectors of Lyme disease in the U.S.A. In this study, adult ticks were observed on public trails exhibiting increasing levels of terrain complexity with a potential host nearby. The goal of this study was to (a) examine the extent to which adult ticks may actively search (vs. sit-and-wait) for a nearby host, (b) determine whether or not ticks could locate the position of the host in natural conditions and (c) determine the role of terrain complexity on the distances ticks travelled in a short period of time (30 min). Aversion of the invasive Asian longhorned tick to the white-footed mouse, the dominant reservoir of tick-borne pathogens in the U.S.A.

The Asian longhorned tick (Haemaphysalis longicornis) was reported for the first time in the U.S.A. in 2017 and has now spread across 12 states. The potential of this invasive tick vector to transmit pathogens will be determined through its association to hosts, such as the white-footed mouse (Peromyscus leucopus), which is the primary reservoir for the causative agent of Lyme disease (Borrelia burgdorferi) and other zoonotic pathogens. Larval H. longicornis were placed on P. leucopus; 65% of the larvae (n = 40) moved off the host within a short period of time, and none engorged. By contrast,

Effects of Insecticides on Oviposition and Host Discrimination Behavior in Trichogramma chilonis (Hymenoptera: Trichogrammatidae).

Trichogramma chilonis Ishii is an important natural enemy of many lepidopterous pests on various crops. Effects of two conventional insecticides on oviposition and host discrimination behavior in T. chilonis were evaluated in the laboratory through video tracking. During the oviposition period, when the number of host eggs was limited (only one host egg), females exposed to beta-cypermethrin LC1 or LC20 exhibited significantly higher feeding and re-drilling rate and significantly longer post-oviposition duration than control females. Spinosad LC20 treatment not only decreased the wasp oviposit

Carbon Dioxide, Methane, and Synthetic Cattle Breath Volatiles Attract Host-Seeking Stable Flies, Stomoxys calcitrans.

Stable flies, Stomoxys calcitrans (L.), are blood-feeding ectoparasites of cattle. Host-seeking stable flies respond to various cattle host cues, but a potential role of cattle breath gases [carbon dioxide (CO2), methane (CH4)] and cattle breath volatiles (acetone, isoprene, 2-butanone, 2-propanol, propionic acid, 3-methyl butyric acid, phenol), alone or in combination, on host-seeking behavior of stable flies has not yet been comprehensively investigated. In laboratory and greenhouse experiments, we tested the hypotheses that (1) CO2 and CH4 interactively attract stable flies, (2) CO2 'gates'

Steroid hormone pathways coordinate developmental diapause and olfactory remodeling in Pristionchus pacificus.

Developmental and behavioral plasticity allow animals to prioritize alternative genetic programs during fluctuating environments. Behavioral remodeling may be acute in animals that interact with host organisms, since reproductive adults and the developmentally arrested larvae often have different ethological needs for chemical stimuli. To understand the genes that coordinate the development and host-seeking behavior, we used the entomophilic nematode Pristionchus pacificus to characterize dauer-constitutive mutants (Daf-c) that inappropriately enter developmental diapause to become dauer larva

Semiochemical-Mediated Host-Searching and Biological Control Potential of Trichogramma Wasps: Mechanisms, Behavioral Plasticity, and Pest Management Applications.

Globally, Trichogramma Westwood (Hymenoptera: Trichogrammatidae) is known as the most effective biological control agent due to its ability to parasitize insect pest eggs. However, identifying an appropriate host is vital for Trichogramma to prosper. Therefore, this study delves into the complex role of semiochemicals in shaping the host-seeking behavior of Trichogramma parasitoids, with a particular focus on their responses to both plant-derived and host-derived cues. The mechanism of semiochemical reception in Trichogramma wasps relies on a highly specialized, sensitive olfactory and gustato

Dengue infection modulates locomotion and host seeking in Aedes aegypti.

Pathogens may manipulate their human and mosquito hosts to enhance disease transmission. Dengue, caused by four viral serotypes, is the fastest-growing transmissible disease globally resulting in 50-100 million infections annually. Transmission of the disease relies on the interaction between humans and the vector Aedes aegypti and is largely dependent on the odor-mediated host seeking of female mosquitoes. In this study, we use activity monitors to demonstrate that dengue virus-1 affects the locomotion and odor-mediated behavior of Ae. aegypti, reflecting the progression of infection within t

Hold your breath - Differential behavioral and sensory acuity of mosquitoes to acetone and carbon dioxide.

Host seeking in the yellow fever mosquito, Aedes aegypti, and the African malaria mosquito, Anopheles coluzzii, relies on specific and generic host-derived odorants. Previous analyses indicate that the behavioral response of these species depends differentially on the presence of carbon dioxide (CO2) and other constituents in human breath for activation and attraction. In this study, we use a flight tube assay and electrophysiological analysis to assess the role of acetone, a major component of exhaled human breath, in modulating the behavioral and sensory neuronal response of these mosquito s

Carbon dioxide shapes parasite-host interactions in a human-infective nematode.

Skin-penetrating nematodes infect nearly one billion people worldwide. The developmentally arrested infective larvae (iL3s) seek out hosts, invade hosts via skin penetration, and resume development inside the host in a process called activation. Activated infective larvae (iL3as) traverse the host body, ending up as parasitic adults in the small intestine. Skin-penetrating nematodes respond to many chemosensory cues, but how chemosensation contributes to host seeking, intra-host development, and intra-host navigation - three crucial steps of the parasite-host interaction - remains poorly under

Sensory appendage protein triggers alarm to pyrethroid in Indian malarial vector Anopheles culicifacies.

Sensory appendage proteins (SAPs), members of the odorant-binding protein family, mediate chemical communication that is vital for mosquito survival and vector competence. Among the six identified SAP members, AcSAP1 and AcSAP2 exhibited dominant expression in olfactory and leg tissues of Anopheles culicifacies. Our findings reveal that the expression of AcSAP1, and to a lesser extent AcSAP2, is significantly modulated by the circadian rhythm, with peak expression coinciding with the nocturnal host-seeking behavior of female mosquitoes. A delayed host attraction response by the AcSAP1-silenced

Recurrent connectivity supports carbon dioxide sensitivity in Aedes aegypti mosquitoes.

The mosquito Aedes aegypti's human host-seeking behavior depends on the integration of multiple sensory cues. One of these cues, carbon dioxide (CO2), gates odorant and heat pathways and activates host-seeking behavior. The neuronal circuits underlying processing of CO2 information remain unclear. We used automated serial-section transmission electron microscopy (EM) to image and reconstruct the circuitry of the glomeruli that are innervated by the Ae. aegypti maxillary palp, including the glomerulus that responds to CO2. Notably, CO2-sensitive olfactory sensory neurons (OSNs) make high levels

Odorant receptor Or117 detects camphor to induce floral seeking by the mosquito Aedes aegypti.

Female mosquitoes display distinct, staged, and dynamic resource-seeking behaviors to sustain energy and reproductive demands. Teneral females are attracted to floral odors due to innate energy requirements. During maturation, females develop the competence to take a blood meal, a process expedited following mating and linked to a shift to vertebrate host cue attraction. The change in resource-seeking behaviors is correlated with an overall increase in abundance of chemosensory receptors, with select odorant receptor genes, including odorant receptor Or117, displaying an opposite dynamic expre

The Strongyloides stercoralis nervous system.

The skin-penetrating, human-parasitic nematode Strongyloides stercoralis infects over 600 million people worldwide, causing extensive morbidity and socioeconomic burden. Outside the host body, these nematodes usually live in moist soil as infective third-stage larvae (iL3s), a life stage that is considered to be evolutionarily homologous to the dauer stage of free-living nematodes such as Caenorhabditis elegans. S. stercoralis iL3s navigate their local environment and seek out hosts using sensory cues such as heat, host-emitted odorants, and carbon dioxide. Once host contact is made, iL3s acti

Geographical distribution patterns and habitat suitability models for presence of host-seeking ixodid ticks in dense woodlands of Mendocino County, California.

We used drag sampling to examine the geographical distribution patterns of ixodid ticks engaging in open (non-nidicolous) host-seeking behavior in dense woodland habitats of the climatically and ecologically diverse Mendocino County in north coastal California. The findings based on this sampling methodology reflect risk of human exposure to host-seeking ticks rather than the true distribution of the ticks. Drag sampling in 78 sites yielded 7,860 nymphal or adult Ixodes pacificus Cooley & Kohls, 220 Dermacentor occidentalis Marx, 150 Ixodes spinipalpis Hadwen & Nuttall, 15 Hemaphysalis leporis

The neural bases of host plant selection in a Neuroecology framework.

Understanding how animals make use of environmental information to guide behavior is a fundamental problem in the field of neuroscience. Similarly, the field of ecology seeks to understand the role of behavior in shaping interactions between organisms at various levels of organization, including population-, community- and even ecosystem-level scales. Together, the newly emerged field of "Neuroecology" seeks to unravel this fundamental question by studying both the function of neurons at many levels of the sensory pathway and the interactions between organisms and their natural environment. Using Videography to Study the Biomechanics and Behavior of Freely Moving Mosquitoes.

Female mosquitoes of most species require a blood meal for egg development. When biting a human host to collect this blood meal, they can spread dangerous diseases such as malaria, yellow fever, or dengue. Researchers use videography to study many aspects of mosquito behavior, including in-flight host-seeking, takeoff, and landing behaviors, as well as probing and blood feeding, and more. Here, we introduce protocols on how to use videography to capture and analyze mosquito movements at high spatial and temporal resolution, in two and three dimensions.

Effect of temperature on mosquito olfaction.

Mosquitoes use a wide range of cues to find a host to feed on, eventually leading to the transmission of pathogens. Among them, olfactory cues ( e.g. , host emitted odors, including CO 2 , and skin volatiles) play a central role in mediating host seeking behaviors. While mosquito olfaction can be impacted by many factors, such as the physiological state of the insect ( e.g. , age, reproductive state), the impact of environmental temperature on the olfactory system remains unknown. In this study, we quantified the behavioral responses of Aedes aegypti mosquitoes, vectors of dengue, yellow fever

Parasites in space and time: a novel method to assess and illustrate host-searching behaviour of trematode cercariae.

The transmission from one host to another constitutes a challenging obstacle for parasites and is a key determinant of their fitness. Due to their complex life histories involving several different hosts, the free-living dispersal stages (cercariae) of digenean trematodes show a huge diversity in morphology and behaviour. On a finer scale, we still have an extremely limited understanding of the inter- and intraspecific variation in transmission strategies of many trematode species. Here, we present a novel method to study the movement patterns of cercariae of four New Zealand trematode species

Medical students' healthcare seeking-behavior: A cross-sectional study from Sudan.

Healthcare-seeking behavior plays a crucial role in early diagnosis and effective treatment. In conflict-affected low-income communities like Sudan, factors affecting healthcare-seeking behavior and the rationale for engaging in self-medication practices can vary tremendously in comparison to stable communities. Responsibilities placed upon medical students who faced displacement are substantial within their host communities. This study aims to explore the role of armed conflict and the displacement crisis in reshaping the factors affecting the healthcare-seeking behavior and self-medication p
