Flavescence dorée phytoplasma enters insect cells by a clathrin-mediated endocytosis allowing infection of its insect vector.

To perform its propagative and circulative cycle into its insect vector, the flavescence dorée phytoplasma invades different cell types. Clathrin-mediated endocytosis is used by a wide range of bacteria to infect eukaryote cells. Among the insect proteins interacting with the phytoplasma adhesin VmpA, we identified the adaptor protein complex AP-1 and AP-2 suggesting that phytoplasmas could enter the insect cells via clathrin-mediated endocytosis. By infection assays of insect cells in culture, we showed that phytoplasmas entry into Drosophila S2 cells was more efficient than infection of the

Viral and cellular determinants of polarized trafficking of viral envelope proteins from insect-specific and insect-vectored viruses in insect midgut and salivary gland cells.

UNLABELLED: Systemic viral infection of insects typically begins with the primary infection of midgut epithelial cells (enterocytes) and subsequent transit of the progeny virus in an apical-to-basal orientation into the hemocoel. For insect-vectored viruses, an oppositely oriented process (basal-to-apical transit) occurs upon secondary infection of salivary glands and is necessary for virus transmission to non-insect hosts. To examine this inversely oriented virus transit in these polarized tissues, we assessed the intracellular trafficking of two model viral envelope proteins (baculovirus GP6

Active but not passive passengers: Persistent-propagative plant viruses manipulate insect vectors for efficient transmission.

Most plant viruses rely on insect vectors for transmission. In general, nonviruliferous vectors acquire viruses from infected plants, whereas viruliferous vectors subsequently move to uninfected plants for virus inoculation. However, they are not merely passive passengers within their vector. Persistent-propagative plant viruses, which invade and replicate in insect vectors, can actively and directly reshape their physiology, morphology, and feeding behavior to facilitate efficient transmission. Recent studies have uncovered both indirect plant-mediated mechanisms and direct vector-targeted me

Haemosporidian infections in the Tengmalm's Owl (Aegolius funereus) and potential insect vectors of their transmission.

Sedentary bird species are suitable model hosts for identifying potential vectors of avian blood parasites. We studied haemosporidian infections in the Tengmalm's Owl (Aegolius funereus) in the Ore Mountains of the Czech Republic using molecular detection methods. Sex of owl nestlings was scored using molecular sexing based on fragment analysis of PCR-amplified CHD1 introns. Observed infection prevalences in nestlings and adult owls were 51 and 86 %, respectively. Five parasite lineages were detected. Most of the infections comprised the Leucocytozoon AEFUN02 and STOCC06 lineages that probably

Key role of exportin 6 in exosome-mediated viral transmission from insect vectors to plants.

Exosomes play a key role in virus exocytosis and transmission. The exportin family is usually responsible for cargo nucleocytoplasmic trafficking, and they are frequently found in exosomes. However, the function of exportins sorted in exosomes remains unknown. Here, we successfully isolated "cup holder"-like exosomes from the saliva of ∼30,000 small brown planthoppers, which are vectors of rice stripe virus (RSV). RSV virions were packed in comparatively large exosomes. Four viral genomic RNAs at a certain ratio were identified in the saliva exosomes. The virions contained in the saliva exosom

Exploring the immunogenicity of an insect-specific virus vectored Zika vaccine candidate.

Zika virus (ZIKV) is an important re-emerging flavivirus that presents a significant threat to human health worldwide. Despite its importance, no vaccines are approved for use in humans. Insect-specific flaviviruses (ISFVs) have recently garnered attention as an antigen presentation platform for vaccine development and diagnostic applications. Here, we further explore the safety, immunogenicity, and efficacy of a chimeric ISFV-Zika vaccine candidate, designated Aripo-Zika (ARPV/ZIKV). Our results show a near-linear relationship between increased dose and immunogenicity, with 1011 genome copies

[Effect of a sublethal dose of an insect growth regulator on the physiology of onchocercosis vectors].

In most of the large scale vector control programs, the larviciding operations are not fully successful, resulting in a surviving vector population due to the use of sublethal doses. This problem leads the authors to study the effect of such doses upon the future of the residual population in order to know if it represents an epidemiologically dangerous population. One insect growth regulator was selected to study this phenomenon using Onchocerca volvulus as the parasite and Simulium damnosum s.l. as the vector. The experiment was conducted under field conditions by treating a river of a savan

Construction of modular and versatile plasmid vectors for the high-level expression of single or multiple genes in insects and insect cell lines.

We have constructed a series of plasmid vectors for the expression of foreign genes in insects or insect cell lines. We incorporated the Drosophila hsp70 and actin 5C promoters, as well as the hr5 enhancer-driven baculovirus ie1 promoter, into plasmids that allow convenient cloning of heterologous genes into multiple cloning sites. We combined these promoters with either a short, double poly-adenylation site derived from the Heliothis virescens p63 chaperonin gene, or with a fusion of the small t intron with the early 3' untranslated region and poly-adenylation sites of SV40. Unique eight base

CO2 drives the pine wood nematode off its insect vector.

Insects have developed special organs, spiracles and the trachea, for oxygen-carbon dioxide exchange to adapt to terrestrial life. The plant-parasitic nematode Bursaphelenchus xylophilus, also known as pine wood nematode (PWN), is vectored by pine sawyer beetles (Monochamus spp.) and causes destructive pine wilt disease, threatening the safety and stability of pine forest ecosystems. Unlike the free-living nematode model species Caenorhabditis elegans, PWN have two distinct life stages (dispersive and propagative), each requiring a unique host relationship ranging from symbiotic/commensal to p

The oligosaccharides of influenza virus hemagglutinin expressed in insect cells by a baculovirus vector.

The hemagglutinin of fowl plague virus has been expressed in Spodoptera frugiperda (SF) cell cultures using a baculovirus vector. To elucidate the structure of the carbohydrate side chains, radioactively labeled oligosaccharides were liberated by treatment with endoglucosaminidase H and glycopeptidase F. Sequential degradation with exoglycosidases and chromatographic analyses revealed the presence of oligomannosidic side chains, predominantly of the structures Man5-9GlcNAc2, and the truncated oligosaccharide cores Man3GlcNAc2 and Man3[Fuc]GlcNAc2. Insect-Specific Flaviviruses: Novel Strategies for Indonesian Vector-Borne Disease Control.

Arboviral diseases in Indonesia remain a public health problem and a threat. The annual increase in cases is caused by changes in bioclimatological factors, increased reports of resistance to insecticides and larvicides, and low community participation in vector control, resulting in an uncontrolled abundance of the species. Therefore, an alternative, safe, effective, and evidence-based vector control approach is needed. This narrative review, conducted via the SANRA protocol encompassing PubMed, Scopus, and Web of Science (2014-2024), aims to (1) synthesize current Insect-Specific Flavivirus

Emergent insects as both prey and neonicotinoid vectors for aerial insectivores in prairie agroecosystems.

Aerially insectivorous birds in North America have declined since the 1980s for reasons that are hypothesized to relate to changes in their insect prey. Agricultural practices have been implicated in these declines, as shifts toward more intensive cultivation alter insect communities and contaminate non-target insects with insecticides. To examine how agricultural intensity affects diet composition and insecticide exposure among aerial insectivores, we evaluated differences in diet via metabarcoding of 309 fecal samples and analysis of seven neonicotinoid insecticides in 389 blood plasma sampl

Challenges in the control of neglected insect vector diseases of human importance in the Anglo-Caribbean.

INTRODUCTION: Neglected tropical diseases (NTDs) in developing countries like the Caribbean, negatively affect multiple income-generating sectors, including the tourism industry upon which island states are highly dependent. Insect-transmitted NTDs include, but are not limited to, malaria, dengue and lymphatic filariasis. Control measures for these disease, are often ignored because of the associated cost. Many of the developing country members are thus retained in a financially crippling cycle, balancing the cost of prophylactic measures with that of controlling an outbreak. The purpose of the

Weather, host and vector--their interplay in the spread of insect-borne animal virus diseases.

The spread of insect-borne animal virus diseases is influenced by a number of factors. Hosts migrate, move or are conveyed over long distances: vectors are carried on the wind for varying distances in search of hosts and breeding sites; weather and climate affect hosts and vectors through temperature, moisture and wind. As parasites of host and vector, viruses are carried by animals, birds and insects, and their spread can be correlated with the migration of hosts and the carriage of vectors on winds associated with the movements of the Intertropical Convergence Zone (ITCZ) and warm winds to t

Purification of Recombinant Human Tyrosinase from Insect Larvae Infected with the Baculovirus Vector.

The purification of an enzyme from insect larvae infected with a baculovirus vector is described. The enzyme tyrosinase is of biomedical importance and catalyzes the first rate-limiting steps in melanin production. Tyrosinase mutations can result in oculocutaneous albinism type 1 (OCA1), an inherited eye disease associated with decreased melanin pigment production and vision defects. To simplify expression and subsequent purification, the extracellular domain is expressed in insect cells, produced in Trichoplusia ni larvae, and purified using affinity and size-exclusion chromatography. "Fleaing" the Plague: Adaptations of Yersinia pestis to Its Insect Vector That Lead to Transmission.

Interest in arthropod-borne pathogens focuses primarily on how they cause disease in humans. How they produce a transmissible infection in their arthropod host is just as critical to their life cycle, however. Yersinia pestis adopts a unique life stage in the digestive tract of its flea vector, characterized by rapid formation of a bacterial biofilm that is enveloped in a complex extracellular polymeric substance. Localization and adherence of the biofilm to the flea foregut is essential for transmission. Here, we review the molecular and genetic mechanisms of these processes and present a com

Zoonotic cutaneous leishmaniasis: reservoir host and insect vector in north Sinai, Egypt.

Cutaneous leishmaniasis (ZCL) is a protozoan disease well documented not only in Egypt, but in nearly all the East Mediterranean Countries. Sinai Peninsula was a sparsely populated area where sporadic cases of ZCL were reported with the reconstruction of Sinai and people coming in and out, it was indicated to study the status of ZCL in North Sinai Governorate, the reservoir host(s) and insect vector(s) in Sinai. In the present study, the six species of rodents were trapped from areas or nearby areas where human ZCL cases were detected. Rodents (50) were Mus musculus (10), Rattus r. alexandrinu

The effect of isometamidium chloride on Trypanosoma vivax occurring within the insect vector (Glossina).

The effect of chemoprophylaxis on developing and mature Trypanosoma vivax in Glossina tachinoides and G. palpalis palpalis was evaluated. Newly emerged G. tachinoides and G.p. palpalis flies were infected with T. vivax by allowing them to feed on parasitaemic animals. Three experiments were conducted and in each the flies were divided into two groups. One group of infected flies was fed on animals treated 1 day previously with isometamidium (1 mg/kg body wt. in a 4% soln.) after which the surviving flies were dissected and examined for trypanosomes. The other group was fed on untreated animals

Targeted deletion of the gp72 gene decreases the infectivity of Trypanosoma cruzi for mice and insect vectors.

The infective behavior of a mutant Trypanosoma cruzi clone, carrying a targeted deletion of the gp72 gene, was studied in the insect vector Triatoma infestans and in mice. After feeding T. infestans with complement-resistant forms (CRF) of Ynull and wild-type clones, it was observed that the number of parasites released in the bug's feces was reduced to less than 1% in the mutant clone. Both gp72-null and wild-type clones had a low infectivity for mice in comparison with other T. cruzi isolates, probably as a consequence of prolonged in vitro culture. Therefore, the behavior of both clones was

Plant Virus-Induced Inheritable Apoptosis Drives Reproductive Costs in Female Insect Vectors to Balance Viral Biparental Transmission.

Insect-borne pathogens often reduce the reproductive fitness of insect vectors. Rice gall dwarf virus (RGDV) is biparentally transmitted to the offspring of its leafhopper vector. However, maternal transmission of RGDV decreases female fecundity and disrupts egg development via an unknown mechanism. This study reveals that RGDV induces mitochondria-dependent apoptosis in leafhopper ovaries, promoting viral infection but impairing ovary development. This apoptosis is transmitted to eggs during maternal RGDV transmission, promoting viral infection while harming eggs. RGDV in the ovary activates
