Research progress on Hippo signaling pathway effector molecules in rheumatic immune system diseases.

The core components of the Hippo signaling pathway encompass upstream regulatory molecules, core kinase cascade complexes, and downstream transcriptional regulation complexes. This pathway modulates cellular behaviors by influencing the effector molecules of its core components and plays a pivotal role in immune regulation. Effector molecules, such as Yes-associated protein (YAP), transcriptional coactivator with PDZ-binding motif (TAZ), transcriptional enhanced associate domain transcriptional factor (TEAD), monopolar spindle-one binder (MOB1), large tumor suppressor (LATS), can stimulate fibr

[Diagnostic and prognostic value of humoral immune status indices for alimentary system diseases].

The paper describes the results of studying the immune status of 1,960 patients with stomach, pancreas, liver, gall bladder, small and large intestine disorders, who were treated in the Central Research Institute of Gastroenterology. The results of the study demonstrate that alimentary system diseases are concomitant with changes in the functional activity of the immune system and development of the systemic immune response aimed at the neutralization and elimination of pathogenic agents. Impaired regulatory and efferent lymphocyte capacities, increased synthesis of cytokines, immunoglobulins,

Therapeutic immune tolerance for central nervous system autoimmune diseases - Prospects, challenges and pitfalls.

Autoimmune diseases of the central nervous system (CNS), including multiple sclerosis (MS), neuromyelitis optica spectrum disorders (NMOSD), and myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD), are characterized by chronic inflammation driven by autoreactive immune responses against CNS antigens. Successes in the development of efficacious immunotherapies are hampered by the risks of broad immunosuppression and the consequences of compromising physiological immune functions. Antigen-specific immune tolerance offers a promising alternative by selectively silencing pathog

cGAS-STING Signaling in Central Nervous System Diseases: Neuroinflammatory Mechanisms and Immune Regulation.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway has emerged as a critical cytosolic DNA-sensing mechanism that orchestrates innate immune activation in response to cellular stress. In the central nervous system (CNS), this pathway demonstrates highly context-specific and cell-type-dependent functions, ranging from promoting neuroinflammation in neurodegenerative diseases such as Alzheimer's disease (AD) and Parkinson's disease (PD), to modulating immune surveillance and therapeutic responsiveness in glioma. This review systematically delineates the molecular mec

Metabolic regulation of the immune system in health and diseases: mechanisms and interventions.

Metabolism, including glycolysis, oxidative phosphorylation, fatty acid oxidation, and other metabolic pathways, impacts the phenotypes and functions of immune cells. The metabolic regulation of the immune system is important in the pathogenesis and progression of numerous diseases, such as cancers, autoimmune diseases and metabolic diseases. The concept of immunometabolism was introduced over a decade ago to elucidate the intricate interplay between metabolism and immunity. The definition of immunometabolism has expanded from chronic low-grade inflammation in metabolic diseases to metabolic r

Specificity of Adaptive Immune Responses in Central Nervous System Health, Aging and Diseases.

The field of neuroimmunology endorses the involvement of the adaptive immune system in central nervous system (CNS) health, disease, and aging. While immune cell trafficking into the CNS is highly regulated, small numbers of antigen-experienced lymphocytes can still enter the cerebrospinal fluid (CSF)-filled compartments for regular immune surveillance under homeostatic conditions. Meningeal lymphatics facilitate drainage of brain-derived antigens from the CSF to deep cervical lymph nodes to prime potential adaptive immune responses. During aging and CNS disorders, brain barriers and meningeal

The interplay between immune system and microbiota in gynecological diseases: a narrative review.

OBJECTIVE: The vaginal microbiome is a dynamic environment, depending on the results of a complex interplay between microbiota and the host. In physiological conditions, Lactobacillus species are the most represented, regulating glycogen metabolism in order to maintain normal pH. Vaginal flora has been divided into five subtypes. Pattern recognition receptors are present on both squamous epithelial cells lining the vagina and columnar cells lining the upper female genital tract. They respond directly to bacterial product expressed by vaginal microbiome. The vagina contains different immune rel

The Immune Status of Patients with Inflammatory Diseases of the Male Reproductive System.

Immune disorders are very important in pathogenesis of chronic prostatitis. They lead to more prolonged illness and decrease the effect of traditional therapy. 32 healthy men and 51 patients with chronic prostatitis were examined. Combined immunodeficiency was revealed in all patients. T-, B cells count in venous blood decreased, while 0-cells increased. We suggest it is connected to the disturbance of maturation and the differentiation of precursor cells. According to the results of functional tests the early E-rosette-forming cells, the subpopulations of theophylline-sensitive and theophylli

[Auto-immune diseases of the skeletal muscle-system. Immune-pathogenetic mechanisms in myasthenia gravis and polymyositis (author's transl)].

Intensive clinical as well as experimental research over the past decade in the pathogenesis of myasthenia gravis suggest the imporatance of involved immunological reactions. The thymus as well as the muscle endplates appeare to be possible target organs of autoaggressive reactions, although the real trigger mechanismus of autoimmune reactivity are not yet understood. Different pathogenetic hypothesis for myasthenia gravis are discussed. Regarding the pathogenesis of polymyositis more direct evidence for the possible involvement of viruses are offered by electronmicroscopy. Beside clinical and

The role of the immune system in Alzheimer's disease.

Alzheimer's disease (AD) is the most common neurodegenerative disorder where the accumulation of amyloid plaques and the formation of tau tangles are the prominent pathological hallmarks. Increasing preclinical and clinical studies have revealed that different components of the immune system may act as important contributors to AD etiology and pathogenesis. The recognition of misfolded Aβ and tau by immune cells can trigger a series of complex immune responses in AD, and then lead to neuroinflammation and neurodegeneration. In parallel, genome-wide association studies have also identified seve

The role of the immune system in Huntington's disease.

Huntington's disease (HD) is characterized by a progressive course of disease until death 15-20 years after the first symptoms occur and is caused by a mutation with expanded CAG repeats in the huntingtin (htt) protein. Mutant htt (mhtt) in the striatum is assumed to be the main reason for neurodegeneration. Knowledge about pathophysiology has rapidly improved discussing influences of excitotoxicity, mitochondrial damage, free radicals, and inflammatory mechanisms. Both innate and adaptive immune systems may play an important role in HD. Activation of microglia with expression of proinflammato

The innate immune system in Alzheimer's disease.

Alzheimer's disease (AD) is the leading cause for dementia in the world. It is characterized by two biochemically distinct types of protein aggregates: amyloid β (A β ) peptide in the forms of parenchymal amyloid plaques and congophilic amyloid angiopathy (CAA) and aggregated tau protein in the form of intraneuronal neurofibrillary tangles (NFT). Several risk factors have been discovered that are associated with AD. The most well-known genetic risk factor for late-onset AD is apolipoprotein E4 (ApoE4) (Potter and Wisniewski (2012), and Verghese et al. (2011)). Recently, it has been reported by

The humoral immune system in coeliac disease.

IgA is transported into intestinal secretions to perform exclusion of luminal antigens. The prerequisites are antigen sampling by the Peyer's patch M cells, antigen processing by antigen-presenting cells, and presentation of antigenic peptides by HLA class II molecules to immunocompetent T-cells. The basis for intestinal immunity is the maturation cycle of specifically primed T and B cells from the gut-associated lymphoid tissue via mesenteric lymph nodes and peripheral blood back to the intestinal lamina propria. In coeliac disease, patients are sensitized against gluten and serum gliadin ant

HTLV-1 and the host immune system: how the virus disrupts immune regulation, leading to HTLV-1 associated diseases.

Human T-cell leukemia virus type 1 (HTLV-1) was the first retrovirus shown to cause human diseases, such as adult T-cell leukemia (ATL) and HTLV-1 associated myelopathy/tropic spastic paraparesis (HAM/TSP). Despite extensive study for three decades, it remains elusive how HTLV-1 induces these diseases. HTLV-1 mainly infects CD4 T cells, inducing dysregulation of the host immune system. Recent studies have uncovered the mechanisms of differentiation and function of CD4 T cells at the cellular and molecular levels, extending our understanding of the pathological conditions associated with HTLV-1

The Innate Immune System in Cardiovascular Diseases and Its Role in Doxorubicin-Induced Cardiotoxicity.

Innate immune cells are the early responders to infection and tissue damage. They play a critical role in the initiation and resolution of inflammation in response to insult as well as tissue repair. Following ischemic or non-ischemic cardiac injury, a strong inflammatory response plays a critical role in the removal of cell debris and tissue remodeling. However, persistent inflammation could be detrimental to the heart. Studies suggest that cardiac inflammation and tissue repair needs to be tightly regulated such that the timely resolution of the inflammation may prevent adverse cardiac damag
