In vivo antimalarial activity of a probiotic bacterium Lactobacillus sakei isolated from traditionally fermented milk in BALB/c mice infected with Plasmodium berghei ANKA.

ETHNOPHARMACOLOGICAL RELEVANCE: Milk production, processing and consumption are integral part of traditional practices in Fulani tribe of Cameroon. It has been observed that Fulani are resistant to malaria. Dairy products traditionally processed by Fulani are intensively used in the ritual treatment of malarial, inflammations and behavioural disorders. Many studies have demonstrated that fermented milk is a rich source of probiotic bacteria. However, the antimalarial activity of probiotics isolated from this natural source has not been experimentally tested. AIM OF THE STUDY: Hence, this stud

Bacterial metataxonomic analysis of the Algerian traditional dried-salted meat 'El Kaddid' and characterization of its lactic acid bacteria.

This study investigated the indigenous bacterial diversity of 'El Kaddid', a traditional Algerian dried-salted meat, using a culture-independent approach. In parallel, culture-dependent methods were applied specifically to isolate and identify the lactic acid bacteria (LAB) population. Microbial fingerprinting of ten 'El Kaddid' samples using 16S rRNA gene (V3-V4 region) sequencing and subsequent sequence analysis revealed complex bacterial communities dominated by species of Staphylococcus. LAB were isolated on selective agar plates from 32 'El Kaddid' samples, followed by basic phenotypic ch

Isolation of a deuA-deficient strain of Aspergillus oryzae and its application to soy sauce fermentation.

The thermostable protease DeuA from Aspergillus oryzae contributes to undesirable proteolysis in processed foods prepared with heated soy sauce. To obtain a deuA-deficient A. oryzae strain without recombinant DNA techniques or genome editing, we developed a high-throughput screening method utilizing the bacteriostatic effect of protamine on Latilactobacillus sakei. Loss of DeuA activity reduced protamine degradation, thereby suppressing bacterial acid production and enabling rapid identification of mutants. Screening of approximately 20,000 UV-mutagenized colonies from the industrial strain A.

Fermentation of Youlk, an Australian Native Root Vegetable, Using Defined Lactic Acid Bacterial Strains.

Youlk (Platusace deflexa) is a native root vegetable in Australia that is primarily consumed fresh, baked, or roasted. This study aimed to produce a fermented product using youlk with added lactic acid bacteria (LAB), in comparison with spontaneous fermentations. Three LAB strains were selected for their high in vitro antioxidant activity, leading to eight formulations that featured individual strains or a 1:1:1 combination. The strains were Latilactobacillus sakei F1, Lacticaseibacillus paracasei D2 and Lacticaseibacillus rhamnosus JL, all previously isolated from fermented vegetables. Two sa

Gassericins from Lactobacillus Paragasseri K7: Preparative Isolation and Discovery of Dose-Dependent Anti-Inflammatory Effects.

Gassericins K7 are heterodimeric bacteriocins produced by Lactobacillus paragasseri K7. Their biological characterisation has been limited by challenges in obtaining these compounds in sufficient purity and quantity. In this study, we used a preparative isolation and purification workflow for gassericins K7, combining ammonium sulphate precipitation, Amberlite XAD-16 extraction, and reversed-phase chromatography. Mass spectrometry confirmed the presence of all four peptide components GasK7A α, GasK7A β, GasK7B α, and GasK7B β in the pooled preparation, including GasK7A β, identified here for t

Lactobacillus sakei CVL-001 Inhibits Osteoclast Differentiation Through TLR2 and GPCR-Dependent Pathways and Prevents Bone Loss in Ovariectomized Mice.

Postmenopausal osteoporosis is a common bone disease characterized by bone loss due to the disruption of bone homeostasis caused by decreased estrogen levels. It is estimated that approximately one third of women over the age of 50 will experience osteoporosis in their lifetime. Therefore, developing functional foods or treatments that can be safely consumed to prevent bone disease is essential. Recent studies have reported that gut microbiota is closely related to the development of osteoporosis. In this study, we confirmed that Lactobacillus sakei CVL-001 (CVL-001) and its metabolites (CVL-0

Lactobacillus sakei OK67 ameliorates high-fat diet-induced blood glucose intolerance and obesity in mice by inhibiting gut microbiota lipopolysaccharide production and inducing colon tight junction protein expression.

A high-fat diet (HFD) induces obesity and the associated increases in blood glucose and inflammation through changes in gut microbiota, endotoxemia, and increased gut permeability. To counteract this, researchers have suggested that the use of probiotics that suppress production of proinflammatory lipopolysaccharide (LPS). Here, we tested whether Lactobacillus sakei OK67, which inhibits gut microbiota LPS production selected from among the lactic acid bacteria isolated from kimchi, exerted antihypoglycemic or anti-inflammatory effects in HFD-fed mice. Mice were randomly divided into 2 groups a

Adjunctive administration of parabiotic Lactobacillus sakei CVL-001 ameliorates drug-induced toxicity and pulmonary inflammation during antibiotic treatment for tuberculosis.

Tuberculosis (TB) treatment requires a long therapeutic duration and induces adverse effects such as hepatotoxicity, causing discontinuation of treatment. Reduced adherence to TB medications elevates the risk of recurrence and the development of drug resistance. Additionally, severe cavitary TB with a high burden of Mycobacterium tuberculosis (Mtb) and inflammation-mediated tissue damage may need an extended treatment duration, resulting in a higher tendency of drug-induced toxicity. We previously reported that the administration of Lactobacillus sakei CVL-001 (L. sakei CVL-001) regulates infl

Antimicrobial Activity, Genetic Diversity and Safety Assessment of Lactic Acid Bacteria Isolated from European Hakes (Merluccius merluccius, L.) Caught in the Northeast Atlantic Ocean.

Background/Objectives: The overuse and misuse of antibiotics has contributed significatively to the growing problem of the emergence and spread of antibiotic resistance genes among bacteria, posing a serious global challenge to the treatment of bacterial infectious diseases. For these reasons, there is a current and growing interest in the development of effective alternative or complementary strategies to antibiotic therapy for the prevention of fish diseases, which are mainly based on the use of probiotics-in particular, those belonging to the Lactic Acid Bacteria (LAB) group. In this contex
