Glycol chitosan stabilized nanomedicine of lapatinib and doxorubicin for the management of metastatic breast tumor.

Advanced breast cancer is known to be highly evasive to conventional therapeutic regimes with a 5-year survival rate of less than 30% compared to over 90% for early stages. Although several new approaches are being explored to improve the survival outcome, there is still some room for equipping existing drugs such as lapatinib (LAPA) and doxorubicin (DOX) to fight the systemic disease. LAPA is associated with poorer clinical outcomes in HER2-negative patients. However its ability to also target EGFR has warranted its use in recent clinical trials. Nevertheless, the drug is poorly absorbed post

ErbB expression, activation, and inhibition with lapatinib and tyrphostin (AG825) in human vestibular schwannomas.

BACKGROUND: Pharmacologic agents targeted against the ErbB family, or the intracellular pathways that mediate their effects, could slow clinical progression of vestibular schwannoma (VS) in patients where other modalities carry a high risk-to-benefit ratio. OBJECTIVE: Determine the identity of the predominant ErbB dimer partners in VS tumors and assess the effects of targeted inhibition of the ErbB molecules on VS growth and proliferation, as well as apoptosis. SETTING: Academic tertiary referral center. METHODS: Coimmunoprecipitation and Western blotting of VS tissue, in vitro BrdU assays

Mechanism of lapatinib-mediated radiosensitization of breast cancer cells is primarily by inhibition of the Raf>MEK>ERK mitogen-activated protein kinase cascade and radiosensitization of lapatinib-resistant cells restored by direct inhibition of MEK.

BACKGROUND AND PURPOSE: We recently showed that lapatinib, an EGFR/HER2 inhibitor, radiosensitized breast cancer cells of the basal and HER2+ subtypes. The purpose of this study was to identify the downstream signaling pathways responsible for lapatinib-mediated radiosensitization in breast cancer. MATERIALS AND METHODS: Response of EGFR downstream signaling pathways was assessed by Western blot and clonogenic cell survival assays in breast tumor cells after irradiation (5Gy), lapatinib, CI-1040, or combined treatment. RESULTS: In SUM102 cells, an EGFR+ basal breast cancer cell line, exposur

Dermatological Adverse Events Associated With Lapatinib Treatment in Dogs With Urothelial Carcinoma: A Retrospective Study.

BACKGROUND: Lapatinib is widely used in human oncology; however, dermatological adverse events (DAEs) are common and have been correlated with treatment efficacy. In veterinary medicine, lapatinib use in combination with piroxicam has been shown to be effective in treating canine urothelial carcinoma (UC); however, the incidence and prognostic significance of DAEs in dogs remain unknown. OBJECTIVE: To evaluate the DAEs in dogs with UC treated with lapatinib and piroxicam. ANIMALS: Eighty-five dogs with UC were treated with lapatinib/piroxicam and 42 were treated with piroxicam alone. MATERI

Investigation of the antitumor activity of lapatinib and NNC 55-0396 dihydrochloride combination in gastric adenocarcinoma.

Targeted drugs such as Lapatinib and Everolimus have limited efficacy in the treatment of gastric cancer due to heterogeneous tumor structure. Combination therapies may offer a promising alternative to overcome this limitation. In this study, the potential synergistic effects of the combination of T-type calcium channel blocker NNC 55-0396 dihydrochloride (NNC) with Lapatinib and Everolimus were investigated in gastric cancer cell lines. In the first stage of the experiment, all three drugs were applied to AGS and SNU-1 cell lines at different doses, and IC50 values were calculated. Since Lapa

PI3K independent activation of mTORC1 as a target in lapatinib-resistant ERBB2+ breast cancer cells.

Therapies targeting the ERBB2 receptor, including the kinase inhibitor lapatinib (Tykerb, GlaxoSmithKline), have improved clinical outcome for women with ERBB2-amplified breast cancer. However, acquired resistance to lapatinib remains a significant clinical problem, and the mechanisms governing resistance remain poorly understood. We sought to define molecular alterations that confer an acquired lapatinib resistance phenotype in ER-/ERBB2+ human breast cancer cells. ERBB2-amplified SKBR3 breast cancer cells were rendered resistant to lapatinib via culture in increasing concentrations of the dr

Interfacial lipid digestion-responsive injectable nanoemulsions enhance lapatinib efficacy in triple-negative breast cancer.

Lapatinib (LPT), a brick-dust kinase inhibitor, shows slow oral bioavailability and dose-limiting gastrointestinal side effects that frequently lead to therapeutic discontinuation. Increasing oral bioavailability does not mitigate these adverse effects, and developing complex LPT delivery systems is challenging due to its poor solubility in organic or lipid solvents. Here, we identified a synergistic lipid-surfactant nanoemulsion system comprising Miglyol 812-oleic acid (1:1) oil phase, Solutol HS-15-TPGS (1:2) surfactant, and PEG-400 cosurfactant as a solubilization space for LPT and formulat

Lapatinib-loaded reductive-responsive hyaluronic acid-cholesterol nanoparticles for inhibiting metastasis of uveal melanoma.

Uveal melanoma (UM) is the most common intraocular primary malignancy in adults with highly metastatic characteristics. Currently, there are no effective therapies to prevent metastasis formation in UM, resulting in a poor prognosis. Herein, we report a novel lapatinib-loaded reductive-responsive nanoparticle platform prepared via the self-assembly of amphiphilic hyaluronic acid-cystamine-cholesteryl hemisuccinate conjugate to suppress the distant metastasis of UM. The platform can maintain a stable nanosphere structure in the physiological environment and effectively deliver the drug to UM tu

Design, synthesis, and antiviral activity of fragmented-lapatinib aminoquinazoline analogs towards SARS-CoV-2 inhibition.

The severe impact of COVID-19 on global health and economies highlights the critical need for innovative treatments. Recently, lapatinib, a drug initially used for breast cancer, has been identified as a potential inhibitor of the main protease (Mpro) of SARS-CoV-2, meriting further investigation. Utilizing rational design strategies and guided by MD simulations, we developed novel aminoquinazoline analogs based on fragmented lapatinib's structure. Preliminary computational screenings identified promising candidates, which were synthesized using a concise 3-4 step process. In vitro assays demo

Comparative pharmacokinetics of tyrosine kinase inhibitor, lapatinib, in dogs and cats following single oral administration.

Lapatinib is an orally administered tyrosine kinase inhibitor used to treat human epidermal growth factor receptor 2 (HER2) -overexpressing breast cancers in humans. Recently, the potential of lapatinib treatment against canine urothelial carcinoma or feline mammary tumor was investigated. However, the pharmacokinetic studies of lapatinib in dogs and cats are not well-defined. In the present study, the pharmacokinetic characteristics of lapatinib in both cats and dogs after a single oral administration at a dose of 25 mg/kg were compared with each other. Lapatinib was administered orally to fo

The efficacy of lapatinib and nilotinib in combination with radiation therapy in a model of NF2 associated peripheral schwannoma.

Neurofibromatosis type 2 (NF2), a neurogenetic condition manifest by peripheral nerve sheath tumors (PNST) throughout the neuroaxis for which there are no approved therapies. In vitro and in vivo studies presented here examine agents targeting signaling pathways, angiogenesis, and DNA repair mechanisms. In vitro dose response assays demonstrated potent activity of lapatinib and nilotinib against the mouse schwannoma SC4 (Nf2 -/-) cell line. We then examined the efficacy of everolimus, nilotinib, lapatinib, bevacizumab and radiation (RT) as mono- and combination therapies in flank and sciatic n

Effective Treatment of Solitary Pituitary Metastasis with Panhypopituitarism in HER2-Positive Breast Cancer by Lapatinib.

Brain metastasis affects one third of patients with HER2-positive breast cancer after treatment with trastuzumab. Surgical resection and radiation therapy are often unsuccessful at accomplishing complete control of metastasis. Lapatinib is presumed to cross the blood-brain barrier, and exhibits clinical activities for treatment of HER2-positive breast cancer. A 43-year-old woman was treated for early breast carcinoma with total mastectomy, axillary lymph-node dissection, and adjuvant chemotherapy with cyclophosphamide plus doxorubicin. After the end of adjuvant trastuzumab therapy, she was dia

Nuclear-Targeted Boron-Lapatinib Hybrid for Enhanced Boron Neutron Capture Therapy in Advanced Thyroid Carcinoma.

To enhance the effectiveness of boron neutron capture therapy (BNCT) for advanced thyroid carcinoma (ATC), we evaluated two previously synthesized boron-enriched derivatives of Lapatinib incorporating high-boron clusters. Lapatinib, a tyrosine kinase inhibitor (TKI), is clinically used in targeted therapy against tumor proliferation and progression. The compounds consisted of Lapatinib-pharmacophore linked either to an ortho-carborane cluster (Cmpd-1) or to a metallacarborane cobalta-bis-dicarbollide (COSAN) anion (Cmpd-2). Trefoil Factor 1 contributes to Paclitaxel and Lapatinib resistance in brain metastatic breast cancer cells.

Late occurrence of breast cancer brain metastases in patients pretreated with systemic therapies is often associated with therapy resistant brain metastatic lesions. Prior studies targeting previously identified mediators of resistance have yielded limited success, especially in the case of brain metastatic breast cancer (BMBC). Trefoil Factor 1 (TFF1) has been associated with therapy resistance in Estrogen Receptor (ER) positive breast cancer. However, its role in therapy resistance of triple negative and HER2+ breast cancer remains elusive. Herein, we assessed the involvement of TFF1 in ther

Rapamycin Partially Reverts Cavernoma Endothelial Cell Phenotype and, When Combined With Lapatinib, Ameliorates Chronic Lesions.

This study investigates the impact of rapamycin and propranolol on cerebral cavernous malformations (CCMs). Employing an unbiased transcriptomic analysis, we aimed to comprehensively elucidate the molecular mechanisms underlying these drug effects. Mouse Brain Microvascular Endothelial Cells (mBMEC) deficient in Ccm3 were treated with propranolol or rapamycin and were analysed by RNA-seq and immunofluorescence. While propranolol shows limited efficacy in modulating the CCM transcriptomic phenotype in mBMEC, rapamycin demonstrates a significant impact. Rapamycin partially reverses gene expressi

Integrative proteo-genomic profiling uncovers key biomarkers of lapatinib resistance in HER2-positive breast cancer.

INTRODUCTION: Drug resistance is a major obstacle to the long-term effectiveness of cancer therapies. Approximately 70% of breast cancer patients relapse after 5 years of treatment, and the lack of biomarkers associated with drug resistance translates to poor prognosis in the clinic. Previous research has utilised omics approaches to uncover biomarkers driving drug resistance, with a strong emphasis on genetic mutations. METHODS: Here, we identified a nine-marker signature associated with resistance to lapatinib in a HER2-positive breast cancer model using a target discovery approach by emplo
