Organophosphate-Pesticide-Mediated Immune Response Modulation in Invertebrates and Vertebrates.

Organophosphate pesticides (OPs) have greatly facilitated food production worldwide, and their use is not limited to agriculture and the control of pests and disease vectors. However, these substances can directly affect the immune response of non-target organisms. In this sense, exposure to OPs can have negative effects on innate and adaptive immunity, promoting deregulation in humoral and cellular processes such as phagocytosis, cytokine expression, antibody production, cell proliferation, and differentiation, which are crucial mechanisms for host defense against external agents. Ecology and environmental characteristics influence methylmercury bioaccumulation in coastal invertebrates.

Quantifying mercury (Hg) concentrations in invertebrates is fundamental to determining risk for bioaccumulation in higher trophic level organisms in coastal food webs. Bioaccumulation is influenced by local mercury concentrations, site geochemistry, individual feeding ecologies, and trophic position. We sampled seven species of invertebrates from five coastal sites in the Minas Basin, Bay of Fundy, and determined body concentrations of methylmercury (MeHg), total mercury (THg), and stable isotopes of nitrogen (δ15N) and carbon (δ13C). To evaluate the effects of environmental chemistry on Hg pr

Organic wastes as soil amendments - Effects assessment towards soil invertebrates.

Using organic wastes, as soil amendments, is an important alternative to landfilling with benefits to soil structure, water retention, soil nutrient and organic matter concentrations. However, this practice should be monitored for its environmental risk due to the frequent presence, of noxious substances to soil organisms. To evaluate the potential of eight organic wastes with different origins, as soil amendments, reproduction tests with four soil invertebrate species (Folsomia candida, Enchytraeus crypticus, Hypoaspis aculeifer, Eisenia fetida) were performed using gradients of soil-waste mi

Photoautotrophic microorganisms as a carbon source for temperate soil invertebrates.

We tested experimentally if photoautotrophic microorganisms are a carbon source for invertebrates in temperate soils. We exposed forest or arable soils to a (13)CO2-enriched atmosphere and quantified (13)C assimilation by three common animal groups: earthworms (Oligochaeta), springtails (Hexapoda) and slugs (Gastropoda). Endogeic earthworms (Allolobophora chlorotica) and hemiedaphic springtails (Ceratophysella denticulata) were highly (13)C enriched when incubated under light, deriving up to 3.0 and 17.0%, respectively, of their body carbon from the microbial source in 7 days. Earthworms assim

Pesticide runoff from energy crops: A threat to aquatic invertebrates?

The European Union aims to reach a 10% share of biofuels in the transport sector by 2020. The major burden is most likely to fall on already established annual energy crops such as rapeseed and cereals for the production of biodiesel and bioethanol, respectively. Annual energy crops are typically cultivated in intensive agricultural production systems, which require the application of pesticides. Agricultural pesticides can have adverse effects on aquatic invertebrates in adjacent streams. We assessed the relative ecological risk to aquatic invertebrates associated with the chemical pest manag

Some highlights of research on aging with invertebrates, 2006-2007.

The invertebrate model organisms continue to be engines of discovery in aging research. Recent work with Drosophila stem cells has thrown light on their human equivalents, and on the role of stem cells and their niches in the decline in fecundity with age. Inspired by observations of aging in bacteria and yeast, a new theoretical study has revealed evolutionary forces that could favour asymmetry in the distribution of damaged cell constituents at division, and hence pave the way for the evolution of aging and selective maintenance of integrity of the germ line. Mechanisms of nutrient sensing a

Quantitative metabarcoding for invertebrate pest monitoring and management.

Invertebrate pests pose one of the most significant threats to global agriculture. Trap-based surveillance is widely used to monitor the presence and abundance of pests, beneficial taxa and broader agroecosystem communities; however nonselective traps often collect hundreds or even thousands of individuals per sample, making conventional sorting and morphological identification labor-intensive and delaying the delivery of actionable information. Metabarcoding offers a scalable alternative for rapidly identifying agriculturally significant taxa in mixed trap samples, while providing more precis

Detection, movement and persistence of invertebrate eDNA in groundwater.

Sampling groundwater biodiversity is difficult because of limited access and issues with species identification. Environmental DNA (eDNA) provides a viable alternative to traditional sampling approaches, however limited knowledge of the abundance and fate of DNA in groundwater hinders the interpretation of data from these environments. Groundwater environments are dark and have lower oxygen concentrations and microbial activity than surface waters. Consequently, assumptions about DNA fate in surface ecosystems may not apply to groundwaters. Here, we test the longevity and transport of eDNA in

Efficacy of voriconazole in vitro and in invertebrate model of cryptococcosis.

Cryptococcosis is a common opportunistic infection in patients with advanced HIV infection and may also affect immunocompetent patients. The available antifungal agents are few and other options are needed for the cryptococcosis treatment. In this work, we first analyzed the virulence of twelve C. neoformans and C. gattii strains assessing capsule thickness, biofilms formation, and survival and morbidity in the invertebrate model of Galleria mellonella and then we evaluated the antifungal activity of voriconazole (VRC) in vitro and in vivo also using G. mellonella. Our results showed that all

Coexistence of gap and sseptate junctions in an invertebrate epithelium.

The intercellular junctions of the epithelium lining the hepatic caecum of Daphnia were examined. Electron microscope investigations involved both conventionally fixed material and tissue exposed to a lanthanum tracer of the extracellular space. Both septate junctions and gap junctions occur between the cells studied. The septate junctions lie apically and resemble those commonly discerned between cells of other invertebrates. They are atypical in that the high electron opacity of the extracellular space obscures septa in routine preparations. The gap junctions are characterized by a uniform 3

Invertebrate genetic models of amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is a common adult-onset neurodegenerative disease characterized by the progressive death of motor neurons in the cerebral cortex, brain stem, and spinal cord. The exact mechanisms underlying the pathogenesis of ALS remain unclear. The current consensus regarding the pathogenesis of ALS suggests that the interaction between genetic susceptibility and harmful environmental factors is a promising cause of ALS onset. The investigation of putative harmful environmental factors has been the subject of several ongoing studies, but the use of transgenic animal model

On the Spread of Microbes That Manipulate Reproduction in Marine Invertebrates.

AbstractBacterial symbionts are functionally integral to animal reproduction and development, some of which have evolved additional mechanisms to override these host programs. One habitat that is increasingly recognized to contain phylogenetically related lineages of reproductive manipulators is the ocean. The reproduction of marine invertebrates often occurs by free spawning instead of by the physical contact of copulation in terrestrial systems. We developed an integrated model to understand whether and when microbes that manipulate host reproduction by cytoplasmic incompatibility, feminizat

Limited capacity for rapid acclimation to marine heatwaves in coastal invertebrates.

Marine heatwaves are increasing in frequency and intensity worldwide and represent a growing threat to coastal ecosystems and the animals inhabiting them. The vulnerability of marine species to these events depends on several factors, including upper thermal tolerance, the thermal sensitivity of physiological performance, and the capacity for rapid physiological acclimation. We examined upper thermal tolerance and short-term acclimation capacity in 10 mobile macroinvertebrate species inhabiting shallow coastal ecosystems in the Northeast Atlantic. Individuals were exposed either to ambient the

Invertebrates gut viromes mediate microbial adaptation to pharmaceutical diversity under warming.

BACKGROUND: Pharmaceutical pollution is an emerging environmental concern that can disrupt microbial communities and ecological processes, while climate warming adds further stress with broad ecological consequences. Soil invertebrates such as collembolans harbor gut microbiomes essential for host health and ecosystem stability, yet the responses of these communities-particularly viral communities-to combined pharmaceutical and warming pressures remain unclear. Here, we used controlled microcosm experiments with Folsomia candida to investigate how pharmaceutical diversity and fluctuating warmi
