Rho GAP myosin IXa is a regulator of kidney tubule function.

Mammalian class IX myosin Myo9a is a single-headed, actin-dependent motor protein with Rho GTPase-activating protein activity that negatively regulates Rho GTPase signaling. Myo9a is abundantly expressed in ciliated epithelial cells of several organs. In mice, genetic deletion of Myo9a leads to the formation of hydrocephalus. Whether Myo9a also has essential functions in the epithelia of other organs of the body has not been explored. In the present study, we report that Myo9a-deficient mice develop bilateral renal disease, characterized by dilation of proximal tubules, calyceal dilation, and

Response of primary rabbit kidney proximal tubule cells to estrogens.

The effects of estrogens on the growth and function of primary rabbit kidney proximal tubule (RPT) cells have been examined in hormonally defined phenol red-free medium. 17beta-estradiol was observed to stimulate growth at dosages as low as 10(-10) M. The growth stimulatory effects of 17beta-estradiol were mitigated in the presence of hydrocortisone, suggesting that these two steroid hormones acted at least in part by common mechanisms. The effects of other steroids known to interact with the estrogen receptor were examined. Alpha estradiol was found to be growth stimulatory over a concentrati

[Fixation of tubules with radioactive technetium dimercaptosuccinate in parenchymal diseases of the kidney].

In order to evaluate tubular function 99mTc-DMS dynamic kidney scintigraphy and 99mTc-DMS uptake determinations were performed in 55 patients with endemic nephropathy, diabetic nephropathy and glomerulo nephritis. From 64-656th segment of computerized radiorenograms 99mTc-DMS uptake and curve slope were assessed and compared with the corresponding parameters of 20 healthy subjects. Both parameters were decreased in patients of all three groups. Tubular uptake of 99mTc-DMS measured during 60 sec. 4 hours after 99mTc-DMS administration, was decreased in endemic nephropathy and diabetic nephropat

The distribution of leptospires in the kidney tubules of some British wild mammals.

Histological examination of 69 pairs of infected kidneys from 12 species of Rodentia, Lagomorpha, Carnivora and Insectivora revealed that leptospires were confined mainly to the proximal and distal convoluted tubule, were less often found in the thick loop of Henle and only rarely in the collecting duct. On no occasion were the organisms present in the thin loop of Henle. Preliminary observations on the relationship of leptospires to tubule epithelium indicate some degree of physical attachment. It is suggested that the avoidance of the thin loop of Henle might be a reflection of its structura

The kidney distal tubule potassium switch, modern diet and hypertension.

Hypertension is the leading contributor to premature death and disability in the world. Genetic susceptibility and high dietary salt (NaCl) consumption have long been considered to be the primary culprits but growing evidence indicates that low dietary potassium consumption has an equally important role. Although the underlying mechanisms are complex and multifactorial, the potassium switch signalling mechanism in the kidney distal convoluted tubule represents a crucial pathway with implications for preventing and treating hypertension. Comprising a Kir4.1 and Kir5.1 channel potassium-sensing

Proximal Tubule Secretory Clearance, Injury, and Kidney Viability in Cirrhosis.

INTRODUCTION: Cirrhosis affects all structures of the kidney, in particular the tubules, which are responsible for secretion of protein-bound metabolites and electrolyte/water homeostasis. Yet, prevailing assessments of kidney function focus solely on glomerular filtration rate (GFR), which may incompletely reflect these processes. We sought to characterize markers of tubular function, injury, and viability in patients with and without cirrhosis. METHODS: We recruited outpatients undergoing liver transplantation evaluation for a collection of plasma and 24-hour urine, matching by GFR to contr

Guided tissue organization and disease modeling in a kidney tubule array.

Three-dimensional (3D) in vitro kidney tubule models have either largely relied on the self-morphogenetic properties of the mammalian cells or used an engineered microfluidic platform with a monolayer of cells cultured on an extracellular matrix (ECM) protein coated porous membrane. These systems are used to understand critical processes during kidney development and transport properties of renal tubules. However, high variability and lack of kidney tubule-relevant geometries among engineered structures limit their utility in disease research and pre-clinical drug testing. Here, we report a no

A protein with anion exchange properties found in the kidney proximal tubule.

One important mechanism for reabsorption of chloride in the kidney proximal tubule involves anion exchange of chloride for a base. Anion exchange transport systems in general demonstrate sensitivity to inhibition by disulfonic stilbenes, probenecid, furosemide, and the arginyl amino group modifier phenylglyoxal. Using disulfonic stilbene affinity chromatography, we have identified and partially purified a protein with anion exchanger properties in luminal membrane vesicles isolated from rabbit kidney cortex. This protein has a molecular weight of 162 kD. The binding of the 162 kD protein to th

Microvilli-Mediated Transport and Internalization of Extracellularly Assembled Nanoparticles in Kidney Proximal Tubules.

Proximal tubules, with their unique microenvironment and specialized brush-border microvilli, actively reabsorb, metabolize, and process filtered molecular cargos, yet how they interact with renal-clearable nanomaterials after glomerular filtration remains poorly understood. Here, using reactive oxygen species (ROS)-responsive glutathione-coated Au11 nanoclusters (∼0.6 nm) as a model, we show that renal-filtered Au11 undergoes extracellular ROS-mediated biotransformation into 2-3 nm gold nanoparticles, followed by their assembly into large nanostructures (∼50-500 nm) within the proximal tubula

Hydrogen sulfide accelerates the recovery of kidney tubules after renal ischemia/reperfusion injury.

BACKGROUND: Progression of acute kidney injury to chronic kidney disease (CKD) is associated with inadequate recovery of damaged kidney. Hydrogen sulfide (H2S) regulates a variety of cellular signals involved in cell death, differentiation and proliferation. This study aimed to identify the role of H2S and its producing enzymes in the recovery of kidney following ischemia/reperfusion (I/R) injury. METHODS: Mice were subjected to 30 min of bilateral renal ischemia. Some mice were administered daily NaHS, an H2S donor, and propargylglycine (PAG), an inhibitor of the H2S-producing enzyme cystath

Hyaline droplet accumulation in rodent kidney proximal tubules: an association with histiocytic sarcoma.

Since recognition during the last decade that certain renal carcinogens can initially cause an accumulation of hyaline (protein) droplets in proximal tubules of male rats, it has become appropriate to establish whether this phenomenon of protein overload can also occur in rodent kidneys unrelated to chemical treatment. Kidney tissue from a number of selected rodent studies held in the National Toxicology Program (NTP) or Food and Drug Administration (FDA) archives were evaluated for hyaline droplet accumulation in proximal tubules. The survey concentrated on rats and mice of both sexes bearing

PCO2 measurements in surface proximal tubules and peritubular capillaries of the rat kidney.

PCO2 was measured in surface proximal tubules and peritubular capillaries in the rat under normal acid-base conditions and in three settings with decreased HCO3(-) reabsorption: benzolamide administration, respiratory alkalosis, and metabolic acidosis. Under normal conditions, PCO2 in the early proximal tubule (EP) was 10.5 mmHg higher than PaCO2 (P less than 0.001) and 3-4 mmHg higher than late proximal (LP) and peritubular capillary (PC) PCO2 (P less than 0.001). PCO2 in LP and PC was 7 mmHg higher than PaCO2 (P less than 0.001). Benzolamide (3 mg/kg) had no effect on the difference between

Characteristics of glutamine metabolism by rat kidney tubules: a carbon and nitrogen balance.

The metabolism of glutamine by a suspension of rat kidney tubules was studied in vitro. The influence of duration of incubation, glutamine concentration, and metabolic state of the donor animals was investigated. The relative importance of glucose synthesis, amino acid production, and oxidation to CO2 was estimated by drawing a complete balance of the nitrogens and the carbon chains of the extracted glutamine. It was found that the initial (first 15 min) rate of glutamine utilization was significantly greater than the subsequent rate due to an initial, but transient, extracellular accumulation

Stimulation by human calcitonin of electrolyte transport in distal tubules of rat kidney.

The effects of human calcitonin (HCT) on the distal tubule were investigated by micropuncture in hormone-deprived rats, i.e. in the absence of parathyroid hormone, antidiuretic hormone and glucagon, which might have masked these effects. Two groups of rats were studied: hormone-deprived and hormone-deprive + HCT, infused at 1.0 mU/min X 100 g.b.w. In the urine, HCT markedly reduced Ca and Mg excretion whereas excretion of water, Na and K was not significantly affected. Along the distal tubule, HCT strongly enhanced Na, Cl, Mg, Ca and total solute reabsorption, decreased K secretion but did not

A Newly Identified Protective Role of C5a Receptor 1 in Kidney Tubules against Toxin-Induced Acute Kidney Injury.

Acute kidney injury (AKI) remains a major reason for hospitalization with limited therapeutic options. Although complement activation is implicated in AKI, the role of C5a receptor 1 (C5aR1) in kidney tubular cells is unclear. Herein, aristolochic acid nephropathy (AAN) and folic acid nephropathy (FAN) models were used to establish the role of C5aR1 in kidney tubules during AKI in germline C5ar1-/-, myeloid cell-specific, and kidney tubule-specific C5ar1 knockout mice. After aristolochic acid and folic acid injection, C5ar1-/- mice had increased AKI severity and a higher degree of tubular inju

Plasma Biomarkers of Kidney Tubule Health during Ambulatory AKI and Kidney Function Recovery in SPRINT.

KEY POINTS: AKI frequently occurs in the ambulatory setting, but there are no available tools to distinguish whether kidney function will recover. Plasma biomarkers reflecting kidney tubular injury and dysfunction measured at the time of AKI are associated with nonrecovery in kidney function. Measuring plasma biomarkers of kidney tubule health during ambulatory AKI episodes could be helpful in guiding treatment decisions. BACKGROUND: AKI frequently occurs in the ambulatory setting, but few tools are available to distinguish whether an individual's kidney function will subsequently recover. M

Immune-mediated tubule atrophy promotes acute kidney injury to chronic kidney disease transition.

Incomplete repair after acute kidney injury can lead to development of chronic kidney disease. To define the mechanism of this response, we compared mice subjected to identical unilateral ischemia-reperfusion kidney injury with either contralateral nephrectomy (where tubule repair predominates) or contralateral kidney intact (where tubule atrophy predominates). By day 14, the kidneys undergoing atrophy had more macrophages with higher expression of chemokines, correlating with a second wave of proinflammatory neutrophil and T cell recruitment accompanied by increased expression of tubular inju

Dysfunction of the circadian clock in the kidney tubule leads to enhanced kidney gluconeogenesis and exacerbated hyperglycemia in diabetes.

The circadian clock is a ubiquitous molecular time-keeping mechanism which synchronizes cellular, tissue, and systemic biological functions with 24-hour environmental cycles. Local circadian clocks drive cell type- and tissue-specific rhythms and their dysregulation has been implicated in pathogenesis and/or progression of a broad spectrum of diseases. However, the pathophysiological role of intrinsic circadian clocks in the kidney of diabetics remains unknown. To address this question, we induced type I diabetes with streptozotocin in mice devoid of the circadian transcriptional regulator BMA

Selective depletion of mouse kidney proximal straight tubule cells causes acute kidney injury.

The proximal straight tubule (S3 segment) of the kidney is highly susceptible to ischemia and toxic insults but has a remarkable capacity to repair its structure and function. In response to such injuries, complex processes take place to regenerate the epithelial cells of the S3 segment; however, the precise molecular mechanisms of this regeneration are still being investigated. By applying the "toxin receptor mediated cell knockout" method under the control of the S3 segment-specific promoter/enhancer, Gsl5, which drives core 2 β-1,6-N-acetylglucosaminyltransferase gene expression, we establi

Mapping the cytoskeletal architecture of renal tubules and surrounding peritubular capillaries in the kidney.

The human kidney includes ~1 million nephrons which are long U-shaped tubules with convoluted segments that serve as filtration units. During the passage of the ultrafiltrate through a nephron, electrolytes and nutrients are re-absorbed into peritubular capillaries. The fluid remaining in the distal end of the renal tubules flows through the collecting ducts into the ureter. In this study, we generated high-resolution images of mouse kidney sections using confocal microscopy with only two fluorescently tagged biomarkers, F-actin binding phalloidin and CD34 antibodies as a marker for blood vess
