Evaluation of antifungal volatile compounds on the basis of the elongation rate of a single hypha.

A novel method is proposed for the evaluation of the activity of an antifungal agent administered as a gas. This system is composed of a batch-flow type reaction vessel, a gas flow system, and a microscopic observation system. The agar plate was prepared on the ceiling of the reaction vessel, and the mycelium of a fungus (Aspergillus niger or Rhizoctonia solani) was inoculated onto it. After preincubation at 25 degrees C for 24 h, the reaction vessel was connected to the gas flow system. An appropriate hypha was selected, and its elongation rate was measured. Then a sample holder containing an

Evolution of the Immune Response to Chronic Airway Colonization with Aspergillus fumigatus Hyphae.

Airway colonization by the mold Aspergillus fumigatus is common in patients with underlying lung disease and is associated with chronic airway inflammation. Studies probing the inflammatory response to colonization with A. fumigatus hyphae have been hampered by the lack of a model of chronic colonization in immunocompetent mice. By infecting mice intratracheally with conidia embedded in agar beads (Af beads), we have established an in vivo model to study the natural history of airway colonization with live A. fumigatus hyphae. Histopathological examination and galactomannan assay of lung homog

Carbohydrate and lipid components of hyphae and conidia of human pathogen Fonsecaea pedrosoi.

The carbohydrate and lipid components of mycelium and conidia of Fonsecaea pedrosoi (Brumpt) were analysed by paper, thin-layer and gas-chromatography, mass spectrometry and ultraviolet spectroscopy. Glucose, mannose, galactofuranose, rhamnose and glucosamine were polysaccharide components identified in F. pedrosoi. Significant changes in the carbohydrate pattern occurred during the conversion of mycelium into conidia. Rhamnose was predominant in conidia whereas galactose was prominent in mycelium. Palmitic, stearic, oleic, linoleic, and arachidonic acids were the fatty acids identified in the

Iron starvation confers azole tolerance in Aspergillus fumigatus hyphae via mitochondrial function modulation.

Antifungal tolerance, a transient survival state distinct from genetic resistance, poses a significant challenge to antifungal therapy. However, the factors that induce antifungal tolerance and their underlying mechanisms remain poorly understood. Here, we demonstrate that iron starvation exerts a paradoxical effect on azole susceptibility in the human pathogenic filamentous fungus Aspergillus fumigatus by specifically increasing azole tolerance in hyphae while reducing the minimal inhibitory concentration against both conidia and hyphae. This hyphal tolerance could be quantified using a newly

Lacticaseibacillus Paracasei Regulates Vaginal Microecology and Inhibits Hyphae Formation for Candida Vaginitis Therapy.

Vulvovaginal candidiasis (VVC) is a prevalent global health issue that significantly impairs the quality of life for millions of women. The rising incidence of azole resistance and high recurrence rates underscore the urgent need for alternative therapeutic strategies. This study investigates a novel probiotic approach targeting the key virulence trait of Candida albicans-the yeast-to-hyphal transition-using Lacticaseibacillus paracasei CPU-Lps0708, a strain isolated from healthy human vaginal secretions. The cell-free culture supernatant (CFCS) of L. paracasei CPU-Lps0708 exhibited potent ant

Caging giants: Characterizing the molecular mechanisms of neutrophil swarming against Candida albicans hyphae.

Neutrophils utilize many mechanisms to restrict fungal growth. When phagocytosis occurs, neutrophils can create many toxic antimicrobials including reactive oxygen species and the products of myeloperoxidase. If a pathogen is too large to phagocytose, neutrophils can also resort to the release of neutrophil extracellular traps or it can engage in the behavior of "swarming," in which the recruitment and antimicrobial action of many neutrophils are coordinated against a single target. Here we optimized an assay to study the behavior of swarming directly against live Candida albicans hyphae. We f

Hydroquinones Including Tetrachlorohydroquinone Inhibit Candida albicans Biofilm Formation by Repressing Hyphae-Related Genes.

Candida albicans is an opportunistic pathogenic fungus responsible for candidiasis. The pathogen readily forms antifungal agent-resistant biofilms on implanted medical devices or human tissue. Morphologic transition from yeast to filamentous cells and subsequent biofilm formation is a key virulence factor and a prerequisite for biofilm development by C. albicans. We investigated the antibiofilm and antifungal activities of 18 hydroquinones against fluconazole-resistant C. albicans. Tetrachlorohydroquinone (TCHQ) at subinhibitory concentrations (2 to 10 μg/mL) significantly inhibited C. albican

Real-time visualization of immune cell clearance of Aspergillus fumigatus spores and hyphae.

Invasive aspergillosis (IA) is a disease of the immunocompromised host and generally caused by the opportunistic fungal pathogen Aspergillus fumigatus. While both host and fungal factors contribute to disease severity and outcome, there are fundamental features of IA development including fungal morphological transition from infectious conidia to tissue-penetrating hyphae as well as host defenses rooted in mechanisms of innate phagocyte function. Here we address recent advances in the field and use real-time in vivo imaging in the larval zebrafish to visually highlight conserved vertebrate inn

Extracellular Vesicles Regulate Biofilm Formation and Yeast-to-Hypha Differentiation in Candida albicans.

In this study, we investigated the influence of fungal extracellular vesicles (EVs) during biofilm formation and morphogenesis in Candida albicans. Using crystal violet staining and scanning electron microscopy (SEM), we demonstrated that C. albicans EVs inhibited biofilm formation in vitro. By time-lapse microscopy and SEM, we showed that C. albicans EV treatment stopped filamentation and promoted pseudohyphae formation with multiple budding sites. The ability of C. albicans EVs to regulate dimorphism was further compared to EVs isolated from different C. albicans strains, Saccharomyces cerev

Practical Diagnostic Approach to the Presence of Hyphae in Neuropathology Specimens With Three Illustrative Cases.

OBJECTIVES: Early and accurate diagnosis remains crucial in the therapeutic management of invasive central nervous system fungal infections. Different molds have intrinsic resistance to antifungal agents; thus, morphologic differentiation is helpful to clinicians. METHODS: Using three examples, we present a guide on how to approach neuropathology specimens where hyphae are identified on initial histologic examination. RESULTS: Hyphae can be classified into three basic groups: hyaline pauciseptated, hyaline septated, and pigmented or dematiaceous. The hyaline pauciseptated group includes the

Diagnostic Importance of Hyphae on Heart Valve Tissue in Histoplasma Endocarditis and Treatment With Isavuconazole.

A patient who never resided in an endemic area for dimorphic fungi was diagnosed with Histoplasma capsulatum endocarditis. His diagnosis was suggested by yeast and hyphae on cardiac valve tissue pathology. Isavuconazole was an optimal therapeutic option due to renal dysfunction and anticoagulation with warfarin for mechanical valve replacement.

Pulmonary sporotrichosis with hyphae in a human immunodeficiency virus-infected patient. A case report.

BACKGROUND: Pulmonary sporotrichosis is a rare event. Sporothrix schenckii is a dimorphic fungus and develops at 37 degrees C in yeast form. Usually hyphae are not observed in tissues, although their presence has been occasionally demonstrated in biopsies. CASE: A 37-year-old man, human immunodeficiency virus-1 positive, with a CD4 cell count of 345/mm3, developed a productive cough. A sputum smear revealed the presence of a large amount of long, thin, septated micelia. The hyphae bore oval, sessile conidia. Cultures of sputum yielded numerous colonies of S schenckii. CONCLUSION: This is the

Intrahyphal hyphae in corneal tissue from a case of keratitis due to Lasiodiplodia theobromae.

Lasiodiplodia theobromae, a recognized plant pathogen, was isolated in culture from a case of human mycotic keratitis. Chemotherapy with a variety of azoles was unsuccessful and the lesion was removed surgically. Electron microscopy of thin sections of the excised corneal tissue revealed several examples of intrahyphal hyphae, a unique process described previously in in vitro cultures of various zoopathogenic fungi. We believe this to be the first report of the presence of intrahyphal hyphae in parasitized animal or human tissue. The demonstration of this process in vivo is thought to be consi

Ultrastructural observations of an unusual osmiophilic body in the hyphae of Sporothrix schenckii and Ceratocystis stenoceras.

Electron microscopy of 8 strains of Sporothrix schenckii and 1 strain each of Ceratocystis stenoceras, C. pluriannulata, C. ulmi, and C. minor revealed the presence of unusual osmiophilic structures (EOB) which appeared as normal organellar components of young cells of these fungi. In S. schenckii and C. stenoceras, these structures were markedly osmiophilic, reacted strongly with thiocarbohydrazide, could be partially solubilized with the lipid solvent sodium methoxide, and appeared to possess lipase activity. On subsequent cellular ageing, lipid bodies were commonly seen in intimate associat

Switching off the yeast-to-hyphae transition in Yarrowia lipolytica through histone deacetylase inhibitors.

Fungi can develop a variety of morphotypes to survive, colonize, adapt and prevail under different environmental conditions. In general, two morphological shapes encompass the others: yeast (unicellular) and hyphae (multicellular). Under specific conditions, some fungi can adopt these two cellular morphologies, and for this reason, they are called dimorphic. Histone acetylation and deacetylation are well-known important mechanisms of chromatin remodelling that control cell differentiation processes as dimorphism. The reactions involved are catalysed by histone acetyltransferases (HATs) and his

Homobifunctional imidoester-modified zinc nano-spindle attenuated hyphae growth of Aspergillus against hypersensitivity responses.

Fungi cause various forms of invasive fungal disease (IFD), and fungal sensitization can contribute to the development of asthma, asthma severity, and other hypersensitivity diseases, such as atopic dermatitis (AD). In this study, we introduce a facile and controllable approach, using homobifunctional imidoester-modified zinc nano-spindle (HINS), for attenuating hyphae growth of fungi and reducing the hypersensitivity response complications in fungi-infected mice. To extend the study of the specificity and immune mechanisms, we used HINS-cultured Aspergillus extract (HI-AsE) and common agar-cu

Albumin Enhances Caspofungin Activity against Aspergillus Species by Facilitating Drug Delivery to Germinating Hyphae.

The modest in vitro activity of echinocandins against Aspergillus implies that host-related factors augment the action of these antifungal agents in vivo. We found that, in contrast to the other antifungal agents (voriconazole, amphotericin B) tested, caspofungin exhibited a profound increase in activity against various Aspergillus species under conditions of cell culture growth, as evidenced by a ≥4-fold decrease in minimum effective concentrations (MECs) (P = 0. 0005). Importantly, the enhanced activity of caspofungin against Aspergillus spp. under cell culture conditions was strictly depend

Mycorrhizal hyphae as ecological niche for highly specialized hypersymbionts - or just soil free-riders?

Mycorrhizal fungi interconnect two different kinds of environments, namely the plant roots with the surrounding soil. This widespread coexistence of plants and fungi has important consequences for plant mineral nutrition, water acquisition, carbon allocation, tolerance to abiotic and biotic stresses and interplant competition. Yet some current research indicates a number of important roles to be played by hyphae-associated microbes, in addition to the hyphae themselves, in foraging for and acquisition of soil resources and in transformation of organic carbon in the soil-plant systems. We criti

Inhibition of Yeast-to-Hypha Transition and Virulence of Candida albicans by 2-Alkylaminoquinoline Derivatives.

A rapid increase in Candida albicans infection and drug resistance has caused an emergent need for new clinical strategies against this fungal pathogen. In this study, we evaluated the inhibitory activity of a series of 2-alkylaminoquinoline derivatives against C. albicans isolates. A total of 28 compounds were assessed for their efficacy in inhibiting the yeast-to-hypha transition, which is considered one of the key virulence factors in C. albicans Several compounds showed strong activity to decrease the morphological transition and virulence of C. albicans cells.
