VigorBaby dietary supplement administration to mice during pregnancy and lactation alleviated ovarian disorders induced by Zearalenone in offsprings.

Zearalenone (ZEN) is a mycotoxin with estrogenic activity that is widely present in cereals. We exposed pregnant and lactating mice to 40 μg/kg body weight (bw) of ZEN to confirm and extend previous observational reports. The current study demonstrated that ZEN exposure increased reactive oxygen species (ROS), DNA damage, and mitochondrial dysfunction in the ovaries of neonatal offspring, leading to primordial follicle (PF) impairment. Moreover, before puberty, such dysfunction resulted in impairment of oocyte maturation (evaluated as their capacity to resume and complete meiosis) and ability

Association of gestational diabetes with other lactation-related disorders: A 2-sample Mendelian randomization analysis of causal effects.

Gestational diabetes mellitus (GDM) is associated with adverse metabolic outcomes and may also be related to postpartum breast and lactation disorders. Observational studies are susceptible to confounding and reverse causation. We used a 2-sample Mendelian randomization design to examine the association between genetic liability to GDM and other disorders of the breast and lactation associated with childbirth. Genetic liability to GDM was associated with higher odds of a composite phenotype of breast and lactation disorders associated with childbirth. Replication using independent samples and

Exposure to Kalach, a Glyphosate-Based Herbicide, During Pregnancy and Lactation Induces Hypothyroidism and Bone Disorders in Rat Offspring.

Kalach (KL) is a glyphosate (G)-based herbicide extensively used in agricultural and urban areas in Tunisia. It has been reported that G crosses the placenta in pregnant rats, potentially disrupting organ function in offspring. The present study examined the effects of prenatal and lactational exposure to KL on thyroid function, bone integrity, and phosphocalcic homeostasis in rat offspring. Pregnant rats were divided into two groups, group A (control group) and group B, exposed to KL (each mother rat received 0.07 mL of KL diluted in 1 mL of water by gavage). On postnatal day 14, plasma sampl

Protein Restriction During Pregnancy and Lactation Triggers Steatohepatitis and Adrenal Disorders in Rat Dams: Insights into Maternal Metabolic Health.

BACKGROUND: The Developmental Origins of Health and Disease (DOHaD) concept is strongly supported by epidemiological and experimental data demonstrating that exposure to adverse conditions early in life can impact offspring health. Despite numerous studies showing the early and long-term impacts of maternal malnutrition on offspring, little research has been conducted on its effects on mothers. METHODS: We investigated the effects of protein restriction (6%) during gestation and lactation on the metabolism, as well as the structural and molecular parameters of the livers and adrenals of dams.

Lactate Sensors for Early Detection of Metabolic Disorders in Dynamic Muscle Tissue Models.

Lactate monitoring is essential for the diagnosis and management of metabolic disorders, such as mitochondrial myopathies. To detect these conditions, assessment of lactate levels during dynamic physiological states such as exercise is a promising approach. However, conventional metabolite sensors can be limited by poor stability under mechanical stress, including muscle contractions, and slow response times, hindering their utility in capturing real-time lactate dynamics. We report a biocompatible, acutely implantable lactate sensor on a flexible substrate embedded with a lactate oxidase enzy

Automated detection of health disorders in lactating dairy cattle on pasture: a preliminary study.

Since previous health monitoring systems have shown themselves to be unsuccessful in predicting health disorders in dairy cows managed on pasture, the aim of this study was to evaluate the performance of automated health monitoring integrated in an accelerometer-based oestrus detection system (ODS) for dairy cows on pasture. Mixed-breed lactating dairy cows (n=109) in a seasonal-calving herd managed at pasture were fitted with an ODS that provided automated health monitoring. The ODS performed multimetric analysis of behavioural patterns to generate health alerts. Data were collected during th

Abnormal activity-dependent brain lactate and glutamate+glutamine responses in panic disorder.

BACKGROUND: Prior evidence suggests panic disorder (PD) is characterized by neurometabolic abnormalities, including increased brain lactate responses to neural activation. Increased lactate responses could reflect a general upregulation of metabolic responses to neural activation. However, prior studies in PD have not measured activity-dependent changes in brain metabolites other than lactate. Here we examine activity-dependent changes in both lactate and glutamate plus glutamine (glx) in PD. METHODS: Twenty-one PD patients (13 remitted, 8 symptomatic) and 12 healthy volunteers were studied.

Distinct panicogenic activity of sodium lactate and cholecystokinin tetrapeptide in patients with panic disorder.

BACKGROUND: The validity of experimentally induced panic attacks as a model to study the pathophysiology of panic disorder has been questioned. Unspecific, unpleasant and aversive effects as well as specific patterns of psychovegetative symptoms pointing to different subtypes of panic disorder patients have been observed. These findings raise the question of challenge paradigms as a valuable tool to identify different vulnerabilities in patients with panic disorder. METHODS: We compared the two most widely studied panicogenic drugs sodium lactate and cholecystokinine tetrapeptide (CCK-4) with

[Warning signals in acute abdominal disorders. Lactate is the best marker of mesenteric ischemia].

An increased plasma lactate concentration (PLC) is a recognized danger signal often found in cases of shock, septicaemia, hepatic and renal failure, and diabetic ketoacidosis. In 120 patients with abdominal complaints, we found the PLC to be above normal limits in 96 per cent (24/25) of the mesenteric ischaemia subgroup, in all 20 of the general bacterial peritonitis subgroup, in 30 per cent (6/20) of the acute pancreatitis subgroup, and in about half of the 25 cases of intestinal obstruction. In all other abdominal conditions represented (n = 30), comprising various inflammatory or infectious

The emerging role of lactate in skeletal homeostasis and disorders: Integrated mechanisms and translational opportunities.

UNLABELLED: Lactate has evolved from a metabolic byproduct to a central regulator of skeletal homeostasis and disorders, integrating metabolic flux with epigenetic reprogramming. Within the skeletal system, it modulates the processes of osteogenesis, osteoclastogenesis, chondrogenesis, and matrix synthesis by regulating the physiological activities of bone marrow mesenchymal stem cells, osteoblasts, osteoclasts, chondrocytes, and intervertebral disc cells, thereby maintaining skeletal homeostasis. Dysregulated lactate signaling is increasingly recognized as a pivotal pathological mechanism in

Diagnostic Utility of Sodium Lactate Infusion and CO2-35% Inhalation for Panic Disorder.

Laboratory measures have played an integral role in diagnosing pathology; however, compared to traditional medicine, psychiatric medicine has lagged behind in using such measures. A growing body of literature has begun to examine the viability and development of different laboratory measures in order to diagnose psychopathologies. The present review examines the current state of development of both sodium lactate infusion and CO2-35% inhalation as potential ancillary measures to diagnose panic disorder (PD). A previously established 3-step approach to identifying laboratory-based diagnostic te

Elevated serum lactate following hyperventilation during glucose infusion in panic disorder.

Early investigators reported that patients with anxiety syndromes associated with panic attacks produced more lactate during exercise than control subjects. These studies suggested a metabolic difference between patients and controls. However, the possibility that patients were simply less fit than controls could not be excluded. In this study, serum lactate was measured in panic disorder patients in response to a metabolic challenge not involving exercise. Voluntary hyperventilation during iatrogenic hyperglycemia led to an increase in serum lactate. The increase in serum lactate was signific

Lactate increase and oxygen desaturation in mitochondrial disorders--evaluation of two diagnostic screening protocols.

BACKGROUND: Mitochondrial disorders are characterized by an accumulation of lactate and an insufficient oxygen extraction from blood during exercise. Therefore, both parameters (lactate and oxygen saturation) can be used as screening tests in mitochondrial disorders. However, conflicting results regarding sensitivities and specifities of both tests have been reported. METHOD: We examined 27 patients with genetically defined mitochondrial disorders (single deletions n = 15, multiple deletions n = 5, A3243G mutation n = 7), patients with other neuromuscular disorders, and healthy controls. Impact of postpartum health disorders on metabolic status and reproductive performance of lactating dairy cows.

This study evaluated the impact of the presence of diseases within the first 21 days in milk (DIM) in dairy cows on metabolic status after the voluntary waiting period (VWP), conception at first service, infectious disease seroconversion, and the association between metabolic status at first artificial insemination (AI) and conception at first service. Cows were classified as sick (n = 48) or healthy (n = 75) based on diseases within 21 DIM. Blood samples were obtained at first AI (D0), pregnancy diagnosis (D30), and pregnancy confirmation (D60) after the first AI. Samples were analyzed for gl

Association of Lactation Initiation and Duration with Blood Pressure Recovery in Individuals with a Hypertensive Disorder of Pregnancy.

BACKGROUND: Hypertensive disorders of pregnancy (HDPs) are associated with an increased risk of long-term maternal cardiovascular disease. Lactation is associated with favorable cardiometabolic profiles, but its impact on blood pressure (BP) recovery following an HDP is less clear. OBJECTIVE: We sought to assess the relationship of lactation initiation and duration with BP among individuals with prior HDP. MATERIALS AND METHODS: We used prospectively collected data from individuals with prepregnancy overweight or obesity with HDP and no prepregnancy hypertension who participated in a postpar

Insufficient Lactation Leading to Postpartum Diagnosis of Placenta Accreta Spectrum Disorder in a Primigravid Patient.

BACKGROUND: Placenta accreta spectrum is most commonly diagnosed antenatally or at the time of delivery, but it may also present in the postpartum period. CASE: A 29-year-old primigravid patient without risk factors for placenta accreta spectrum had an uncomplicated vaginal birth with normal blood loss and delivery of an intact-appearing placenta. Five days postpartum, she was not lactating and uterine imaging to evaluate for retained products of conception was suspicious for placenta accreta spectrum. She began to develop bleeding in the following days and elected for definitive management.

Lactate in the brain: an update on its relevance to brain energy, neurons, glia and panic disorder.

Lactate is considered an important metabolite in the human body, but there has been considerable debate about its roles in brain function. Research in recent years has suggested that lactate from astrocytes may be crucial for supporting axonal function, especially during times of high metabolic demands or hypoglycemia. The astrocyte-neuron lactate transfer shuttle system serves a protective function to ensure a supply of substrates for brain metabolism, and oligodendrocytes appear to also influence availability of lactate. There is increasing evidence for lactate acting as a signaling molecule

Physiological and behavioral effects of naloxone and lactate in normal volunteers with relevance to the pathophysiology of panic disorder.

This study investigates whether naloxone, an opioid receptor antagonist, could render normal controls, normally nonresponsive to panic inducing stimuli, sensitive to the physiological and behavioral effects of sodium lactate, a robust panicogen in panic disorder patients. Twelve normal controls received intravenous naloxone followed by sodium lactate. Four of these subjects underwent a separate infusion with naloxone followed by saline. Respiratory physiological symptoms were measured throughout. Clinical symptoms, assessed by the Acute Panic Inventory (API), an Anxiety Scale, and the Borg Bre

High-fat intake during lactation ameliorates cardiac fatty acid metabolic disorders and dysfunction in mouse offspring undergoing prenatal poly (I:C) stimulation.

Prenatal inflammation exposure (PIE) is associated with increased prevalence of cardiovascular diseases (CVDs) in offspring, including heart failure and hypertension. In this study, we investigated the molecular mechanisms underlying the prenatal programming of cardiac function. Pregnant mice were injected with poly (I:C) (20 mg/kg, i.p.) on day 10.5 of gestation. Mothers and pubs were fed with high-fat diet during lactation. Cardiac tissues of the offspring were collected for analysis. We found that prenatal poly (I:C) exposure significantly reduced fatty acid metabolism and impaired the home
