Characterization of antibody 444 using chromatographically purified enantiomers of juvenile hormones I, II, and III: implications for radioimmunoassays.

Optically pure (> 99.5%) enantiomers of insect juvenile hormones (JH) I, II, and III were obtained by injection of racemic mixtures onto a chiral HPLC column using hexane:2-propanol (99.5:0.5) as the mobile phase. The enantiomers of JH III were the best resolved (R = 4.26), followed by those of JH II (R = 2.29) and JH I (R = 1.47). These purified natural and unnatural enantiomers were used to further characterize an antiserum (444) developed for JH radioimmunoassays (RIAs). Based on ED50 values generated using optically pure [methyl-3H]-10R,11S-JH II as a tracer, the natural isomers of JH I, J

Correlations between environmental salinity levels, blood biochemistry parameters, and steroid hormones in wild juvenile American alligators (Alligator mississippiensis).

American alligators (Alligator mississippiensis) inhabit freshwater wetlands that are vulnerable to salinization caused by anthropogenic alterations to freshwater flow, in addition to storm surges, sea level rise, and droughts. Salinization of coastal freshwater habitats is a growing concern in a changing climate due to increased frequency and intensity of storm surges and drought conditions. This study opportunistically sampled juvenile male and female wild alligators in various salinities each month excluding November, December, and January for one year at Rockefeller Wildlife Refuge in coas

Simultaneous preparation of both enantiomers of juvenile hormones labeled at C-10 with tritium at high specific activity.

We report an improved method for the synthesis of high specific activity insect [10-(3)H]juvenile hormones (JH) I, II, and III which affords both enantiomers of each in high optical purity. A synthetic route for JH I was modified to give higher yields and purity. We increased the specific activity of the synthetic [10-(3)H]JHs using normal phase liquid chromatography optimized to give near baseline resolution of [10-(3)H]JHs and unlabeled JHs. Racemic [10-(3)H]JHs and their corresponding diol metabolites were enantiomerically separated using a chiral column eluted with 2-propanol:hexane. Acidi

Serum steroid hormones including 11-ketotestosterone, 11-ketoandrostenedione, and dihydroprogesterone in juvenile and adult bonnethead sharks, Sphyrna tiburo.

Previous studies in the placental viviparous bonnethead shark, Sphyrna tiburo, have correlated 17 beta-estradiol, progesterone, testosterone, and dihydrotestosterone with reproductive events in both males and females. However, several key reproductive events, including implantation, maintenance of pregnancy, and parturition, did not correlate with these four steroid hormones. Therefore, the present study investigated three steroid hormones, 11-ketotestosterone, 11-ketoandrostenedione, and dihydroprogesterone, which have demonstrably important roles in the reproductive cycles of teleosts. It wa

Cloning and functional analysis of the juvenile hormone receptor gene CsMet in Coccinella septempunctata.

The potential role of the juvenile hormone receptor gene (methoprene-tolerant, Met) in reproduction of Coccinella septempunctata L. (Coleoptera: Coccinellidae)(Coleoptera: Coccinellidae), was investigated by cloning, analyzing expression profiles by quantitative real-time PCR, and via RNA interference (RNAi). CsMet encoded a 1518-bp open reading frames with a predicted protein product of 505 amino acids; the latter contained 2 Per-Arnt-Sim repeat profile at amino acid residues 30-83 and 102-175. CsMet was expressed in different C. septempunctata larvae developmental stages and was most highly

Cloning and Characterization of Drosophila melanogaster Juvenile Hormone Epoxide Hydrolases (JHEH) and Their Promoters.

Juvenile hormone epoxide hydrolase (JHEH) plays an important role in the metabolism of JH III in insects. To study the control of JHEH in female Drosophila melanogaster, JHEH 1, 2 and 3 cDNAs were cloned and sequenced. Northern blot analyses showed that the three transcripts are expressed in the head thorax, the gut, the ovaries and the fat body of females. Molecular modeling shows that the enzyme is a homodimer that binds juvenile hormone III acid (JH IIIA) at the catalytic groove better than JH III. Analyses of the three JHEH promoters and expressing short promoter sequences behind a reporte

Anti-Müllerian hormone and Oestradiol as markers of future reproductive success in juvenile gilts.

There is a need for an early marker for reproductive success in gilts as the traditional process for selecting breeding females is inefficient. There is evidence that circulating anti-Müllerian hormone (AMH) is indicative of ovarian reserve, antral follicle populations, gonadotropin responsiveness and fertility in various species other than the pig. Additionally, oestradiol (E2) has been shown to mark antral follicle populations in cattle and pregnancy outcomes in women, after gonadotropin treatment. The aims of this study were to determine whether 1) serum levels of AMH or E2, prior to or aft

Allatotropin inhibits juvenile hormone biosynthesis by the corpora allata of adult Bombyx mori.

Juvenile hormone (JH) synthesis by the corpora allata (CA) does not occur during the pupal stage in both male and female Bombyx mori but begins shortly before adult ecdysis and thereafter only in females. JH biosynthesis in female adults was prevented by allatotropin (AT) through the corpora cardiaca (CC) and the tightly attached oesophagus before adult ecdysis, but after ecdysis, removal of the CC had little effect and only the oesophagus was necessary for AT to prevent JH synthesis. AT could not prevent JH synthesis by the CA alone in either stage. Short neuropeptide F (sNPF) acted directly

Transient hypothyroxinemia in juvenile glycoprotein hormone subunit B5 knock-out mice.

The heterodimer thyrostimulin, comprised of two novel glycoprotein hormone subunits GPA2 and GPB5, activates the TSH receptor. To understand its role in the regulation of the hypothalamus-pituitary-thyroid (HPT-) axis, we evaluated juvenile and adult GPB5 knock-out (GPB5(-/-)) and wild type mice (WT) during euthyroidism, hypothyroidism and thyrotoxicosis. Surprisingly, juvenile euthyroid GPB5(-/-) mice displayed marked hypothyroxinemia (25% lower serum T(4), unchanged TSH) and also during thyrotoxicosis juvenile GPB5(-/-) mice had 25% lower serum T(4), compared to WT. During hypothyroidism, de

Effects of growth hormone treatment in juvenile idiopathic arthritis: bone and body composition.

BACKGROUND: Inflammation and glucocorticoid therapy are major factors influencing growth and bone maturation in patients with juvenile idiopathic arthritis (JIA). In addition to alterations in total bone mineral density and bone geometry, longitudinal data confirm that the main contributors to errant bone maturation in patients with JIA are reductions in muscle mass and force. Growth hormone (GH) therapy, which has shown efficacy in controlling disease, may also positively influence body composition. For several years, GH therapy has been used to treat growth retardation in patients with JIA r

Allatostatic regulation of juvenile hormone production in vitro by the ring gland of Drosophila melanogaster.

A factor(s) extracted from the third instar larval brains of the dipteran species Sarcophaga bullata and Drosophila melanogaster causes a dose-dependent reduction of juvenile hormone (JHB3) biosynthesis by isolated ring glands in vitro. In situ, this factor is presumably neuronally transmitted from the brain to ring gland. The allatostatic effect of the brain factor is reversible in vitro and may be overcome partially by the JHB3 precursor farnesoic acid. Agents which act to increase the intracellular levels of cAMP (3-isobutyl-1-methylxanthine (MIX), forskolin, 8-benzoyl cAMP) all caused the

Effects of recombinant human growth hormone in juvenile Nile crocodiles (Crocodylus niloticus).

1. Recombinant human growth hormone (hGH) showed somatotropic activity in juvenile Nile crocodiles (Crocodylus niloticus). 2. Body weight of crocodiles receiving 3.25 micrograms hGH/g body weight twice a week was increased by 49% after five weeks of treatment, compared to 31% increase in controls. 3. Total length was increased by 15 and 5%, respectively, in the two groups. 4. Food conversion efficiency increased from 28% in the controls to 36% in the hormone injected animals. 5. Cessation of hormone treatment was followed by reduced appetite and decreasing body growth.

Juvenile hormone degradation enzymes have shared and unique requirements in Drosophila development.

UNLABELLED: Precise control of hormones is essential for animal development. Hormone bioavailability is regulated by their synthesis, transport, sequestration, natural turnover, and programmed degradation. Here, we use Drosophila melanogaster to investigate developmentally programmed degradation of the retinoid-like juvenile hormones (JHs). JHs promote juvenile growth and timely development by functionally opposing the steroid hormone, ecdysone. Despite well-characterized biochemistry, the developmental requirements for programmed JH degradation are poorly understood due to paralogue expansion

Juvenile hormone receptor relies on conserved domain interfaces for dimerization-dependent activity.

Juvenile hormone (JH) is vital to insects. In Drosophila, JH signals through a basic helix-loop-helix-PER-ARNT-SIM (bHLH-PAS) protein methoprene-tolerant (MET) and its paralog germ cell-expressed (GCE). Binding of JH to the PAS-B domain of MET or GCE triggers the assembly of a DNA-bound, transcriptionally active JH receptor (JHR) heterodimer comprising either MET or GCE and their partner taiman (TAI). Molecular mechanisms of the JHR complex activation remain unclear. Here, we use the Drosophila GCE protein to address the presumably critical but as yet unexplored function of the other PAS domai

Reproduction and Wing Differentiation of Gynoparae Are Regulated by Juvenile Hormone Signaling in Aphis gossypii.

Gynopara is a specific winged type in the life history of Aphis gossypii. As a key reproduction mode between parthenogenesis and sexual reproduction, it lays sexual females in late autumn. However, little is known about gynoparae because of its rare presence in the wild and the lack of its detailed descriptions. In this study, we investigated the morphological characteristics, ovary maturation process, and key signaling pathways during the development of gynoparae of A. gossypii. With the extension of development stage, the gynoparae exhibited gradually enlarged and thickened abdomen, deepened

Juvenile hormone affects the splicing of Culex quinquefasciatus early trypsin messenger RNA.

The full length of Culex quiquefasciatus early trypsin has been cloned and sequenced and a three-dimensional (3D) model of the enzyme was built showing that the enzyme has the canonical trypsin's active pocket containing H78, D123, S129, and D128. The biosynthesis of juvenile hormone (JH) III by the corpora allata (CA) in female Cx. quiquefasciatus is sugar-dependent. Females that were maintained on water after emergence synthesize very little JH III, JH III bisepoxide, and methyl farnesoate (MF) (3.8, 1.1, and 0.8 fmol/4 hr/CA, respectively). One hour after sugar feeding, the synthesis of JH

Cardiac changes induced by excess exogenous growth hormone in juvenile miniature poodles.

The transient elevated plasma growth hormone (GH) levels that occur at a young age in giant breed dogs may have consequences in adult life. The aim of this study was to investigate whether excess juvenile GH has consequences for cardiac function and morphology. To simulate the naturally occurring juvenile hypersomatotropism in giant breed dogs, elevated plasma GH and insulin-like growth factor-I (IGF-I) concentrations were induced in six miniature poodles (GH dogs) by daily administration of supraphysiological doses of GH starting at 12 weeks of age. Eight miniature poodles of the same age tha

Target of rapamycin (TOR) mediates the transduction of nutritional signals into juvenile hormone production.

Anautogeny is a reproductive strategy by which females do not reproduce until they feed. Therefore, nutritional signals must inform the reproductive tissues, and cells that the organism has reached a nutritional status suitable for triggering reproductive processes. One of the possible pathways involved in anautogeny is the "target of rapamycin" (TOR) pathway, which has been described as connecting the nutritional status with growth, proliferation, and cancer. The German cockroach, Blattella germanica, is an anautogenous species whose vitellogenesis is governed by juvenile hormone. In the pres

Effect of fumigation and juvenile hormone on oxygen consumption of the larvae of Corcyra cephalonica (Stainton).

Reduced effect of JH-I, JH-II and JH-III on oxygen consumption of II-V instars and increased effect on the oxygen consumption of VI instar larvae suggested that control of corcyra with juvenile hormones could be brought about only when applied to just emerged VI instar larvae. Similarly fumigation of juvenile hormone treated larvae could prove beneficial only at VI instar stage. The possibility of enhancing the effect of fumigant with pretreatment of JH will be futile as even only JH treated VI instar larvae develop into abnormal individuals which die later.
