Confronting the opioid crisis with consumer health information: a look at East Tennessee.

BACKGROUND: Starting in the 1990s, health care providers began prescribing opioids to patients as pain relievers, believing they were safe. However, many patients became addicted to these pills. In 2017, the US Department of Health and Human Services declared a public health emergency to fight the opioid epidemic. This crisis was prevalent in East Tennessee, where many residents were prescribed opioids. CASE PRESENTATION: Librarians at an academic medical center library in East Tennessee analyzed the health information requests related to pain, mental health, and addiction over the last fifte

A Newly Discovered Obolenskvirus Phage with Sustained Lytic Activity Against Multidrug-Resistant Acinetobacter baumannii.

Background: Acinetobacter baumannii is a highly concerning pathogen in hospital settings, responsible for severe infections such as ventilator-associated pneumonia, urinary tract infections, and meningitis. Its remarkable genetic plasticity facilitates the rapid acquisition of antibiotic resistance, significantly complicating treatment and increasing mortality rates. As multidrug-resistant (MDR) infections continue to rise, phage therapy emerges as a viable alternative. Methods: This study reports the isolation and characterization of Acinetobacter phage vB_AbaM_A72 from stagnant water in Jali

Potential of Therapeutic Bacteriophages in Nosocomial Infection Management.

Nosocomial infections (NIs) are hospital-acquired infections which pose a high healthcare burden worldwide. The impact of NIs is further aggravated by the global spread of antimicrobial resistance (AMR). Conventional treatment and disinfection agents are often insufficient to catch up with the increasing AMR and tolerance of the pathogenic bacteria. This has resulted in a need for alternative approaches and raised new interest in therapeutic bacteriophages (phages). In contrast to the limited clinical options available against AMR bacteria, the extreme abundance and biodiversity of phages in n

Identification of broadly protective human antibodies to Pseudomonas aeruginosa exopolysaccharide Psl by phenotypic screening.

Pseudomonas aeruginosa is a leading cause of hospital-associated infections in the seriously ill, and the primary agent of chronic lung infections in cystic fibrosis patients. A major obstacle to effective control of P. aeruginosa infections is its intrinsic resistance to most antibiotic classes, which results from chromosomally encoded drug-efflux systems and multiple acquired resistance mechanisms selected by years of aggressive antibiotic therapy. These factors demand new strategies and drugs to prevent and treat P. aeruginosa infections. Herein, we describe a monoclonal antibody (mAb) sele

Pangenome profiling of novel drug target against vancomycin-resistant Enterococcus faecium.

Enterococcus faecium is a frequent causative agent of nosocomial infection mainly acquired from outgoing hospital patients (Hospital Acquired Infection-HAIs). They are largely involved in the outbreaks of bacteremia, UTI, and endocarditis with a high transmissibility rate. The recent emergence of VRE strain (i.e. vancomycin resistant enterococcus) turned it into high priority pathogen for which new drug research is of dire need. Therefore, in current study, pangenome and resistome analyses were performed for available antibiotic-resistant genomes (n = 216) of E. faecium. It resulted in the pre

Is there still room for novel viral pathogens in pediatric respiratory tract infections?

Viruses are the most frequent cause of respiratory disease in children. However, despite the advanced diagnostic methods currently in use, in 20 to 50% of respiratory samples a specific pathogen cannot be detected. In this work, we used a metagenomic approach and deep sequencing to examine respiratory samples from children with lower and upper respiratory tract infections that had been previously found negative for 6 bacteria and 15 respiratory viruses by PCR. Nasal washings from 25 children (out of 250) hospitalized with a diagnosis of pneumonia and nasopharyngeal swabs from 46 outpatient chi

Clinical Characteristics and Molecular Profiling of SF3B1-Mutated Myelodysplastic Syndrome (MDS) in a Real-World Practice.

SF3B1-mutated myelodysplastic syndrome (MDS) is a distinct entity associated with a favorable prognosis. Recent data suggest that certain SF3B1 variants portend a worse prognosis. Our study aims to (1) describe SF3B1-MDS patients from a single tertiary center in Singapore and (2) determine if variant type holds prognostic value. We identified MDS patients with SF3B1 variants via next-generation sequencing (NGS) performed from 1 November 2021 to 31 October 2025 at Singapore General Hospital. Extracted genomic material from marrow or blood samples was amplified. Libraries were prepared, sequence

Hospital 3D-printed cyclophosphamide printlets for paediatric patients.

Cyclophosphamide (CPA) remains a cornerstone of paediatric oncology, including in metronomic regimens, but its oral formulations are limited, necessitating the development of dose-adapted formulations. Recently, the World Health Organization (WHO) defined a Target Product Profile (TPP) advocating age-appropriate solid oral dosage forms with flexible dosing and multi-month stability for paediatric cyclophosphamide. Here, the development and characterisation of a 3D-printed chewable cyclophosphamide "printlet" designed to meet this TPP and local prescribing practices are reported. Retrospective

Estimation of the Demand for Hospital Care After a Possible High-Magnitude Earthquake in the City of Lima, Peru.

UNLABELLED: Introduction A model prepared by National Civil Defense (INDECI; Lima, Peru) estimated that an earthquake with an intensity of 8.0 Mw in front of the central coast of Peru would result in 51,019 deaths and 686,105 injured in districts of Metropolitan Lima and Callao. Using this information as a base, a study was designed to determine the characteristics of the demand for treatment in public hospitals and to estimate gaps in care in the hours immediately after such an event. METHODS: A probabilistic model was designed that included the following variables: demand for hospital care;
