Use of animal testing for evaluating glycoconjugate vaccine immunogenicity.

Most animal species respond with high antibody levels to polysaccharide antigens after they have been covalently linked to a protein carrier, converting a T-cell independent to a T-cell dependent antigen. This chemical modification has enabled the development of glycoconjugate vaccines for Haemophilus influenzae type b, Neisseria meningitidis, and multivalent Streptococcus pneumoniae. This new generation of vaccines can be well characterized physically and chemically to ensure consistent vaccine manufacture. Such analytical tests provide an alternative to animal models for Quality Control Labo

Immunogenicity and feasibility of purified chick embryo cell vaccine.

Two hundred seventy-one children reported at the WHO Collaborative Centre for Rabies Epidemiology for South-East Asia at National Institute of Communicable Diseases, Delhi, for advice and antirabies antibody assessment after post-exposure prophylaxis with purified chick embryo cell (PCEC) antirabies vaccine from January 1986 to October 1992. Vaccine was very well tolerated by these children and only 7% complained about mild to moderate side reactions like pain, induration, fever or rash. On an average, every year 50-55 children had reported at this Centre after post-exposure vaccination with P

Immunogenicity of hepatitis B vaccine in renal transplant recipients.

To evaluate the immunogenicity of hepatitis B vaccine in renal transplant recipients, we administered three 40-microgram doses of vaccine to 17 patients who had previously undergone transplantation and were on immunosuppressive therapy. Life-table analysis revealed a cumulative antibody response rate of only 17.6% at 12 months, and the three responders had low titers of antibody to hepatitis B surface antigen. There were no serious adverse effects and no episodes of graft rejection in responders or nonresponders. In addition, the ratio of helper/inducer (T4) to suppressor/cytotoxic (T8) T cell

The immunogenicity and safety of adenoviral-based vaccines.

PURPOSE OF REVIEW: Human adenoviruses (HAdVs) have shown promise as versatile and effective delivery vectors for vaccine development. This was highlighted during the COVID-19 pandemic, which demonstrated the importance of effective vaccines but also provided scope to explore potential limitations of current viral-vector based strategies. This review summarizes the current applications of adenoviral vectors, in addition to discussing the immunological mechanisms underpinning immunogenicity and safety of viral vector-based vaccines. RECENT FINDINGS: Clinical trials involving HAdVs have been und

Mucoadhesive Antioxidant Nanoadjuvant Delivers Enhanced Intranasal Influenza Vaccine Immunogenicity.

Influenza viruses remain a persistent and significant threat to global public health. However, currently available whole inactivated virus (WIV) vaccines fail to elicit robust mucosal immune responses, weakening the first line of defense against respiratory viral infection. Intranasal vaccination represents a promising strategy for achieving both mucosal and systemic immunity against influenza, but safe and highly immunostimulatory adjuvants are required for good efficacy. Here, we developed a multifunctional nanocomposite adjuvant, GO-IONPs-PEI (GIP), which was designed to enhance immunogenic

Nanoparticle Delivery of Alu RNA Adjuvants Enhances Vaccine Immunogenicity.

Vaccine adjuvants stimulate innate immunity to enhance and shape adaptive immune responses. However, approved adjuvants typically elicit weak CD8+ T cell responses to protein- and peptide-based vaccines, motivating an investigation into the discovery and testing of new adjuvants. Unedited forms of endogenous Alu RNAs are sensed by pattern recognition receptors (PRRs) to trigger sterile inflammation, and therefore we hypothesized that synthetic Alu RNA molecules could be harnessed as vaccine adjuvants. To enhance their intracellular delivery, Alu RNA was copackaged with a model antigen into pol

Immunogenicity of varicella zoster vaccine in pediatric liver transplantation.

BACKGROUND: Pediatric liver transplant (LT) candidates often miss complete varicella-zoster virus (VZV) vaccination before LT. We aimed to evaluate the immunogenicity of two doses of VZV vaccines in pediatric LT candidates younger than 2 years and persistence of its immunogenicity after LT. METHODS: Patients aged 9-24 months were enrolled before LT. The first dose of VZV vaccine was given at 9 months, and the second dose was given at between 1 to 3 months later, and at least 4 weeks before LT. Varicella-zoster IgG (VZG) was used to detect immunoglobulin G antibodies to VZV and was reported as

Scalable production and immunogenicity of a cholera conjugate vaccine.

There is a need to develop cholera vaccines that are protective in young children under 5 years of age, which induce long-term immunity, and which can be incorporated into the Expanded Programme of Immunization (EPI) in cholera-endemic countries. The degree of protection afforded by currently available oral cholera vaccines (OCV) to young children is significantly lower than that induced by vaccination of older vaccine recipients. Immune responses that protect against cholera target the O-specific polysaccharide (OSP) of Vibrio cholerae, and young children have poor immunological responses to

Immunogenicity of a DNA vaccine candidate for COVID-19.

The coronavirus family member, SARS-CoV-2 has been identified as the causal agent for the pandemic viral pneumonia disease, COVID-19. At this time, no vaccine is available to control further dissemination of the disease. We have previously engineered a synthetic DNA vaccine targeting the MERS coronavirus Spike (S) protein, the major surface antigen of coronaviruses, which is currently in clinical study. Here we build on this prior experience to generate a synthetic DNA-based vaccine candidate targeting SARS-CoV-2 S protein. The engineered construct, INO-4800, results in robust expression of th

Immunogenicity of varicella zoster virus glycoprotein E DNA vaccine.

In the present study a eukaryotic expression vector of varicella zoster virus (VZV) glycoprotein E (gE) was constructed and enabled to express in COS7 cells. Furthermore, a specific immune response against the VZV gE eukaryotic expression plasmid was induced in BALB/c mice. The VZV gE gene was amplified using polymerase chain reaction (PCR) and cloned into a eukaryotic expression vector, pcDNA3.1. The recombinant vector was subsequently transfected into COS7 cells using a liposome transfection reagent. The recombinant protein was instantaneously expressed by the transfected cells, as detected

Safety and immunogenicity of a candidate parvovirus B19 vaccine.

Parvovirus B19 is an important human pathogen causing erythema infectiosum, transient aplastic crisis in individuals with underlying hemolytic disorders and hydropsfetalis. We therefore evaluated a parvovirus B19 virus like particle (VLP) vaccine. The safety and immunogenicity of a 25 μg dose of parvovirus B19 recombinant capsid; 2.5 and 25 μg doses of the recombinant capsid given with MF59; and saline placebo were assessed in healthy adults. Because of 3 unexplained cutaneous events the study was halted after enrollment of 43 subjects and before any subject received their third scheduled dose

Immunogenicity of a recombinant hepatitis B vaccine in hemophiliacs.

Yeast-recombinant vaccines against hepatitis B virus (HBV) are now available, but there is no information about whether or not they are immunogenic in patients with hemophilia and other congenital bleeding disorders. Twenty micrograms of a recombinant vaccine expressing the adw serotype of the hepatitis B surface antigen (HBsAg) were given to 41 patients negative for HBV markers and again after 1 and 6 months. Ten percent of the vaccinees had anti-hepatitis B surface antibody (anti-HBs) responses, with titers of 10 mIU/ml or more, 1 month after the first dose of vaccine. The percentage of anti

Pneumococcal conjugate vaccine followed by pneumococcal polysaccharide vaccine; immunogenicity in patients with ataxia-telangiectasia.

The immunodeficiency in Ataxia-telangiectasia (A-T) is characterised by low T and B cell counts, low levels of IgE, IgA and/or IgG2, and especially low levels of pneumococcal antibodies. The 23-valent pneumococcal polysaccharide vaccine (PPV23) has previously been shown not to be effective in A-T, but these patients are capable of making protective antibodies to other vaccines such as diphtheria and tetanus toxin, promising effect of the seven-valent pneumococcal conjugated vaccine (PCV7). Nine A-T patients and 25 age and sex matched controls were vaccinated with both PCV7 and PPV23, and three

Immunogenicity and protective efficacy of a Streptococcus suis vaccine composed of six conserved immunogens.

A vaccine protecting against different Streptococcus suis serotypes is highly needed in porcine practice to improve animal welfare and reduce the use of antibiotics. We hypothesized that immunogens prominently recognized by convalescence sera but significantly less so by sera of susceptible piglets are putative protective antigens. Accordingly, we investigated immunogenicity and protective efficacy of a multicomponent vaccine including six main conserved immunogens, namely SSU0934, SSU1869, SSU0757, SSU1950, SSU1664 and SSU0187. Flow cytometry confirmed surface expression of all six immunogens

Ionizable lipid impact on mRNA vaccine formulation properties and immunogenicity.

The COVID-19 pandemic disrupted global supply chains, causing shortages of critical lipid-based raw materials for mRNA vaccine production. To enhance formulation resilience, we systematically evaluated the impact of five ionizable lipids (ILs) on lipid nanoparticle (LNP) organization, mRNA delivery and immunogenicity. The physiochemical analysis revealed the potential coexistence of liquid crystalline lamellar (Lα) and inverse hexagonal (HII) phases, or a mixture of nucleotide-loaded and non-loaded HII phases within developed LNP formulations. Furthermore, ILs with saturated, multi-tail struct

Redispersible nanoalum-adjuvanted hepatitis B vaccine microneedles with enhanced immunogenicity.

Nanoalum has been widely investigated as an advanced aluminum-based adjuvant; however, its performance in solid vaccine formulations remains poorly understood because administration depends on reconstitution after storage and delivery. Here, a hepatitis B surface antigen (HBsAg) vaccine was formulated with nanoalum, freeze-dried, and delivered using a powder-attached microneedle array patch (P-MAP). Formulations were screened based on redispersion in PBS after lyophilization to identify a lead composition (HBsAg: Alhydrogel: PAA: Trehalose = 1:5:5:50, w/w/w/w; F12), and the effects of preparat

ACAM2000 vaccine safety and immunogenicity considerations for smallpox and mpox.

INTRODUCTION: Smallpox, eradicated globally in 1980, remains a major biodefense concern due to retained Variola virus stocks and the absence of population immunity following cessation of routine vaccination. Monkeypox virus (MPXV), a related orthopoxvirus, has reemerged as a global public health threat, with major outbreaks in 2022 and renewed spread from 2024 onward. AREAS COVERED: This review evaluates ACAM2000®, a second-generation live smallpox vaccine, summarizing its development, immunogenicity, efficacy, safety, regulatory status, and potential role in mpox prevention. Assessment of Salmonella spp. Lipopolysaccharide immunogenicity for candidate vaccine development.

BACKGROUND: Salmonella infections impose considerable global health challenges, mostly due to the variable serovar efficacy of existing vaccines. Salmonella's lipopolysaccharide (LPS), an integral surface component that potently stimulates and activates the innate immune system, is a prime candidate for the development of universal vaccine formulations. Evaluating the immunogenic potential of LPS requires meticulous and complete molecular typing and characterization of the Salmonella subspecies. AIM: This study aimed to isolate, purify, and characterize LPS from Salmonella spp. isolates, conf

A modular vaccine platform for optimized lipid nanoparticle mRNA immunogenicity.

Certain messenger RNA antigens in mRNA vaccines elicit an insufficient immune response due to challenges in cell surface translocation (CST) of the antigens. Here we develop a modular vaccine platform (MVP) to enhance the immunogenicity of challenging mRNA antigens by optimizing antigen expression and presentation. MVPs enable the modular assembly of chimeric antigens. Our platform comprises diverse modules capable of generating >2,500 combinations with any antigen and displaying distinct antigen epitopes on the cell surface. We quantify the CST efficacy of various modules using multiple antig
