Enzymatic synthesis of some sugar-lauric acid esters by lipase from Candida antarctica and their functionalities as emulsifiers and antibacterial agents.

To investigate the regioselectivity of lipase-catalyzed synthesis of important sugar-laurate esters and their functionalities as emulsifiers and antimicrobial agents, glucose, galactose, mannose, maltose and trehalose were used. 6-O-lauryl glucose (Glu-L), 6-O-lauryl galactose (Gal-L), 6-O-lauryl mannose (Man-L), 6'-O-lauryl maltose (Mal-L) and 6-O-lauryl trehalose/6'-O-lauryl trehalose (Tre-L) were synthesized using lipase from Candida antarctica. The Glu-L and Man-L achieved the highest yields (65.49 % and 58.16 %, respectively). The synthesized esters produced smaller-particle-size (1.9-3.1

In Silico and Wet Lab Studies Reveal the Cholesterol Lowering Efficacy of Lauric Acid, a Medium Chain Fat of Coconut Oil.

The coconut oil (CO) contains 91 % of saturated fatty acids in which 72 % are medium chain fatty acids (MCFAs) like lauric, capric and caprylic acids. In contrast to animal fat, coconut oil has no cholesterol. Despite this fact, CO is sidelined among other vegetable oils due to the health hazards attributed to the saturated fatty acids. Though various medicinal effects of CO have been reported including the hypolipidemic activity, people are still confused in the consumption of this natural oil. In silico analyses and wet lab experiments have been carried out to identify the hypolipidemic prop

Vancomycin hydrochloride-loaded stearic acid/lauric acid in situ forming matrix for antimicrobial inhibition in patients with joint infection after total knee arthroplasty.

Knee joint infection following total knee arthroplasty (TKA) is a serious condition and the treatments are complicated. The intra-articular solvent exchange-induced in situ forming matrix is of interest for modulating the release of antibiotics with a high drug concentration and a long period of exposed time at the target site. Stearic acid (S) and lauric acid (L) at various ratios were used as matrix formers by dissolving them in biocompatible solvents such as N-methyl pyrrolidone (NMP) and dimethyl sulfoxide (DMSO). Their matrix formation behaviors in phosphate buffer (pH7.4) and hyaluronic

A retrospective assessment and comparison of the effectiveness of benzoyl peroxide; the combination of topical niacinamide, gallic acid, and lauric acid; and the combination of benzoyl peroxide and erythromycin in acne vulgaris.

Acne vulgaris is a chronic inflammatory skin disease that mostly develops during adolescence and continues throughout adulthood. It affects the face, the main location of cosmetic appearance. Despite many developments in acne treatment, various combination therapies are needed to create the best option. Ninety patients were included in this study. We used the global acne grading system (GAGS) and the lesion counting and photographic standards that were used by Hayashi et al., to assess acne severity. The patients were randomly divided into three groups as group 1 (using only 5% BPO, twice a da

Comparative effects of intraduodenal infusions of lauric and oleic acids on antropyloroduodenal motility, plasma cholecystokinin and peptide YY, appetite, and energy intake in healthy men.

BACKGROUND: The regulation of gastrointestinal function and energy intake by fatty acids depends on their chain length. Animal studies suggest that lauric acid (C12) may have more potent suppressive effects on energy intake than does oleic acid (C18). OBJECTIVE: We compared the effects of equicaloric loads of C12 and C18 on antropyloroduodenal (APD) motility, plasma concentrations of cholecystokinin (CCK) and peptide YY (PYY), appetite, and energy intake. DESIGN: Thirteen healthy men (aged 20-46 y) were studied on 3 occasions in double-blind, randomized fashion. APD pressure waves, plasma ho

Effect of lauric acid on efflux pump inhibition in Staphylococcus aureus K2068: Regulation of the mepA gene and membrane permeability alterations.

MepA efflux protein found in Staphylococcus aureus belongs to the MATE (Multidrug and Toxic Compound Extrusion) family and plays an important role in bacterial resistance. We investigated the potential of lauric acid as an inhibitor of the MepA efflux pump by means of Minimum Inhibitory Concentration (MIC) assays and modulation of the MICs of ciprofloxacin and ethidium bromide (EtBr) against S. aureus; the compound's ability to increase the fluorescence of EtBr and Sytox Green, as well as its ability to inhibit the expression of the mepA gene. The results showed that lauric acid had no antibac

Lauric acid alleviates deoxynivalenol-induced intestinal stem cell damage by potentiating the Akt/mTORC1/S6K1 signaling axis.

Intestinal stem cell (ISC)-driven intestinal homeostasis is subjected to dual regulation by dietary nutrients and toxins. Our study investigated the use of lauric acid (LA) to alleviate deoxynivalenol (DON)-induced intestinal epithelial damage. C57BL/6 mice in the control, LA, DON, and LA + DON groups were orally administered PBS, 10 mg/kg BW LA, 2 mg/kg BW DON, and 10 mg/kg BW LA + 2 mg/kg BW DON for 10 days. The results showed that LA increased the average daily gain and average daily feed intake of the mice exposed to DON. Moreover, the DON-triggered impairment of jejunal morphology and bar

Effects of Duodenal Infusion of Lauric Acid and L-Tryptophan, Alone and Combined, on Fasting Glucose, Insulin and Glucagon in Healthy Men.

The fatty acid, lauric acid ('C12'), and the amino acid, tryptophan ('Trp'), when given intraduodenally at loads that individually do not affect energy intake, have recently been shown to stimulate plasma cholecystokinin, suppress ghrelin and reduce energy intake much more markedly when combined. Both fatty acids and amino acids stimulate insulin secretion by distinct mechanisms; fatty acids enhance glucose-stimulated insulin secretion, while amino acids may have a direct effect on pancreatic β cells. Therefore, it is possible that, by combining these nutrients, their effects to lower blood gl

Fat malabsorption in cystic fibrosis: comparison of quantitative fat assay and a novel assay using fecal lauric/behenic acid.

OBJECTIVES: The gold standard for the diagnosis of fat malabsorption, the 72-hour fat balance study, requires a 3-day collection to generate a coefficient of fat absorption (CFA). We hypothesized that a new test using behenic acid (behenate test) as a nonabsorbable lipid marker may provide a facile means to assess fat absorption. The study proposed to answer 2 questions: first, whether the behenate test correlated with the gold standard and, second, whether the CFA improved when taking pancreatic enzymes during meals instead of taking them before meals. PATIENTS AND METHODS: The study compare

Provision of Lauric Acid Results in Endogenous Production of the Liver Receptor Homolog-1 Ligand, Dilauroylphosphatidylcholine, and Evidence of LRH-1 Transactivation.

BACKGROUND: The unusual phosphatidylcholine species dilauroylphosphatidylcholine (DLPC) has been reported to bind and activate the orphan nuclear receptor liver receptor homolog-1 (LRH-1). To date, DLPC has not been reported endogenously. OBJECTIVE: Herein, we test the hypothesis that the provision of the acyl constituent of DLPC, lauric acid (C12:0), a saturated fatty acid rich in coconut oil, will 1) result in endogenous DLPC production and 2) enhance LRH-1 transcriptional activity. METHODS: We measured DLPC following provision of C12:0 to HepG2 cells, C57/BL6J mice, and to healthy human p

Lauric acid modulates the cyclooxygenases and nitric oxide pathways and reduces oxidative stress in preventing tracheal hyperresponsiveness in asthmatic Wistar rats.

Lauric acid, or dodecanoic acid, a medium-chain fatty acid, prevents alterations in pulmonary ventilation and tracheal hyperresponsiveness in Wistar rats with allergic asthma induced by ovalbumin (OVA). Therefore, the aim was to evaluate the mechanism of action of lauric acid (LA) in its preventive effect on changes caused by asthma. Rats were randomly divided into a control group (CG), an asthmatic group (AG), and an asthmatic lauric acid 25-mg/kg group (ALA25G). Rats in the AG and ALA25G groups were sensitized and challenged with OVA. For the experimental protocols, the trachea and lungs wer

Assessment of Scalable Fractionation Methodologies to Produce Concentrated Lauric Acid from Black Soldier Fly (Hermetia illucens) Larvae Fat.

In the present study, different methodologies with potential scalability and environmental friendliness, such as winterization, supercritical fluid extraction, and multistage distillation, were evaluated for lauric acid concentration. In all cases, to facilitate fractionation, the transformation of triacylglycerols into free fatty acids or fatty acid ethyl esters was required as a previous step. For the winterization experimental assays, the amount and type of solvent was studied, resulting in a product containing ~65% lauric acid with a recovery of ~81% using a 1:10 oil-to-solvent ratio with

Lauric acid with or without levodopa ameliorates Parkinsonism in genetically modified model of Drosophila melanogaster via the oxidative-inflammatory-apoptotic pathway.

BACKGROUND: Parkinson's disease (PD), the most prevalent type of Parkinsonism, is a progressive neurological condition characterized by a range of motor and non-motor symptoms. The complicated etiology of PD is thought to involve a summation of aging, genetic predisposition, and environmental variables. However, the α-synuclein protein plays a significant role in the disease's pathophysiology. MATERIALS AND METHODS: The UAS-α-Syn and Ddc-Gal4 strains were crossed to produce offspring referred to as PD flies. The entire population of flies was divided into five groups, each having about 100 fl

Potential Role of Lauric Acid in Milk Fat Synthesis in Chinese Holstein Cows Based on Integrated Analysis of Ruminal Microbiome and Metabolome.

The composition and metabolic profile of the ruminal microbiome have an impact on milk composition. To unravel the ruminal microbiome and metabolome affecting milk fat synthesis in dairy cows, 16S rRNA and internal transcribed spacer (ITS) gene sequencing, as well as ultra-high performance liquid chromatography-mass spectrometry (UHPLC-MS/MS) methods were used to investigate the significant differences in ruminal bacterial and fungal communities as well as metabolome among Chinese Holstein cows with contrasting milk fat contents under the same diet (H-MF 5.82 ± 0.41% vs. L-MF 3.60 ± 0.12%). Effects of lauric acid on upper gut motility, plasma cholecystokinin and peptide YY, and energy intake are load, but not concentration, dependent in humans.

Animal studies suggest that the effects of fatty acids on gastric emptying and pancreatic secretion are both concentration and load dependent, while their suppressive effect on energy intake is only load dependent. We postulated that, in humans, the modulation of antropyloroduodenal pressure waves, plasma cholecystokinin (CCK) and peptide YY (PYY) concentrations and energy intake by intraduodenal lauric acid, a fatty acid with 12 carbon atoms ('C12') would be load, but not concentration, dependent. Microsomal lauric acid hydroxylase activities after treatment of rats with three classical cytochrome P450 inducers and peroxisome proliferating compounds.

In order to investigate a proposed relationship between induction of hepatic microsomal lauric acid hydroxylase activity and peroxisome proliferation in the liver, male Wistar rats were treated with peroxisome proliferating compounds, and the lauric acid hydroxylase activity, the immunochemical detectable levels of cytochrome P450 4A1 and the activities of peroxisomal enzymes were determined. In addition, the levels of cytochrome P450 4A1 and lauric acid hydroxylase activities were studied after treatment of rats with three cytochrome P450 inducers. Cytochrome P-450 induction by clofibrate. Purification and properties of a hepatic cytochrome P-450 relatively specific for the 12- and 11-hydroxylation of dodecanoic acid (lauric acid).

Hypolipidaemic drugs induce peroxisomal proliferation in the liver and many induce the formation of the hepatic endoplasmic reticulum in general and the formation of cytochrome P-450 in particular. We have induced the formation of rat liver microsomal cytochrome P-450 by the administration of the hypolipidaemic drug clofibrate, isolated the endoplasmic reticulum, solubilized the cytochrome P-450 from these membranes and subdivided the cytochrome P-450 into four fractions by the use of hydrophobic, anionic, cationic and adsorption chromatography. One of these fractions (cytochrome P-450 fractio

Lauric acid attenuates hepatic lipid metabolism by modulating the AMPK-SREBP pathway in high-fat diet-induced obese mice.

Insulin resistance, characterized by a diminished cellular response to insulin signaling, is a central hallmark of metabolic disorders such as obesity and type 2 diabetes mellitus. This impaired responsiveness disrupts glucose and lipid homeostasis, leading to the progression of metabolic dysfunction. This study investigated the effects of lauric acid on lipid metabolism in male C57BL/6J mice. The mice were fed a high-fat diet for eight consecutive weeks to induce obesity, while the control group was fed a normal chow diet. Three experimental groups were co-treated with HFD and lauric acid (12
