Development of a CRISPR/Cas12a genome editing toolbox in Kluyveromyces marxianus and its application in succinic acid biosynthesis.

Kluyveromyces marxianus is a promising thermotolerant yeast for industrial biotechnology, but lacks efficient genome engineering tools. A CRISPR/Cas12a genome editing toolbox for K. marxianus was developed for the first time in this study. A plasmid-free transient system achieved single-gene knockout efficiencies of about 50 %-100 % in Δku70 strain. Even with homology arms as short as 35 bp, the knockout efficiency remained 66.67 %. Chromosomal integration of Cas12a enabled single-to-triple fragment knock-ins efficiency of 82.93-85.70 % and 94.50 % for large fragment (>5 kb) integrations. Appl

The hypoxic transcription factor KlMga2 mediates the response to oxidative stress and influences longevity in the yeast Kluyveromyces lactis.

Hypoxia is defined as the decline of oxygen availability, depending on environmental supply and cellular consumption rate. The decrease in O2 results in reduction of available energy in facultative aerobes. The response and/or adaptation to hypoxia and other changing environmental conditions can influence the properties and functions of membranes by modifying lipid composition. In the yeast Kluyveromyces lactis, the KlMga2 gene is a hypoxic regulatory factor for lipid biosynthesis-fatty acids and sterols-and is also involved in glucose signaling, glucose catabolism and is generally important f

Kluyveromyces marxianus: a promising alternative to baker's yeast Saccharomyces cerevisiae for enhanced oven rise performance during bread baking.

For millennia, bread making has relied on Saccharomyces cerevisiae, valued for its efficient sugar fermentation, robustness under processing stresses and production of a desirable flavour profile. Nevertheless, there is growing interest in exploring alternative yeast species that show advantageous traits beyond the physiological boundaries of S. cerevisiae. For example, a yeast's ability to stimulate dough oven rise is of particular interest because it determines loaf volume. It was recently shown for S. cerevisiae that dough oven rise strongly correlates with yeast viability and thermotoleran

Engineering Kluyveromyces marxianus for 3-hydroxypropionic acid production at elevated temperature from Jerusalem artichoke tubers and crude glycerol.

This study engineered the thermotolerant yeast Kluyveromyces marxianus to produce 3-hydroxypropionic acid (3-HP), a key precursor for biodegradable plastics, via the malonyl-CoA pathway using non-food feedstocks. The 3-HP titer was further increased through deleting Adh2A and Ach1, which prevents the synthesis of byproducts ethanol and acetic acid. Using Jerusalem artichoke tuber powder, engineered strain produced 27.32 and 32.31 g/L of 3-HP at 37 °C and 42 °C through fed-batch fermentation. Metabolic reconstruction replaced the native FADH2-dependent glycerol pathway (GUT1/GUT2) with an NADH-

The lysine-rich C-terminal repeats of the centromere-binding factor 5 (Cbf5) of Kluyveromyces lactis are not essential for function.

The gene coding for the centromere-binding factor 5 (CBF5) of Kluyveromyces lactis has been isolated by hybridization of a Saccharomyces cerevisiae CBF5 DNA probe to a K. lactis library. The amino acid sequence of KlCbf5 is highly homologous, 88% identity, to ScCbf5, but also to the rat protein Nap57 (64% identity). The main difference between both yeast proteins and the rat protein is the presence of a lysine-rich domain with KKE/D repeats in the C-terminal part of the protein. These repeats are thought to be involved in binding of the protein to microtubules. Deletion of the KKE/D domain in

Production of mannitol from molasses and Jerusalem artichoke tubers at elevated temperatures using engineered Kluyveromyces marxianus through consolidated bioprocessing.

In this study, an engineered Kluyveromyces marxianus strain with enhanced mannitol biosynthesis capacity was developed, enabling efficient conversion of molasses and Jerusalem artichoke tubers into mannitol via consolidated bioprocessing. An efficient mannitol-producing strain was constructed by screening of five mannitol synthesis-related genes, with mannitol dehydrogenase (CmMDH) from Candida magnoliae identified as the most effective enzyme. Mannitol production was further enhanced by increasing the copy number of CmMDH. Among the four tested NADPH synthesis-related genes, overexpression of

Physiological characterization of Kluyveromyces marxianus strains for the development of a microbial platform to obtain bioethanol from cheese whey permeate.

The increasing interest in renewable feedstocks for biochemicals production includes the valorization of cheese whey (CW), a by-product of the dairy industry. CW contains a high concentration of lactose, accounting for most of its organic load, making it a major environmental pollutant if untreated. A valorization approach of CW is based on the separation of valuable proteins by ultrafiltration, and the obtainment of a lactose-rich cheese whey permeate (CWP). The conversion of lactose in CWP into bioethanol is considered a sustainable solution for the valorization of this waste. However, lacto

The Biotherapeutic Potential of a Novel Probiotic Kluyveromyces marxianus Isolated from a Sourdough Starter Against Vaginal Candida albicans Strains.

There is an increasing interest in yeasts isolated from natural sources to be used as probiotics. Saccharomyces-based probiotics have been proposed as a valid alternative to the conventional drug therapy for the prevention and treatment of vulvovaginal candidiasis, also considering the resistance of some Candida strains to many antifungals. Here, we isolated from an artisanal sourdough a new yeast strain which was identified as Kluyveromyces marxianus and assessed its probiotic and safety properties, which resulted in comparable properties to all those exhibited by the commercial probiotic Sac

Reprogramming a Hgt-family Kluyveromyces marxianus sugar transporter by site-directed mutagenesis to enable co-consumption of glucose and xylose.

Efficient utilization of lignocellulosic hydrolysates in yeast-based biorefineries requires simultaneous consumption of glucose and xylose, which is often limited by preferential glucose uptake. In Kluyveromyces marxianus, we kinetically characterized two native xylose transporters, KMAR_10 531 and KMAR_60 179, identifying medium- and low-affinity xylose transporters, respectively. KMAR_10 531 also mediated high-affinity glucose uptake (Km 0.28 ± 0.1 mM), limiting xylose utilization in mixed-sugar media. Guided by structural modelling, we engineered the KMAR_10 531 N325V variant, which reduced

Glutathione S-transferase ϴ-subunit as a phenotypic suppressor of pmr1Δ strain, the Kluyveromyces lactis model for Hailey-Hailey disease.

BACKGROUND: Hailey-Hailey disease (HHD), also known as familial benign chronic pemphigus, is a rare, chronic and recurrent blistering disorder, histologically characterized by suprabasal acantholysis. HHD has been linked to mutations in ATP2C1, the gene encoding the human adenosine triphosphate (ATP)-powered calcium channel pump. METHODS: In this work, the genetically tractable yeast Kluyveromyces lactis has been used to study the molecular basis of Hailey-Hailey disease. The K. lactis strain depleted of PMR1, the orthologue of the human ATP2C1 gene, was used to screen a Madin-Darby canine ki

Enhancing ascorbic acid delivery and stability by yeast microencapsulation in vitro and food matrices: A comparative study of Candida, and Kluyveromyces species.

Ascorbic acid (vitamin C) is highly unstable and susceptible to degradation under heat, oxygen, and moisture, which significantly limits its shelf life and functional efficiency in food and pharmaceutical systems. Therefore, developing natural and efficient delivery systems to improve its protection and controlled release have a critical role in food science. This study aimed to enhance the stability and release behavior of ascorbic acid by entrapping it into plasmolysed (P) and non-plasmolysed (NP) yeast cells of Candida utilis and Kluyveromyces lactis. Within these systems, non-plasmolysed K

Functional analysis of the single Est1/Ebs1 homologue in Kluyveromyces lactis reveals roles in both telomere maintenance and rapamycin resistance.

Est1 and Ebs1 in Saccharomyces cerevisiae are paralogous proteins that arose through whole-genome duplication and that serve distinct functions in telomere maintenance and translational regulation. Here we present our functional analysis of the sole Est1/Ebs1 homologue in the related budding yeast Kluyveromyces lactis (named KlEst1). We show that similar to other Est1s, KlEst1 is required for normal telomere maintenance in vivo and full telomerase primer extension activity in vitro. KlEst1 also associates with telomerase RNA (Ter1) and an active telomerase complex in cell extracts. Both the te

Sugar reduction and associated biochemical changes in pomegranate (Punica granatum L.) fruit juice during non-alcoholic fermentation by Kluyveromyces marxianus.

The reduction of high fructose and glucose content without affecting other important biochemicals is a challenge for healthier pomegranate juice, which was attempted using non-alcoholic fermentation by Kluyveromyces marxianus (isolated from kefir grains). For optimization of sugar reduction, static fermentation was undertaken with varying temperature (20 to 37C), pH (3.5, 4.0), and duration (up to 21 days). The reduction of fructose, glucose and formation of ethanol was measured periodically using HPLC for 21 days. The maximum sugar reduction by 34% (glucose 42% and fructose 18%) without ethan

Enhanced expression of glycolysis regulators KmGcr1p and KmGcr2p in Kluyveromyces marxianus: An efficient strategy for high-temperature ethanol production from inulin.

Bioethanol production from non-food biomass has emerged as a sustainable alternative to satisfy future energy demands. Kluyveromyces marxianus stands out as a potential consolidated bioprocessing (CBP) strain for high-temperature ethanol production from inulin. Nevertheless, enhancing its fermentation efficiency remains crucial. Here, we characterized its glycolysis regulator KmGcr2p and demonstrated that KmGcr2p boosts the expression of target genes controlled by another glycolysis regulator, KmGcr1p, through their interaction. Furthermore, we engineered the strain Ogcr1 + 2 through co-overex

Highly efficient transactivation by the yeast Kluyveromyces lactis transcription factor LAC9 and its inhibition by the negative regulator GAL80 in mammalian cells.

Expression of the LAC9 gene from the yeast Kluyveromyces lactis in HepG2 human hepatoblastoma cells efficiently induced luciferase expression from reporter plasmids containing the four LAC9 binding sites from the K. lactis GAL1-GAL10 gene linked to a basal promoter. Induction was approximately 100fold and was dependent on the presence of the UAS sequence and an intact reading frame in the LAC9 gene. Additional cotransfection of constructs expressing the K. lactis GAL80 gene reduced luciferase activity by up to 98%. This inhibition was not affected by addition of 14mM galactose to the medium. N

Development of a multi-copy integration platform in Kluyveromyces marxianus enabled by a computational method for genome-wide identification of multi-copy integration loci.

Multi-copy integration is a core strategy for redirecting metabolic flux toward target compounds. However, its application has been hampered by the absence of methods for systematically identifying native multi-copy genomic loci. To overcome this, we developed a computational procedure for genome-wide identification of such loci. Theoretically, this method is potentially applicable to any genome-sequenced species as it only requires the genomic assembly of the target species as input. Applying the procedure to Kluyveromyces marxianus, we identified four groups of loci (KmCS1-4). Combining thes

Analyzing the effect of glucose availability on the ethyl acetate production by Kluyveromyces marxianus under iron limitation conditions by metabolic flux analysis approach.

To address waste management challenges, lignocellulosic industrial co-products can be valorized microbially to propose sustainable and economically viable alternatives to fossil routes. This study advances a bio-based ethyl acetate microbial production by the yeast Kluyveromyces marxianus by investigating glucose availability under iron-limiting fed-batch conditions. Two feeding strategies were compared: one that maintained an excess of glucose and one that operated at zero residual glucose to understand their respective effects on ethyl acetate synthesis dynamics.
