H9N2 and H5N2 avian influenza A viruses (IAVs) are two of the most widespread and economically damaging subtypes affecting the poultry industry. Controlling these viruses in poultry remains challenging, highlighting the need for a vaccine that can confer broad and effective protection across multiple avian IAV subtypes. Previously, we fused two copies of the matrix 2 ectodomain (M2e) of human and avian IAV (2xhuM2e-2xavM2e) at the C-terminus of Macrobrachium rosenbergii nodavirus capsid protein (NvC). This chimeric protein (NvC-2xhuM2e-2xavM2e) self-assembled into virus-like particles (VLPs),

The first detection of a Highly Pathogenic Avian Influenza (HPAI) H5N8 virus in Bulgaria dates back to December 2016. Since then, many outbreaks caused by HPAI H5 viruses from clade 2.3.4.4B have been reported in both domestic and wild birds in different regions of the country. In this study, we characterized the complete genome of sixteen H5 viruses collected in Bulgaria between 2019 and 2021. Phylogenetic analyses revealed a persistent circulation of the H5N8 strain for four consecutive years (December 2016-June 2020) and the emergence in 2020 of a novel reassortant H5N2 subtype, likely in a

Between December 2014 and June 2015, an outbreak of H5N2 HPAI caused the largest and most expensive agriculture emergency in U.S. Department of Agriculture-Animal and Plant Health Inspection Service history. The outbreak affected 21 states; 232 poultry farms (211 commercial and 21 backyard) were affected, and approximately 49.6 million birds were depopulated on poultry farms. The majority of affected farms were commercial turkey operations (n = 160). This report is a case series describing 104 H5N2 HPAI-affected turkey farms in Iowa, Minnesota, Missouri, North Dakota, South Dakota, and Wiscons

100 years after the infamous "Spanish flu" pandemic, the 2017-2018 flu season has been severe, with numerous infections worldwide. In between, there have been continuous, relentless attacks from (re-)emerging viruses. To fully understand viral pathogenesis and develop effective medical countermeasures, we must strengthen current surveillance and basic research efforts.

UNLABELLED: During 2014, a subclade 2.3.4.4 highly pathogenic avian influenza (HPAI) A(H5N8) virus caused poultry outbreaks around the world. In late 2014/early 2015, the virus was detected in wild birds in Canada and the United States, and these viruses also gave rise to reassortant progeny, composed of viral RNA segments (vRNAs) from both Eurasian and North American lineages. In particular, viruses were found with N1, N2, and N8 neuraminidase vRNAs, and these are collectively referred to as H5Nx viruses. In the United States, more than 48 million domestic birds have been affected. Here we pr

BACKGROUND: Rapid identification and subtype determination of influenza virus is important in managing infected patients. Rapid influenza diagnostic tests (RIDTs) are widely used in this manner, but most can only detect influenza A and B viruses without subtyping. A new RIDT, GENEDIA Multi Influenza Ag Rapid Test (GENEDIA), was developed for detection of influenza A and B viruses and also subtyping of influenza A to H1, H3, H5 which has not been possible with other RIDTs.
OBJECTIVES: Assess the performance of GENEDIA.
STUDY DESIGN: Nasopharyngeal swabs were collected from 274 clinically susp

BACKGROUND: In the fall of 2014, highly pathogenic avian influenza (HPAI) subtype H5N8 clade 2.3.4.4 was introduced into North America by migrating waterfowl from Asia where, through reassortment, novel HPAI H5N2 and H5N1 viruses emerged.
OBJECTIVES: Assess the susceptibility of pigs to HPAI H5N1, H5N2, and H5N8 clade 2.3.3.3 from North America.
METHODS: Pigs and trachea explants were inoculated with a representative panel of H5NX clade 2.3.4.4 HPAI viruses from North America. Nasal swabs, BALF, and sera were collected to assess replication and transmission in challenged and direct contact p

Low pathogenic avian influenza (LPAI) viruses are a source of sporadic human infections and could also contribute to future pandemic outbreaks but little is known about inter-species differences in the host responses to these viruses. Here, we studied host gene expression signatures of cell lines from three species (human, chicken, and canine) in response to six different viruses (H1N1/WSN, H5N2/F59, H5N2/F118, H5N2/F189, H5N3 and H9N2). Comprehensive microarray probe set re-annotation and ortholog mapping of the host genes was necessary to allow comparison over extended functionally annotated

A combination of in vitro and in vivo selection procedures was used to examine the possibility that certain mildly pathogenic field isolates of avian influenza (AI) virus may contain minority subpopulations of highly pathogenic virus. Two mildly pathogenic H5N2 isolates, A/chicken/New Jersey/12508/86 (NJ12508) and A/chicken/Florida/27716/86 (FL27716), recovered from chickens epidemiologically associated with urban live-bird markets, were cloned in trypsin-free chicken embryo fibroblast cultures. Selected clones were inoculated intranasally and intratracheally (IN/IT) into specific-pathogen-fre

The influenza A virus (A/Chick/Pennsylvania/83 (H5N2) that caused up to 80% mortality in chickens provided a model system for testing the efficacy of chemotherapeutic agents against highly virulent influenza virus. Amantadine and rimantadine administered to chickens in drinking water were efficacious both prophylactically and therapeutically. However, under conditions simulating natural transmission of virus amantadine and rimantadine-resistant viruses arose and were transmitted to contact birds, causing mortality. Simultaneous administration of inactivated H5N2 vaccine and amantadine provided

Avian influenza (AI) currently poses a serious problem for poultry farming worldwide. Its prevalence in Mexico, despite vaccination, has highlighted the need for new approaches to control AI and reduce the economic losses associated with its occurrence in susceptible birds. The different interactions between AI viruses (AIV) and cellular receptors have been described, along with the affinity of some viruses for certain types of species-specific receptors. This receptor-ligand specificity, combined with an understanding of viral interference processes and their relevance in different viral mode

BACKGROUND: The emergence and spread of highly pathogenic avian influenza (H5N1) viruses have raised global concerns of a possible human pandemic, spurring efforts towards H5N1 influenza vaccine development and improvements in vaccine administration methods. We previously showed that a prime-boost vaccination strategy induces robust and broadly cross-reactive antibody responses against the hemagglutinin globular head domain. Here, we specifically measure antibodies against the conserved hemagglutinin stem region in serum samples obtained from the prior study to determine whether stalk-reactive

An avian influenza outbreak in 2002 affected 197 poultry farms in Virginia and cost an estimated $130 million in losses and cleanup. In 2004-2005, researchers initiated a project to investigate the feasibility and practicality of in-house composting of turkey mortalities (heavy hens and toms) as a method of disposal and disease containment. Occurrences of low pathogenic avian influenza (LPAI) in West Virginia and Virginia in 2007 provided an opportunity for first responders to verify composting as an effective carcass disposal method. Many lessons learned from these experiences have led to imp

Outbreaks of avian influenza virus (AIV) cause substantial economic losses in the poultry industry, highlighting the need for sensitive, reliable and field-adaptable detection platforms. In this study, we report a signal-on electrochemical aptasensor for avian influenza virus H5N2(K08-404) by translating a previously reported J3/JH4 cognate aptamer pair into an electrode-confined sandwich sensing format. The simultaneous binding capability of this aptamer pair enabled target-mediated recruitment of the HRP-labeled reporter aptamer and generated a signal-on amperometric response. This signal-on

The development of a universal influenza vaccine that provides broad cross protection against existing and unforeseen influenza viruses is a critical challenge. In this study, we constructed and expressed conserved sM2 and HA2 influenza antigens with cholera toxin subunit A1 (CTA1) on the surface of Lactobacillus casei (pgsA-CTA1sM2HA2/L. casei). Oral and nasal administrations of recombinant L. casei into mice resulted in high levels of serum immunoglobulin G (IgG) and their isotypes (IgG1 & IgG2a) as well as mucosal IgA. The mucosal administration of pgsA-CTA1sM2HA2/L. casei may also signific

In November 2010, an outbreak of avian influenza (AI) due to the H5N2 subtype virus occurred in a turkey breeder farm in northern Manitoba, Canada. The only clinical signs observed were depression, decrease in food consumption, and loss of egg production. The hemagglutinin (HA) cleavage (HA(0)) site of the isolated H5N2 virus was PQRETR/GLF, consistent with low pathogenic AI viruses. The intravenous pathogenicity index of this virus was zero. Whole-genome sequencing of two isolates that originated from two different barns was performed, and both isolates had 100% identical protein sequence in

Infection of backyard and poultry with low pathogenicity avian influenza LPAI A(H5N2) viruses has occurred in Mexico since 1994, and the first human infection caused by this influenza virus was detected in 2024. Since its emergence in the Americas, frequent reassortments between high pathogenicity avian influenza HPAI A(H5N1) and LPAI viruses has occurred. In September 2025, the Instituto Nacional de Enfermedades Respiratorias of Mexico City identified an unsubtypeable influenza A virus infection in a young adult patient later determined to be a reassortant HPAI (H5N2) virus with a clade 2.3.4

Epimedium koreanum Nakai has been extensively used in traditional Korean and Chinese medicine to treat a variety of diseases. Despite the plant's known immune modulatory potential and chemical make-up, scientific information on its antiviral properties and mode of action have not been completely investigated. In this study, the broad antiviral spectrum and mode of action of an aqueous extract from Epimedium koreanum Nakai was evaluated in vitro, and moreover, the protective effect against divergent influenza A subtypes was determined in BALB/c mice. An effective dose of Epimedium koreanum Naka
