Systemic positive feedback regulation of ecdysone biosynthesis via sugar metabolism in insects.

Insects are recognized as the most diverse species in the world. They are classified into three separate groups based on their developmental characteristics: ametabola, hemimetabola, and holometabola. Holometabolous insects have attracted the attention of many developmental biologists because of their dramatic metamorphosis as a key model for studying regulatory mechanisms of development. Central to this process is ecdysone, a steroid hormone that governs insect growth and maturation. Extensive research has clarified ecdysone's structure, functions, and signaling pathways. In particular, the r

Gene Expression of GqBursicon in Gynaephora qinghaiensis (Lepidoptera: Lymantriidae) and Its Impact on Wing Expansion.

The tanning hormone (Bursicon) is a heterodimer secreted by the insect nervous system, playing a crucial role in the process of cuticle darkening and hardening and wing expansion in insects. In this study, we knocked out the GqBurs gene using RNAi, thereby impairing the flight ability of adult males by affecting wing development, laying the groundwork for further control of the damage they cause to alpine meadows. The GqBurs gene was successfully characterized and expressed at all developmental stages of Gynaephora qinghaiensis, with higher expression in 5th instar larvae and wing primordia. I

The essential role of bursicon during Drosophila development.

BACKGROUND: The protective external cuticle of insects does not accommodate growth during development. To compensate for this, the insect life cycle is punctuated by a series of molts. During the molt, a new and larger cuticle is produced underneath the old cuticle. Replacement of the smaller, old cuticle culminates with ecdysis, a stereotyped sequence of shedding behaviors. Following each ecdysis, the new cuticle must expand and harden. Studies from a variety of insect species indicate that this cuticle hardening is regulated by the neuropeptide bursicon. However, genetic evidence from Drosop

Influenza Neuraminidase Virus-Like Particle-Based Nanocarriers as a New Platform for the Delivery of Small-Peptide Antigens.

A new and simple platform to produce a nanocarrier for small-peptide antigen delivery was developed. Virus-like particles (VLPs) were of interest due to their good cell-penetrating properties and ability to protect target molecules from degradation. In this study, the VLP that was entirely formed by influenza neuraminidase (NA), NA-VLPs, was employed. The platform construction includes the genetic engineering of target peptides into the NA structure immediately above its stalk, at the bottom of the NA head, by an overlap extension PCR. The resulting chimeric gene is next expressed in stably tr

A co-regulatory switch for immunity and yeast-like symbiont release: Pleiotropic mechanism of the JH-Met signaling pathway in Nilaparvata lugens (Stål).

Hormonal signaling pleiotropically regulates insect physiological processes, including development, reproduction, and immunity. Juvenile hormone (JH) and its receptor Methoprene-tolerant (Met) modulate immune responses across many insect species. Our previous studies demonstrated that mating behavior in Nilaparvata lugens induces the expression of defensin genes NlDfA/B, enhancing resistance to pathogens. Furthermore, the abundance of yeast-like symbionts (YLS) is closely linked to the reproduction-immunity trade-off. However, the regulatory mechanism underlying the dynamic changes in NlDfA/B

Mutations in the Drosophila glycoprotein hormone receptor, rickets, eliminate neuropeptide-induced tanning and selectively block a stereotyped behavioral program.

Adult insects achieve their final form shortly after adult eclosion by the combined effects of specialized behaviors that generate increased blood pressure, which causes cuticular expansion, and hormones, which plasticize and then tan the cuticle. We examined the molecular mechanisms contributing to these processes in Drosophila by analyzing mutants for the rickets gene. These flies fail to initiate the behavioral and tanning processes that normally follow ecdysis. Sequencing of rickets mutants and STS mapping of deficiencies confirmed that rickets encodes the glycoprotein hormone receptor DLG

Compounds Inhibiting Noppera-bo, a Glutathione S-transferase Involved in Insect Ecdysteroid Biosynthesis: Novel Insect Growth Regulators.

Glutathione S-transferases (GSTs) are conserved in a wide range of organisms, including insects. In 2014, an epsilon GST, known as Noppera-bo (Nobo), was shown to regulate the biosynthesis of ecdysteroid, the principal steroid hormone in insects. Studies on fruit flies, Drosophila melanogaster, and silkworms, Bombyx mori, demonstrated that loss-of-function mutants of nobo fail to synthesize ecdysteroid and die during development, consistent with the essential function of ecdysteroids in insect molting and metamorphosis. This genetic evidence suggests that chemical compounds that inhibit activi

Diverse reactions catalyzed by cytochrome P450 and biosynthesis of steroid hormone.

Steroid hormones modulate numerous physiological processes in various higher organisms. Research on the physiology, biosynthesis, and metabolic degradation of steroid hormones is crucial for developing drugs, agrochemicals, and anthelmintics. Most steroid hormone biosynthetic pathways, excluding those in insects, have been elucidated, and the roles of several cytochrome P450s (CYPs, P450s), heme (iron protoporphyrin IX)-containing monooxygenases, have been identified. Specifically, P450s of the animal steroid hormone biosynthetic pathways and their three dimensional structures and reaction mec

Supreme glutathione-dependent ketosteroid isomerase in the yellow-fever transmitting mosquito Aedes aegypti.

The steroid hormone ecdysone is essential for the reproduction and survival of insects. The hormone is synthesized from dietary sterols such as cholesterol, yielding ecdysone in a series of consecutive enzymatic reactions. In the insect orders Lepidoptera and Diptera a glutathione transferase called Noppera-bo (Nobo) plays an essential, but biochemically uncharacterized, role in ecdysteroid biosynthesis. The Nobo enzyme is consequently a possible target in harmful dipterans, such as disease-carrying mosquitoes. Flavonoid compounds inhibit Nobo and have larvicidal effects in the yellow-fever tr

Ecdysteroids elicit a rapid Ca2+ flux leading to Akt activation and increased protein synthesis in skeletal muscle cells.

Phytoecdysteroids, structurally similar to insect molting hormones, produce a range of effects in mammals, including increasing growth and physical performance. In skeletal muscle cells, phytoecdysteroids increase protein synthesis. In this study we show that in a mouse skeletal muscle cell line, C(2)C(12), 20-hydroxyecdysone (20HE), a common phytoecdysteroid in both insects and plants, elicited a rapid elevation in intracellular calcium, followed by sustained Akt activation and increased protein synthesis. The effect was inhibited by a G-protein coupled receptor (GPCR) inhibitor, a phospholip

Lipid metabolism in insect disease vectors.

More than a third of the world population is at constant risk of contracting some insect-transmitted disease, such as Dengue fever, Zika virus disease, malaria, Chagas' disease, African trypanosomiasis, and others. Independent of the life cycle of the pathogen causing the disease, the insect vector hematophagous habit is a common and crucial trait for the transmission of all these diseases. This lifestyle is unique, as hematophagous insects feed on blood, a diet that is rich in protein but relatively poor in lipids and carbohydrates, in huge amounts and low frequency. Another unique feature of

Function and composition of male accessory gland secretions in Anopheles gambiae: a comparison with other insect vectors of infectious diseases.

Human malaria, a major public health burden in tropical and subtropical countries, is transmitted exclusively by the bite of a female Anopheles mosquito. Malaria control strategies aimed at inducing sexual sterility in natural vector populations are an attractive alternative to the use of insecticides. However, despite their importance as disease vectors, limited information is available on the molecular mechanisms regulating fertility in Anopheles mosquitoes. In the major malaria vector, An. gambiae, the full complement of sperm and seminal fluid required for a female's lifelong egg productio

The Adipokinetic Hormone (AKH) and the Adipokinetic Hormone/Corazonin-Related Peptide (ACP) Signalling Systems of the Yellow Fever Mosquito Aedes aegypti: Chemical Models of Binding.

Neuropeptides are the main regulators of physiological, developmental, and behavioural processes in insects. Three insect neuropeptide systems, the adipokinetic hormone (AKH), corazonin (Crz), and adipokinetic hormone/corazonin-related peptide (ACP), and their cognate receptors, are related to the vertebrate gonadotropin (GnRH) system and form the GnRH superfamily of peptides. In the current study, the two signalling systems, AKH and ACP, of the yellow fever mosquito, Aedes aegypti, were comparatively investigated with respect to ligand binding to their respective receptors. To achieve this, t

Growth-Promoting and Quality-Enhancing Effects of Insect-Derived Serratia marcescens BRC-CXG2 on Romaine Lettuce.

To explore the application potential of insect-derived functional microorganisms in short-cycle leafy vegetable production, we evaluated the effects of Serratia marcescens BRC-CXG2, isolated from larvae of Monochamus alternatus, on romaine lettuce in a pot experiment. Plant growth traits, biomass accumulation, nutritional quality, endogenous hormones, and rhizosphere microbial communities were systematically evaluated. The results demonstrated that inoculation significantly promoted seedling development. Plant height and root length increased by 48.7% and 29.1%, respectively, while shoot and r

The role of apolipoproteins RPRC010099 and RPRC015421 in male fertility of the Chagas disease vector, Rhodnius prolixus.

The kissing bug Rhodnius prolixus, a primary vector of Chagas disease, serves as a model for understanding insect physiology and vector-pathogen interactions. While the role of male reproductive factors in influencing female fertility is well-established in many insects, little is known about male fertility in R. prolixus. Our study examines the role of two apolipoproteins, RPRC010099 and RPRC015421, in male fertility and their potential function as carriers of eicosanoid hormones. Using RNA interference (RNAi), we silenced the genes for both apolipoproteins in male R. prolixus. The silencing

SOX12 and DELTA mediate ecdysone-driven adult neural cell proliferation during metamorphosis.

During metamorphic development from larva to adult, insect brains exhibit cell proliferation; however, the molecular mechanisms governing this process remain poorly understood. Employing the lepidopteran insect Helicoverpa armigera (cotton bollworm), an agricultural pest, as a model system, this study demonstrates that the steroid hormone 20-hydroxyecdysone (20E) regulates neural cell proliferation during imaginal brain development by upregulating the expression of SOX12 and DELTA via nuclear receptor EcRA. SOX12 exhibits significantly elevated expression and is localized in the brain during m

Novel insect orcokinins: characterization and neuronal distribution in the brains of selected dicondylian insects.

Orcokinins are a family of myotropic neuropeptides identified in various decapod crustaceans and recently in a cockroach. Their presence in the crustacean nervous system and hemolymph suggests that they act as hormones and as locally acting neuromodulators. To provide further evidence for the existence of orcokinins in insects, we identified a novel orcokinin-related peptide in the locust Schistocerca gregaria and used an antiserum against Asn13-orcokinin for immunostaining in the brains of selected dicondylian insects, including a silverfish, three polyneopteran species (a cockroach and two l

Partiti-like viruses from African armyworm increase larval and pupal mortality of a novel host: the Egyptian cotton leafworm.

BACKGROUND: The general principle of using microbes from one species to manage a different pest species has a clear precedent in the large-scale release of mosquitoes carrying a Wolbachia bacterium derived from Drosophila flies. New technologies will facilitate the discovery of microbes that can be used in a similar way. Previously, we found three novel partiti-like viruses in the African armyworm (Spodoptera exempta). To investigate further the utility and consistency of host shift of insect viruses as a potential pest management tool, we tested the interaction between the partiti-like viruse

Dynamics of release and activity of select neuropeptides post-bloodmeal in the female mosquito, Aedes aegypti.

Female Aedes aegypti secrete urine rapidly post-bloodmeal ingestion, with diuresis beginning immediately for removal of excess salts and water. This post-prandial diuresis includes a peak, post-peak and late phase, involving the combined actions of multiple hormones, including diuretic and anti-diuretic factors. Calcitonin-like diuretic hormone 31 (DH31) and kinin peptides stimulate diuresis through actions on their cognate receptors localized in the Malpighian 'renal' tubules (MTs). In contrast, the anti-diuretic neurohormone CAPA inhibits secretion by MTs stimulated by select diuretic hormon
