Results obtained with the immunoblot technique and indirect immunofluorescence on the specific circulating autoantibodies in 20 patients with bullous pemphigoid and in 15 patients with pemphigus vulgaris were compared. In 17 of the 20 patients with bullous pemphigoid, autoantibodies against the 230 kD major antigen could be demonstrated, and in 9 of these 17 patients more specific autoantibodies against "salt-split" epidermis antigen (220, 180, 130, 100 and 75 kD) were shown. Mitochondrial antigens were purified from rat liver and characterized by immunoblotting. Sera from 19 well defined patients with primary biliary cirrhosis (PBC) reacted with two mitochondrial polypeptides of 68 Kd and 45 Kd. We have found that kallikrein-like proteins differ in their isoelectric point but share antigenic determinants. For identification of kallikrein-like proteins an initial separation was carried out in flat-bed isoelectrofocusing gels. The kallikrein-like nature was demonstrated by an immunological similarity to kallikrein-like proteins by immunoblotting using antiserum against a kallikrein family member for staining. Partially purified but unfixed C. burnetii cells were submitted to sodium dodecylsulfate polyacrylamide gel electrophoresis, Western blotted and used as antigens for evaluation of antibody responses against the agent in mice, cattle and humans. Increased specificity was achieved by using glutaraldehyde as additional blocking agent. Gels as well as membranes were subjected to laser densitometry and data analysed with an IBM personal computer. This technique offers the possibility to objectively quantify and document the results of immunoblotting.

Four antigen preparations from Rhizopus arrhizus were made and analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and column chromatography.
