Effects of constituent amino acids on tubular handling of microinfused angiotensin II.

[14C]angiotensin II ([14C]AII) was microinjected alone or with excess L-isoleucine (IIe) or L-aspartic acid (Asp) into renal tubules of anesthetized rats. Urinary excretion of 14C-labeled material was measured, and the intact peptide and its metabolites were identified and quantified. When isoleucine was administered with [14C]AII, urinary recovery of the 14C-labeled material increased directly with distance of infusion site from glomerulus, and most of the radioactivity in urine was [14C]Ile. The data suggest that isoleucine interfered with reabsorption of [14C]Ile derived from hydrolysis of

Localization of aristolochic acid in mouse kidney tissues by immunohistochemistry using an anti-AA-I and AA-II monoclonal antibody.

Aristolochic acids (AAs) are found in herbal medicines of Aristolochiaceae plants, including Aristolochia and Asarum species. AAs are associated with a rapidly progressive interstitial nephritis, which is called aristolochic acid nephropathy (AAN). However, the in-situ localization of AAs in the target organ, the kidney, has not been investigated yet. In the present study, the accumulation of aristolochic acid I (AA-I) in mouse kidney was revealed by immunoperoxidase light microscopy as well as colloidal gold immunoelectron microscopy (IEM) based on an anti-AA-I and AA-II monoclonal antibody (

Atubular glomeruli in renal artery stenosis.

The aim of the present study was to investigate and quantitate the structural renal changes developing after stenosis of the renal artery. Twelve kidneys removed at operation from patients with elevated blood pressure due to stenosis of the renal artery were investigated together with three kidneys with renal artery stenosis but no clinical evidence of renovascular hypertension. Eight age-matched autopsy kidneys served as controls. Stereologic methods were used to estimate the volume fractions of different structural parameters, the volume of individual glomeruli, and the structural integrity

Distal tubular epithelial cells of the kidney: Potential support for proximal tubular cell survival after renal injury.

The tubular epithelium of the kidney is susceptible to injury from many causes, such as ischemia-reperfusion and the associated oxidative stress, nephrotoxins, inflammatory and immune disorders and many others. The outcome is often acute kidney injury, which may progress to chronic kidney disease and fibrosis. Acute kidney injury involves not only direct injury to the distal tubular (DT) and proximal tubular (PT) epithelium during and immediately following the injurious event, but the closely-associated and sometimes dysfunctional renal vascular endothelium also plays an important part in modu

Spatio-temporal patterning of different connexins in developing and postnatal human kidneys and in nephrotic syndrome of the Finnish type (CNF).

Connexins (Cxs) are membrane-spanning proteins which enable flow of information important for kidney homeostasis. Changes in their spatiotemporal patterning characterize blood vessel abnormalities and chronic kidney diseases (CKD). We analysed spatiotemporal expression of Cx37, Cx40, Cx43 and Cx45 in nephron and glomerular cells of developing, postnatal kidneys, and nephrotic syndrome of the Finnish type (CNF) by using immunohistochemistry, statistical methods and electron microscopy. During kidney development, strong Cx45 expression in proximal tubules and decreasing expression in glomeruli w

[Bilateral renal hypoplasia with oligomeganephronie. A morphometric, light- and electronmicroscopic study (author's transl)].

Kidney biopsy specimens of three patients with a special form of hypoplasia, the oligomeganephronic hypoplasia (Oligomeganephronie) were studied with light- and electron-microscopy as well as morphometric methods. The morphometric measurements demonstrated that there were no quantitative differences of the glomerular components between normal and oligomeganephronic glomeruli of patients of the same age. All of the structures measured--glomerular capillary surface, glomerular volume, volume of the glomerular capillary lumina and volume of Bowman's capsular space--were increased about 5.4 times.

Role of inwardly rectifying K+ channel 5.1 (Kir5.1) in the regulation of renal membrane transport.

PURPOSE OF REVIEW: Kir5.1 interacts with Kir4.2 in proximal tubule and with Kir4.1 in distal convoluted tubule (DCT), connecting tubule (CNT) and cortical collecting duct (CCD) to form basolateral-K+-channels. Kir4.2/Kir5.1 and Kir4.1/Kir5.1 play an important role in regulating Na+/HCO3--transport of the proximal tubule and Na+/K+ -transport in the DCT/CNT/CCD. The main focus of this review is to provide an overview of the recent development in the field regarding the role of Kir5.1 regulating renal electrolyte transport in the proximal tubule and DCT. RECENT FINDINGS: Loss-of-function-mutati

Crystallisation properties in stone forming and normal subjects' urine diluted using a standardised procedure to match the composition of urine in the distal part of the distal tubule and the middle part of the collecting duct.

Using a standardised procedure, we assessed the crystallisation properties of calcium phosphate in urine with a composition matching that in the distal part of the distal tubules (DTd) and of calcium oxalate in urine with a composition matching that in the mid-collecting duct (CDm). We used 8-h urine samples collected between 2200 h and 0600 h with sodium azide as preservative. Urine from ten patients with recurrent CaOx stone formation and from ten normal subjects was used for the measurements. The DTd and CDm samples were obtained by diluting the voided 8-h urine to 3000 ml and 1750 ml per 1

Diuretic effects on calcium metabolism.

Diuretics have numerous effects on calcium metabolism. The loop diuretics promote, and the thiazide diuretics inhibit, renal calcium excretion. In this review we detail the basic mechanisms of renal calcium excretion and then explain how diuretics influence this excretion. Finally we review how these agents can be used to alter calcium homeostasis in a clinically efficacious manner.

Characterization of distal hydrogen ion secretion in acute respiratory alkalosis.

The effect of acute respiratory alkalosis (ARA) on distal nephron H+ secretion was evaluated by measuring urine-to-blood (U-B) Pco2 in dogs with highly alkaline urine (urine pH greater than 7.8). ARA led to a significant decrease in U-B Pco2 and in urine HCO3 concentration; urine pH, however, increased significantly, indicating that the decrease in urine Pco2 was of greater magnitude than the decrease in urine HCO3 concentration. For any given urine HCO3 concentration urine Pco2 was lower (i.e., urine pH was higher) in ARA than in controls. Administration of tris(hydroxymethyl)aminomethane (Tr

Distinction between proximal and distal regulations of sodium and potassium excretion in humans.

The clearances of creatinine (CCr), lithium (CLi), sodium (CNa), and potassium (CK) were determined during three consecutive 2-hour periods in a large number of physically healthy persons who were in lithium treatment for manic-depressive illness. CLi was used as a measure of the proximal tubular fluid output (Vprox). CLi showed considerable spontaneous variations from one 2-hour period to another, and neither bladder emptying errors nor changes of CCr could account for these. The variations of CLi, which reflected variations of Vprox, resulted in joint variations of CNa and CK. Also the ratio

Incomplete Distal Renal Tubular Acidosis and Kidney Stones.

Renal tubular acidosis (RTA) is comprised of a diverse group of congenital or acquired diseases with the common denominator of defective renal acid excretion with protean manifestation, but in adults, recurrent kidney stones and nephrocalcinosis are mainly found in presentation. Calcium phosphate (CaP) stones and nephrocalcinosis are frequently encountered in distal hypokalemic RTA type I. Alkaline urinary pH, hypocitraturia, and, less frequently, hypercalciuria are the tripartite lithogenic factors in distal RTA (dRTA) predisposing to CaP stone formation; the latter 2 are also commonly encoun

Micropuncture studies of glucose transport in the dog: mechanism of renal glycosuria.

Clearance and micropuncture studies were performed in 23 dogs without glucose loading to examine the tubule mechanism of renal glycosuria. Studies were carried out in three groups of animals before and after 10% extracellular volume expansion, and administration of maleic acid in low dose at 150 mumol/kg and in high dose at 300 mumol/kg. Specific hexokinase methods were used for the determination of glucose in tubule fluid and urine. Under control conditios, glucose reabsorption occurred predominantly in the proximal tubule. In all three groups, proximal tubule reabsorption of both sodium and

Mechanisms and regulation of renal magnesium transport.

Magnesium's most important role is in the release of chemical energy. Although most magnesium is stored outside of the extracellular fluid compartment, the regulated value is blood magnesium concentration. Cellular magnesium and bone magnesium do not play a major role in the defense of blood magnesium concentration; the major role is played by the kidney, where the renal tubule matches the urinary magnesium excretion and the net entry of magnesium into the extracellular fluid. In the kidney, magnesium is reabsorbed in the proximal tubule, the thick ascending limb of the loop of Henle, and the

Stem cell marker TRA-1-60 is expressed in foetal and adult kidney and upregulated in tubulo-interstitial disease.

The kidney has an intrinsic ability to repair itself when injured. Epithelial cells of distal tubules may participate in regeneration. Stem cell marker, TRA-1-60 is linked to pluripotency in human embryonic stem cells and is lost upon differentiation. TRA-1-60 expression was mapped and quantified in serial sections of human foetal, adult and diseased kidneys. In 8- to 10-week human foetal kidney, the epitope was abundantly expressed on ureteric bud and structures derived therefrom including collecting duct epithelium. In adult kidney inner medulla/papilla, comparisons with reactivity to epithe

"Histological and Immunohistochemical Characterization of the Kidney and Adrenal Gland in Nile Monitor Lizards (Varanus niloticus)".

This study provides a detailed histological and immunohistochemical analysis of the kidney and adrenal gland in Varanus niloticus (Nile monitor lizard), highlighting their structural features and adaptive mechanisms. Ten adult female Nile monitors were collected from Qena Province, Egypt, and their kidneys and adrenal glands were examined. Kidney tissues were processed for histological analysis, and immunohistochemistry was performed to evaluate the expression of key markers, including vimentin, E-cadherin, CK7, and NSE. In contrast to other reptiles, the kidney was divided into the cortex and

Measurement of nephron filtration from distal and proximal tubules.

Nephron filtration rate (SNGFR), measured proximally by micropuncture using the technique of total collection of tubular fluid, may be spuriously high as the fall in intratubular pressure during collection may increase the net filtration pressure. Alternatively, the interrupted distal delivery of tubular fluid during proximal collection may up-regulate SNGFR by activating the tubulo-glomerular feedback. The accuracy of nephron filtration measurements was tested in 65 rats, by sampling from the distal and then from the proximal tubule of the same nephrons. Collections were also made in the pres
