Chemical profiling, molecular docking, and harnessing Manuka honey's antimicrobial and prebiotic potential in probiotic milk and yoghurt.

The nutritional and therapeutic properties of Manuka honey (MH) constituted a major interest of the scientific community. The current study characterized the chemical composition of New Zealand MH via LC-HRMS/MS unveiling its richness in 59 compounds. MH antimicrobial activity was assessed against 7 microbial strains where E. coli was the most sensitive followed by P. aeruginosa. Two longitudinal studies were conducted, one examined MH potential use as a prebiotic with Bifidobacterium bifidum in functional probiotic milk, while the other examined its feasibility as a natural antimicrobial addi

Leptospermone acts as a predominantly non-competitive β-triketone inhibitor of tyrosinase.

Leptospermone, a β-triketone derived from Leptospermum scoparium seed oil, was evaluated as a predominantly non-competitive inhibitor of tyrosinase. The oil inhibited mushroom tyrosinase at 100 µg/mL and retained strong activity at 50 µg/mL (L-tyrosine 75.2%, L-DOPA 71.4%). GC/MS profiling identified ten volatiles spanning six chemical classes, with leptospermone (23.8%) as the active. Guided fractionation confirmed leptospermone as the inhibitor of both substrates with comparable intensity (IC50, 30.3–31.5 µg/mL), whereas most co-constituents were inactive at ≤ 100 µg/mL; naphthalene showed m

Mode of toxicity of the β-triketone leptospermone to Aedes aegypti mosquitoes.

Leptospermone, a natural β-triketone and major constituent of manuka oil (Leptospermum scoparium), is an established inhibitor of plant HPPD and was identified to induce rapid knockdown and induce high toxicity to Aedes aegypti adults via topical and tarsal contact exposure with LD50 values of 150 ng/mg of mosquito and 357 ng/cm2, respectively. Although toxic to mosquitoes, leptospermone was non-toxic to ticks, the honey bee, or the fruit fly indicating a high degree of insect specificity. Importantly, leptospermone was equally toxic to non-blood fed and blood-fed mosquitoes suggesting the mod

The efficacy of Australian essential oils for the treatment of head lice infestation in children: A randomised controlled trial.

BACKGROUND: The increase in resistance of head lice to neurotoxic pediculicides and public concern over their safety has led to an increase in alternative treatments, many of which are poorly researched or even untested. METHODS: A multicentre, randomised, assessor-blind, parallel-group trial (Trial 1) was conducted to compare the safety and efficacy of a head lice treatment containing Australian eucalyptus oil and Leptospermum petersonii (EO/LP solution; applied thrice with 7-day intervals between applications) with a neurotoxic treatment containing pyrethrins and piperonyl butoxide (P/PB mo

Unique Mānuka Factor (UMF)-dependent antimicrobial activity of Mānuka honey against respiratory pathogens cannot be explained by sugar and methylglyoxal alone.

Respiratory bacterial infections remain a major cause of morbidity and mortality worldwide, and their treatment is increasingly complicated by antimicrobial resistance. Mānuka honey has attracted interest as a potential alternative antimicrobial agent, although the extent to which antibacterial activity varies with Unique Mānuka Factor (UMF™) grade and the relative contribution of methylglyoxal (MGO) remain unclear. In this study, the antimicrobial activity of five UMF™ grades of Mānuka honey (5+, 10+, 12+, 15+ and 20+) was evaluated against clinically relevant respiratory pathogens: methicill

Essential oils, asthma, thunderstorms, and plant gases: a prospective study of respiratory response to ambient biogenic volatile organic compounds (BVOCs).

Purpose: Prevailing opinion is that wind-pollinated plants affect asthma negatively and that insect- pollinated ones do not. "Thunderstorm" asthma, too, is attributed to bursting grass pollens. Additional biogenic volatile organic compounds (BVOCs) are identified here. Essential oils' BVOCs are inhaled from plants, oil diffusers, candles, room "fresheners", perfumes, and hygiene products. Claims of BVOC "safety" for sensitive respiratory systems are questioned. Methods: Fourteen volunteers, of mixed-age and gender, with seasonal asthma recorded peak expiratory flow (PEF) and 11 symptom scores.

Standardised antibacterial Manuka honey in the management of persistent post-operative corneal oedema: a case series.

BACKGROUND: Corneal oedema is a common post-operative problem that delays or prevents visual recovery from ocular surgery. Honey is a supersaturated solution of sugars with an acidic pH, high osmolarity and low water content. These characteristics inhibit the growth of micro-organisms, reduce oedema and promote epithelialisation. This clinical case series describes the use of a regulatory approved Leptospermum species honey ophthalmic product, in the management of post-operative corneal oedema and bullous keratopathy. METHODS: A retrospective review of 18 consecutive cases (30 eyes) with corn

Manuka Honey in Craniofacial Wound Management: A Narrative Review.

BACKGROUND: Craniofacial wounds present unique management challenges owing to the aesthetic significance of the facial region, proximity to vital structures, risk of contamination from oral flora, and the frequent presence of exposed bone or hardware. Manuka honey, derived from the Leptospermum scoparium plant, possesses well-documented antimicrobial, anti-inflammatory, and wound healing properties attributable primarily to its methylglyoxal content, osmotic activity, and low pH. However, no review has specifically addressed its application in the craniofacial context. METHODS: A narrative re

Trade-offs between environmental and economic factors in conversion from exotic pine production to natural regeneration on erosion prone land.

BACKGROUND: Some of New Zealand's exotic pine (Pinus radiata D.Don) forests were planted for erosion mitigation but cultural, legislative, environmental, and profitability limitations in some parts of the landscape have led to reassessment of their suitability. There is limited information to support landowner decisions on the viability of natural regeneration of native forest post-pine-harvest. METHODS: We evaluated scenarios of post-harvest natural regeneration, compared to remaining in pine production, using erosion susceptibility determined from historical occurrence of landslides, gullie

[Mouthwash solutions with microencapsuled natural extracts: Efficiency for dental plaque and gingivitis].

Mouthwash solutions are mainly used for their antiseptic properties. They currently include synthetic agents (chlorhexidine, triclosan, etc.) or essential oils (especially Listerine). Many natural extracts may also be used. These associate both antiseptic effects and direct action on host response, due to their antioxidant, immunoregulatory, analgesic, buffering, or healing properties. The best known are avocado oil, manuka oil, propolis oil, grapefruit seed extract, pycnogenol, aloe vera, Q10 coenzyme, green tea, and megamin. The development of new technologies, such as microencapsulation (Gi

An ex vivo, assessor blind, randomised, parallel group, comparative efficacy trial of the ovicidal activity of three pediculicides after a single application--melaleuca oil and lavender oil, eucalyptus oil and lemon tea tree oil, and a "suffocation" pediculicide.

BACKGROUND: There are two components to the clinical efficacy of pediculicides: (i) efficacy against the crawling-stages (lousicidal efficacy); and (ii) efficacy against the eggs (ovicidal efficacy). Lousicidal efficacy and ovicidal efficacy are confounded in clinical trials. Here we report on a trial that was specially designed to rank the clinical ovicidal efficacy of pediculicides. Eggs were collected, pre-treatment and post-treatment, from subjects with different types of hair, different coloured hair and hair of different length. METHOD: Subjects with at least 20 live eggs of Pediculus c
