Previous efforts to use adsorption techniques to enhance the specificity of enzyme-linked immunosorbent assays for antibodies to the spirochete that causes Lyme disease resulted in unacceptable reductions in assay sensitivity. We present here evidence that preadsorption of serum with Escherichia coli can enhance test specificity without significantly reducing test sensitivity.

An indirect-enzyme immune-linked immunosorbent assay (IEA) was compared to a sandwich-enzyme-linked immunosorbent assay (SEA) to diagnose fasciolosis in naturally infected cattle. By means of a rabbit polyclonal IgG antibody to F. hepatica antigens, captured circulating antigens were detected by an SEA technique. A total of 85.1% of the samples examined was considered positive by using the IEA-test, whereas this percentage was of 37.3% by the SEA test. Using the two enzymatic probes, only the 38.3% resulted positive to both immunoassay probes. Our data indicate that circulating antigen detecti

We evaluated the ability of epitope-blocking enzyme-linked immunosorbent assays (ELISAs) to detect West Nile virus (WNV) antibodies in domestic mammals. Sera were collected from experimentally infected horses, cats, and pigs at regular intervals and screened in ELISAs and plaque reduction neutralization tests. The diagnostic efficacies of these techniques were similar.

The main steps of a sensitive and quantitative technique for the simultaneous determination, in the same microplate, of antigen, antibody and specific immune complexes has been described. The assay involved principles of the double sandwich technique used in the "Enzyme-Linked Immunosorbent Assay" (ELISA). The technique proved to be useful for the estimation of antigen, antibody and specific immune complexes in sera of patients with human chronic hepato-intestinal schistosomiasis.

The principle of extracorporal immunoadsorption (IA) is based on affinity adsorption of pathogenic (auto-)antibodies and circulating immune complexes (CIC) which reversibly bind to an immobilized ligand of the adsorber. In pemphigus, a blistering autoimmune disease affecting skin and mucous membranes, autoantibodies, mainly of the IgG subclass are directed against desmosomal adhesion molecules and other non-desmosomal antigens on the surface of epidermal keratinocytes, such as acetylcholine receptors. The pathogenicity of these autoantibodies has been shown in various in vitro and in vivo syst

Three techniques--an indirect haemagglutination test, a fluorescent antibody test and enzyme-linked immunosorbent assay (ELISA)--have been established for the detection of cysticercosis antibodies in the serum or cerebrospinal fluid of patients with cysticercosis. Results obtained using these techniques have been compared to determine the most successful serodiagnostic method. In each test there was a marked difference between the detection of active and of calcified cysts. The results obtained using the three tests were remarkably similar, and none had a statistically significant advantage ov

The aggregation of proteins into insoluble amyloid fibrils coincides with the onset of numerous diseases. An array of techniques is available to study the different stages of the amyloid aggregation process. Recently, emphasis has been placed upon the analysis of oligomeric amyloid species, which have been hypothesized to play a key role in disease progression. This paper reviews techniques utilized to study aggregation of the amyloid-β protein (Aβ) associated with Alzheimer's disease. In particular, the review focuses on techniques that provide information about the size or quantity of oligom

Acute lymphoblastic leukemia (ALL) is the most common cancer in children and is also seen in adults. Currently, no plasma-based test for the detection of ALL is available. We have cultured the home of a patient with ALL and isolated a mycovirus containing Aspergillus flavus. This culture was subjected to electron microscopy, purification, and mass spectrometry. Using enzyme-linked immunosorbent assay technique, plasma of patients with ALL and long-term survivors of this disease were tested for antibodies, utilizing supernatant of the culture of this organism. The results were compared with 3 g

A homogeneous extract of a 28-kDa cysteine protease of Fasciola gigantica adult worms was used as the antigen for immunodiagnosis of fasciolosis in cattle, sheep and goats using the Falcon assay screening test enzyme-linked immunosorbent assay technique. This antiprotease assay technique was found to be very rapid and sensitive and may be useful as a supplementary method for the diagnosis of fasciolosis in ruminants.

Wild caught rock pigeons (Columba livia) with antibodies to West Nile virus were monitored for 15 months to determine antibody persistence and compare results of three serologic techniques. Antibodies persisted for the entire study as detected by epitope-blocking enzyme-linked immunosorbent assay and plaque reduction neutralization test. Maternal antibodies in squabs derived from seropositive birds persisted for an average of 27 days.

Two established immunodiagnostic techniques, immunofluorescence and indirect haemagglutination, were compared with ELISA (enzyme-linked immunosorbent assay) using intact promastigotes as antigen for the detection of specific antibodies against Leishmania in the serum of patients with visceral or mucosal leishmaniasis from the Sudan. The ELISA was found to be more sensitive and more specific than either of the other two tests.

Autoimmune bullous dermatoses are defined by autoantibodies directed against adhesion proteins in the epidermis or basement membrane zone, resulting in blister formation on the skin and mucosa. Diagnosis depends on lesional biopsy for histopathology and perilesional biopsy for direct immunofluorescence. Additional diagnostic methods include indirect immunofluorescence, enzyme-linked immunosorbent assay, and immunoblot (Western blot), which may be selected in specific clinical scenarios due to improved sensitivity and/or specificity. This contribution reviews the available evidence supporting t

This review is an update on the subject of ocular toxocariasis. An introductory section is designed to familiarize the clinician with the epidemiology, characteristics of the organism, the life cycle in dogs and the systemic human disease known as visceral larval migrans ( VLM ). The more comprehensive second section considers the historical aspects of the ocular involvement, the clinical variations of ocular toxocariasis, pathology, differential diagnosis, diagnostic approaches, and treatment. Emphasis is placed upon new diagnostic techniques such as the enzyme-linked immunosorbent assay (ELI

An enzyme-linked immunosorbent assay (ELISA), and radioimmunoassays using antigen bound to plastic tubes or microcrystalline particles, were adapted for the indirect serological measurement of anti-schistosome antibodies. These alkaline phosphatase and 125I-labeled anti-immunoglobulin techniques were found to be highly quantitative and adaptable. Comparison showed them to be considerably more sensitive than the indirect hemagglutination and indirect fluorescent antibody techniques. Using adult S. mansoni freeze-thaw antigen, anti-schistosome antibodies were detected in sera from patients infec

The enzyme-linked immunosorbent assay and immunoblot technique were used to investigate the humoral immune response following intradermal infection of rabbits with Haemophilus ducreyi. Basically, the kinetics of IgM and IgG antibody synthesis was the same as in other bacterial infections. However, antibody titres in the enzyme-linked immunosorbent assay were remarkably low. In the immunoblot experiments, antibodies of both immunoglobulin classes were mainly directed against Haemophilus ducreyi antigens with relative molecular masses of 79,000, 62,000, 55,000, 49,000, and 26,000, suggesting tha

BACKGROUND: The potential that Strongyloides stercoralis infection has to cause major morbidity and high mortality when the disseminated form occurs in transplant patients is of particular concern.
METHODS: In this study, the objective was to observe S. stercoralis infection in patients who are candidates for transplantation by using parasitological, serological, and molecular techniques and to propose an algorithm for the detection of that infection in transplant candidates.
RESULTS: By parasitological techniques, 10% of fecal samples were positive. Anti-Strongyloides antibodies immunoglobu

Recent methods for detection of viruses in clinical specimens include immunofluorescence, immunoperoxidase, immune adherence hemagglutination, radioimmunoassay, enzyme-linked immunosorbent assay (ELISA), and immunoelectron microscopy. Some are useful for the detection of traces of viral antigens but are more complicated and time-consuming than others. Simple techniques of immunofluorescence and negative stain electron microscopy are used for the rapid detection of viruses in human adenoviral, herpetic, rubella, molluscum contagiosum, and vaccinial infections.

Four methods employed in the diagnosis of experimental porcine trichinellosis (trichinoscopy, digestion method, immunofluorescence and enzyme linked immunosorbent assay (ELISA)) were compared by eleven laboratories in the countries of the European Economic Community and Sweden. The aim of this study was to test the reliability of ELISA during the onset of T. spiralis infection. Material from conventionally raised pigs infected with 1500 to 10000 larvae was compared to uninfected controls at Day 17 and Day 21 post infection. The serological techniques gave higher percentages of positive results

Anti-endoplasmic reticulum antibody positive autoimmune hepatitis in children is characterized by the recognition of a single 50,000 MW protein of the endoplasmic reticulum in liver microsomal fractions by their sera. We have developed an enzyme-linked immunosorbent assay technique with rat liver microsomal preparations as the antigen to be used for detection of this disease. Titers obtained may be useful in following the course of the disease and as an aid in determining when therapy can be discontinued. The technique is rapid, sensitive, reproducible, and simple to perform and is easier to m

Removing β2-microglobulin (β2M) from blood circulation is considered to be the most effective method to delay the occurrence of dialysis-related amyloidosis (DRA). The ideal extracorporeal β2M removal system should be cost-effective, highly specific and having a high capacity. However, the traditional technologies based on size exclusion do not have an adequate specificity, and alternative immunosorbents have limited applications due to low capacity and their high cost. Nanobodies (Nbs), the smallest functional recombinant antibody fragments, offer several advantages to overcome these obstacle
