Assessing Intraoperative Laser Speckle Contrast Imaging of Parathyroid Glands in Relation to Total Thyroidectomy Patient Outcomes.

Background: Accurate assessment of parathyroid gland vascularity is important during thyroidectomy to preserve the function of parathyroid glands and to prevent postoperative hypocalcemia. Laser speckle contrast imaging (LSCI) has been shown to be accurate in detecting differences in parathyroid vascularity. In this surgeon-blinded prognostic study, we evaluate the relationship between intraoperative LSCI measurements and postoperative outcomes of total thyroidectomy patients. Methods: Seventy-two thyroidectomy patients were included in this study. After thyroid resection, an LSCI device was u

Effects on perfusion by stretching and rotation of forehead skin flaps, as measured by laser speckle contrast imaging.

PURPOSE: Flaps are used for repair of defects after tumor excision and often need stretching and rotating to cover the defects. Manipulation of flaps may affect blood perfusion, which is crucial for healing. The aim was to examine the impact on perfusion by stretching and rotating forehead skin flaps. METHODS: Blood perfusion was monitored using laser speckle contrast imaging in 19 flaps, raised as part of a direct brow lift. Perfusion was measured before and after stretching (1 and 2 N), and after 90° rotation. RESULTS: Perfusion decreased along the flap, reaching a minimum plateau at 13 mm

Hyperspectral and Laser Speckle Contrast Imaging for Monitoring the Effect of Epinephrine in Local Anesthetics in Oculoplastic Surgery.

PURPOSE: Epinephrine is used in local anesthetics to induce vasoconstriction and thus reduce bleeding and prolong the anesthetic effect. Finding the optimal delay between the administration of the anesthetic and skin incision to ensure vasoconstriction and minimize bleeding is important and has recently become the subject of debate. This is the first study to assess blood perfusion and oxygen saturation (sO 2 ) simultaneously in response to a local anesthetic containing epinephrine in human oculoplastic surgery. METHODS: A local anesthetic consisting of lidocaine and epinephrine (20 mg/ml + 1

DASHING-LSCI: deep learning-based anatomical multiclass segmentation for human intraoperative neurosurgical guidance - laser speckle contrast imaging.

Laser speckle contrast imaging (LSCI) provides real-time, label-free visualization of cerebral blood flow in vessels and perfusion in cortical tissue during neurosurgery. At present, surgeons interpret LSCI-derived flow and perfusion changes qualitatively between microsurgical maneuvers, which limits continuous quantitative analysis. Automated delineation of blood vessels and cortical tissue is a prerequisite for such analysis. Here we evaluate a deep learning approach for multiclass segmentation of intraoperative LSCI images into blood vessels, cortical tissue, and background (e.g., surgical

The effect of foot temperature on laser speckle contrast imaging (LSCI) and transcutaneous oxygen pressure (TcPO2) measurements.

BACKGROUND/AIM: Assessment of foot microcirculation is increasingly implemented into clinical practice to assess vascular status and wound-healing potential in patients with vascular disease when conventional assessment techniques provide limited or unreliable information. Transcutaneous oxygen pressure (TcPO2) and laser speckle contrast imaging (LSCI) are techniques used to assess microcirculation in the foot. However, foot temperature is currently not considered during TcPO2 and LSCI measurements, despite substantial variation between patients and across measurements. Determining its influen

Protocol for Standardized Assessment of Systemic Microvascular Function in Human Cutaneous Microcirculation Using Laser Speckle Contrast Imaging.

Laser speckle contrast imaging (LSCI) is a high-resolution, non-invasive optical technique that enables real-time, full-field visualization of microvascular blood perfusion. This protocol presents a standardized methodology for evaluating systemic microvascular function in human cutaneous microcirculation using LSCI. Because measurements obtained with this technique are highly sensitive to environmental and physiological confounders, the protocol emphasizes strict standardization procedures to improve reproducibility and experimental reliability. The protocol details essential environmental co

Time-resolved laser speckle contrast imaging (TR-LSCI) of cerebral blood flow response to intracranial pressure elevation.

SIGNIFICANCE: Cerebral autoregulation (CA) reflects the dynamic coupling among cerebral blood flow (CBF), intracranial pressure (ICP), and arterial blood pressure (ABP); its failure contributes to secondary brain injury. Existing bedside methods rely on indirect or spatially limited CBF surrogates and cannot resolve microvascular flow dynamics across space, depth, and time. AIM: To develop, optimize, and apply a scalable, noncontact time-resolved laser speckle contrast imaging (TR-LSCI) platform for depth-sensitive, high-speed, wide-field CBF imaging during controlled ICP perturbations. Real-time quantification of bowel perfusion using Laparoscopic Laser Speckle Contrast Imaging (LSCI) in a porcine model.

BACKGROUND/PURPOSE: Real-time quantification of tissue perfusion can improve intraoperative surgical decision making. Here we demonstrate the utility of Laser Speckle Contrast Imaging as an intra-operative tool that quantifies real-time regional differences in intestinal perfusion and distinguishes ischemic changes resulting from arterial/venous obstruction. METHODS: Porcine models (n = 3) consisted of selectively devascularized small bowel loops that were used to measure the perfusion responses under conditions of control/no vascular occlusion, arterial inflow occlusion, and venous outflow o

Real-time monitoring of cerebral blood flow by laser speckle contrast imaging after cardiac arrest in rat.

Cardiac arrest (CA) results in global brain ischemia. To explore the role of cerebral blood flow (CBF) during ischemia, laser speckle contrast imaging (LSCI), a full-field high-resolution optical imaging technique, was used for real-time monitoring of the fluctuations of CBF in a rat model of asphyxial-CA. The temporal changes of CBF were characterized and the relationship between CBF and mean arterial pressure (MAP) was evaluated. Asphyxial-CA led to transient CBF dysregulation, manifested by changes in CBF velocity were significantly impacted by MAP. Hyperemia is aligned with a bolus injecti

The monitoring of microvascular liver blood flow changes during ischemia and reperfusion using laser speckle contrast imaging.

OBJECTIVE: The recovery of microvascular liver blood flow (LBF) after ischemia is an important determinant of the degree of hepatocellular injury. Laser speckle contrast imaging (LSCI) was recently suggested to be a suitable instrument for monitoring the LBF. This study was designed to evaluate LSCI in monitoring the LBF changes during liver ischemia and reperfusion (IR). METHODS: A rat model with 120-min ischemia and 60-min reperfusion to 90% of the liver (entire liver except the caudate lobe, which was kept as portal blood bypass) was used. The LBF of the sham operation (SO) group and the I

[Effects of high salt diet on renal vascular reactivity in mice detected by laser speckle contrast imaging].

The aim of this study was to conduct in vivo experiments using laser speckle contrast imaging (LSCI) technology to investigate the effects of high salt diet on renal vascular reactivity in mice. LSCI is a technology for monitoring blood flow based on the laser speckle principle. It has been widely used to detect microcirculatory functions in tissues such as the skin and brain. The kidneys are located behind the peritoneum, and their position is easily affected by the movement of abdominal organs. Measuring renal microcirculation in a living individual is difficult. The present study used a sel

Laser speckle contrast imaging for assessment of abdominal visceral microcirculation in acute peritonitis: does sequential impairments exist?

OBJECTIVE: It is believed that the microcirculation of multiple organs is impaired during acute peritonitis, however whether distinct susceptibilities of visceral microvasculature exist is still unknown. The present study aims to verify whether the microcirculatory alterations occur sequentially among multiple abdominal viscera during acute peritonitis. MATERIALS AND METHODS: Acute peritonitis was achieved on 29 Sprague-Dawley rats through colon ascendens stent peritonitis (CASP) model. With laser speckle contrast imaging (LSCI), the microcirculation of the liver, ileum and renal cortex was m

Laser speckle contrast imaging for intraoperative assessment of intestinal microcirculation in normo- and hypovolemic circulation in a porcine model.

INTRODUCTION: Healing is essential for successful colorectal surgery. Optimal microcirculation is needed to ensure this; however, this is only subjectively assessed by the surgeon. Laser Speckle Contrast Imaging (LSCI) is an objective non-contact, image-based method to quantify microcirculation in bowel ends. This study aimed to evaluate the application of LSCI in an open surgery porcine model, determine differences between normal and impaired microcirculation, and test the LSCI applicability to repeated measurements. METHOD: A midline laparotomy was made in ten healthy female pigs to expose

Paired Cisterna Magna Nanoinjection and Laser Speckle Contrast Imaging Assay to Study Cerebral Blood Flow Regulation In Vivo.

Cerebral blood flow (CBF) regulation is crucial for maintaining cognitive functions, but its underlying mechanisms remain poorly understood. Investigating the cellular and molecular processes involved in this regulation requires a pharmacological approach that allows precise drug delivery into cerebral spinal fluid (CSF) while enabling real-time monitoring of cerebral blood flow dynamics. In this paper, we present a method for studying cerebral blood flow regulation in vivo by combining nanoinjector-based intracisternal magna (ICM) injection with laser speckle contrast imaging. The nanoliter i

Laser Speckle Contrast Imaging-based diagnosis of severe mesenteric traction syndrome: Hemodynamics and prostacyclin - A prospective cohort study.

BRIEF ABSTRACT: Today, the diagnosis and grading of mesenteric traction syndrome relies on a subjective assessment of facial flushing. However, this method has several limitations. In this study, Laser Speckle Contrast Imaging and a predefined cut-off value are assessed and validated for the objective identification of severe mesenteric traction syndrome. BACKGROUND: Severe mesenteric traction syndrome (MTS) is associated with increased postoperative morbidity. The diagnosis is based on an assessment of the developed facial flushing. Today this is performed subjectively, as no objective metho

Laser speckle contrast imaging for blood flow monitoring in predicting outcomes after cerebral ischemia-reperfusion injury in mice.

BACKGROUND: In the treatment of ischemic cerebral stroke (ICS), most conventional treatments, including carotid endarterectomy and carotid artery stenting, may cause cerebral ischemia-reperfusion injury (CIRI). For treated ICS patients, changes in cerebral blood flow are directly related to brain function. At present, computed tomography perfusion, dynamic susceptibility contrast-enhanced perfusion weighted imaging and magnetic resonance arterial spin labeling perfusion imaging are used to monitor cerebral blood flow, but they still have some limitations. Our study aimed to monitor the changes

Laser speckle contrast imaging for quantitative assessment of facial flushing during mesenteric traction syndrome in upper gastrointestinal surgery.

The mesenteric traction syndrome (MTS) is associated with prostacyclin (PGI2) facilitated systemic vasodilatation during surgery and is identified by facial flushing. We hypothesized that severe facial flushing would be related to the highest concentrations of plasma PGI2 and accordingly to the highest levels of skin blood flow measured by laser speckle contrast imaging (LSCI). Patients scheduled for major upper abdominal surgery were consecutively included. Within the first hour of the procedure, facial flushing was scored according to a standardized scale, and skin blood flow (LSPU) was cont

Characterization of age-related penile microvascular hemodynamic impairment using laser speckle contrast imaging: possible role of increased fibrogenesis.

Current technology for penile hemodynamic evaluations in small animals is invasive and has limitations. We evaluated a novel laser speckle contrast imaging (LSCI) technique to determine age-related changes in penile microvascular perfusion (PMP) and tested the role of cavernosal muscle (CC) fibrosis mediated by Wnt-TGF β1 signaling pathways in a mouse model. Ten young (2-3 months) and old (24-28 months) wild-type C57BL6 male mice were subjected to PMP measured using a LSCI system. Penile blood flow (PBF, peak systolic velocity, PSV) was also measured using a color Doppler ultrasound for compar
