Small molecule screening identifies cytotoxic endoplasmic reticulum-associated degradation inhibitors in multiple myeloma.

Multiple myeloma (MM) is an incurable plasma cell neoplasm that is highly reliant on endoplasmic reticulum-associated degradation (ERAD) to maintain protein homeostasis. Disrupting ERAD has been proposed as a therapeutic strategy to overcome proteasome inhibitor resistance; however, the identification of novel inhibitors has been limited. To address this, we conducted a cell-based high-throughput screen using the FDA repurposing library and identified omaveloxolone (RTA408) as a potent ERAD inhibitor that selectively impairs the degradation of ER luminal and membrane substrates, without affect

Development of a Fluorescence Polarization Assay for p300/CBP and Its Application Using a Direct-to-Biology Approach.

The histone acetyltransferases E1A binding protein of 300 kDa (p300) and its homologue cyclic AMP response element binding protein (CREB) binding protein (CBP) are potential targets for cancer treatment. However, no drugs targeting p300/CBP have yet been approved. Various bioassay methods have been developed to evaluate the inhibitory potency of the corresponding inhibitors. However, suitable assays for high-throughput screening (HTS) of novel inhibitors targeting the p300/CBP bromodomain are lacking. To address this shortcoming, we developed a fluorescence polarization (FP) assay. This employ

Potential of Small Molecule TAAR-1 Agonists for the Therapy of Neurodegenerative Psychosis: A Medicinal Chemistry Perspective.

This comprehensive review critically evaluates the emerging therapeutic potential of small-molecule Trace Amine-Associated Receptor 1 (TAAR-1) agonists as a novel, diseasemodifying strategy for neurodegenerative psychosis. From a medicinal chemistry perspective, we assess Structure-Activity Relationship (SAR) data across a broad spectrum of chemotypes, including thyronamine analogues, pyrimidinone-benzimidazoles, guanfacine derivatives, thiophenedihydropyran (Ulotaront), piperidine-carboxamides, sulfonamides, and biguanides. Our analysis establishes a unified strutural model centered on four e

Direct small-molecule inhibitors of the Keap1-Nrf2 protein-protein interaction: Scaffold evolution, PROTAC strategies, and therapeutic implications.

The Keap1-Nrf2 signaling pathway is recognized as a central regulator of cellular protection against oxidative and electrophilic stress, and its pharmacological modulation has attracted considerable attention in drug discovery. Disruption of the Keap1-Nrf2 protein-protein interaction (PPI) has been proposed as an effective strategy to stabilize and activate Nrf2, thereby enhancing the expression of cytoprotective genes. In this review, we provide a comprehensive overview of recent progress in developing direct small-molecule Keap1-Nrf2 PPI inhibitors, spanning diverse chemotypes such as tetrah

Newly identified properties of known pharmaceuticals and myxobacterial small molecules revealed by screening for autophagy modulators.

Autophagy is a cellular degradation and recycling process important for maintaining cellular health and function. It is constitutively active at a low level in eukaryotic cells and can be induced by conditions of cellular stress, such as nutrient starvation. Moreover, autophagy plays an important role in diverse processes such as immunobiology, pathogen infection, ageing, and neurodegenerative and other diseases. Using a high-content fluorescence assay for microtubule-associated protein 1 light chain 3 beta (LC3B), a major player in the autophagic pathway, we screened a library of commercial d

Bruxism, Sleep Quality, Anxiety Disorders, and Tension-Type Headache in Temporomandibular Joint Disorders: A Systematic Review.

Temporomandibular joint disorders (TMD) involve pain and dysfunction of the temporomandibular joint and associated muscles. Emerging evidence suggests associations with bruxism, poor sleep quality, anxiety disorders, and tension-type headache (TTH). To systematically review the associations between bruxism, sleep quality, anxiety disorders, and TTH in patients with TMD. Systematic searches were conducted in PubMed, Scopus, Web of Science, and Cochrane Library up to May 2025. Studies included adults with TMD and reported associations with bruxism, sleep quality, anxiety, or TTH. Risk of bias wa

Neuroinflammation inhibition by small-molecule targeting USP7 noncatalytic domain for neurodegenerative disease therapy.

Neuroinflammation is a fundamental contributor to progressive neuronal damage, which arouses a heightened interest in neurodegenerative disease therapy. Ubiquitin-specific protease 7 (USP7) has a crucial role in regulating protein stability in multiple biological processes; however, the potential role of USP7 in neurodegenerative progression is poorly understood. Here, we discover the natural small molecule eupalinolide B (EB), which targets USP7 to inhibit microglia activation. Cocrystal structure reveals a previously undisclosed covalent allosteric site, Cys576, in a unique noncatalytic HUBL

Gene-wide analyses of genome-wide association data sets: evidence for multiple common risk alleles for schizophrenia and bipolar disorder and for overlap in genetic risk.

Genome-wide association (GWAS) analyses have identified susceptibility loci for many diseases, but most risk for any complex disorder remains unattributed. There is therefore scope for complementary approaches to these data sets. Gene-wide approaches potentially offer additional insights. They might identify association to genes through multiple signals. Also, by providing support for genes rather than single nucleotide polymorphisms (SNPs), they offer an additional opportunity to compare the results across data sets. We have undertaken gene-wide analysis of two GWAS data sets: schizophrenia a

The ATC12 small molecule inhibits the Aurora-A/TPX2 interaction and impairs the proliferation of breast cancer cells.

The Aurora-A kinase and its major regulator TPX2 act as key players during mitosis. Both are overexpressed in tumors, and the Aurora-A/TPX2 complex has been proposed as a potential oncogenic holoenzyme. Evidence of Aurora-A non-mitotic roles in cancer, some of which depend on its nuclear accumulation in interphase and are independent from the kinase activity, is emerging. Indeed, many Aurora-A ATP-competitive inhibitors have shown limited efficacy in clinical trials so far, highlighting the need for novel strategies to inhibit Aurora-A. Interestingly, our recent results suggest an involvement

Genome-wide transcriptome analysis of Chinese pollination-constant nonastringent persimmon fruit treated with ethanol.

BACKGROUND: The persimmon Diospyros kaki Thunb. is an important commercial and deciduous fruit tree. The fruits have proanthocyanidin (PA) content of >25% of the dry weight and are astringent. PAs cause astringency that is often undesirable for human consumption; thus, the removal of astringency is an important practice in the persimmon industry. Soluble PAs can be converted to insoluble PAs by enclosing the fruit in a polyethylene bag containing diluted ethanol. The genomic resource development of the persimmon is delayed because of its large and complex genome. Second-generation sequencing i

Do different adverse childhood experiences lead to specific symptoms of psychosis in adulthood? A systematic review of the current literature.

Previous research has demonstrated significant associations between adverse childhood experiences (ACEs) and risks of psychosis. Further research has examined underlying mechanisms to understand the relationship between these variables. This review aimed to explore the associations between various ACEs and the development of different psychotic symptoms in adulthood. The Cochrane Library, Cinahl, PsychINFO, Medline, Embase, and Web of Science were searched from January 1980 to November 2021 to ensure a systematic review of relevant literature. Poverty, fostering, adoption, childhood emotional

Context-Dependent Associations of the APOA1 rs670 Polymorphism with Apolipoprotein A1 and HDL Cholesterol: A Systematic Review and Meta-Analysis.

The APOA1 rs670 (-75 G>A) polymorphism has been implicated in lipid metabolism and susceptibility to metabolic syndrome (MetS). However, individual studies have reported inconsistent associations with circulating apolipoprotein A1 (Apo A1) and lipid parameters, and no meta-analysis has comprehensively evaluated these associations stratified by metabolic disorder status. We therefore quantified the associations between APOA1 rs670 and serum Apo A1 and lipid parameters, and determined whether these associations are modified by metabolic disorder status. Childhood Obesity and Age-Related Diseases: A Systematic Review and Meta-Analysis of Mendelian Randomization Evidence.

BACKGROUND: Childhood obesity poses a global health threat, with emerging evidence suggesting distinct adipocyte senescence patterns compared to adult-onset obesity. We systematically explored whether childhood obesity may be causally associated with age-related diseases through a meta-analysis of Mendelian randomization (MR) evidence. METHODS: We screened six databases (MEDLINE, PubMed, EMBASE, Web of Science, Cochrane Library and CNKI) and identified 45 MR articles examining paediatric adiposity and major age-related diseases. For each disease, we performed a meta-analysis using results fro

Use of surveillance systems for occupational cancer: data from the Danish National system.

In Denmark the 1970-census population has been followed up for deaths, emigrations, and cancer cases during the ten-year period 1970-80. The linked register allows tabulation of cancer incidence by occupation, however no information is available on specific exposures. It is therefore an indication of the validity of the register that classic associations known from in-depth epidemiological studies are found also in the register. Examples are cancer of the lip in farmers, RR = 1.85, and fishermen, RR = 3.17; and cancer of the nasal cavities and sinuses in skilled furniture makers, RR = 12.25. T

Therapeutic Potential of TAAR1 Agonists in Schizophrenia: Evidence from Preclinical Models and Clinical Studies.

Trace amine-associated receptor 1 (TAAR1) has emerged as a promising therapeutic target for neuropsychiatric disorders due to its ability to modulate monoaminergic and glutamatergic neurotransmission. In particular, agonist compounds have generated interest as potential treatments for schizophrenia and other psychoses due to TAAR1-mediated regulation of dopaminergic tone. Here, we review unmet needs in schizophrenia, the current state of knowledge in TAAR1 circuit biology and neuropharmacology, including preclinical behavioral, imaging, and cellular evidence in glutamatergic, dopaminergic and

Using the Bacterial Ribosome as a Discovery Platform for Peptide-Based Antibiotics.

The threat of bacteria resistant to multiple antibiotics poses a major public health problem requiring immediate and coordinated action worldwide. While infectious pathogens have become increasingly resistant to commercially available drugs, antibiotic discovery programs in major pharmaceutical companies have produced no new antibiotic scaffolds in 40 years. As a result, new strategies must be sought to obtain a steady supply of novel scaffolds capable of countering the spread of resistance. The bacterial ribosome is a major target for antimicrobials and is inhibited by more than half of the a
