Abnormalities of Oocyte Maturation: Mechanisms and Implications.

The elucidation of oocyte maturation mechanisms is paramount for advancing embryo development within the scope of assisted reproductive technologies (ART). Both cytoplasmic and nuclear maturation represent intricate processes governed by tightly regulated cellular pathways, which are essential for ensuring the oocyte's competence for fertilization and subsequent embryogenesis. A comprehensive grasp of these mechanisms is vital, as the maturation stage of the oocyte significantly influences chromosomal integrity, spindle formation, and its ability to support the initial stages of embryonic deve

In vitro maturation (IVM) of human immature oocytes: is it still relevant?

In vitro maturation (IVM) of human immature oocytes has been shown to be a viable option for patients at risk of ovarian hyperstimulation syndrome (OHSS), those seeking urgent fertility preservation and in circumstances where controlled ovarian stimulation is not feasible. Moreover, IVM techniques can be combined with ovarian tissue cryobanking to increase the chances of conception in cancer survivors. The clinical applications of IVM in the field of reproductive medicine are rapidly expanding and the technique is now classified as non-experimental. In contrast to conventional IVF (in vitro fe

Prior exposure to chemotherapy does not reduce the in vitro maturation potential of oocytes obtained from ovarian cortex in cancer patients.

STUDY QUESTION: Does chemotherapy exposure affect IVM potential of immature oocytes retrieved from the ovarian cortex following ovarian tissue cryopreservation (OTC) for fertility preservation? SUMMARY ANSWER: The IVM potential of oocyte retrieved from ovarian cortex following OTC is not affected by prior exposure to chemotherapy but primarily dependent on patient's age, while successful retrieval of immature oocytes from the ovarian tissue is negatively affected by chemotherapy and its timing. WHAT IS KNOWN ALREADY: The potential and feasibility of IVM in premenarche patients was previously

Season and oxygen tension during maturation and fertilization impact the developmental competence of bovine oocytes.

Seasonal heat stress (HS) can reduce the developmental competence of bovine oocytes used for in vitro embryo production. Atmospheric oxygen tension (20% O2), commonly used for in vitro maturation and fertilization (IVM/IVF), may also influence embryo development and quality. This study investigated whether oxygen tension during IVM/IVF affects the developmental competence of bovine oocytes collected in summer and winter. Oocytes were matured and fertilized under physiological (5% O2) or atmospheric oxygen tension. Summer conditions showed a significantly higher temperature-humidity index than

Is day 7 culture necessary for in vitro fertilization of cryopreserved/warmed human oocytes?

BACKGROUND: Human embryos are usually cultured to blastocyst stage by Day 5 or 6 after insemination. However, some embryos grow slowly and reach blastocyst stage at Day 7. Acceptable live birth rates have been reported after transfer of Day 7 blastocysts resulted from fresh oocyte in vitro fertilization (IVF). It is unknown whether an extended embryo culture to Day 7 is necessary for cryopreserved oocyte IVF to obtain more transferrable blastocysts. METHODS: In this study, 455 oocytes from 57 cycles were warmed, inseminated, and the resulting embryos were cultured by Day 7 to examine blastocy

Human oocyte maturation in vitro.

Oocytes from medium-sized antral follicles have already completed their growth phase and, if released from the follicular environment and cultured in vitro, are able to resume the meiotic process and mature. However, in vitro maturation (IVM) does not entirely support all the nuclear and cytoplasmic changes that occur physiologically as an effect of the ovulatory stimulus. Regardless, oocyte IVM is widely applied for the breeding of agriculturally important species. In assisted reproduction technology, IVM has been proposed as an alternative treatment to circumvent the drawbacks of standard ov

Effects of in vitro growth culture duration and prematuration culture on maturational and developmental competences of bovine oocytes derived from early antral follicles.

Bovine ovaries offer a large pool of oocytes that could be used for in vitro production of embryos of genetically valuable animals. The effects of in vitro growth (IVG) culture duration (10, 12, and 14 days) on the viability and growth of bovine oocytes derived from early antral follicles (0.5-1 mm diameter) in this study. In addition, the effect of pre-IVM culture with phosphodiesterase inhibitor (3-isobutyl-1-methylxanthine) on nuclear maturation of IVG oocytes was also evaluated. In experiment 1, oocyte viability observed after 10 or 12 days of IVG culture was greater (P < 0.05) than that o

Retinoic Acid Action in Cumulus Cells: Implications for Oocyte Development and In Vitro Fertilization.

In the field of human in vitro fertilization (IVF), selecting the best oocyte for freezing or embryo for transfer remains an important focus of clinical practice. Although several techniques are and have been used for this goal, results have generally not been favorable and/or are invasive such that damage to some embryos occurs, resulting in a reduced number of healthy births. Therefore, the search continues for non-invasive oocyte and embryo quality markers that signal the development of high-quality embryos. Multiple studies indicate the important positive effects of retinoic acid (RA) on o

Present state and future outlook for the application of in vitro oocyte maturation in human infertility treatment.

In vitro oocyte maturation is an assisted reproductive technology in which a meiotically immature oocyte (prophase I or germinal vesicle stage) is recovered from an antral follicle and matured in vitro prior to fertilization. This technology, although in widespread use in domestic livestock, is not typically implemented during human in vitro fertilization cycles. This review examines how in vitro oocyte maturation is currently used in the clinical setting, including the various ways in vitro oocyte maturation is defined in practice. The role of in vitro oocyte maturation in patient care and th

Involvement of nitric oxide during in vitro oocyte maturation, sperm capacitation and in vitro fertilization in pig.

The importance of porcine species for meat production is undeniable. Due to the genetic, anatomical, and physiological similarities with humans, from a biomedical point of view, pig is considered an ideal animal model for the study and development of new therapies for human diseases. The in vitro production (IVP) of porcine embryos has become widespread as a result of these qualities and there is significant demand for these embryos for research purposes. However, the efficiency of porcine embryo IVP remains very low, which hinders its use as a model for research. The high degree of polyspermi

Comparison of the Developmental Potential and Clinical Results of In Vivo Matured Oocytes Cryopreserved with Different Vitrification Media.

BACKGROUND: Oocyte vitrification is widely used throughout the world, but its clinical efficacy is inconsistent and depends on the vitrification media. This study compared the developmental potential and clinical results of in vivo matured oocytes cryopreserved with different vitrification media. METHODS: This retrospective study involved vitrified-warmed oocytes at one in vitro fertilization laboratory. Vitrification media kits comprised the MC kit (ethylene glycol [EG] plus 1,2-propanediol [PROH]), the KT kit (EG plus dimethyl sulphoxide [DMSO]), and the Modified kit (EG plus DMSO and PROH

To rescue or not to rescue immature oocytes: prospects and challenges.

In vitro maturation (IVM) of immature oocytes has been explored for research and clinical purposes since the dawn of assisted reproductive technologies. Oocyte maturation is a highly specific process, on the basis of complex mutual relationships between the germ and somatic cell compartments. The complexity of this relationship has made the quest for achieving oocyte maturation in vitro arduous. In its classical form, in which intact cumulus-enclosed oocytes are collected after very mild or no ovarian stimulation, oocyte IVM is nonexperimental and has been proposed as a more friendly treatment

Control of sperm penetration using stereumamide A derived from Trichaptum fuscoviolaceum in the in vitro fertilization of pig oocytes.

Fungal metabolites are known to have potent and diverse properties such as antiviral, antidiabetic, antitumour, antioxidant, free radical scavenging, and antibacterial effects which can be utilized to treat diseases. In this study, we investigated the functional activity of stereumamide A (StA) derived from a culture broth of Trichaptum fuscoviolaceum during the in vitro fertilization (IVF) of pig oocytes, to determine its effects on sperm penetration. Oocytes matured in vitro were fertilized in the absence or presence of varying concentrations of StA (0-50 μg/ml StA). When StA was directly ad

Effects of individual or in-combination antioxidant supplementation during in vitro maturation culture on the developmental competence and quality of porcine embryos.

The oocyte maturation process requires a high supply of energy, which generates reactive oxygen species (ROS), adversely affecting oocyte and embryo development. Balancing ROS by antioxidant supplementation is essential for maintaining oocyte maturation and embryonic quality in vitro. This study aimed to evaluate the impact of four antioxidants: β-mercaptoethanol (β-ME), chlorogenic acid (CGA), curcumin and sericin, when applied individually or in combinations, during oocyte maturation on development of porcine oocytes. Cumulus-oocyte complexes were collected, cultured in maturation medium sup

Effects of pre-maturational culture duration on developmental competence of bovine small-sized oocytes.

We investigated the effects of pre-maturational (pre-IVM) culture on the developmental competence of small-sized bovine oocytes (110 and < 115 µm). Oocytes were cultured with 3-isobutyl-1-methylxanthine (IBMX) for 0, 5, or 10 h and subjected to in vitro maturation, fertilization, and culture. The cleavage rate (73%) of small-sized oocytes with 5 h pre-IVM was higher than those with 0 and 10 h pre-IVM (61 and 62%, respectively). The blastocyst rate (16%) of embryos derived from small-sized oocytes with 5 h pre-IVM was higher than those with 0 and 10 h pre-IVM (9 and 8%, respectively). In additi

Effects of storing queen ovaries in saline solution at 22 C on ovarian follicle integrity and oocyte quality and maturation.

This study evaluated the effects of storing queen ovaries in saline at 22 °C for up to 12 h. Two experiments were conducted. In the first experiment, ovaries from five queens were sectioned into four fragments; stored for 4, 8, or 12 h; and histologically analyzed to assess follicular morphology (Grades I-IV). In the second experiment, ovaries from 15 additional queens were stored under the same conditions, after which cumulus-oocyte complexes (COCs) were retrieved and graded (I-IV); only Grades I and II underwent in vitro maturation to assess cumulus expansion and meiotic status. The effects

Ovarian Tissue Oocyte-In Vitro Maturation for Fertility Preservation.

Mature oocyte vitrification is the standard of care to preserve fertility in women at risk of infertility. However, ovarian tissue cryopreservation (OTC) is still the only option to preserve fertility in women who need to start gonadotoxic treatment urgently or in prepubertal children. During ovarian cortex preparation for cryopreservation, medullar tissue is removed. Growing antral follicles reside at the border of the cortex-medullar interface of the ovary and are broken during this process, releasing their cumulus-oocyte complex (COC). By thoroughly inspecting the medium and fragmented medu

New aspect on the regulation of in vitro oocyte maturation: role of the obesity, neuropeptides and adipokines.

Oocyte quality determinants and nuclear and cytoplasmic maturation establish essential processes for fertilization and further development of the conceptus. Moreover, female fertility is strongly dependent on the metabolic status of the organism. Numerous sources indicate that obesity impairs ovarian function including oocyte physiology by inhibiting nuclear maturation, stimulating lipotoxicity and inflammation, enabling cumulus cells apoptosis, promoting reactive oxygen species formation and ultimately imposing pathogenic effects on mitochondria leading to infertility. Whereas, the number of

Transcriptome analysis of rhesus monkey failed-to-mature oocytes: deficiencies in transcriptional regulation and cytoplasmic maturation of the oocyte mRNA population.

STUDY QUESTION: Which different pathways and functions are altered in rhesus monkey oocytes that fail to mature after an ovulatory stimulus? SUMMARY ANSWER: Failed to mature (FTM) oocytes complete a large portion of the transition in transcriptome composition associated with normal maturation, but also manifest numerous differences that indicate incomplete transcriptional repression and cytoplasmic maturation affecting multiple processes. WHAT IS KNOWN ALREADY: Oocyte maturation defects contribute to unexplained female infertility. Failure of some oocytes to undergo germinal vesicle breakdow
