An Integrative ATAC-Seq and RNA-Seq Analysis of Spleen Tissues from Largemouth Bass (Micropterus salmoides) Infected with Iridovirus (LMBV).

In this study, we systematically analyzed the dynamic changes in chromatin accessibility and the transcriptional responses in the spleen of largemouth bass (Micropterus salmoides) following infection with iridovirus (LMBV) using the assay for transposase-accessible chromatin with sequencing (ATAC-seq) and transcriptome sequencing (RNA-seq). Based on post-infection survival status, largemouth bass were classified into a resistant group (SR) and a susceptible group (SS). A total of 11,317 differentially accessible regions were identified between the two groups, among which the chromatin accessib

Apoptosis in spotted sea bass brain and astroglia-like cell line implies a possible pathogenesis of Lateolabrax maculatus iridovirus infection.

Brain undergoing apoptosis often results in serious dysfunction, especially when subjected to viral infection. Our previous findings revealed that Lateolabrax maculatus iridovirus (LMIV), genetically closely related to Red Sea Bream Iridovirus (RSIV), was able to invade the brain tissue of L. maculatus. The analysis of consequent brain injury may bring us explanations for the pathogenesis of LMIV infection. In the present study, viral loads in different brain segments (telencephalon, optic tectum, cerebellum, hypothalamus, medulla oblongata, optic nerve) of L. maculatus were measured by qPCR.

Epitope screening of the major capsid protein within grouper iridovirus of Taiwan and the immunoprotective effect with SWCNTs as the vaccine carrier.

Iridovirus can cause a mass of death in grouper, leading to huge economic loss in recent years. At present, practical vaccine is still the best way to control the outbreak of this virus. Many researches had indicated that the major capsid protein (MCP) of grouper iridovirus of Taiwan (TGIV) is an effective antigen to induce a specific immune response in grouper. However, these traditional vaccines that based on large proteins or whole organisms are faced with challenges because of the unnecessary antigenic load. Thus, in this study, we screened the dominant linear epitope within the MCP of TGI

Immune responses and seasonal red sea bream iridovirus dynamics following formalin-inactivated vaccination in farmed rock bream (Oplegnathus fasciatus).

Red sea bream iridovirus (RSIV) remains a major constraint on rock bream aquaculture, but vaccine-associated immune responses during natural field exposure are poorly understood. We monitored formalin-inactivated RSIV-vaccinated and unvaccinated rock bream from post-vaccination land-based rearing through a seasonal RSIV episode in commercial marine net pens. Splenic RSIV load and the mRNA expression of MyD88, IL-1β, IRF3, Mx, PKR, membrane-bound IgT (mIgT), and secretory IgT (sIgT) were assessed longitudinally. Full-length major capsid protein sequences obtained from RSIV-positive fish in both

Insights into red sea bream iridovirus pathogenesis: Unveiling host-pathogen interactions using membrane yeast two-hybrid and molecular dynamics.

Red sea bream iridovirus (RSIV), a member of the genus Megalocytivirus, poses a substantial threat to global aquaculture through its infection of marine fish species. The major capsid protein (MCP) represents the principal immunogenic component of RSIV, yet the mechanisms governing its pathogenicity is unclear. Here, we employed a comprehensive approach combining protein interaction screening with artificial intelligence-based structural prediction to elucidate the interface between RSIV and its host. In brief, by constructing a yeast cDNA library of membrane protein in red snapper and using a

Ferritin middle-subunit (LcFerM) from large yellow croaker (Larimichthys crocea): molecular features and immune roles against iridovirus (LYCIV) infection.

Increasing evidence indicates that viral infections can disrupt iron metabolism, thereby contributing to the onset and progression of viral infection-related diseases. Ferritin exerts immunosuppressive and proinflammatory functions during viral infection. Previous integrated transcriptomics and proteomics analyses, aimed at elucidating the resistance mechanisms of Larimichthys crocea against large yellow croaker iridovirus (LYCIV), have suggested that ferritin middle-subunit may confer a potential advantage. To elucidate the immunity-protecting roles of ferritin middle-subunit from large yello

Effect of rock bream iridovirus (RBIV) contained tissue intake on rock bream (Oplegnathus fasciatus) mortality and blood cell distribution.

Rock bream (Oplegnathus fasciatus) is one of the highly priced cultured marine fish in Korea. Rock bream iridovirus (RBIV) outbreaks in aquaculture farms may involve environmental factors, co-infection with other pathogenic microorganisms and grounded (raw) fish feed. This study evaluated the effects of RBIV-containing tissue intake on mortality and oral transmission in rock bream. Virus-containing tissues administered to rock bream [50 mg (1.53 × 108/major capsid protein, MCP gene copies) to 2400 mg (7.34 × 109)] held at 23 °C lead to 100 % mortality by 27 days post administration. Identification of Inhibitory Compounds Against Singapore Grouper Iridovirus Infection by Cell Viability-Based Screening Assay and Droplet Digital PCR.

Singapore grouper iridovirus (SGIV) is one of the major causative agents of fish diseases and has caused significant economic losses in the aquaculture industry. There is currently no commercial vaccine or effective antiviral treatment against SGIV infection. Annually, an increasing number of small molecule compounds from various sources have been produced, and many are proved to be potential inhibitors against viruses. Here, a high-throughput in vitro cell viability-based screening assay was developed to identify antiviral compounds against SGIV using the luminescent-based CellTiter-Glo reage

Entry of tiger frog virus (an Iridovirus) into HepG2 cells via a pH-dependent, atypical, caveola-mediated endocytosis pathway.

Tiger frog virus (TFV), in the genus Ranavirus of the family Iridoviridae, causes high mortality of cultured tiger frog tadpoles in China. To explore the cellular entry mechanism of TFV, HepG2 cells were treated with drugs that inhibit the main endocytic pathways. We observed that TFV entry was inhibited by NH(4)Cl, chloroquine, and bafilomycin, which can all elevate the pH of acidic organelles. In contrast, TFV entry was not influenced by chlorpromazine or overexpression of a dominant-negative form of Esp15, which inhibit the assembly of clathrin-coated pits. These results suggested that TFV

Red sea bream interleukin (IL)-1β and IL-8 expression, subcellular localization, and antiviral activity against red sea bream iridovirus (RSIV).

Interleukin-1 beta (IL-1β) is transcribed by monocytes, macrophages, and dendritic cells in response to activation of toll-like receptors (TLRs) by pathogen-associated molecular patterns (PAMPs) or cytokine signalling and causes a rapid inflammatory response to infection. IL-8, also known as chemokine C-X-C motif ligand (CXCL)-8, is regulated by IL-1β and affects the chemotaxis of macrophages and neutrophils upon pathogen infection. In healthy red sea bream, rsbIL-1β is most highly distributed in the liver, and rsbIL-8 is most highly distributed in the head kidney. In response to RSIV infectio

Protective immunity against iridovirus disease in mandarin fish, induced by recombinant major capsid protein of infectious spleen and kidney necrosis virus.

Infectious spleen and kidney necrosis virus (ISKNV) is the causative agent of a disease causing high mortality and economic losses in mandarin fish, Siniperca chuatsi in China. But little information about vaccine development against ISKNV disease is available. In this study the gene encoding the major capsid protein (MCP), which is predominant structural component of the iridovirus particles, was cloned into a temperature induction prokaryotic expression vector pBV220 and a recombinant protein was detected about 50 kDa in molecular weight and accounted for 23% of total proteins of whole cell.

The TLR9/NF-κB and cGAS/STING pathways mediate the assembly of the NLRP3 inflammasome induced by Singapore grouper iridovirus DNA.

Groupers are economically significant fish in southern China and Southeast Asia, where viral diseases severely hinder the sustainable development of aquaculture. Singapore grouper iridovirus (SGIV) is a highly pathogenic virus in groupers that can cause mass mortality upon infection. NOD-like receptor thermal protein domain associated protein 3 (NLRP3), a crucial component of innate immunity, plays vital roles in various viral infections. However, whether SGIV infection induces NLRP3 inflammasome activation and the precise mechanisms involved remain to be elucidated. In this study, we investig

Singapore grouper iridovirus utilizes Rab1 to facilitate viral trafficking and evade the immune response by targeting the STING-IRF3 pathway.

BACKGROUND: The phylum Nucleocytoviricota comprises diverse nucleocytoplasmic large DNA viruses (NCLDVs) that infect a wide range of eukaryotic hosts, yet the mechanisms underlying their intracellular transport and immune evasion remain poorly understood. Singapore grouper iridovirus (SGIV), a representative NCLDV, is a major pathogen in marine aquaculture. Rab GTPases are central regulators of vesicular trafficking and frequent targets of viral manipulation, but their roles in SGIV infection have not been elucidated. Here, we investigated the roles of Rab1 from Epinephelus coioides (EcRab1) d

Oral transmission of rock bream iridovirus (RBIV) drives delayed systemic infection and organ specific immune reprogramming in rock bream (Oplegnathus fasciatus).

Rock bream iridovirus (RBIV) is a major threat in marine fish aquaculture, but the role of oral transmission in disease progression remains poorly understood. Here, we compared intraperitoneal (i.p.) and oral infection routes in rock bream (Oplegnathus fasciatus) to evaluate viral replication, pathology, and immune responses. Fish infected via the i.p. route showed rapid systemic spread and 100 % mortality within 17-35 days post infection (dpi). In contrast, oral infection caused delayed mortality, occurring 15-20 days later, reflecting the additional mucosal barriers encountered during viral

Dual protection against grouper Rana-Iridovirus (GIV-R) and nervous necrosis virus (NNV) by novel GIV-R-Δ51-NNV-CP chimeric vaccine candidates.

Grouper Rana-iridovirus (GIV-R) and nervous necrosis virus (NNV) are two distinct yet very important causative agents in several fish species. However, as of now, no effective treatment has been developed to simultaneously prevent these two pathogens. In this study, we developed GIV-R-NNV chimeric vaccine candidates by using GIV-R to chimerically express the full length and P domain of the coat protein of NNV, and designated as GIV-R-Δ51-NNV-CP-FL, GIV-R-Δ51-NNV-CP-P, respectively. Firstly, the orf51 deletion recombinant GIV-R (GIV-R-Δ51) was constructed and characterized. The results showed t

Epigallocatechin Gallate (EGCG) Inhibits Singapore Grouper Iridovirus (SGIV) Infection by Interfering With Viral Life Cycle and Modulating Host Cell Cycle G2/M Checkpoint.

Singapore grouper iridovirus (SGIV) is a highly pathogenic virus that causes substantial economic losses in marine grouper aquaculture, and effective antiviral agents are currently lacking. In this study, the anti-SGIV activity of epigallocatechin gallate (EGCG), a natural compound from green tea, was evaluated and its underlying mechanisms were investigated. Cytotoxicity assays showed that EGCG at concentrations up to 12.5 μg/mL was safe for grouper spleen (GS) cells. EGCG treatment reduced viral MCP and VP19 mRNA expression in a dose-dependent manner, indicating its anti-SGIV activity. Mecha
