Long-term treatment of perennial allergic rhinitis with ipratropium bromide nasal spray 0.06%.

The purpose of this study was to assess the safety and efficacy of ipratropium bromide nasal spray 0.06% (aqueous solution), 84 micrograms per nostril three times a day, in reducing nasal hypersecretion in the long-term treatment of patients with perennial allergic rhinitis (PAR). This was an open-label 1-year trial. In the first 6 months all patients were treated with two puffs ipratropium bromide nasal spray 0.06%, 84 micrograms per nostril three times per day, unless they were unable to tolerate the dose. In the last 6 months the dose could be reduced to the lowest amount required to contro

Effect of ipratropium bromide on the interfacial organization of model lung surfactant membranes.

The interactions of inhaled drugs with lung surfactant are important for understanding their behavior at the first biological barrier of the respiratory tract. In this work, we investigated the effect of the anticholinergic bronchodilator, ipratropium bromide (IPB), on model lung surfactant membranes composed of dipalmitoylphosphatidylcholine (DPPC), dipalmitoylphosphatidylglycerol (DPPG), and their 8:2 molar mixture. Langmuir monolayer studies showed that ipratropium bromide has little influence on zwitterionic DPPC, while it interacts strongly with negatively charged DPPG, leading to concent

Unilateral mydriasis: an unexpected effect of ipratropium bromide inhalation-a brief report.

UNLABELLED: Anisocoria often raises concerns about potential underlying conditions such as intracranial hemorrhage, brain tumor, or Horner syndrome. However, iatrogenic exposures may also lead to unilateral mydriasis. A six-month-old infant was hospitalized due to acute bronchiolitis with a history of prematurity, bronchopulmonary dysplasia, and periventricular leukomalacia (PVL). He received non-invasive respiratory support, salbutamol, and ipratropium bromide by inhalation. Four days after admission, the infant exhibited an episode of sudden unilateral mydriasis with no other new signs or sy

Utilization of intranasal ipratropium bromide in the prevention of recurrent croup events: Is it effective?

OBJECTIVE: Recurrent croup (RC) is a common problem in the pediatric population. We theorize that reduced rhinorrhea and post-nasal drip as well as suppressed cough receptor activity by the anticholinergic, intranasal ipratropium bromide (IB), may lead to reduced inflammation and edema of the subglottis, decreasing RC symptoms. The aim of this study is to determine the effectiveness of IB in improving symptoms of RC and in reducing the need for alternative forms of management. METHOD: A retrospective chart review combined with survey data of patients with RC was conducted to assess demographi

A comparison of the effect of ipratropium and albuterol in the treatment of chronic obstructive airway disease.

Twenty-five subjects with moderate to severe chronic obstructive airway disease were studied to compare the effect of ipratropium bromide, albuterol, and placebo on the forced expiratory volume in 1 s, (FEV1), forced vital capacity, heart rate, and blood pressure during six hours. Ipratropium produced a significantly greater improvement than albuterol in the FEV1 at 30 minutes and at 3, 4, and 5 hours and in the forced vital capacity at one through six hours. Fifteen subjects did not demonstrate a 15% improvement and at least a 200-mL increase in the FEV1, 15 minutes after metaproterenol sulfa

Prevention of fog-induced bronchospasm by high doses of ipratropium bromide in asthmatics.

In 12 adult asymptomatic, relatively stable adult asthmatics the protective effect of 3 doses (200, 1,000 and 2,000 micrograms) of ipratropium bromide against bronchospasm induced by an ultrasonic mist of distilled water was functionally (sRaw, FEV1) assessed. Ipratropium bromide and placebo were administered as an inhalation powder via a special inhaler in random order and under double-blind conditions at the same time on 4 usually consecutive days 90 min prior to the challenge. A considerable protective effect (clearly inferior, in any case, to that previously observed by us with fenoterol)

Effect of ipratropium bromide and fenoterol on airway obstruction in chronic pulmonary tuberculosis.

Single-blind, cross-over comparison of standard doses of ipratropium bromide (2 puffs of 0.02 mg each) and fenoterol (2 puffs of 0.2 mg each) in 36 pairs of experiments in 6 patients with partially reversible airway obstruction, presumably secondary to chronic widespread pulmonary tuberculosis, revealed no difference between the drugs in bronchodilating effect, evaluated by peak-flow measurements and spirometry. The inhalation of isoprenaline at the end of the experiment induced no further improvement in the lung function variables measured. The results suggest that a vagal reflex mechanism pl

A clinical trial of ipratropium bromide nasal spray in patients with perennial nonallergic rhinitis.

Intranasal ipratropium bromide has been shown to significantly reduce rhinorrhea. Use of a freon-propelled intranasal preparation has resulted in side effects associated with the drying properties of the propellant. The purpose of the present trial was to study the safety and efficacy of a new isotonic aqueous ipratropium bromide nasal spray pump, specifically in patients with perennial nonallergic rhinitis. Two hundred thirty-three patients participated in an 8-week double-blind parallel comparison of ipratropium bromide nasal spray with its vehicle, a saline solution. Treatment with the ipra

Effect of ipratropium on nasal reactivity to histamine and eosinophil influx in perennial allergic rhinitis.

In a double-blind, placebo-controlled study nasal saline and histamine provocation tests were performed in patients with perennial allergic rhinitis in order to assess changes in eosinophil influx and non-specific nasal reactivity after 8 days of treatment with ipratropium bromide. A "nasal pool" method was used to trace changes in protein level and eosinophil influx into nasal secretions. Treatment with ipratropium 80 mg q.i.d. caused a significant decrease in the albumin and total protein level in saline washings and induced a five-fold increase in eosinophils as compared to the placebo trea

Effect of ipratropium bromide aerosol on respiratory function in patients under ventilator treatment.

Twenty patients, 36-78 years old, with a history of chronic obstructive lung disease, and immediately after operation mostly for cardiac disease, were treated with ipratropium bromide (IB, 15 cases) or placebo (5 cases). A metered dose inhaler with a new adapter was used during postoperative ventilator treatment. In the IB group the heart rate did not change, but the inspiratory resistance decreased and the arterial oxygen tension increased. This is considered to indicate an effect not only on large but also on small airways, or an improvement of the ventilation/perfusion relationship. The inv

[Hemodynamic effects in antagonism of neuromuscular blockage: atropine-pyridostigmine versus ipratropium bromide-pyridostigmine].

Antagonism of neuromuscular block using cholinesterase inhibitors and atropine is charged with several risks, which are at least partly caused by pharmacological characteristics of the anticholinergic drugs, e.g. short duration of action causing secondary bradycardia. Compared to atropine, ipratropium bromide, a new anticholinergic drug, is--due to its quarternary ammonium compound--characterized by longer duration of action. In contrast to atropine, this substance does not penetrate the blood-brain and placental barrier. Our present study was designed to compare the haemodynamic effects of bo

Evaluation of artificial intelligence identified ipratropium bromide for the treatment of coronavirus disease 2019.

Ipratropium bromide (IB), identified through an artificial intelligence (AI)-aided drug screening platform, was evaluated for its safety and efficacy in treating severe coronavirus disease 2019 (COVID-19) using a hamster model in this study. The mortality rates in the vehicle, remdesivir-treated, IB-treated, and IB-remdesivir combination treatment groups were 70%, 15%, 5%, and 50%, respectively. The IB-treated group showed significantly lower blood D-dimer levels and fibrin degradation product when compared to that in the vehicle group, indicating prevention of thrombosis-related complications

[Effect of ipratropium bromide on histamine-induced bronchoconstriction in subjects with chronic airway obstruction].

Ipratropium bromide (IB), a quaternary derivative of atropine has been extensively recommended as the first bronchodilator to be tried in chronic obstructive pulmonary disease (COPD). Despite the large information concerning IB use, a controversy still persists about the lack of non bronchodilator effects (preventive) of inhaled IB. Therefore, the purpose was: to study the effects of IB (80 micrograms) on histamine-induced bronchoconstriction in moderate airway obstruction due to COPD. From outpatient clinic 9 men aged (mean +/- SEM) 57.9 +/- 2.4 yr with smoking history of 54.6 +/- 5.1 pack-yr
