A dual regulatory circuit consisting of S-adenosylmethionine decarboxylase protein and its reaction product controls expression of the paralogous activator prozyme in Trypanosoma brucei.

Polyamines are essential for cell growth of eukaryotes including the etiologic agent of human African trypanosomiasis (HAT), Trypanosoma brucei. In trypanosomatids, a key enzyme in the polyamine biosynthetic pathway, S-adenosylmethionine decarboxylase (TbAdoMetDC) heterodimerizes with a unique catalytically-dead paralog called prozyme to form the active enzyme complex. In higher eukaryotes, polyamine metabolism is subject to tight feedback regulation by spermidine-dependent mechanisms that are absent in trypanosomatids. Instead, in T. brucei an alternative regulatory strategy based on TbAdoMet

Pathophysiology of Gout.

Multiple interacting checkpoints are involved in the pathophysiology of gout. Hyperuricemia is the key risk factor for gout and is considered a prerequisite for monosodium urate (MSU) crystal formation. Urate underexcretion through renal and gut mechanisms is the major mechanism for hyperuricemia in most people. Multiple genetic, environmental, and metabolic factors are associated with serum urate and alter urate transport or synthesis. Urate supersaturation is the most important factor for MSU crystal formation, and other factors such as temperature, pH, and connective tissue components also

Editing of the myosin phosphatase regulatory subunit suppresses angiotensin II induced hypertension via sensitization to nitric oxide mediated vasodilation.

Alternative splicing of exon 24 (E24) of the myosin phosphatase regulatory subunit (Mypt1) tunes smooth muscle sensitivity to NO/cGMP-mediated vasorelaxation and thereby controls blood pressure (BP) in otherwise normal mice. This occurs via the toggling in or out of a C-terminal leucine zipper (LZ) motif required for hetero-dimerization with and activation by cGMP-dependent protein kinase cGK1α. Here we tested the hypothesis that editing (deletion) of E24, by shifting to the LZ positive isoform of Mypt1, would suppress the hypertensive response to angiotensin II (AngII). To test this, mice und

Proteomics and metabolomics analyses of Streptococcus agalactiae isolates from human and animal sources.

Streptococcus agalactiae (S. agalactiae), group B Streptococcus (GBS), a major cause of infection in a wide variety of diseases, have been compared in different human and animal sources. We aimed to compare the bacterial proteome and metabolome profiles of human and animal S. agalactiae strains to delineate biological interactions relevant to infection. With the innovative advancement in mass spectrometry, a comparative result between both strains provided a solid impression of different responses to the host. For instance, stress-related proteins (Asp23/Gls24 family envelope stress response p

Leucyl tRNA synthetase ameliorates cholestatic liver injury by inhibiting integrated stress response in mice.

Aminoacyl-tRNA synthetases, as pivotal enzymes in protein biosynthesis, have been linked to the development of certain diseases when specific isoforms are dysregulated. However, the involvement of leucyl-tRNA synthetase (LARS) in hepatic pathologies remains unexplored. In this study, a marked upregulation of LARS was observed in cholestatic liver tissues. Mice with liver-specific LARS knockdown manifested substantially attenuated hepatic injury, evidenced by reductions in serum alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase levels, as well as dysregulated bile a

Targeting the NLRP3 inflammasome in sepsis: Molecular mechanisms and therapeutic strategies.

Sepsis is a life-threatening organ dysfunction triggered by a dysregulated host response to infection, with immune homeostasis imbalance at the core of its pathological mechanism. The NACHT, leucin-rich repeat (LRR) and pyrin domain (PYD)-containing protein 3 (NLRP3) inflammasome, as an innate immune sensor, plays a critical role in the development and progression of sepsis. Upon activation, the NLRP3 inflammasome triggers caspase-1 activation, which mediates the maturation and release of IL-1β and IL-18, as well as pyroptosis. Studies have demonstrated that inhibiting NLRP3 inflammasome activ

Coupling signalling pathways to transcriptional control: nuclear factors responsive to cAMP.

Several endocrine and neuronal functions are governed by the cAMP-dependent signalling pathway. In eukaryotes, transcriptional regulation upon stimulation of the adenylyl cyclase signalling pathway is mediated by a family of cAMP-responsive nuclear factors. This family consists of a large number of members that may act as activators or repressors. These factors contain the basic domain/ leucine zipper motifs and bind as dimers to cAMP-response elements (CRE). The function of CRE-binding proteins (CREBs) is modulated by phosphorylation by several kinases. Direct activation of gene expression by

Unveiling EXOC4/SEC8: a key player in enhancing antiviral immunity by inhibiting the FBXL19-STING1-SQSTM1 signaling axis.

As a core aptamer for anti-DNA viral immunity, STING1 (stimulator of interferon response cGAMP interactor 1) is tightly regulated to ensure the proper functioning of the natural antiviral immune response. However, many mechanisms underlying the regulation of STING1 remain largely unknown. In this study, we identify EXOC4/SEC8 (exocyst complex component 4) as a novel positive regulator of DNA virus-triggered type I interferon signaling responses through stabilizing STING1, thereby inhibiting DNA viral replication. Mechanistically, EXOC4 suppresses K27-linked ubiquitination of STING1 at K338, K3

Targeting Exportin-1 in Cancer: From Nuclear Export to Oncogenic Transcriptional Hubs.

Maintaining cellular homeostasis requires precise coordination of molecular trafficking between the nucleus and the cytoplasm. Central to this process is the nucleocytoplasmic transport system, which governs the spatial distribution of proteins and RNA. Exportin-1 (XPO1), also known as chromosome region maintenance 1 (CRM1), is the principal nuclear export receptor responsible for the transport of hundreds of cargo proteins bearing leucine-rich nuclear export signals. In a wide range of malignancies, XPO1 is frequently dysregulated through overexpression or recurrent somatic mutations. This dy

Thioredoxin improves contact dermatitis through an anti-inflammatory mechanism different from glucocorticoids.

Thioredoxin (TRX), a redox-regulatory protein of 12 kDa, plays an essential role in modulating oxidative stress and mediating inflammatory processes. In this study, we compared and analyzed the anti-inflammatory effects of topically applied recombinant human TRX (rhTRX), hydrocortisone, and their combination on murine models of contact dermatitis (CD). Topical application of rhTRX, hydrocortisone, and their synergistic combination notably ameliorated ear edema, reduced neutrophilic infiltration within the ear tissues and suppressed the production of cytokines. We explored the distinct anti-inf

FcRav2, a gene with a ROGDI domain involved in Fusarium head blight and crown rot on durum wheat caused by Fusarium culmorum.

Fusarium culmorum is a soil-borne fungal pathogen which causes foot and root rot and Fusarium head blight on small-grain cereals, in particular wheat and barley. It causes significant yield and quality losses and results in the contamination of kernels with type B trichothecene mycotoxins. Our knowledge of the pathogenicity factors of this fungus is still limited. A transposon tagging approach based on the mimp1/impala double-component system has allowed us to select a mutant altered in multiple metabolic and morphological processes, trichothecene production and virulence. The flanking regions

The RNA chaperone protein ProQ is a pleiotropic regulator in enteropathogenic Escherichia coli.

Enteropathogenic Escherichia coli (EPEC) is a gastrointestinal pathogen that affects individuals of all age groups, with infections ranging from subclinical colonization to acute or persistent diarrhea. The bacterium's ability to cause diarrhea depends on the locus of enterocyte effacement (LEE) pathogenicity island. Although regulation of the LEE has been systematically characterized, until the last decade, studies mainly focused on its transcriptional control. Posttranscriptional regulation of the LEE continues to be an underappreciated and understudied area of gene regulation. In the past f

Regulation of Expression of Uropathogenic Escherichia coli Nonfimbrial Adhesin TosA by PapB Homolog TosR in Conjunction with H-NS and Lrp.

Urinary tract infections (UTIs) are a major burden to human health. The overwhelming majority of UTIs are caused by uropathogenic Escherichia coli (UPEC) strains. Unlike some pathogens, UPEC strains do not have a fixed core set of virulence and fitness factors but do have a variety of adhesins and regulatory pathways. One such UPEC adhesin is the nonfimbrial adhesin TosA, which mediates adherence to the epithelium of the upper urinary tract. The tos operon is AT rich, resides on pathogenicity island aspV, and is not expressed under laboratory conditions. Because of this, we hypothesized that t

Coping with stress: eIF2 kinases and translational control.

In response to environmental stresses, a family of protein kinases phosphorylate eIF2 (eukaryotic initiation factor 2) to alleviate cellular injury or alternatively induce apoptosis. Phosphorylation of eIF2 reduces global translation, allowing cells to conserve resources and to initiate a reconfiguration of gene expression to effectively manage stress conditions. Accompanying this general protein synthesis control, eIF2 phosphorylation induces translation of specific mRNAs, such as that encoding the bZIP (basic leucine zipper) transcriptional regulator ATF4 (activating transcription factor 4).

Elucidating the ancestral role of Class I HD-Zip transcription factors in land plants.

Class I homeodomain-leucine zipper (C1HDZ) transcription factors regulate diverse developmental and stress-related processes in land plants. While extensively studied in angiosperms, their functions in other lineages are poorly understood. In the liverwort Marchantia polymorpha, the sole C1HDZ orthologue (MpC1HDZ) controls herbivore defense by regulating oil body cell formation. This suggests that the function of C1HDZ genes may have diverged during the evolution of land plants. Here, we further investigated the function of MpC1HDZ in M. polymorpha and its integration into Arabidopsis (Arabido

Receptor-Like Kinase ZmRLK9 Functions as an Upstream ABA Signaling Factor Conferring Drought Resilience in Maize.

The phytohormone abscisic acid (ABA) plays a pivotal role in plant drought responses. Although a substantial amount of research has concentrated on intracellular ABA receptors and SnRK2 protein kinases as the central signaling components, how the extracellular ABA signal is transduced to this core pathway remains largely unknown. Here, through a genome-wide association study mapping drought resistance in maize at the flowering stage, we identify a 4,052-bp hAT transposon insertion upstream of a leucine-rich repeat receptor-like kinase gene, named ZmRLK9, that suppresses gene expression and com

Mechanistic insights into iron oxide nanoparticles-induced iron deregulation and mitochondrial dysfunction in yellow catfish Pelteobagrus fulvidraco: Comparison with ferrous sulphate.

Despite the widespread use of iron oxide nanoparticles (Fe2O3NPs) in various applications, their differential effects on iron metabolism and iron form-dependent toxicity relative to ferrous sulphate (FeSO4) remain unclear. In this study, we demonstrate that, Fe2O3NPs, in contrast to FeSO4, cause liver structural damage, promote hepatic iron accumulation, impair iron-sulfur cluster (ISC) biogenesis and induce mitochondrial dysfunction in yellow catfish Pelteobagrus fulvidraco. Mechanistically, Fe2O3NPs-induced oxidative stress suppresses NRF2 expression, thereby reducing transcription of fratax

Key regulatory elements of the TGFβ-LRRC15 axis predict disease progression and immunotherapy resistance across cancer types.

Transforming growth factor-beta (TGFβ) has dual roles in cancer, initially suppressing tumors but later promoting metastasis and immune evasion. Efforts to inhibit TGFβ have been largely unsuccessful due to significant toxicity and indiscriminate immunosuppression. Leucine-rich repeat-containing protein 15 (LRRC15) is a TGFβ-regulated antigen expressed by cancer cells of mesenchymal origin and cancer-associated fibroblasts (CAFs). In preclinical studies, ablation of TGFβ-driven LRRC15+ CAFs enhances effector functions of CD8+ T cells. However, the pathobiological mechanisms associated with TGF

NLRP12 Regulates Anti-viral RIG-I Activation via Interaction with TRIM25.

Establishing the balance between positive and negative innate immune mechanisms is crucial for maintaining homeostasis. Here we uncover the regulatory crosstalk between two previously unlinked innate immune receptor families: RIG-I, an anti-viral cytosolic receptor activated type I interferon production, and NLR (nucleotide-binding domain, leucine repeat domain-containing protein). We show that NLRP12 dampens RIG-I-mediated immune signaling against RNA viruses by controlling RIG-I's association with its adaptor MAVS. The nucleotide-binding domain of NLRP12 interacts with the ubiquitin ligase T

Co-expression analysis provides a new strategy for mining key metabolites and genes in response to drought stress in Agropyron mongolicum.

Drought is a major natural disaster that affects plant growth. Agropyron mongolicum possesses a wide range of drought tolerance genes acquired during its long evolution and adaptation to harsh environments. However, the regulatory mechanisms for drought resistance in A. mongolicum are complex, limiting the development and utilization of gene resources in response to drought stress. In this study, we examined differences in morphological, physiological, metabolite and transcript levels between the drought-tolerant (T) and drought-sensitive (S) genotypes of A. mongolicum to identify key metaboli
