Advances in endogenous hypochlorous acid-mediated regulation of tumor cell fates.

Hypochlorous acid (HOCl), an important reactive oxygen species generated by myeloperoxidase (MPO), is widely involved in the pathogenesis of various conditions, including cardiovascular, renal, and neurodegenerative diseases, as well as cancer. This article reviews recent progress regarding the roles of HOCl in determining cell fate and driving disease progression. We first introduce the mechanisms of HOCl formation and its explicit mechanistic links to these pathologies. Subsequently, we summarize the oxidative modification of biological macromolecules, focusing specifically on proteins. Furt

Bactericidal, fungicidal, and antiviral activity of highly purified hypochlorous acid water.

This study demonstrates that high-purity hypochlorous acid water (HP-HAW) produced via reverse osmosis (RO) membrane filtration exhibits rapid, broad-spectrum antimicrobial and antiviral activity, comparable to that of hypochlorous acid water (HAW) prepared without RO membrane treatment. Common vegetative bacteria, dermatophytes, and Candida albicans were completely inactivated within 10 s, indicating excellent performance for routine disinfection. HP-HAW also showed moderate efficacy against Mycobacterium abscessus but limited activity against M. avium, M. kansasii, and spore-forming bacteria

A sensitive fluorescent probe for monitoring hypochlorous acid levels in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disease. Hypochlorous acid (HClO) is a signature reactive oxygen species (ROS) closely associated with the progression of RA. Here, we report a novel fluorescent probe, ZCP1, which exhibits high sensitivity to HClO. In the presence of HClO, ZCP1 demonstrates a rapid detection time of 20 s and a low detection limit of 19.1 nM, allowing for fast and sensitive reactions with HClO, with a 150-fold fluorescence enhancement. ZCP1 can be employed for fluorescent detection of both exogenous and endogenous HClO levels in live cells. Furthermore, ZCP1 ha

Visualizing hypochlorous acid production by human neutrophils with fluorescent graphene quantum dots.

In living organisms, redox reactions play a crucial role in the progression of disorders accompanied by the overproduction of reactive oxygen and reactive chlorine species, such as hydrogen peroxide and hypochlorous acid, respectively. We demonstrate that green fluorescence graphene quantum dots (GQDs) can be employed for revealing the presence of the hypochlorous acid in aqueous solutions and cellular systems. Hypochlorous acid modifies the oxygen-containing groups of the GQD, predominantly opens epoxide ring C-O-C, forms excessive C=O bonds and damages the carbonic core of GQDs. These change

Activation of Nrf2-mediated oxidative stress response in macrophages by hypochlorous acid.

Hypochlorous acid (HOCl), a potent oxidant generated when chlorine gas reacts with water, is important in the pathogenesis of many disorders. Transcription factor Nrf2-mediated antioxidant response represents a critical cellular defense mechanism that serves to maintain intracellular redox homeostasis and limit oxidative damage. In the present study, the effect of HOCl on Nrf2 activation was investigated in macrophages, one of the target cells of chlorine gas exposure. Exposure of RAW 264.7 macrophages to HOCl resulted in increased protein levels of Nrf2 in nuclear extractions, as well as a ti

Topical Hypochlorous Acid for Acne Vulgaris: Mechanisms, Clinical Evidence, and Therapeutic Potential.

Acne vulgaris is driven by Cutibacterium acnes, sebaceous activity, and innate immune activation, leading to inflammation, scarring, and reduced quality of life. Although standard topical and systemic therapies are effective, their use may be limited by irritation, adverse effects, and antimicrobial resistance. Hypochlorous acid (HOCl) exhibits antimicrobial, biofilm-disruptive, and anti-inflammatory activity through inhibition of nuclear factor kappa B (NF-κB) and mitogen-activated protein kinase (MAPK) signaling. Preliminary studies suggest HOCl provides lesion reductions comparable to benzo

Hypochlorous Acid-Gated Hydrolysis of a Phosphinate Ester Dye in Living Cells.

Theranostic fluorescent platforms are capable of the selective delivery of small molecules to target cells with simultaneous optical monitoring. Such technologies promise to significantly reduce off-target effects compared with cytotoxic chemotherapy. However, small-molecule approaches are often hindered by relatively complex designs that are required to incorporate a fluorescent reporter, reactive linker, targeting ligand, and cargo into a single molecule. Herein, we provide the first direct evidence for the ability to gate the delivery of small-molecule cargos from phosphinate ester-containi

Near-Infrared Ratiometric Fluorescent Probe for Monitoring Hypochlorous Acid in Ferroptosis Models.

Ferroptosis is an iron-dependent, regulated cell death caused by the disruption of the oxidation-reduction balance. The hypochlorous acid (HClO) levels are closely correlated with the onset and progress of ferroptosis. Hence, investigating the underlying relationships between HClO changes and ferroptosis is particularly vital. To this end, we presented a new near-infrared (NIR) ratiometric fluorescent probe (BV-HD) based on the oxidation of the phenothiazine moiety for imaging HClO variations in ferroptosis models. Employing this BV-HD, the exogenous and endogenous HClO concentrations were con

A smart hypochlorous acid fluorescent probe enabling Ibuprofen-release for osteoarthritis theranostics.

Background: Osteoarthritis (OA) standing as the most prevalent form of arthritis, closely associates with heightened levels of reactive oxygen species, particularly hypochlorous acid (HOCl). Although there are numerous probes available for detecting HOCl in the OA region, probes with dual functions of diagnostic and therapeutic capabilities are still significantly lacking. While this type of probe can reduce the time gap between diagnosis and treatment, which is clinically needed. Methods: We developed a fluorescent probe (DHU-CBA1) toward HOCl with theranostics functions through the release o

Interleukin-10 Attenuates Hypochlorous Acid-Mediated Cytotoxicity to HEI-OC1 Cochlear Cells.

Inflammatory reaction plays a crucial role in the pathophysiology of acquired hearing loss such as ototoxicity and labyrinthitis. In our earlier work, we showed the pivotal role of otic fibrocytes in cochlear inflammation and the critical involvement of proinflammatory cytokines in cisplatin ototoxicity. We also demonstrated that otic fibrocytes inhibit monocyte chemoattractant protein 1 (CCL2) upregulation in response to interleukin-10 (IL-10) via heme oxygenase 1 (HMOX1) signaling, resulting in suppression of cochlear inflammation. However, it is still unclear how IL-10 affects inflammation-

The myeloperoxidase product hypochlorous acid generates irreversible high-density lipoprotein receptor inhibitors.

OBJECTIVE: Elevated levels of advanced oxidation protein products have been described in several chronic inflammatory diseases, like chronic renal insufficiency, rheumatoid arthritis, and atherosclerosis. Recent findings revealed that advanced oxidation protein products are inhibitors of the major high-density lipoprotein receptor, scavenger receptor class B, type 1 (SR-BI). Here, we investigated which oxidation-induced structural alterations convert plasma albumin into a high-density lipoprotein-receptor inhibitor. APPROACH AND RESULTS: Exposure of albumin to the physiological oxidant, hypoc

Fluorescent probe for the detection of hypochlorous acid in water samples and cell models.

Hypochlorous acid (HClO) is a special kind of reactive oxygen species, which plays an important role in resisting pathogen invasion and maintaining cell redox balance and other physiological processes. In addition, HClO is commonly used in daily life as a bleaching and disinfectant agent. Its excessive use can also lead to death of water animals and serious respiratory and skin diseases in humans. Therefore, it is of great significance to develop a quick and convenient tool for detecting HClO in the environment and organisms. In this paper, we utilize the specific reaction of HClO with dimethy

Degradation of vincristine by myeloperoxidase and hypochlorous acid in children with acute lymphoblastic leukemia.

Vincristine (VCR) is an effective drug against acute lymphoblastic leukemia (ALL), many solid tumors, but not acute myeloid leukemia. It has been hypothesized that resistance of myeloblasts to VCR is related to myeloperoxidase (MPO) and production of hypochlorous acid (HOCl). We investigated the relationship between VCR degradation and MPO expression and serum HOCl concentrations in pediatric patients with ALL, lymphoma and solid tumors. We studied the sera from 43 children, of which 23 were newly diagnosed and as yet untreated cancer patients, 10 on chemotherapy and 10 healthy control subject

A Novel ESIPT-Based Probe for Sensitive Detection and Bioimaging of Hypochlorous Acid.

Hypochlorous acid (HOCl), a key reactive oxygen species in the innate immune system, is closely implicated in various inflammatory diseases and tissue injuries when its concentration becomes dysregulated. To enable highly selective and sensitive detection and imaging of HOCl, we developed a novel HOCl probe BCPT, engineered on an excited-state intramolecular proton transfer (ESIPT) fluorophore platform. BCPT is constructed by conjugating a 4-pyridineacetonitrile unit to a 2-(2′-hydroxyphenyl)benzothiazole (HBT) core, with its phenolic hydroxyl group protected by a highly specific dimethylthioc

Hypochlorous acid triggered fluorescent probes for in situ imaging of a psoriasis model.

Psoriasis is a common skin disease with complex pathogenesis that lacks diagnostic methods. Typically, psoriasis is an inflammation-related disease accompanied by high expression of reactive oxygen species (ROS) in the infected part. However, due to the lack of suitable tools, it is difficult to identify the ROS, especially certain types of ROS (e.g., HOCl) in the psoriasis model. Here, two HOCl-specific fluorescent probes, G1 and G2, were designed and synthesized based on oxazine 1. Both probes could react with HOCl with high selectivity among other ROS under physiological conditions. The sel
