Local anesthetic lidocaine decelerates cytoplasmic streaming in Egeria densa via altered actin dynamics and reactive oxygen species generation.

Local anesthetics (LAs) block voltage-gated Na+ channels in mammals, but they also affect cytoplasmic streaming in plant cells. Although certain LAs inhibit streaming in algae such as Chara, the underlying mechanisms remain unclear. This study aimed to elucidate how LAs reduce cytoplasmic streaming velocity, focusing on the actomyosin system and oxidative stress responses, using lidocaine as a representative LA. Streaming velocity in Egeria densa leaves decreased reversibly after lidocaine treatment and recovered following a washout procedure. Confocal laser scanning microscopy of fluorescentl

Turtlegrass (Thalassia testudinum) undergoes a coordinated hypersensitive response when challenged with pathogenic Labyrinthula sp.

Infection of the marine subtropical seagrass Thalassia testudinum Banks ex König by pathogenic Labyrinthula sp. was found to induce lesion progression, alterations to the host's oxidative metabolism, and production of defense metabolites over the early stages of infection (monitored over a 72-hr time course). By 48-hr post-infection, host oxygen consumption, internal reactive oxygen concentrations, and caspase-3 proteolytic activity reached their highest levels. The formation of reactive oxygen species (ROS) and nitric oxide (NO) were detected in T. testudinum lesions by use of redox-sensitive

Sediment contamination alters the submersed macrophyte Vallisneria natans and root-associated microbiome profiles during phytoremediation.

The submerged plant Vallisneria natans plays an important role in the remediation of polycyclic aromatic hydrocarbon (PAH)-contaminated sediments. In this study, V. natans and sediments were collected from different V. natans natural vegetation zones, and sediment mesocosms were set up for phytoremediation tests. In addition, commercial-grade V. natans were obtained from the Fish-Bird-Flower market for comparison with phytoremediation. Phytoremediation using V. natans from natural growth significantly increased the degradation of PAHs in Dashui Harbor (0.0148±0.0015 d-1) and Taihu Lake bay sed

Glufosinate-ammonium increased nitrogen and phosphorus content in water and shaped microbial community in epiphytic biofilm of Hydrilla verticillata.

Glufosinate-ammonium (GLAM) can be released into adjacent water bodies with rainfall runoff and return water from farmland irrigation. However, impacts of GLAM on aquatic organisms remain unclear. In this study, changes in water quality, plant physiological parameters and epiphytic microbial community were investigated in wetlands with Hydrilla verticillata exposed to GLAM for 24 days. We found GLAM addition damaged cell and reduced chlorophyll a content in Hydrilla verticillata leaves, and increased ammonium and phosphorus in water (p < 0.001). The α-diversity increased in bacterial community

Calm after the storm? Similar patterns of genetic variation in a riverine foundation species before and after severe disturbance.

In summer 2011, Tropical storms Lee and Irene caused an estimated 90% decline of the submersed aquatic plant Vallisneria americana Michx. (Hydrocharitaceae) in the Hudson River of New York (USA). To understand the genetic impact of such large-scale demographic losses, we compared diversity at 10 microsatellite loci in 135 samples collected from five sites just before the storms with 239 shoots collected from nine sites 4 years after. Although 80% of beds sampled in 2011 lacked V. americana in 2015, we found similar genotypic and genetic diversity and effective population sizes in pre-storm ver

Transmission loss in manatee habitats.

The Florida manatee is regularly exposed to high volumes of vessel traffic and other human-related noise because of its coastal distribution. Quantifying specific aspects of the manatee's acoustic environment will allow for a better understanding of how these animals respond to both natural and human-induced changes in their environment. Transmission loss measurements were made in 24 sampling sites that were chosen based on the frequency of manatee presence. The Monterey-Miami Parabolic Equation model was used to relate environmental parameters to transmission loss in two extremely shallow wat

Application of microchip-CE electrophoresis to follow the degradation of phenolic acids by aquatic plants.

In this paper, we describe the separation and detection of six phenolic acids using an electrophoretic microchip with pulsed amperometric detection (PAD). The selected phenolic acids are particularly important because of their biological activity. The analysis of these compounds is typically performed by chromatography or standard CE coupled with a wide variety of detection modes. However, these methods are slow, labor intensive, involve a multistep solvent extraction, require skilled personnel, or use bulky and expensive instrumentation. In contrast, microchip CE offers the possibility of per

Elodea nuttallii exposure to mercury exposure under enhanced ultraviolet radiation: Effects on bioaccumulation, transcriptome, pigment content and oxidative stress.

The hypothesis that increased UV radiation result in co-tolerance to Hg toxicity in aquatic plants was studied at the physiological and transcriptomic level in Elodea nuttallii. At the transcriptomic level, combined exposure to UV+Hg enhanced the stress response in comparison with single treatments, affecting the expression level of transcripts involved in energy metabolism, lipid metabolism, nutrition, and redox homeostasis. Single and combined UV and Hg treatments dysregulated different genes but with similar functions, suggesting a fine regulation of the plant to stresses triggered by Hg, U

Cadmium-induced biochemical responses of Vallisneria spiralis.

The following study was carried out to investigate the cadmium (Cd) accumulating potential of Vallisneria. After subjecting plants to different concentrations of Cd, it was observed that plants are able to accumulate ample amount of metal in their roots (5,542 microg g(-1) dw) and leaves (4,368 microg g(-1) dw) in a concentration- and duration-dependent manner. Thus, it is evident that the accumulation in roots was 1.3 times higher than the shoots. It was also noted that with increasing Cd accumulation, roots of the plant appeared darker in color and harder in texture. In response to metal exp

Going With the Flow? Relative Importance of Riverine Hydrologic Connectivity Versus Tidal Influence for Spatial Structure of Genetic Diversity and Relatedness in a Foundational Submersed Aquatic Plant.

Genetic connectivity in rivers is generally high, and levels of genotypic and genetic diversity of riverine species are expected to accumulate in downstream locations. Genetic structure of marine and estuarine species is less predictable, even though hydrologic connectivity is also expected to be relatively high in those ecosystems. These observations have been generated across different species and locations such that our understanding of the effects of hydrologic connectivity in the same river, spanning tidal and nontidal habitats, remains incomplete. To control for species and location, we

Detection of the free-living forms of sulfide-oxidizing gill endosymbionts in the lucinid habitat (Thalassia testudinum environment).

Target DNA from the uncultivable Codakia orbicularis endosymbiont was PCR amplified from sea-grass sediment. To confirm that such amplifications originated from intact bacterial cells rather than free DNA, whole-cell hybridization (fluorescence in situ hybridization technique) with the specific probe Symco2 was performed along with experimental infection of aposymbiotic juveniles placed in contact with the same sediment. Taken together, the data demonstrate that the sulfide-oxidizing gill endosymbiont of Codakia orbicularis is present in the environment as a free-living uncultivable form.

In Silico Analysis of Bioactive Peptides in Invasive Sea Grass Halophila stipulacea.

Halophila stipulacea is a well-known invasive marine sea grass in the Mediterranean Sea. Having been introduced into the Mediterranean Sea via the Suez Channel, it is considered a Lessepsian migrant. Although, unlike other invasive marine seaweeds, it has not demonstrated serious negative impacts on indigenous species, it does have remarkable invasive properties. The present in-silico study reveals the biotechnological features of H. stipulacea by showing bioactive peptides from its rubisc/o protein. These are features such as antioxidant and hypolipideamic activities, dipeptidyl peptidase-IV

Fate and removal of fluoroquinolone antibiotics in mesocosmic wetlands: Impact on wetland performance, resistance genes and microbial communities.

The fate of fluoroquinolone antibiotics norfloxacin and ofloxacin were investigated in mesocosmic wetlands, along with their effects on nutrients removal, antibiotic resistance genes (ARGs) and epiphytic microbial communities on Hydrilla verticillate using bionic plants as control groups. Approximately 99% of norfloxacin and ofloxacin were removed from overlaying water, and H. verticillate inhibited fluoroquinolones accumulation in surface sediments compared to bionic plants. Partial least squares path modeling showed that antibiotics significantly inhibited the nutrient removal capacity (0.55

Trace metal partitioning in Thalassia testudinum and sediments in the Lower Laguna Madre, Texas.

Seagrass communities dominate the Laguna Madre, which accounts for 25% of the coastal region of Texas. Seagrasses are essential to the health of the Laguna Madre (LM) and have experienced an overall decline in coverage in the Lower Laguna Madre (LLM) since 1967. Little is known on the existing environmental status of the LLM. This study focuses on the trace metal chemistry of four micronutrient metals, Fe, Mn, Cu, and Zn, and two non-essential metals, Pb and As, in the globally important seagrass Thalassia testudinum. Seasonal trends show that concentrations of most essential trace metals incr

C4-like metabolism and HCO3- use in submerged leaves of Ottelia cordata lacking Kranz anatomy at both low and high CO2 concentrations.

To date, only a few submerged plants have been reported to perform C4 and CAM. Ottelia cordata is a heteroblastic aquatic plant developing both submerged and floating leaves throughout its life cycle. Previous research found that, besides HCO3- use, the submerged leaves of O. cordata can also perform C4 metabolism. However, it remains unclear how the HCO3- use or the C4 pathway, and the anatomical structure, respond to varying CO2 environments. Therefore, we examined the anatomical structures and carbon-concentrating mechanisms in O. cordata submerged leaves under high (HC) and low CO2 (LC) co

Trophic fate of inorganic and methyl-mercury in a macrophyte-chironomid food chain.

Dietary transfer of mercury (Hg) is central for its effects on higher trophic animals, nonetheless, its driving parameters and characteristics are not well understood. Here we measured Hg species transfer (uptake) from the macrophyte Elodea nuttallii -mimicking tissues incorporation in sediments after decay- to Chironomus riparius. Methyl-Hg (MMHg) was more transferable than inorganic Hg (IHg) from plant's intracellular and cell wall compartments. After 10-d-long exposure, MMHg was predominantly found in MMHg form in the cytosolic compartment (S) of chironomids, while IHg showed similar concen
