Finding a suitable rootstock for Cucumis sativus L. is an important area of research, as it is aimed at extending the fruiting period and increasing the yield. In such studies, it is important to evaluate various biochemical parameters in addition to morphometric data, such as amino acid and auxin content, which will reflect the effectiveness of the rootstock for scion growth. In this research, we tested 56 genotypes and lines from four species of the Cucurbitaceae family as potential candidates for grafting cucumber rootstock: Cucurbita ficifolia Bouché, Cucurbita moschata L., Cucurbita pepo

Gut microbiome dysbiosis has been implicated in the pathogenesis of systemic lupus erythematosus (SLE). However, microbiota-targeted therapeutic strategies have been lacking. Here, we report the potential of Faecalibacterium prausnitzii (strain UT1) to ameliorate gut dysbiosis and alleviate disease progression in the B6.Sle1.Yaa male mouse model of SLE. Fecal metagenomes of patients with SLE shifted carbohydrate catabolism from dietary fibers to host glycans, coinciding with depletion of F. prausnitzii. Oral administration of UT1 partially reversed lupus-associated microbiome alterations and r

The use of seaweed or macroalgae as plant biostimulants has increased in recent years. This study discusses the effect of seasonal sea temperatures on the phytohormone and flavonoid contents, and the antioxidant activity of Acanthophora spicifera and Ulva intestinalis from Ecuador. For the extraction, aqueous digestion (AD), ethanol digestion (ED), ethanol sonication (ES), and methanol digestion (MD) were used. Metabolites and antioxidant activity were quantified by liquid chromatography and spectrophotometry. Multivariate analysis revealed that U. intestinalis exhibited enhanced flavonoid (up

Tryptophan hydroxylase (Tph), the rate-limiting enzyme of serotonin (5-hydroxytryptophan; 5-HT) synthesis, exists in two isoforms, Tph1 and Tph2. Tph1 upregulates peripheral 5-HT synthesis and blood 5-HT levels, whereas Tph2 upregulates brain 5-HT synthesis. Here, we show that plasma and brain levels of tryptophan and its metabolites, including 5-HT, 5-hydroxy indoleacetic acid, indolepropionic acid, indole-3-acetic acid, kynurenine, and xanthurenic acid, are decreased in young (8-week-old) Tph1-mutant mice compared with age-matched wild-type mice. In older (7-month-old) Tph1-mutants, the decr

MiRNAs play an important role in regulating plant growth and immune response. Mosaic diseases are recognized as the most important plant diseases in the world, and mosaic symptoms are recovery tissues formed by plants against virus infection. However, the mechanism of the formation of mosaic symptoms remains elusive. In this study, two typical mosaic systems consisting of Nicotiana tabacum-cucumber mosaic virus (CMV) and N. tabacum-tobacco mosaic virus (TMV) were used to investigate the relevance of miRNAs to the appearance of mosaic symptoms. The results of miRNA-seq showed that there were si

Cortisol concentrations in CSF were measured by radioimmunoassay in healthy controls, depressed patients, patients who had attempted suicide but were not depressed, and obsessive-compulsive patients. The factors that contributed most to the variance in CSF cortisol levels were a diagnosis of depression, height, and important life changes during the six months preceding the investigation. Depression was by far the most important factor. The depressed patients had significantly higher CSF cortisol levels than the controls. In obsessive-compulsive and depressed patients treated with clomipramine

The effect of two serotonin precursors, 5-hydroxytryptophan (5-OH-TRP) and indolepyruvic acid (IPA), a tryptophan ketoanalogue, on rat pineal indole metabolism during the light-dark cycle was investigated. 5-OH-TRP drastically increased the production of 5-hydroxyindoleacetic acid at a dose of only 10 mg/kg, whereas 50-100 mg/kg was needed to reach higher serotonin levels. It had no effect on the pathway leading to the production of N-acetylserotonin and melatonin. IPA, on the other hand, led to a marked dose-related increase in tryptophan, 5-OH-TRP, serotonin and 5-OH-indoleacetic acid, and w

Targeted protein degradation is a favorable strategy for studying the immediate downstream effects of protein loss-of-function, such as the auxin-inducible degron (AID) system. Although this system has been applied extensively to cell and animal models, degradation resistance to long-term auxin treatment has not been studied. With the advent of the new AID2 system, cellular toxicity caused by the high auxin concentrations required in the original AID1 system is no longer a concern, enabling the study of protein degradation over extended periods. In this study, we derived multiple miniAID-tagge

In the present study, the effects of long-term pinealectomy on tryptophan, 5-hydroxytryptamine (5-HT or serotonin), 5-hydroxy-3-indoleacetic acid (5-HIAA), and tryptophan hydroxylase and monoamine oxidase activities were studied in preoptic area-anterior hypothalamus (POA-AH) and in the medial and posterior hypothalamus of the rat. After pinealectomy, 5-HT levels decreased significantly in medial hypothalamus but increased in the POA-AH. The levels of 5-HIAA decreased significantly in the POA-AH and medial hypothalamus. Tryptophan levels remained unchanged while tryptophan hydroxylase activity

The involvement of excitatory amino acids (EAA) in the pathogenesis of hypoxic-ischemic states is well-documented. Information on the role of overexcitation by EAA in perinatalasphyxia (PA), however, is limited and data from adult models cannot be directly extrapolated to immature systems. Moreover, most adult models of ischemia are representing stroke rather than PA. We decided to study long term effects in a non-invasive rat model of PA resembling the clinical situation three months following the asphyctic insult. Morphometry on Nissl - stained sections was used to determine neuronal death i

While Alismatis Rhizoma polysaccharides demonstrated promising anti-obesity potential, several research gaps remain, including extraction methods, chemical composition, and gut microbiota interactions. Herein, a new method termed the salt concentration saturation Oil Red O staining method (SCS-ORO) was developed to screen 7 salts capable of forming biphasic systems with ethanol from a pool of 28 salts, of which the K2HPO4/ethanol system was suitable for extracting A. Rhizoma crude polysaccharides (ARP). Meanwhile, ARP obtained from the K2HPO4/ethanol system alleviated lipid accumulation in Cae

INTRODUCTION: Influenza A virus (IAV) remains a major respiratory pathogen that causes substantial morbidity, mortality, and socioeconomic burden worldwide. Although several antiviral agents are available for clinical use, their effectiveness is often compromised by the emergence of drug-resistant variants, limited therapeutic windows, and potential adverse effects. Small-molecule antivirals represent an important strategy for influenza treatment; however, improving antiviral efficacy while minimizing toxicity remains a major challenge in drug development. In this study, we sought to identify

Inflammatory bowel disease (IBD) remains an incurable condition, often accompanied by high rates of anxiety and depression, further diminishing the quality of life of patients. Endogenous gasotransmitters, such as carbon monoxide (CO) and hydrogen sulfide (H₂S), exhibit potent anti-inflammatory and immunomodulatory effects. However, their therapeutic application is limited by challenges in targeted delivery to affected tissues. Here, we propose a novel strategy for targeted gut delivery of CO/H2S through engineering Escherichia coli Nissle 1917 (EcN) with CO/H2S-releasing copolymer (POSR) load

Analysis of the interactions between two representatives of plant hormones: synthetic (1-naphthaleneacetic acid, NAA) as well as natural (indole-3-acetic acid, IAA) and phospholipids occurring in biological membrane of both plant and animal cells was the subject of present studies. The aim of undertaken experiments was to elucidate the problem of direct influence of these plant growth regulators on phosphatidylcholines (PCs) and phosphatidylethanolamines (PEs) in monolayers at the air/water solution interface. The studied phospholipids differ not only as regards the structure of polar head-gro

INTRODUCTION: Alcoholic-associated liver diseases (ALD) are now widespread issues worldwide. Alcoholic-induced chronic dysbiosis of the gut microbiota is one of the factors in the pathophysiology of ALD.
METHODS: In this work, we employed a chronic-binge ethanol feeding mice model, as described in a previous report.
RESULTS: Our findings demonstrate that hepatic inflammatory injury damage and accumulation of fat can be effectively reduced in mice with ALD by altering the gut microbiota utilizing Bacillus coagulans. Treatment with B. coagulans significantly modulates the levels of TNF-α, IL-1

Leaf elongation is a crucial factor controlling forage production and is sensitive to environmental factors including CO2 concentration ([CO2]) and nitrogen (N). The objectives of this study were to examine the effects of elevated [CO2] on leaf elongation and clarify the underlying regulatory mechanism related to auxin metabolism in tall fescue (Festuca arundinacea. cv. 'Barlexas') under different N levels. Plants treated with low N level (0.25 mM) and moderate N level (4 mM) were exposed to ambient [CO2] (400 μmol⋅mol-1) or elevated [CO2] (800 μmol⋅mol-1) in environment-controlled growth cham

Strigolactones are plant hormones that regulate the development of different plant parts. In the shoot, they regulate axillary bud outgrowth and in the root, root architecture and root-hair length and density. Strigolactones are also involved with communication in the rhizosphere, including enhancement of hyphal branching of arbuscular mycorrhizal fungi. Here we present the role and activity of strigolactones under conditions of phosphate deprivation. Under these conditions, their levels of biosynthesis and exudation increase, leading to changes in shoot and root development. At least for the

The interactions of Fe3+ with some ligands (Tranexamic (TXA-), Indole-3-acetic (IAA-), and Aminomethylphosphonic (AMPA2-) acids) of biological and environmental interest were studied. The speciation studies were performed in NaNO3(aq) and NaCl(aq) using potentiometric and, only for IAA-, spectrophotometric titrations at T = 298.15 K and 0.01 ≤ I/mol dm-3 ≤ 1.0. The proposed speciation models are as follows: Fe(TXA)H3+, Fe(TXA)2+, Fe(TXA)(OH)+, and Fe(TXA)(OH)2(aq) for TXA-; Fe(IAA)2+ for IAA-; and Fe(AMPA)H23+, Fe(AMPA)H2+, and Fe(AMPA)+ for AMPA2-.
