Preparation and characterization of hybridomas secreting monoclonal antibodies to tick-borne encephalitis virus.

Monoclonal antibodies (MA) were prepared to two strains of tick-borne encephalitis (TBE) virus: strain 4072 isolated from a patient in the U.S.S.R. and low-pathogenic for mice strain Skalica, isolated from a bank vole (Clethrionomys glareolus) in Slovakia. MA specific to the 4072 and Skalica strains were produced by hybridomas of the KEN (60 clones) and NEK (65 clones) series, respectively. Chromosomal analysis of MA producing 114 hybridoma clones of both series revealed a great variability in the number of chromosomes either in the range of given clones or between individual clones. The hybri

Antibody Development Strategies for SFTSV: From Hybridoma to Emerging Technologies.

Severe fever with thrombocytopenia syndrome (SFTS) is an emerging zoonotic disease caused by the tick-borne SFTS virus (SFTSV), with a case fatality rate of 16.2-32.6% in East Asia. Currently, no approved vaccines or antiviral treatments exist. Monoclonal antibody (mAb) therapy offers rapid immune protection and is a promising strategy against SFTSV. This review outlines advances in SFTSV neutralizing mAb research, covering conventional generation methods (hybridoma, phage display) and innovative approaches such as single B cell sequencing. We also introduce computational tools like artificial

An anti-ganglioside antibody-secreting hybridoma induces neuropathy in mice.

Immune responses against gangliosides are strongly implicated in the pathogenesis of some variants of Guillain-Barré syndrome (GBS). For example, IgG antibodies against GM1, GD1a, and related gangliosides are frequently present in patients with post-Campylobacter acute motor axonal neuropathy (AMAN) variant of GBS, and immunization of rabbits with GM1 has produced a model of AMAN. However, the role of anti-ganglioside antibodies in GBS continues to be debated because of lack of a passive transfer model. We recently have raised several monoclonal IgG anti-ganglioside antibodies. We passively tr

Differentiation of a human B cell hybridoma with interleukin-2 receptor.

A 6-thioguanine-neo-resistant clone, Raji 6-TGR-neoR was fused with pokeweed mitogen-stimulated B cells (PWM-B cells) obtained from a normal healthy donor, and selected by a new selection medium (HAT-neo). Two hybrid clones, HYNI-1 and HYNI-2, were established. These clones expressed surface IgG and HLA class II antigens derived from PWM-B cells, and they had about 20 more chromosomes than Raji 6-TGR-neoR. More significantly, one clone, HYNI-2, expressed interleukin-2 receptor on the cell surface, and generated IgG when stimulated with recombinant interleukin-2. These findings indicate that th

[Monoclonal antibodies to the Japanese encephalitis virus in ascitic hybridoma preparations].

Among 4 ascitic preparations of hybridomas producing monoclonal antibodies to the Japanese encephalitis virus (JE) hybridomas JE-4 and JE-19 had high multiplication potentials and high levels of reimplantation and transplantability in mice BALB/c. The possibility of using the complete Freund's adjuvant and pristan for priming the mice was shown. The use of pristan promoted a significant decrease in the periods of ascitic tumor development and an increase in accumulation of the cells in the ascitic tumor, in the volume of the ascitic fluid and the titers of the monoclonal antibodies. The serolo

Improved Protocol for Establishing CD4+ Hybridomas Specific for Human Class II MHC/Peptide Complex.

Autoreactive CD4+ T cells are shaped by MHC class II-dependent selection, and HLA-DQ8 is a major susceptibility allele for type 1 diabetes and celiac disease. To define how HLA-DQ8 influences the autoreactive CD4+ T-cell repertoire, we generated T-cell hybridomas from HLA-DQ8 humanized mice using a BW5147 Nur77-GFP (BW-GFP) platform that enables sensitive quantification of antigen-induced T-cell receptor (TCR) signaling. The frequency of autoreactive conventional CD4+ hybridomas observed in HLA-DQ8 mice was higher than previously reported in C57BL/6 mice in our earlier study, suggesting that H

Metabolic and kinetic studies of hybridomas in exponentially fed-batch cultures using T-flasks.

Exponentially fed-batch cultures (EFBC) of a murine hybridoma in T-flasks were explored as a simple alternative experimental tool to chemostats for the study of metabolism, growth and monoclonal antibody (MAb) production kinetics. EFBC were operated in the variable volume mode using an exponentially increasing and predetermined stepwise feeding profile of fresh complete medium. The dynamic and steady-state behaviors of the EFBC coincided with those reported for chemostats at dilution rates below the maximum growth rate. In particular, steady-state for growth rate and concentration of viable ce

Development of a Bacterial Macroarray for the Rapid Screening of Targeted Antibody-Secreted Hybridomas.

Hybridoma screening is a key step for the successful generation of high-affinity analyte-specific monoclonal antibodies (MAbs). This work presents an innovative screening method, known as a bacterial macroarray, generated by contact printing of hybridoma cell supernatant samples on a nitrocellulose (NC) membrane initially coated with fluorescein isothiocyanate (FITC)-labeled bacteria. Given that bacterial fixation will be influenced by complex bacterial surface structures, we selected both gram-positive bacteria ( Staphylococcus aureus and Listeria monocytogenes) and gram-negative bacteria ( E

Mouse autoreactive gamma/delta T cells. I. Functional properties of autoreactive T cell hybridomas.

A minor population of dendritic epidermal T cells (DETC) express the V gamma 1.1C gamma 4V delta 6 T cell receptor and T cell clones and hybridomas derived from this subset constitutively secrete cytokines in culture secondary to recognition of an autoantigen. Activation of these autoreactive cells requires the use of the vitronection receptor (VNR) as an accessory molecule which interacts with the Arg-Gly-Asp-Ser (RGDS) sequence of extracellular matrix (ECM) proteins. We have compared the functional properties of C gamma 4+ hybridomas derived from newborn thymocytes and from adult spleen with

Use of anti-CD4+ hybridoma cells to induce Pneumocystis carinii in mice.

A reduction of peripheral CD4+ cell levels has been correlated with the onset of Pneumocystis carinii pneumonia in AIDS patients. Most in vivo drug discovery and development for P. carinii have been conducted in corticosteroid-treated rats. There is need for the development of new small animal models with more selective methods of immunosuppression. This study outlines a new mouse model in which specific depletion of the CD4+ T-lymphocyte population was achieved by subcutaneous injection of G.K1.5 hybridoma cells into C3HeB/FeJ mice. A significant reduction in splenic CD4+ cells was maintained

Monoclonal antibody production by hybridoma growth in Freund's adjuvant primed mice.

Incomplete Freund's adjuvant was used as a priming agent prior to the injection of hybridoma cells in mice to expand monoclonal antibody production. Two hybridoma cell lines, FDO28B (IgG1) and FDO31C (IgM), which produce monoclonal antibodies reactive towards human placenta, were used. Monoclonal antibody was detected in the ascites fluids by agarose gel electrophoresis. It was found that the time interval between adjuvant priming and cell injection could be reduced to 1 day, allowing collection of ascites fluid containing monoclonal antibody within 2 weeks of priming. In addition, as few as 1

Hybridoma-inspired strategy crafts tailored multifunctional exosomes for precision therapy.

Engineering functional exosomes represents a cutting-edge approach in biomedicine, holding the promise to transform targeted therapy. However, challenges such as achieving consistent modification and scalability have limited their wider adoption. Herein, we introduce a universal and effective strategy for engineering multifunctional exosomes through cell fusion. The hybrid-cell-derived exosomes could combine the functional properties of both parental cells and be readily produced by passaging. This method enables customization and large-scale production of exosomes with specific functionalitie

Increase of immunoglobulin productivity of human-human hybridoma HB4C5 cells by histone.

A histone mixture (H1, H2A, H2B, H3, and H4) derived from calf thymus stimulated IgM production by human-human hybridoma HB4C5 cells. On the contrary, the histone mixture did not increase IgM production by the human Burkitt's lymphoma cell line NAT-30, IgG production by the human B lymphoblastoid cell line HMy-2, and IgE production by the human myeloma cell line U266. The immunoglobulin production-stimulating activity of the histone mixture was inactivated by trypsin or chymotrypsin digestion. In addition, confocal laser microscopic analysis had shown that HB4C5 cells incorporated a lot of his

Quantitative Proteomics and CRISPR/Cas9 Editing Reveal UPR-Mediated Control of Immunoglobulin Homeostasis in Hybridomas.

Despite their tremendous economic value, monoclonal antibodies are often compromised by the loss of immunoglobulin (Ig) chains, which disrupts antibody homeostasis and quality control. Through subclone screening and characterization, we identified that the loss of Ig production impaired the recognition ability of hapten-specific hybridomas. Proteomic analysis further highlighted the critical role of the unfolded protein response (UPR) pathway in regulating aberrant Ig chain production. Using CRISPR/Cas9-mediated knockout and rescue experiments, we revealed the importance of the UPR pathway in

CD19+ B lymphocytes are the major source of human antibody-secreting hybridomas generated by electrofusion.

Human monoclonal antibodies may be generated by electrofusion of human B lymphocytes with a human/mouse heteromyeloma line. In addition to a fusion protocol optimised for the fusion partners, the activation of B lymphocytes is crucial for fusion and hybrid efficiency. In this study, we initially treated peripheral blood mononuclear cells (PBMC) from normal blood donors with a large panel of known stimulants and determined the yield of human antibody-secreting hybridomas after electrofusion with the heteromyeloma cell line H73C11; 3- to 5-day incubation with phytohaemagglutinin L (PHA-L) result

Production and characterization of cloned T-cell hybridomas that are responsive to Rickettsia conorii antigens.

T-cell hybridomas produced by the fusion of Rickettsia conorii immune T cells to the AKR thymoma BW 5147 produced interleukin-2 when stimulated with the antigens of three different R. conorii strains. One cloned hybridoma responded only to R. conorii antigens, whereas a second and third cloned hybridoma also responded to the antigens of Rickettsia rickettsii Sheila Smith and Rickettsia sibirica 246, respectively. Antigen responses required antigen-presenting cells, and this interaction was major histocompatibility complex restricted. Fluorescence-activated cell-sorter analysis demonstrated tha

Effects of BCL-2 over-expression on B cells in transgenic rats and rat hybridomas.

The rat is an important biomedical experimental model that benefited from the recent development of new transgenic and knockout techniques. With the goal to optimize rat mAb production and to analyze the impact of Bcl-2 on B-cell development, we generated bcl-2 transgenic rats. Transgenic rats showed Bcl-2 over-expression in B cells, increased B cell numbers in lymphoid organs, elevated production of immunoglobulins (Igs) and prolonged B-cell survival in vitro. Transgenic rats remained healthy, reproduced normally and did not develop autoimmunity. Fusions with bcl-2 transgenic splenocytes did

Characterization of proteoglycan-reactive T cell lines and hybridomas from mice with proteoglycan-induced arthritis.

Hyperimmunization with chondroitin sulfate-depleted fetal human cartilage proteoglycan (HFPG) leads to the development of peripheral arthritis and spondylitis in BALB/c mice. Chondroitin-sulfate-depleted adult human cartilage proteoglycan (HAPG) is much less effective at inducing arthritis. These observations suggest age differences in the presence of arthritogenic proteoglycan (PG) epitopes. Earlier studies from this laboratory have indicated an important role for PG-reactive T cells in the pathogenesis of this arthritis model. To investigate further the cellular immunity to PG in mice, two T

Retinoic acid induces the differentiation of B cell hybridomas from patients with common variable immunodeficiency.

Human-human B cell hybridomas constructed from B lymphocytes of common variable immunodeficiency (CVI) patients and the nonsecreting cell line WIL2/729 HF consistently secrete low levels of Ig and appear to retain a defect characteristic of the CVI patient's B cells. We assessed the differentiative capacity of retinoic acid (RA) on these hybridomas, as well as on hybridomas constructed from normal B cells and from patients with selective IgA deficiency. RA at concentrations varying between 10(-5) and 10(-9) M augmented IgM secretion 4-20-fold from four of four CVI hybridomas tested, but did no

Regulation of B cell activation and differentiation with factors generated by human T cell hybridomas.

Human T cell hybridomas were generated by several techniques and the supernatants generated were screened for activity on human B cells. Three general activities were noted; B cell proliferation factor ( BCPF ), B cell differentiation factor (BCDF), and an IgA isotype-specific helper factor. BCPF acts on B cells to induce proliferation without differentiation and is distinct from conventional BCGF. This was documented by BCPF 's inability to synergize with anti-mu Ab in a standard BCGF assay ( Muraguchi & Fauci 1982, Howard et al. 1982, Sieckman et al. 1981), as well as its differential effect
