Cortical interneuron development: a tale of time and space.

Cortical interneurons are a diverse group of neurons that project locally and are crucial for regulating information processing and flow throughout the cortex. Recent studies in mice have advanced our understanding of how these neurons are specified, migrate and mature. Here, we evaluate new findings that provide insights into the development of cortical interneurons and that shed light on when their fate is determined, on the influence that regional domains have on their development, and on the role that key transcription factors and other crucial regulatory genes play in these events. We foc

Morphology of interneurons in the procerebrum of the snail Helix aspersa.

Terrestrial snails have a highly developed sense of olfaction. Because the procerebrum has a large number of cells and is located at the entry site of the olfactory nerve into the brain, the structure is thought to have a significant role in the processing of olfactory stimuli. The morphology of the procerebral neurons in the snail Helix aspersa was investigated through intracellular injections of biocytin. No formal categorization of neuronal types was possible, but some cells were seen to have neurites entirely intrinsic to the procerebrum, whereas others had both intrinsic and extrinsic arb

Mechanism of an Intrinsic Oscillation in Rat Geniculate Interneurons.

Depolarizing current injections produced a rhythmic bursting of action potentials, a bursting oscillation, in a set of local interneurons in the lateral geniculate nucleus (LGN) of rats. The current dynamics underlying this firing pattern have not been determined, though this cell type constitutes an important cellular component of thalamocortical circuitry and contributes to both pathologic and nonpathologic brain states. We thus investigated the source of the bursting oscillation using pharmacological manipulations in LGN slices in vitro and in silico. We found that selective blockade of cal

Development of GABAergic Interneurons in the Human Cerebral Cortex.

GABAergic interneurons are critical regulators of information processing in the cerebral cortex. They constitute a heterogeneous group of neurons with unique spatial and temporal capabilities to control information flow and influence neural network dynamics through inhibitory and disinhibitory mechanisms. Interneuron diversity is largely conserved between rodents and primates, which indicates that the addition of new types of GABAergic neurons is not the most critical innovation of the primate cortex. In contrast, interneurons are much more abundant and seem more widely interconnected in the c

Feedback inhibition underlies new computational functions of cerebellar interneurons.

The function of a feedback inhibitory circuit between cerebellar Purkinje cells and molecular layer interneurons (MLIs) was defined by combining optogenetics, neuronal activity recordings both in cerebellar slices and in vivo, and computational modeling. Purkinje cells inhibit a subset of MLIs in the inner third of the molecular layer. This inhibition is non-reciprocal, short-range (less than 200 μm) and is based on convergence of one to two Purkinje cells onto MLIs. During learning-related eyelid movements in vivo, the activity of a subset of MLIs progressively increases as Purkinje cell acti

Spinal premotor interneurons controlling antagonistic muscles are spatially intermingled.

Elaborate behaviours are produced by tightly controlled flexor-extensor motor neuron activation patterns. Motor neurons are regulated by a network of interneurons within the spinal cord, but the computational processes involved in motor control are not fully understood. The neuroanatomical arrangement of motor and premotor neurons into topographic patterns related to their controlled muscles is thought to facilitate how information is processed by spinal circuits. Rabies retrograde monosynaptic tracing has been used to label premotor interneurons innervating specific motor neuron pools, with p

State-Dependent Subnetworks of Parvalbumin-Expressing Interneurons in Neocortex.

Brain state determines patterns of spiking output that underlie behavior. In neocortex, brain state is reflected in the spontaneous activity of the network, which is regulated in part by neuromodulatory input from the brain stem and by local inhibition. We find that fast-spiking, parvalbumin-expressing inhibitory neurons, which exert state-dependent control of network gain and spike patterns, cluster into two stable and functionally distinct subnetworks that are differentially engaged by ascending neuromodulation. One group is excited as a function of increased arousal state; this excitation i

Cholinergic Interneurons Underlie Spontaneous Dopamine Release in Nucleus Accumbens.

The release of dopamine from terminals in the NAc is regulated by a number of factors, including voltage-gated ion channels, D2-autoreceptors, and nAChRs. Cholinergic interneurons (CINs) drive dopamine release through activation of nAChRs on dopamine terminals. Using cyclic voltammetry in mouse brain slices, nAChR-dependent spontaneous dopamine transients and the mechanisms underlying the origin were examined in the NAc. Spontaneous events were infrequent (0.3 per minute), but the rate and amplitude were increased after blocking Kv channels with 4-aminopyridine. Although the firing frequency o

Brain-Wide Maps of Synaptic Input to Cortical Interneurons.

UNLABELLED: Cortical inhibition is mediated by diverse inhibitory neuron types that can each play distinct roles in information processing by virtue of differences in their input sources, intrinsic properties, and innervation targets. Previous studies in brain slices have demonstrated considerable cell-type specificity in laminar sources of local inputs. In contrast, little is known about possible differences in distant inputs to different cortical interneuron types. We used the monosynaptic rabies virus system, in conjunction with mice expressing Cre recombinase in either parvalbumin-positive

Selective inhibition of striatal fast-spiking interneurons causes dyskinesias.

Fast-spiking interneurons (FSIs) can exert powerful control over striatal output, and deficits in this cell population have been observed in human patients with Tourette syndrome and rodent models of dystonia. However, a direct experimental test of striatal FSI involvement in motor control has never been performed. We applied a novel pharmacological approach to examine the behavioral consequences of selective FSI suppression in mouse striatum. IEM-1460, an inhibitor of GluA2-lacking AMPARs, selectively blocked synaptic excitation of FSIs but not striatal projection neurons. Infusion of IEM-146

NO synthase-positive striatal interneurons are decreased in schizophrenia.

The gaseous messenger NO has repeatedly been suggested to play a role in the pathogenesis of psychoses. Following a pilot study, we investigated whether the number of nitrinergic neurons in the putamen of patients suffering from schizophrenia, bipolar disorder or major depression is altered. Post-mortem striatum sections of 15 brains from patients with either disease were examined by NADPH-diaphorase staining, which selectively labels NO synthase-positive interneurons. Quantification of these cells revealed significantly lower numbers of NO synthase-containing neurons in the putamen of schizop

Dopamine-dependent synaptic plasticity in the striatal cholinergic interneurons.

The striatum, the input stage of the basal ganglia, is a critical brain structure for the learning of stimulus-response habits as well as motor, perceptual, and cognitive skills. Roles of dopamine (DA) and acetylcholine (ACh) in this form of implicit memory have long been considered essential, but the underlying cellular mechanism is still unclear. By means of patch-clamp recordings from corticostriatal slices of the mouse, we studied whether the identified striatal cholinergic interneurons undergo long-term synaptic changes after tetanic stimulation of cortico- and thalamostriatal fibers. Brainstem interneurons necessary for vestibular influences on sympathetic outflow.

The objective of this study was to determine which general brainstem regions contain interneurons that are critical for mediating vestibulo-sympathetic responses in decerebrate cats, as a prelude for future cell recording studies. Large injections of kainic acid into the lateral reticular formation at levels caudal to the obex abolished sympathetic nerve responses elicited by electrical stimulation of the vestibular nerve. The same lesions also diminished excitatory and some inhibitory components of somato-sympathetic responses elicited by stimulation of the sciatic nerve, raising the possibil

Adenylate cyclase in striatal cholinergic interneurons regulates acetylcholine release.

Fractional [3H]ACH efflux from dissociated rat striata tested whether tonic inhibition prevents stimulation of acetylcholine (ACH) release by adenylate cyclase. Forskolin stimulated release from the dissociated cells (threshold at 300 nM; EC50 > or = 1 MicroM). Release was also stimulated by 3-isobutyl-1-methylxanthine and was additive with forskolin. The 1,9-dideoxy forskolin analog that lacks cyclase-stimulating activity was ineffective. Thus, stimulation of adenylate cyclase within striatal cholinergic interneurons increases ACH secretion but is tonically inhibited by endogenous striatal tr

Altered patterning of trisomy 21 interneuron progenitors.

Individuals with Down syndrome (DS; Ts21), the most common genetic cause of intellectual disability, have smaller brains that reflect fewer neurons at pre- and post-natal stages, implicating impaired neurogenesis during development. Our stereological analysis of adult DS cortex indicates a reduction of calretinin-expressing interneurons. Using Ts21 human induced pluripotent stem cells (iPSCs) and isogenic controls, we find that Ts21 progenitors generate fewer COUP-TFII+ progenitors with reduced proliferation. Single-cell RNA sequencing of Ts21 progenitors confirms the altered specification of

Synchronous activation of striatal cholinergic interneurons induces local serotonin release.

Striatal cholinergic interneurons (CINs) can drive local dopamine release via nicotinic acetylcholine receptors (nAChRs) expressed on dopaminergic axons, but their role in modulating serotonin (5-HT) signaling is poorly understood. Here, we show that synchronous activation of CINs directly triggers local 5-HT release in the dorsal striatum via nAChRs expressed on serotonergic axons. This CIN-5-HT coupling is not detectable in the ventral striatum, despite its substantially denser serotonergic innervation. The nAChR-dependent release not only increases 5-HT levels in the dorsal striatum, but al

Motor Cortex VIP Interneurons Participate in Dexmedetomidine-Associated Sleep Modulation.

Vasoactive intestinal peptide (VIP)-expressing neurons within the motor cortex (MC) exhibit diurnal variation in excitability and may participate in the modulation of sleep architecture. In this study, we found that MCVIP neurons displayed significantly higher excitability during the light phase. Using chemogenetic approaches, we examined their contribution to sleep-wake modulation. Given the sleep-modulating actions of the α2-adrenergic receptor agonist dexmedetomidine (Dex), we further tested whether Dex engages MCVIP neurons. Dex increased MCVIP neuronal excitability in slice recordings in

Parvalbumin interneurons in Alzheimer's disease: Physiological insights and pathophysiological mechanisms.

Fast-spiking parvalbumin-positive (PV+) interneurons, a specialized class of inhibitory neurons, possess unique morphological and functional properties that govern spatiotemporal precision in local microcircuits, large-scale network synchronization, and memory-related computations. Since their initial identification in the late 19th century, technological innovations in cellular neuroscience have progressively elucidated the multifaceted roles of these neurons. In this review, we first delineate the embryonic origins and developmental trajectory of PV+ interneurons, emphasizing their unique pr

An axonal brake on striatal dopamine output by cholinergic interneurons.

Depolarization of axons is necessary for somatic action potentials to trigger axonal neurotransmitter release. Here we show that striatal cholinergic interneurons (ChIs) and nicotinic receptors (nAChRs) on mouse dopamine axons interrupt this relationship. After nAChR-mediated depolarization, dopamine release by subsequent depolarization events was suppressed for ~100 ms. This suppression was not due to depletion of dopamine or acetylcholine, but to a limited reactivation of dopamine axons after nAChR-mediated depolarization, and is more prominent in dorsal than in ventral striatum. In vivo, nA
