Stearic acid spin labels in lipid bilayers: insight through atomistic simulations.

Spin-labeled stearic acid species are commonly used for electron paramagnetic resonance (EPR) studies of cell membranes to investigate phase transitions, fluidity, and other physical properties. In this paper, we use large-scale molecular dynamics simulations to investigate the position and behavior of nitroxide spin labels attached to stearic acid molecules in dipalmitoylphosphatidylcholine (DPPC) bilayers. The results of these studies are potentially very important for the interpretation of EPR spectra, which rely on assumptions about the position of the label in the membrane. Additionally,

Oxidation-dependent changes in the stability and permeability of lipid bilayers.

Peroxidation-dependent change in the permeability of lipid bilayers was mesaured by using artificial membrane systems, that is, planar lipid bilayers and liposomes. The unsaturated fatty acyl chains of phospholipids in small unilamellar vesicles were peroxidized time-dependently by the hydroxyl radical chemically generated by the reaction of H2O2 and Cu(en)2. In contrast, at the same hydroxyl radical concentration and time ranges, no ionic current through the planar lipid bilayers and no release of K+ from the liposomes were observed. These findings indicate that accumulation of lipid peroxide

In Vitro Wrapping of Non-Enveloped Viral Capsids with Lipid Bilayers.

In vitro reconstituted virus-like particles (VLPs) - particles with structure identical to those of virus particles except for containing non-infectious genetic information instead of a viral genome - are promising nanocarriers for delivery of therapeutic cargos. Among the simplest examples are the VLPs formed from the capsid protein (CP) of either Cowpea Chlorotic Mottle Virus (CCMV) or Brome Mosaic Virus (BMV), which form spontaneously from purified components upon mixing CCMV or BMV CP with single-stranded RNA under the right pH and ionic strength conditions. The structure of these VLPs is

Actin polymerization counteracts prewetting of N-WASP on supported lipid bilayers.

Cortical condensates, transient punctate-like structures rich in actin and the actin nucleation pathway member Neural Wiskott-Aldrich syndrome protein (N-WASP), form during activation of the actin cortex in the Caenorhabditis elegans oocyte. Their emergence and spontaneous dissolution is linked to a phase separation process driven by chemical kinetics. However, the mechanisms that drive the onset of cortical condensate formation near membranes remain unexplored. Here, using a reconstituted phase separation assay of cortical condensate proteins, we demonstrate that the key component, N-WASP, ca

Functional investigations of thromboxane synthase (CYP5A1) in lipid bilayers of nanodiscs.

CYP5A1 is a membrane-associated cytochrome P450 that metabolizes the cyclooxygenase product prostaglandin (PGH2 ) into thromboxane A2 (TXA2 ), a potent inducer of vasoconstriction and platelet aggregation. Although CYP5A1 is an ER-bound protein, the role of membranes in modulating the thermodynamics and kinetics of substrate binding to this protein has not been elucidated. In this work, we incorporated thromboxane synthase into lipid bilayers of nanodiscs for functional studies. We measured the redox potential of CYP5A1 in nanodiscs and showed that the redox potential is within a similar range

Continuous lipid bilayers derived from cell membranes for spatial molecular manipulation.

Progress with respect to enrichment and separation of native membrane components in complex lipid environments, such as native cell membranes, has so far been very limited. The reason for the slow progress can be related to the lack of efficient means to generate continuous and laterally fluid supported lipid bilayers (SLBs) made from real cell membranes. We show in this work how the edge of a hydrodynamically driven SLB can be used to induce rupture of adsorbed lipid vesicles of compositions that typically prevent spontaneous SLB formation, such as vesicles made of complex lipid compositions,

Protein translocation through anthrax toxin channels formed in planar lipid bilayers.

The 63-kDa fragment of the protective antigen (PA) component of anthrax toxin forms a heptameric channel, (PA63)7, in acidic endosomal membranes that leads to the translocation of edema factor (EF) and lethal factor (LF) to the cytosol. It also forms a channel in planar phospholipid bilayer membranes. What role does this channel play in the translocation of EF and LF? We report that after the 263-residue N-terminal piece of LF (LFN) binds to its receptor on the (PA63)7 channel and its N-terminal end enters the channel at small positive voltages to block it, LFN is translocated through the chan

Interaction of purified human proteinase 3 (PR3) with reconstituted lipid bilayers.

Proteinase 3 (PR3), the major target autoantigen in Wegener's granulomatosis is a serine proteinase that is normally stored intracellularly in the primary granules of quiescent neutrophils and monocytes. Upon cell activation, a significant portion of this antigen is detected on the cell surface membrane. The nature of the association of PR3 with the membrane and its functional significance are unknown. We investigated the interaction of purified human PR3 with mixtures of zwitterionic (dimyristoyl-L-alpha-phosphatidylcholine, DMPC) and anionic (dimyristoyl-L-alpha-phosphatidylglycerol, DMPG) p

Reconstitution of expressed KCa channels from Xenopus oocytes to lipid bilayers.

Reconstitution of large conductance calcium-activated potassium (KCa) channels from native cell membranes into planar lipid bilayers provides a powerful method to study single channel properties, including ion conduction, pharmacology, and gating. Recently, KCa channels derived from the Drosophila Slowpoke (Slo) gene have been cloned and heterologously expressed in Xenopus oocytes. In this report, we describe the reconstitution of cloned and expressed Slo KCa channels from Xenopus oocyte membranes into lipid bilayers. The reconstituted channels demonstrate functional properties characteristic

Continuous and Rapid Solution Exchange in a Lipid Bilayer Perfusion System Based on Droplet-Interface Bilayer.

Because of the high sensitivity of lipid bilayers to external pressure fluctuations, a major challenge in functional studies of biological pores or ion channels is the difficulty in exchanging solutions rapidly while maintaining the stability of the lipid bilayer in a model membrane. Here we describe a droplet-interface bilayer-based perfusion system that has been routinely used in our research and is currently the most robust and stable perfusion system that provides prompt solution exchange surrounding a lipid bilayer. In this model membrane system, solutions can be completely exchanged with

The Staphylococcus aureus carotenoid staphyloxanthin modifies the structure of phosphoglycerol lipid bilayers.

Staphyloxanthin (STX) is a carotenoid synthesized by the human pathogen Staphylococcus aureus. The golden color of this bacterium is due to this carotenoid. STX protects Staphylococcus aureus from oxidative stress by scavenging free radical species. Furthermore, STX has been shown to mechanically strengthen the Staphylococcus aureus membrane and to form microdomains that recruit antibiotic-resistance factors. Thus, inhibition of STX is a promising strategy for intervening against multidrug-resistant strains of this pathogen. However, the molecular mechanisms by which STX regulates the membrane

Multimodal Investigation into the Interaction of Quinacrine with Microcavity-Supported Lipid Bilayers.

Quinacrine is a versatile drug that is widely recognized for its antimalarial action through its inhibition of the phospholipase enzyme. It also has antianthelmintic and antiprotozoan activities and is a strong DNA binder that may be used to combat multidrug resistance in cancer. Despite extensive cell-based studies, a detailed understanding of quinacrine's influence on the cell membrane, including permeability, binding, and rearrangement at the molecular level, is lacking. Herein, we apply microcavity-suspended lipid bilayers (MSLBs) as in vitro models of the cell membrane comprising DOPC, DO

Cholesterol-Mediated Clustering of the HIV Fusion Protein gp41 in Lipid Bilayers.

The envelope glycoprotein (Env) of the human immunodeficient virus (HIV-1) is known to cluster on the viral membrane surface to attach to target cells and cause membrane fusion for HIV-1 infection. However, the molecular structural mechanisms that drive Env clustering remain opaque. Here, we use solid-state NMR spectroscopy and molecular dynamics (MD) simulations to investigate nanometer-scale clustering of the membrane-proximal external region (MPER) and transmembrane domain (TMD) of gp41, the fusion protein component of Env. Using 19F solid-state NMR experiments of mixed fluorinated peptides

Triggering actin polymerization in Xenopus egg extracts from phosphoinositide-containing lipid bilayers.

Xenopus egg extracts are a powerful tool to reconstitute complex cell biological processes using a cell-free strategy. When used in conjunction with liposomes and supported lipid bilayers, they can recapitulate the biochemical activities occurring at the cytosol/plasma membrane interface of the cell that underlie remodeling of the actin cytoskeleton. We use these in vitro systems to elucidate how membranes and proteins collaborate to make the appropriate actin structure at a given time and place. We have recently broadened the types of membrane substrate used, and also optimized protocols for

Actin polymerization serves as a membrane domain switch in model lipid bilayers.

The ability of cells to mount localized responses to external or internal stimuli is critically dependent on organization of lipids and proteins in the plasma membrane. Involvement of the actin cytoskeleton in membrane organization has been documented, but an active role for actin networks that directly links internal organization of the cytoskeleton with membrane organization has not yet been identified. Here we show that branched actin networks formed on model lipid membranes enriched with the lipid second messenger PIP(2) trigger both temporal and spatial rearrangement of membrane component

Mutation and phosphorylation change the oligomeric structure of phospholamban in lipid bilayers.

Phospholamban (PLB), a 52-residue protein integral to the cardiac sarcoplasmic reticulum, is a key regulator of the Ca pump. PLB has been shown to form pentamers in the denaturing detergent sodium dodecyl sulfate (SDS), but its oligomeric state in the natural environment of the lipid membrane remains unknown. In order to address this issue, we performed electron paramagnetic resonance (EPR) experiments on two types of lipid-reconstituted, recombinant PLB: wild type (WT PLB) and a mutant substituted with alanine at leucine 37 (L37A PLB), whose propensity to oligomerize in SDS is greatly diminis

Liposome capillary electrophoresis for analysis of interactions between lipid bilayers and solutes.

Liposomes, which mimic biomembranes, were used as a pseudostationary phase in capillary zone electrophoresis. The decrease in the mobility of an analyte owing to the presence of liposomes reflected interaction between the analyte and the liposomes. Equations were derived to calculate the specific capacity factor Ks (the capacity factor, k', normalized to the liposome concentration 1 M) from the migration times and to estimate the difference in free energy, delta(delta G0), of the weak analyte/liposome interactions. The order of Ks values for the drugs tested was aspirin < salicylic acid < warf

Detection of transient capacitance increase associated with channel formation in lipid bilayers.

The insertion of alpha- and beta-latrotoxins and sea anemone (Radianthus macrodactilus) toxin into bilayer lipid membranes (BLMs) was investigated using the method of simultaneous conductance/capacitance measurement. All the toxins investigated induced capacitance changes which preceded toxin-induced conductance increases. The processes that may underlie the observed effect are discussed.

Lipid Chemistry beyond the Bilayer: Roles for Nonbilayer Lipids in Biology, Disease, and Delivery.

Cell membranes are composed of both bilayer and nonbilayer lipids, whose molecular shape and interactions support or oppose their packing into flat bilayer sheets. While the role of the former in maintaining membrane structural integrity is clear, a growing body of data highlights the importance of nonbilayer lipids in membrane function, including fission and fusion processes in trafficking, and the dynamics of membrane-associated proteins. In this review, we provide a comprehensive overview of nonbilayer lipids and their roles in membrane biology, disease, and drug delivery. First, we introdu

A molecular dynamics study of PIM2 lipid bilayer membranes.

Developing simplified yet representative models of the complex Mycobacterium tuberculosis inner membrane is crucial for advancing Molecular Dynamics of this pathogen. Phosphatidyl-myo-inositol dimannosides (PIM2), one of the most abundant lipids in this membrane, provides an ideal basis for such a model. In this study, we proposed a minimal, single-component atomistic PIM2 bilayer for use in M. tuberculosis research. We validate the model by assessing its ability to reproduce distinct temperature-dependent ordering regimes, with a more ordered, gel-like behavior at 310 K and increased fluidity
