Over 30-40% of 1.5 million annual hand flexor tendon injuries in the United States result in peritendinous adhesions which limit range of motion (ROM) and severely impact quality of life. Currently, no widespread solution exists for adhesion prevention in the delicate space of the digit while allowing full ROM quickly following surgery. Here, we develop dynamically crosslinked, bioresorbable supramolecular hydrogels as easy-to-apply peritendinous adhesion barriers. These hydrogels exhibit long-term stability, injectability, and thermally stable viscoelastic properties that enable simple storag

Hydrogels are hydrated polymer networks with promising biomedical applications due to their microstructural similarity to biological tissues at sliding interfaces, such as the articular cartilage. These tissues are highly hydrated, charged, and operate in ionic environments. A fundamental understanding of lubrication at charged hydrogel interfaces is thus essential to advance practical applications. Hydration lubrication has recently been proposed to govern hydrogel lubrication. Here, a thin film of water remains confined under pressure, enabling friction coefficients below 0.01, a condition r

Water in hydrogels has often been overlooked or deemed detrimental to the mechanical properties of hydrogels due to the plasticizing effect of water, which weakens interchain interactions and accelerates crack propagation. Here, we propose a slow-water strategy that imparts enhanced fracture resistance to hydrogels. By converting free water into slowly migrating intermediate water, we suppress excess chain mobility by reducing the free volume around polymer chains, thereby enhancing interchain interactions and substantially extending the load-transfer length. Notably, even with only 13% polyme

Adhesive hydrogels are crucial for interfacing wearable electronics with substrates, yet the inherent trade-off between interfacial adhesion and cohesive strength always limits their application performance. Here, we present an adhesive hydrogel based on lignin-mediated catechol chemistry and salt hydrate phase-transition, which achieves synergistic enhancement of interfacial adhesion and cohesive strength upon 1 V electrostimulated crystallization. The hydrogel is soft and exhibits mild adhesion to various substrates in amorphous state. Electrostimulus-induced phase-transition realizes a sign

We present a new method to obtain tertiary amine-based prodrugs with dual functionality, enabling (i) signal-triggered drug activation and (ii) covalent incorporation in polymer materials through a clickable azido-group unit on the molecular prodrug scaffold. Using nucleophilic substitution on an electron deficient azido-phenyl allyl bromide scaffold, we were able to obtain prodrugs from a variety of amine drug candidates. Subsequent drug activation was initiated by using S or N-terminal biomarker nucleophiles including amino acids, a neurotransmitter, and glutathione as chemical signals. Hydr

Icing forecast provides advanced notification, enabling preemptive anti-icing treatments to prevent facility damage and minimize economic losses from unexpected icing events. However, in real-world environments and practical applications, current technologies struggle to accurately predict ice formation on solid surfaces. This difficulty arises from the random and unpredictable nature of ice nucleation, influenced by variable weather conditions, diverse ice-nucleating agents, complex surface properties, and uncertain material defects or contamination. Herein, inspired by the role of ice-nuclea

Traditional petrochemical-derived plastics are challenging to recycle and degrade, and the existing (re)process methods are organic solvent-based and/or energy-intensive, resulting in significant environmental contamination and greenhouse gas emissions. This study presents a sustainable bioplastic material characterized by multi-closed-loop recyclability and water (re)processability. The bioplastics are derived from abundant polysaccharide sources of dextran, alginic acid, carboxymethyl cellulose, and DNA of plant and living organism waste. The process involves chemical oxidation of polysaccha

Natural enzymes, with their precise structural organization and compartmentalization in metabolic pathways, exhibit remarkable catalytic efficiency, inspiring the design of enzyme-integrated hydrogels for advanced biocatalysis. In this review, we first systematically discuss commonly employed physical strategiesincluding self-assembly, electrostatic adsorption, and direct encapsulationfor constructing enzyme-integrated hydrogel systems. Subsequently, we elaborate on enzyme-mediated self-confined polymerization and cross-linking strategies, encompassing natural enzymes, enzyme cascades, nanoz

Multifunctional hydrogels with integrated antioxidant and antibacterial activities are vital for modulating the wound microenvironment, mitigating oxidative stress, and preventing infection. However, efficient construction of such hydrogels with programmable responsiveness and structural integrity that can be tailored to therapeutic demands remains challenging. Herein, we developed a modular, functionally programmable hydrogel platform constructed via an in situ Schiff base reaction between aldehyde-functionalized polyphenolic nanoparticles and aminoglycoside antibiotics. The resulting nanocom

Granular hydrogels composed of hydrogel microparticles are promising candidates for 3D bioprinting due to their ability to protect encapsulated cells. However, to achieve high print fidelity, hydrogel microparticles need to jam to exhibit shear-thinning characteristics, which is crucial for 3D printing. Unfortunately, this overpacking can significantly impact cell viability, thereby negating the primary advantage of using hydrogel microparticles to shield cells from shear forces. To overcome this challenge, a novel solution: a biphasic, granular colloidal bioink designed to optimize cell viabi

Folding of mucosal tissues, such as the tissue within the epithelium of the upper respiratory airways, is critical for organ function. Studying the influence of folded tissue patterns on cellular function is challenging mainly due to the lack of suitable cell culture platforms that can recreate dynamic tissue folding in vitro. Here, a bilayer hydrogel folding system, composed of alginate/polyacrylamide double-network (DN) and hyaluronic acid (HA) hydrogels, to generate static folding patterns based on mechanical instabilities, is described. By encapsulating human fibroblasts into patterned HA

Hydrogels are important platform materials for in vitro cellular studies. Mechanistic studies on durotaxis, the directional movement of a cell affected by a spatial gradient of stiffness of the underlying substrate, requires materials such as polyacrylamide, polyethylene glycol, or PDMS, in which the stiffness can be controlled in a spatiotemporal manner. Here, we describe the synthesis of an o-nitrobenzyl-based photocleavable cross-linker and its incorporation into a polyacrylamide hydrogel to render it photoresponsive. Precise control over the physical properties of the gel allows observatio

Wound infections slow down the healing process and lead to complications such as septicemia, osteomyelitis, and even death. Although traditional methods relying on antibiotics are effective in controlling infection, they have led to the emergence of antibiotic-resistant bacteria. Hydrogels with antimicrobial function become a viable option for reducing bacterial colonization and infection while also accelerating healing processes. Chitosan is extensively developed as antibacterial wound dressings due to its unique biochemical properties and inherent antibacterial activity. In this review, the

Emulsion hydrogels are structurally composite materials combining the advantages of emulsions and hydrogels with the ability to accommodate hydrophobic and hydrophilic components in one system. It is a promising strategy for the excellent encapsulation and delivery of many bioactive ingredients. In this work, the thyme oil-loaded zwitterionic emulsion hydrogels are constructed by the cellulose acetoacetate-horseradish peroxidase-hydrogen peroxide-initiated (CAA-HRP-H2O2-initiated) ternary enzyme-mediated polymerization of the thyme oil-in-water (O/W) emulsions stabilized by cellulose acetoacet

Inert hydrogels are of a great importance in burn first aid. Hydrogel dressings may be an alternative to cooling burn wounds with streaming water, especially in cases of mass casualty events, lack of clean water, hypothermia, or large extent of burns. Hydrogels that contain mostly water evacuate the heat cumulating in the skin by evaporation. They not only cool the burn wound, but also reduce pain and protect the wound area from contamination and further injuries. Hydrogels are ideally used during the first hours after injury, but as they do not have antimicrobial properties per se, they might

Lipid-based boundary layers formed on liposome-containing hydrogels can facilitate lubrication. However, these boundary layers can be damaged by shear, resulting in decreased lubrication. Here, a shear-responsive boundary-lubricated drug-loaded hydrogel is created by incorporating celecoxib (CLX)-loaded liposomes within dynamic covalent bond-based hyaluronic acid (HA) hydrogels (CLX@Lipo@HA-gel). The dynamic cross-linked network enables the hydrogel to get restructured in response to shear, and the HA matrix allows the accumulation of internal liposome microreservoirs on the sliding surfaces,

Recently, four-dimensional (4D) printing is emerging as the next-generation biofabrication technology. However, current 4D bioprinting lacks biocompatibility or multi-component printability. In addition, suitable implantable targets capable of applying 4D bioprinted products have not yet been established, except theoretical and in vitro study. Herein, we describe a cell-friendly and biocompatible 4D bioprinting system including more than two cell types based on digital light processing (DLP) and photocurable silk fibroin (Sil-MA) hydrogel. The shape changes of 3D printed bilayered Sil-MA hydro

We report an elastase-responsive, H2S-releasing hydrogel prepared by covalently crosslinking a mixture of carboxymethylcellulose and poly(ethylene glycol) with an elastase-degradable peptide functionalized with an H2S-releasing S-aroylthiooxime (SATO) unit. Addition of elastase triggered a gel-to-sol transition, which exposed SATOs, leading to more and longer H2S release compared to untriggered gels.

Soft tissue engineering requires antifouling materials that are biocompatible and mechanically flexible. Conventional hydrogels containing more than 70 wt% water are thus promising antifouling material candidates. However, some hydrogels are difficult to apply in internal body organs because of undesirable protein absorption on their surfaces. Due to the lack of an effective method for observing the true charge densities of hydrogels, the reason why electrostatic interactions dominate protein absorption behavior remains unclear. In this work, we adopt the microelectrode technique (MET) to stud

Since its first clinical application, methotrexate (MTX) has been widely used for the treatment of human diseases. Despite great advantages, some properties such as poor absorption, short plasma half-life and unpredictable bioavailability have led researchers to seek novel delivery systems to improve its characteristics for parenteral and oral administration. Recently, great attention has been directed to hydrogels for the preparation of MTX formulations. This review describes the potential of hydrogels for the formulation of MTX to treat cancer, rheumatoid arthritis, psoriasis and central ner
